1.Lipoprotein lipase activity anti its relationship to serum lipid levels in elderly

Objective To evaluate lipoprotein lipase (LPL) activity and its relationship to serum lipid. Methods Serum triglyceride (TG) ,total cholesterol (TC), low density lipoprotein cholesterol (LDL-C), high density lipoprotein cholesterol (HDL-C) and postheparin LPL activity in 40 elderly subjects and 45 subjects in control group. Results TG,TC and LDL-C levels were much higher in the elderly than in control group (P <0.05 orP <0.01) ;postheparin LPL activity was lower in elderly (P <0.05) ; and showed a negative correlation with serum TG level (r = -0. 513 ,P < 0.01), but was positively correlated with serum HDL-C concentration (r = 0. 390, P < 0.05). Conclusion The decreased activity of LPL in the elderly may contribute to hypertriglyceridemia associated with low level of high density lipoprotein cholesterol, and could be a risk factor for atherosclerosis.
2.Expression of Nitric Oxide Synthase 3 and Vascular Endothelial Growth Factor in Hepatocellular Carcinoma

【Objective】To investigate the expression of nitric oxide synthase 3 (NOS3) and vascular endothelial growth factor (VEGF) in hepatocellular carcinoma (HCC).【Methods】The expression of NOS3 and VEGF were detected by immunohistochemistry in 51 cases of HCC.【Results】The positive of NOS3 and VEGF in HCC were 53% and 63%.The expression rate of NOS3 in the recurrent group was significantly higher than that in the non-recurrent group (P<0.01).The expression rates of VEGF in the group with carcinoma embolus of portal vein and the recurrent group were significantly higher than those in the group without carcinoma embolus and non-recurrent group (P<0.05).The expression of NOS3 was closely related with the expression of VEGF positively in HCC.【Conclusions】NOS3 and VEGF are related with the biological behavior of HCC closely.
3.Effect of tripterygium wilfordii together with prednisone in treatment of elderly primary nephrotic syndrome

Objective To investigate the therapeutic effect of tripterygium wilfordii and prednisone in old patients with primary nephrotic syndrome. Methods 78 elderly primary nephrotic syndrome patients were randomly divided into two groups. 42 patients in treated-group were treated with tripterygium wilfordii and prednisone while 36 patients in control-group were treated with prednisone only. The curative effect,24 hours urinary protein, serum albumin,plasma lipid and renal function were detemined after six months. The responses of the patients were classified as complete remission(CR) ,partial remission(PR) and NO-response. Results After six months treatment, there were 24 patients got to CR,12 patients to PR,and 6 patients remained refractory to treatment group. While there were 13 patients got to CR,10 to PR,and 13 as refractory in the control group. The general effective rate in the treatment group was 85.7% ,which was markedly higher than that of the control group,which is 66. 67% (P<0. 05). The recurrence rate in the treatment group and control group were 14. 3% and 36. 1% respectively (P<0. 05 ). Conclusion The curative effect of tripterygium wilfordii and prednison on elderly primary nephrotic syndrome is markedly better than prednisone only.
4.EFFECTS OF BURN SERA AND SUBESCHAR TISSUE FLUID ON APOPTOSIS OF POLYMORPHONUCLEAR LEUKOCYTES IN VITRO

To elucidate apoptosis of polymorphonuclear leukocytes (PMN) in early postburn stage and the effects of burn sera and subeschar tissue fluid (STF) on apoptosis of PMNs, 24 Wistar rats were divided into scald group and control group (no scald group) randomly. PMNs were harvested at 0, 12, 24h after scald injury,and percentage of apoptotic PMNs was measured by flow cytometry.Apoptosis of PMNs was again assayed with flow cytometer after being incubated with burn sera or STF harvested from scalded rats. The result showed that the percentage of apoptotic PMNs in scalded group were lower than that in control group,indicating burn sera and STF could inhibit apoptosis of PMNs in a dose dependent manner. It suggested that spontaneous apoptosis of PMNs is inhibited in early postburn stage, and burn sera and STF play an important role in this process.

6.Comparative study of clinical and pathological features of atypical lupus nephritis

Objective To explore the clinical value of renal biopsy in the diagnosis of atypical lupus nephri-tis(LN). Methods The clinical and renal pathological data of 28 cases with atypical LN were analyzed retrospective-ly. Results All 28 patients could not fulfill the ACR diagnostic criterion,misdiagnosed as primary nephritic syndrome 21 eases,chronic glomerulone-phritis syndrome 4 cases ,asymptomatic hematuria 2 cases and asymptomatic proteinuria 2 cases,all renal biopsy showed changes consistent to lupus nephritis,pathological types:3 cases were class Ⅱ,4 cases were class Ⅲ,21 cases were class Ⅳ ,7 cases were class Ⅴ,2 cases was class Ⅳ+ Ⅴlupus nephritis,the clinical manifestations of nephritic syndrome were mostly pathological type Ⅳ and Ⅴ, the chronic glomerulonephritis syndrome showed diversification of pathological type, asymptomatic hematuria and/or proteinuria were mostly pathological type Ⅱ and Ⅲ. Conclusion clinical manifestations of atypical LN is no specific and easy to be misdiagnosed. Renal bi-opsy has great value in the diagnosis of atypical LN.
7.Effects of NF-κB activation on pathogenesis of early myocardial dysfunction induced by PMN in burned rats

