1.The Effect of Rb Protein Expression on the Clinical Response of the Colorectal Cancer Patients Perfused with 5-Fluorouracil Before Operation

Objective To investigate the effect of Rb protein expression on the clinical response of the colorectal cancer patients perfused with 5-Fu before operation. Methods The expression of Rb protein in 34 cases of the colorectal cancer sample was determined by streptavidin-peroxidase(S-P) immunohistochemical method and the positive reactions were analysed by ICM-100 Cell DNA Picture Analysis System. The apoptotic rate of colorectal cancer cells was examined by the TUNEL staining. The chemosensistivity of the colorectal cancer cells were tested by the MTT colorimetric assay of agar culture. The clinical response of the colorectal cancer patients perfused with 5-Fu before operation was observed simultaneously. [Objective]To investigate the effect of salvianoli acid B and hyperoxic solution on experimental ischemia reperfusion injury of extremities.[Method]Twenty-four healthy New Zealand rabbits were divided into 4 groups randomly.Equivalent 0.9%Nacl(group A),hyperoxic solution(group B),salvianoli acid B(group C),salvianoli acid B and hyperoxic solution(group D) were injected through ear edge vein before ischemia.The femoral artery and vein were blocked and the model of ischemia reperfusion injury of extremities was set up.Before ischemia and after 4 hour's reperfusion,the blood samples were taken to measure the levels of blood malondialdehyde(MDA) and superoxide dismutase(SOD).The gastrocnemius muscles were observed by pathologic test.[Result]Serum enzymatic activity of MDA improved obviously.Salvianoli acid B and hyperoxic solution could prevent the improvement of it(P

Objective To investigate the stromal cell derived factor 1 α (SDF-1 α) and chemokine CXC motif receptor 4 (CXCR4) axis using acupoint electroacupuncture (EA) on the brains of rats after focal cerebral ischemia and reperfusion.Methods Ninety-eight Sprague-Dawley (SD) rats were randomly divided into a control group (8 rats),a model group (50 rats),and an EA group (40 rats).The animal model of focal brain ischemia-reperfusion was made with all the rats in the model group and EA group by using the filament occlusion technique.The model and EA groups were subdivided into 5 subgroups according to the sampling time points on the 1 st,3rd,7th,14th or 21st day after ischemia-reperfusion.The EA was administered bilaterally to the rat analog of the Hegu point (LI 4) in the EA group.The model and control groups received no special treatment.Immunohistochemical methods were employed to detect CXCR4-positive cells in the model and EA groups.The expressions of SDF-1α and CXCR4 mRNA were detected with RT-PCR methods in the 3rd,7th and 14th day subgroups.Results With the prolongation of reperfusion,SDF-1α mRNA expression in the model group had a single peak-like increase in the ischemic area of the cerebral cortex.It had increased significantly by the 3rd day,reached its peak value at the 7th day and then decreased gradually.SDF-1α mRNA expression in the EA group behaved similarly,but SDF-1 mRNA expression was significantly higher in the EA group than in the other two.In the model and EA groups CXCR4 mRNA relative values were higher at the 7th and 14th day than at day 3,and the expression at day 14 was significantly greater than at day 7.CXCR4 mRNA values in the EA group were significantly higher than in the model group at each time point.The expression of CXCR4-positive cells began to increase on the 1 st day in the model group,reached its peak value at the 7th day,then decreased by day 14,but it was still strongly expressed highly at the 21st day.Compared with the model group,the expression in the EA group showed the same pattern,but the number of CXCR4-positive cells in EA group was significantly higher.Conclusion Point EA can activate the SDF-1α and CXCR4 axis to promote angiogenesis after focal cerebral ischemia and reperfusion,at least in rats.
4.Comparison between ADC and standardized ADC value in diagnosing malignancy degree of prostate cancer

