1.Randomized Controlled Study on the Effects of Intensive Cyclophosphamide Therapy Plus Corticosteroids and Corticosteroids-alone Treatment for Lupus Nephritis

To investigate the short and long-term effects of intensive cyclophosphamide therapy plus corticosteroids on lupus mephritis, 27 patients with severe lupus nephritis were divided into two groups. Patients in group 1 received prednisone alone and patients in group 2 were given intensive cyclophosphamide therapy plus corticosteroids. The treatment course lasted for 1.5 years and the patients were followed up for 4 years. At the end of the treatment, the differences in indicators, single or comprehensive, of effieacy were statistically signifieant(P<0.05) between the two groups. A four-year follow-up showed that the difference in relapse was significant(P<0.05). It is concluded that the intensive cyclophosphamide therapy plus corticosteroids on lupus nephritis was better than that of predinison-alone treatment in our series in terms of short-term and long-term effects on lupus nephritis.

3.Role of ten-eleven translocation methylcytosine dioxygenase 3 in trigeminal ganglion in maxillofacial inflammatory pain in mice

Objective:To evaluate the role of ten-eleven translocation methylcytosine dioxygenase 3 (TET3) in trigeminal ganglion in maxillofacial inflammatory pain in mice.Methods:Forty SPF healthy male C57BL/6J mice, aged 8-10 weeks, weighing 19-23 g, were divided into 5 groups ( n=8 each) using a random number table method: control group (group C), inflammatory pain group (group IP), control+ TET3-siRNA group (group C+ siTET3), inflammatory pain+ TET3-siRNA group (group IP+ siTET3) and inflammatory pain+ negative control Scrambled-siRNA group (group IP+ siNC). Normal saline or complete Freund′s adjuvant (CFA) 10 μl was injected into the temporomandibular joint of mice, respectively, and the mechanical paw withdrawal threshold (MWT) was measured at 1, 4, 8 and 12 days after injection (T 1-4). Before injection of normal saline or CFA, 0.75 μl siTET3 or siNC was injected into the trigeminal ganglion and the animals were then sacrificed and trigeminal ganglion was removed at T 2 for determination of the expression of TET3 by Western blot in C+ siTET3, IP+ siTET3 and IP+ siNC groups. Results:Compared with group C, MWT was significantly decreased at T 1-3 , the expression of TET3 in trigeminal ganglion was up-regulate in group IP ( P<0.05 or 0.01). Compared with IP and IP+ siNC groups, MWT was significantly increased at T 2, 3, and the expression of TET3 in trigeminal ganglion was down-regulate in group IP+ siTET3 ( P<0.05 or 0.01). Conclusion:TET3 in trigeminal ganglion is involved in the development of maxillofacial inflammatory pain in mice.
4.Effect of Long Non-coding RNA SPRY4-IT1 on Invasion and Migration of A549 Cells

Background and objective hTe abnormal expression of human long chain non encoding RNA gene is related to many kinds of tumors. hTe aim of this study is to investigate the expression of long non-coding RNA maternally expressed gene 3 (SPRY4-IT1) in lung cancer (A549) cells, and to observe the effect of SPRY4-IT1 on the invasion and migra-tion of A549 cells.Methods hTe levels of SPRY4-IT1 in A549 was detected by Real-time PCR. hTe effects of SPRY4-IT1 on the invasion and migration of A549 cell were analyzed by MTT and Transwell assay. hTe expression of matrix metalloproteinase (MMP) family proteins was determined by Western blot.Results hTe invasion and migration of A549 cells were increased atfer SPRY4-IT1 over-expression. hTe cell spaces were narrower atfer SPRY4-IT1 over expression in the wound healing assay. Tran-swell assays showed that the numbers of transmembrane A549 cells were signiifcantly higher in SPRY4-IT1 over expression group than that in control group (P<0.05). Meanwhile, over expression of SPRY4-IT1 reduced the expression of MMP-2 and MMP-9.Conclusion Over expression of SPRY4-IT1 enhanced the invasion and migration of A549 cells. MMP-2 and MMP-9 might play an important role in this regulation.
