Sicknesses caused by colonic transit dysfunction are commonly seen in clinical practice.The transit dysfunction of the colon has been studied in recent decades on the primary abnormalitie,the secondary damage of the colon enteric nervous system,the distribution of Cajal interstitial cells with the c-kit protein expression of its surface,the damage of smooth muscle by autoimmune,the absorption of water by colon,viral infections and the genetic aspects.
2.Study on correlation between HCMV DNA and T cell subsets as well as islet function in type 1 diabetic patients

Objective To study the correlation between serum human cytomegalovirus (HCMV) DNA and peripheral T cell subsets as well as islet function in type 1 diabetic patients.Methods HCMV DNA levels in sera from 20 type 1 diabetic patients and 40 controls were measured by quantitative polymerase chain reaction (PCR),then the comparison and correlation analysis were made between HCMV DNA and T cell subsets,blood glucose (BG),insulin (Ins) and C peptide (C P).Results The positive rate and levels of HCMV DNA in type 1 diabetic patients were significantly higher than those of controls respectively.The percentage of CD 8 and the levels of fasting BG and BG 2 hours after meal in type 1 diabetic patients with positive HCMV DNA were by far higher than those in controls,while the percentage of CD 4,the ratio of CD 4/CD 8,the levels of fasting Ins,Ins and C P 2 hours after meal were significantly lower than those of controls.There existed the linear positive correlation between HCMV DNA and CD 8 or fasting BG,negative correlation between HCMV DNA and CD 4/CD 8,fasting Ins or fasting C P.Differences were not so significant between type 1 diabetic patients with negative HCMV DNA and controls compared with those between patients with positive HCMV DNA and controls.Conclusion HCMV infection is associated with dysfunction of islet and T cell mediated immunity in type 1 diabetic patients.
3.Effect of Rheum emodi Wall on express level of TNF-?、IL-1? and IL-6 on acute ischemic stroke at high altitude

AIM:To observe the effect of Rheum emodi Wall on expess level of TNF-?、IL-1? and IL-6 on acute ischemic stroke(ACI)at high altitude. METHODS: 65 patients aith ACI at high altitude(2 260 m)were randomly divided into treatment group(34) and control groups(31).both groups received danshen-safflower injection.Meanwhile treatment group took Rheum emodi decoction orally for 4 weeks,while control group doesn't.Serum levels of TNF-?、IL-1? and IL-6 were measured by radioimmumoassay. RESULTS: The treatment group showed much higher rates of improvement than control group(88.24%;70.97%)after weeks.Serum levels of TNF-?、IL-1? and IL-6 decreased significantly in patients with ACI at treatment group as compared with those of control group. CONCLUSION: Rheum emodi Wall can improve the nervous function of ACI by reducing TNF-?、IL-1? and IL-6 content in serun.
4.Rat Aortic Endothelium Damage Caused by Carbon Nanotubes Exposure

Objective To investigate the effect of nanotubes on rat aortic endothelium damage.Methods Wistar rats were intratracheally instilled with carbon nanotubes(CNTs) at the doses of 0,3.5 and 17.5 mg/kg,and with a carbon black negative control,or a quartz positive control.The rats were euthanized after 7 or 30 days of the single treatment,the oxidative stress(GSH,O_2~-?) was determined by using biochemical kits,the expression of sICAM-1 and sVCAM-1 in serum was determined by using ELISA kits and ICAM-1 and VCAM-1 on aortic endothelium was determined with immunohischemistry.Results The content of GSH and O_2~1?in serum increased with a dose-dependent and time-dependent manner.A up-regulated expression of sICAM-1 and sVCAM-1 in serum and ICAM-1 and VCAM-1 in the aortic endothelium was seen.No vascular damage was seen in the rat treated with carbon black,in those treated with high-dose CNTs obvious inflammation was revealed.Conclusion Carbon nanotubes exposure can induce oxidative stress which may be followed by the up-regulated expression of sICAM-1 and sVCAM-1 in serum and ICAM-1 and VCAM-1 in the aortic endothelium,and then the endothelium disfunction may occur.

Objective To prepare polypeptide nerve growth factor (NGF) thermosentitive gel and observe drug release in vitro,in order to provide scientific information for biopharmaceuticals delivery system design aiming to treat the inner disease.Methods The thermosensitive in situ gel was prepared with NGF as main component and Pluronic F127 as the gel matrixes.The effect of concentration of gel matrix PF127 on lower critical solution temperature(LCST) were investigated.In vitro release kinetic studies were performed using membraneless dissolution method and reversed-phase high performance liquid chromatography (HPLC) method was adopted to determine NGF content in the dissolution medium.Results The average LCST of NGF-loaded gel prepared by different concentration of PF127 was 28.48-36.26℃ and the gels had good stability.The in vitro release kinetics was well characterized by sustained release and can be fitted by zero order kinetics.The in vitro accumulated release ratio of NGF in the thermosensitive gel reached to above 95%.hβ-NGF loaded in higher PF127 gel resulted in a more sustained release of hβ-NGF from the thermosensitive gel.Conclusion Well-prepared NGF thermosensitive gel is a promising inner ear-oriented drug delivery system for treating deafness and deserves to further development.
7.Transcatheter closure of coronary artery fistula

Objective To explore the indication and the methodology of transcatheter closure of coronary artery fistula.Methods Altogether 17 cases for intervention,included 13 using the Cook corp coil(2 detachable and another 11 un-detachable);1 with the Pfm corp detachable coil;3 with the AGA corp Amplatzer-Duct.The other 26 cases underwent surgery. Results Intervention group,showed 3 failure cases because of selecting too small and un-detachable coil in early stage with final displacement.Fourteen cases were successful.Follow-up 3 years showed no residue shunt and other complications.All patients were successful with surgery.Conclusions Transcatheter closure of the coronary artery fistula is mild invasive,short hospitalization,safe,and effective under proper indications.
8.EFFECT OF L-STEPHANINE ON APOM ORPHINE-INDUCED ROTATIONAL BEHAVIOUR IN RATS