Objective To compare the value of apparent diffusion coefficient (ADC) value and the standardized apparent dif‐fusion coefficient (ADC) value in diagnosing malignancy degree of prostate cancer .Methods Diffusion weighted imaging (DWI) results of 34 patients with prostate cancer proved pathologically were retrospectively analyzed .The ADC values in 49 lesions and normal peripheral zones of the prostates of the patients were measured .Then ,ADC values of the lesions were calculated .According to the results of Gleason score ,the lesions were divided into 3 groups:Gleason score≤6 (low‐risk group) ,Gleason score=7 (inter‐mediate‐risk group) and Gleason score≥8 (high‐risk group) .Statistical analysis was performed to evaluate the differences of the ADC values and sADC values in three groups ,and to evaluate the correlation between Gleason score and ADC value or sADC value . Results There were statistically differences overall in ADC values and sADC values of the three groups (P<0 .01) .There was no difference between the intermediate‐risk and high‐risk group in ADC values (P> 0 .05) ,however ,there were difference between sADC values of three groups each other (P< 0 .05) .ADC values (r= -0 .546 ,P=0 .000) and sADC values (r= -0 .575 ,P=0 .000) showed all negative correlation with Gleason score .There were no differences between sADC values and ADC values of three groups in areas under the receiver operating characteristic curve (AUCs) (P>0 .05) .However ,when change the specificity to 100% ,sADC had higher sensitivity than ADC values (47 .5% vs .5 .6% ,78 .2% vs .50 .9% ) in differentiating low‐risk from inter‐mediate‐risk group ,and low‐risk from high‐risk group .Conclusion ADC value and sADC value showed all negative correlation with Gleason score ,but sADC value may be of higher performance in diagnosing malignancy degree of prostate cancer .

Objective To evaluate the value of the Heidelberg retina tomograph - Ⅱ in detection of primary chronic angle-closure glaucoma(CACG) in a Chinese population, and to investigate the difference of parameters detected by HRT-Ⅱ between normal subjects and CACG patients. To evaluate the sensitivity and specificity of optic disk detection using Moorfields regression analysis (MRA) and linear discriminant function(LDF) of the HRT-Ⅱ in differentiating normal from CACG eyes. Design Prospective, comparative case series. Participants 64 eyes of 32 normal subjects and 77 eyes of 51 patients with CACG were studied. The normal subjects were ageand refraction-matched with the glaucoma patients. Methods The interventions consisted of optic disk imaged by means of HRT-Ⅱ and visual field (VF) detected with Oculus Twinfield perimetry. The CACG patients then were divided into two groups according to the VF discriminant standard and mean defect (MD) of Oculus Twinfield perimetry, the group with the glaucomatous VF defect and the group without VF defect. The optic disk was described with 14 optic disk parameters. The optic disc parameters between the normal and the patients were compared. The optic disks were also classified as "normal/borderline/outside normal limits" on the basis of MRA. The results from MRA and from LDF provided in the HRT-Ⅱ were compared. 6.Reconstruction of right ventricular outflow tract with BalMedic pulmonary valved conduit

Objective To evaluate the reconstruction of right ventricle outflow tract (RVOT) with BalMedic pulmonary valved conduit in multiple medical center.Methods Since January 2007,50 patients age (4.90 ± 7.63) years (range 6 month to 39 years),weight (16.20 ± 13.69) kg (range 4.50 to 65.0 kg),had been corrected by reconstruction of RVOT.There were 22 patients with pulmonary atresia and ventricular septal defect (PA/VSD) ; 10 patients with corrected transposition of the great arteries and pulmonary stenosis (C-TGA/PS) ; 7 patients with truncus; 4 patients with double outlet of right ventricle and pulmonary stenosis (DORV/PS) ; 3 patients with tetralogy of Fallot (TOF) ; 2 patients with complete transposition of the great arteries and pulmonary stenosis (TGA/PS) ; and each 1 with aortic stenosis (AS) and pulmonary stenosis (PS).Fifty BalMedic pulmonary valved conduits were implanted between pulmonary and RVOT underwent cardiopulmonary bypass.There were different diameter of pulmonary valved conduit included 10 mm to 24 mm depend on the patients weight and pulmonary size.All patients were followed up after operation on 1 month,3-6 months and more than 12 months.Results There was no death.Three patients were lost followed up after 12 months and one late death.There were no pulmonary valve stenosis about 91.1%,moderate pulmonary regurgitation 16.0%,no RVOT obstruction 95.6%,no main pulmonary artery stenosis 80.0%,and no right and left pulmonary artery stenosis 73.0%.Conclusion These results demonstrated that the BalMedic pulmonary valved conduit is reliable and effective in surgical procedure,but the long-term results should be followed up continually.
7.Effects of Early Rehabilitation on Functional Restoration after Fractures of Elbow

