1.Roles of inflammasome in cerebral ischemia

Innate immunity plays an important role in inflammatory injury after cerebral ischemia,and inflammasome is considered to be a key factor.Inflammasome is a macromolecular protein complex,can recognize pathogen-associated molecular patterns and damage-associated molecular patterns,and mediate immune inflammatory responses.Studies have shown that cerebral ischemia can induce the acute activation of the NLRP1 and NLRP3 inflammasomes.This article reviews the structure,activation and regulation of inflammasome,and the roles of inflammasome in cerebral ischemia.
2.Injecting dexamethasone in oropharynx to reduce the postoperative syndrome of uvulopalatopharyngoplasty

Objective To introduce a method of reducing the postoperative syndrome of the uvulopalatopharyngoplasty(UPPP). Method UPPP was performed for 23 patients with obstructive sleep apnea hypopnea syndrom operation(OSAHS). Before or after the operation, dexamethasone was injected in oropharynx to control the edema of the oropharynx which caused by UPPP. Results Less syndrome had been occurred in these patients. Conclusion The edema of the oropharynx which caused by UPPP can be controlled by injecting dexamethasone in oropharynx. It can reduce the risk of the UPPP, and make the operation safer and more efficient.
3.Imitating the ischemic preconditionging in lung preservation

ObjectiveTo investigate the preventive effect of drugs imitating the ischemic preconditio ning to the donor lung.MethodsAdenosine was used to imitate the early protection of ischemic preconditioning, SOD to imitate the delayed protection and both of them to imitate the integral p rocess of ischemic preconditioning. At the same time, a control group was set up . The rabbits' lungs were flushed, the heart-lung blocs harvested, and adenosin e, SOD, both or none of them wadded respectively. After preservation for 6 h, th e lungs were reperfused for 1 h with the isolated rabbit lung reperfusion model. The PaO 2, airway pressure, pulmonary compliance, wet/dry ratio and content of NOS were measured. The microscopic and electron microscopic examinations were c arried out.ResultsPaO 2 values were the highest in the control group, followed by the SOD group, the adenosine group, the combined use group. The differences among the groups we re significant. The wet/dry ratio was the highest in the control group, followed by the adenosine group, the SOD group, the combined use group with the differen ces among the groups being significant. The airway pressure and NOS content in t he imitating ischemic preconditioning groups was lower than that of the control group. The pulmonary compliance of the imitating ischemic preconditioning groups was higher than that of the control group. However, the differences were not si gnificant. The microscopic and electron microscopic examination revealed that th e pathological changes in the imitating ischemic preconditioning groups was obvi ously lightened as compared with those in the control group.ConclusionsAdministration of the drugs imitating ischemic preconditioning could improve pul monary oxygenation function and ameliorate pulmonary edema. Although the drugs s howed less effect in ameliorating airway pressure, pulmonary compliance and NOS activity, the imitating ischemic preconditioning could protect the donor lung.
4.Quantitative specific detection of Staphylococcus aureus based on recombinant lysostaphin and ATP bioluminescence.

Quantitative specific detection of Staphylococcus aureus is based on recombinant lysostaphin and ATP bioluminescence. To produce recombinant lysostaphin, the lysostaphin gene was chemically synthesized and inserted it into prokaryotic expression vector pQE30, and the resulting expression plasmid pQE30-Lys was transformed into E. coli M15 for expressing lysostaphin with IPTG induction. The recombinant protein was purified by Ni(2+)-NTA affinity chromatography. Staphylococcus aureus was detected by the recombinant lysostaphin with ATP bioluminescence, and plate count method. The results of the two methods were compared. The recombinant lysostaphin was successfully expressed, and a method of quantitative specific detection of S. aureus has been established, which showed a significant linear correlation with the colony counting. The detection method developed has good perspective to quantify S. aureus.

5.Expression and clinical significance of PIM-1 in prostate neoplasm

Objective To study the expression of PIM-1 in prostate cancer (PCa) and its clinical significance. Methods Reverse transcription polymerase chain reaction ( RT-PCR) analysis was used to determine the expression level of PIM-1 mRNA in 2 cases of benign prostatic hyperplasia ( BPH) samples and 5 cases of PCa samples, and immunohistochemical analysis was used to investigate PIM-1 protein expression in 20 cases of BPH, 20 cases of high grade-prostatic intraepithelial neoplasia ( HGPIN) and 42 cases of PCa tissues. The immunohistochemical staining intensity was scored as negative, weak, moderate and strong positive. Results The expression level of PIM-1 mRNA in 5 cases of PCa was 0. 63 , 0. 55 , 0. 42, 0. 91 and 0. 76 ; the level in 2 cases of BPH was 0. 26 and 0. 27 , respectively. The negative rates of expression of PIM-1 protein in BPH, HGPIN and PCa tissues were 60% ( 12/20) , 20% (4/20) and 2% (1/42) ,the weak positive rates of the expression were 40% (8/12) , 20% (4/20) and 12% (5/42) , while the moderate to strong positive rates of the expression of PIM-1 protein was 0 (0/20) , 60% ( 12/20) and 86% (36/42) , respectively. 6.Value of CD5L and PaCO2 in predicting the survival of patients with severe asthma and mechanical ventilation