Objective To investigate the effects of NF-κB activation on paihogenesis of PMN aggregation in myocardium and early myocardial dysfunction induced by polymorphonuclear leukocyte (PMN) in burned rats. Method One hundred and seventy wistar rats were randomly divided into three groups, control group ( n = 20, with isotonic saline solution), bum group ( n = 90, with isotonic saline solution after bum), bum and pyrrolidine dithioncarbamate group ( PDTC group, n = 60, with isotonic saline solution and 250 mg/kg PDTC after bum). The rats in bum group and PDTC group underwent 35% TBSA full-thickness bums on the back. The acb'vaty of myocardial NF-κB was tested by electrophoretic mobility shift assay (EMSA) at 1, 3, 6, 12,24 postbum hours (PBH). Expressions of myocardial IL-8 and ICAM-1 mRNA were assessed by reverse transcription polymerase chain reaction (RT-PCR) at 3,6,12,24 PBH. Meanwhile, the avtivity of myocardial myeloperoxidse(MPO), the left ventricular systolic pressure(LVSP) and the left ventricular end diastolic pressure (LVEDP) as well as maximum positive and negative left ventricular pressure change (± dp/dtmax) were observed at 3,6,12,24 PBH. Results The activity of myocardial NF-κB in bum groups was markedly increased at 1 PBH [(20.27± 3.43) × 104 A] .which was obviously higher than that in control group [(2.18±0.38) × 104 A, P < 0.01], and was still higher than that in control group at 24 PBH ( P < 0.01). The expressions of myocardial IL-8 and ICAM-1 mRNA and avtivity of myocardial myeloperoxidse were obviously higher than that in control group at 3,6,12,24 PBH respectively (P < 0.01), LVSP and ± dp/dtmax significantly were lower, but LVEDP was higher than that in control group during 3-24 PBH (P < 0.01). Compared with that in bum group, these indices were ameliorated in PDTC group. Conclusions Severe bum might activate myocardial NF-κB, which ultimately lead to the production of cytokines, PMN aggregation in myocardium and deterioration of cardiac contractility and relaxation consequently.
8.Effects of early postburn escharectomy on the permeability of cadiac and pulmonary blood vessels in rats

Objective To evaluate the prophylactic-therapeutic role of escharectomy during early postburn stage on internal organ dysfunction. Methods A total of 72 Wistar rats were randomized into 3 groups: burn without escharectomy group (A group), burn with escharectomy during early postburn stage (B group) and normal control. Vascular permeability were measured with 99Tc-labelled albumin. Furthermore, the effect of burn sera on permeability of monolayer of vascular endothelial cell (EC) was observed. Results Vascular permeability in group B was significantly lower than in group A. When exposed to burn sera, EC monolayer permeability in group B is decreased when compared with group A. Conclusion Escharectomy during early postburn stage could restore endothelial permeability
9.Fabrication and properties of nano-hydroxyapatite/poly(3-hydroxybutyrate) ultrafine-fibrous scaffolds for bone tissue engineering

BACKGROUND:Poly(3-hydroxybutyrate) is approved as its excel ent biocompatibility, biodegradability and piezoelectric properties, but there are also some deficiencies, such as high breakability and poor hydrophilicity. METHODS:Poly(3-hydroxybutyrate) was mixed with different mass percentages of nanohydroxyapatite (0, 10%, 20%and 30%) to prepare new composite fibrous scaffolds through electrospinning process. The microstructure, group composition, crystal ine phase distribution, thermal properties and surface wettability of the scaffolds were detected. RESULTS AND CONCLUSION:Under the scanning electron microscope, with the increase of nano-hydroxyapatite content, more and more nano-hydroxyapatite particles were distributed evenly on the composite fiber surface;the fiber surface was basical y covered with nano-hydroxyapatite particles at the content of 30%, and the roughness of the fiber surface also increased. Results from differential scanning calorimetry and X-ray diffraction showed that the nano-hydroxyapatite reduced the crystal inity of poly(3-hydroxybutyrate) and the crystal tacticity, and this phenomenon became more evident with the increase of nano-hydroxyapatite content. Additional y, the higher the content of nano-hydroxyapatite content, the lower the contact angle and the higher the hydrophily. These findings indicate that the nano-hydroxyapatite/poly(3-hydroxybutyrate) ultrafine-fibrous scaffold using electrospinning technology can effectively improve the surface wettability and crystal inity of the material as wel as the material hydrophily and brittleness, and the higher the content of nano-hydroxyapatite, the more obvious the effect.
10.Effects of burn sera on I?B? degradation and NF-?B activation in monocytes in vitro

AIM: To investigate the effects of burn sera on I?B? degradation, NF-?B activation in peripheral blood monocytes (PBMCs) in order to explore the role of burn sera on activation of monocytes. METHODS: PBMCs isolated from healthy volunteers were stimulated by sera from healthy volunteers and burn patients and by burn sera together with PDTC (pyrrolidine dithioncarbamate). Activation of monocytic NF-?B was tested by electrophoretic mobility shift assay (EMSA) and the degradation of monocytic I?B? was determined by Western blotting. RESULTS: When compared to that in control group, cytosolic I?B? degradation occurred within 30 min after PBMCs stimulated by burn sera, and peaked at 60 min. But I?B? gradually recovered in the cytoplasm after 2 h of stimulation. Meanwhile, activity of monocytic NF-?B was markedly increased, reached the peak at 30 min to 60 min after stimulation, and gradually decreased after 2 h of stimulation. PDTC (an antioxidants) effectively inhibited the monocytic I?B? degradation and activation of NF-?B induced by burn sera. CONCLUSION: Burn sera might induce the degradation of I?B?, then activate NF-?B, which ultimately lead to the secretion of cytokines from the monocytes. [