ObjectiveTo compare the effect of the rehabilitation in early and late stage postoperatively for the fractures of the elbow. MethodsAccording to the time the rehabilitation intervened, 57 cases were divided into the early group (29 cases) and the late group (28 cases). After systematic rehabilitation, the two groups were evaluated with Mayo Elbow-Performance and range of motion (ROM). The incidence of elbow disorder was compared between groups a year after fracture. ResultsThe Mayo scores (P<0.01) and ROM (P<0.05) improved more in the early groups than in the late groups, and the incidence of elbow disorder was less a year later (P<0.05). ConclusionEarly postoperative rehabilitation for the fractures of the elbow joint could facilitate function restoration.

Objective To establish written and electronic archives of Schistosoma japonicum antibody indirect hemagglutina-tion(IHA)tests. Methods In the process of schistosomiasis screening by IHA,the written records,electronic records,and se-rum sample bank were combined to make comprehensive archives. Results The S. japonicum antibody IHA test archives can pre-serve the schistosomiasis screening data in the long term and even can trace the source of experiments,and the operation was sim-ple. Conclusion The archives of S. japonicum antibody IHA tests are simple and useful,and worth of popularization.
9.Finite-element investigation on center of resistance of maxillary anterior teeth.

A three-dimensional finite element model of premaxillary bone and anterior teeth was established with ANSYS 13.0. The anterior teeth were fixed with strong stainless labial archwire and lingual frame. In the horizontal loading experiments, a horizontal retraction force of 1.5 N was applied bilaterally to the segment through hooks at the same height between 7 and 21 mm from the incisal edge of central incisor; in vertical loading experiments, a vertical intrusion force of 1.5 N was applied at the midline of lingual frame with distance between 4 and 16 mm from the incisal edge of central incisor. After loading, solution was done and displacement and maximum principle stress were calculated. After horizontal loading, lingual displacement and stress in periodontal membrane (PDM) was most homogeneous when the traction force was 14 mm from the edge of central incisor; after vertical loading, intrusive displacement and stress in PDM were most homogeneous when the traction force was 12 mm from the incisal edge of central incisor. The results of this study suggested that the location of center of resistance (CRe) of six maxillary anterior teeth is about 14 mm gingivally and 12 mm lingually to incisal edge of central incisor. The location can provide evidence for theoretical and clinical study in orthodontics.

10.Proliferation of nasopharyngeal carcinoma SP18 cells promoted by latent membrane protein 1 through TNF receptor-associated death domains

AIM: To investigate the effects of TNF receptor-associated death domains (TRADD) of latent membrane protein 1 (LMP1) on the proliferation of nasopharyngeal cancer SP18 cells.METHODS: The SP18-LMP1 cells and SP18-LMP1TRADD cells, which expressed LMP1 and LMP1 TRADD proteins, respectively, were established.The proliferation of SP18 cells affected by LMP1TRADD was detected by cell counting to analyze the cell growth curve, and by colony formation assay, soft agar formation assay, and flow cytometry.Moreover, the expression profile of differential genes between SP18-LMP1 cells and SP18-LMP1TRADD cells was analyzed by gene chips.RESULTS: The cell growth curve, and the results of colony formation and soft agar formation displayed that the growth velocity and colony forming ability of SP18-LMP1 cells were stronger than those of SP18-LMP1TRADD cells (P<0.01).The results of flow cytometry analysis showed that the proliferation index of SP18-LMP1 cells was higher than that of SP18-LMP1TRADD cells (P<0.01).Sixty-three differentially expressed genes associated with cell proliferation were screened out, in which 33 genes were up-regulated and 30 were down-regulated in the SP18-LMP1TRADD cells.CONCLUSION: TRADD active region is an important functional site of LMP1 to promote the proliferation of SP18 cells.LMP1 may improve the cell proliferation index and induce the proliferation of SP18 cells through TRADD.