Rats receiving unilateral nigral injection of 6 OHDA resulted in distinct reduction of DA (-88% ) , DOPAC(-80%) and HVA(-60%) in striatum of lesioned side, and the contents of 5HT and 5HIAA remained almost constant in comparison with that of contralateral striatum, suggesting selective lesion of nigrostriatal dopaminer-gic pathway induced by 6OHDA. Following 6OHDA lesion the rats exhibited circling behaviour after APO challenge. 1-Stephaniae ( 1-STP ) significantly antagonizied the action of APO in circling model in dose-dependent manner. Our results indicate that 1-STP possesses the ability to block DA receptor of central nervous system.
9.Value of GPC3 and PEG10 mRNA in peripheral blood in diagnosis of metastasis in hepatocellular carcinoma

Objective To explore the value of glypican-3(GPC-3)mRNA and paternally expressed 10(PEG10)mRNA in peripheral blood in diagnosis of metastasis in hepatocellular carcinoma(HCC). Methods With SYBR Green I as fluorescence signal,real-time fluorescent quantitative reverse transcription-polymerase chain reaction was employed to detect the expression of GPC-3 mRNA and PEG10 mRNA in peripheral blood from patients with HCC with metastasis(n=8),HCC without metastasis(n=12)and hepatic cirrhosis(n=11),and receiver operator characteristics curve(ROC)and specific parameters were adopted to analyse their value in predictive and exclusive diagnosis. Results The expression of GPC-3 mRNA and PEG10 mRNA in HCC with metastasis was significantly higher than that in HCC without metastasis and in hepatic cirrhosis(P<0.05),while there was no significant difference in the expression of GPC-3 mRNA and PEG10 mRNA between HCC without metastasis and hepatic cirrhosis.In single test,the sensitivities in the differential diagnosis between HCC with metastasis and HCC without metastasis were 66.7%for GPC-3 mRNA and 72.2%for PEG10 mRNA,and the specificities were 91.7%and 91.7%.respectively.The areas under ROC were 0.748 for GPC-3 mRNA and 0.812 for PEG10 mRNA.With two markers in parallel test,the sensitivity,specificity,negative likelihood and diagnostic accuracy were 90.7%,84.O%,0.11 and 83.3%,respectively.In serial test,the sensitivity,specificity,positive likelihood and diagnostic accuracy were 60.5%,98.7%,45.5 and 73.3%,respectively. Conclusion Detection of GPC-3 mRNA and PEG10 mRNA in peripheral blood may help to predict blood metastasis and extrahepatic metastasis of HCC,and PEG10 mRNA works better than GPC-3 mRNA.The serial test of GPC-3 mRNA and PEG10 mRNA is helpful to the predictive diagnosis of peripheral blood metastasis of HCC.
10.Initial experience with the new Pul-Stent in treating postoperative branch pulmonary artery stenosis.

OBJECTIVEThe Pul-stent is a new cobalt alloys stent, specially licensed for pulmonary artery stenosis. The aim of this study was to investigate the value of the stent implantation as the treatment of postoperative pulmonary artery stenosis.
METHODClinical practice was carried out to evaluate effectiveness, safety and maneuverability of Pul-stent in the defined clinical setting. Transthoracic echocardiography, chest roentgenography and electrocardiography were carried out as follow-up studies at 1 and 3 months after procedures.
RESULTTen patients (7 males and 3 females) received Pul-stent implantation in left or right pulmonary arteries (9 stents in left and 1 stent in right). For 9 cases transcatheter stent implantation was performed and in I case hybrid procedure. The median age was 7. 9 years (range 3. 4 - 13. 7 years). The median weight was 22. 3 kg (range 13 - 32 kg). Among them 6 cases were post surgical repair of tetralogy of Fallot, 2 cases were after Fontan procedures, 1 patient was post surgical repair of pulmonary atresia with ventricular septal defect, and 1 patient had stenosis at Glenn pathway. The pressure gradient at the stenosis decreased from (31. 6 ± 10. 2) mmHg(1 mmHg =0. 133 kPa) to (7. 3 ± 10. 3) mmHg, and the diameter of the narrowest segment increased from (4. 0 ± 2. 3) mm to (9. 6 ± 2. 7) mm, the right ventricle to aortic pressure ratio decreased from 0. 54 to 0. 36, all of these improvements were statistically significant (t = 3. 9, -9. 7, 4. 5; P =0. 008, 0. 000, 0. 004). The total procedure time ranged from 55 to 220 min (median 117 min) , and the fluoroscopy time ranged from 9 to 67 min (median 26 min). There were 2 cases of post-stenting pneumorrhagia. No stent fracture, stent malposition and other severe complications were observed. Initial follow-up of 1 and 3-months showed good results with maintenance of improved caliber of the stented vessel, and the gradient across the stenosis area measured by echocardiography was (32. 0 ± 14. 6) mmHg after 3 months. Compared with before stentifng ((40.6 ± 15. 2) mmHg) and 1 month later ((30. 6 ± 13. 6) mmHg), the difference was not statistically significant (t =2. 2,1. 76; P =0. 07, 0. 10).
CONCLUSIONPul-Stent tracking and delivery was excellent, the initial experience has shown that Pul-stent implantation was effective and safe in treating post-operative branch pulmonary artery stenosis. Further follow-up study should be conducted to make sure whether those good results would be kept constant.