Objective To investigate the value of CD5 molecule-like protein(CD5L) and partial pressure of CO2 in arterial blood(PaCO2) in predicting the survival of patients with severe asthma and mechanical ventilation.Methods From Jan.2013 to Jan.2016,a total of 38 patients with severe asthma and mechanical ventilation were enrolled.Acute Physiology and Chronic Health EvaluationⅡ(APACHE II)were used to assess the severity.Enzyme linked immunosorbent assay was used to detect serum levels of CD5L at admission and 6 h after treatment.PaCO2 were also detected.Pearson correlation analysis was used to analyze the correlation between APACHEⅡ score and CD5L and PaCO2 levels.Receiver operation characteristic(ROC) curve was used to evaluate the value of CD5L and PaCO2 in predicting the survival of patients.Results APACHEⅡscores of survival patients were significantly higher than dead patients(P<0.05).CD5L level of survival patients after treatment was significantly lower than dead patients,while PaCO2 level was significantly higher(P<0.05).APACHEⅡ score was negatively correlated with serum CD5L level(r=-0.347,P<0.05),while positively correlated with PaCO2 level(r=0.573,P<0.05).ROC curve analysis showed that serum CD5L and PaCO2 were with predictive value for prediction the survival of patients,with sensitivity of 93.33%,specificity of 75.00%,accuracy of 89.47%,positive predictive value of 93.33%,and negative predictive value of 75.00% for CD5L,and those for PaCO2 were 90.00%,87.50%,89.47%,97.42% and 70.00%.Conclusion With the decreasing of CD5L level and increasing of PaCO2 level,severity of disease in patients with severe asthma and mechanical ventilation could be more serious condition,indicating poor prognosis.CD5L and PaCO2 could be with fine predictive value of survival of patients with severe asthma and mechanical ventilation.
7.Surgical treatment of gastric cancer concurrent with portal hypertension

Objective To evaluate the surgical approaches for gastric carcinoma accompanied by portal hypertension ( PHT).Methods The clinical data of 22 patients with PHT undergoing operation during 5 years were retrospectively analyzed.The liver function was Child's A in 12 cases, Child's B in 10 cases.Total gastrectomy + pericardial devascularization was performed in 11 cases, distal subtotal gastrectomy in 9 cases, distal subtotal gastrectomy + splenectomy in one, distal subtotal gastrectomy + pericardial devascularization in one.12 cases with Child's A underwent D2 lymph node (LN) dissection and 10 cases with Child's B were treated with D1 LN dissection.Liver biopsy was taken in all patients.Results Postoperative complications developed in 50% and mortality rate was 9%.The rate of liver function deterioration in patients of Child A ungergoing D2 lymph node dissection was 42% , and that of patients with Child B was 70%.The rate of postoperatiave complications in patients with Child A ungergoing D2 lymph node dissection was 25% , while that of patients with Child B was 80%.There was no significant difference in liver function deterioration rate between Child A and Child B (P > 0.05) , but the rate of postoperative complications in Child A is much lower than those in Child B(P < 0.05).The complication rate in patients receiving PHT targeting measures was 77% ,much higher than 11% in those without concurrent treatment of PHT ( P < 0.05 ).Conclusions Individualized surgical approache is crucial for treatment of gastric carcinoma accompanied by PHT.Surgical treatment should be on the basis of liver function and the severity of PHT.
8.The neural mechanism of left hemiparalexia and left hemialexia in reading Chinese characters

Objective To investigate the underlying neural mechanism of left hemiparalexia and left hemialexia in reading Chinese characters. Methods A patient with reading disorders caused by brain infarctions at the left ventralis medialis occipitotemporal lobe and the splenium of the corpus callosum was studied. A series of neuropsychological tests, such as reading Chinese characters presented in the central foveal field or in the left and right half of the foveal field, were conducted with the patient, and neuroimaging techniques including high spatial resolution 3D-MRI and diffusion tensor tractography (DTT) were used to examine whether or not there were lesions of the neural pathway. Results The patient showed left hemiparalexia, which was characterized by making substitution or omission mistakes, mostly in the left parts of Chinese characters, and also left hemialexia(alexia for characters presented in left visual field). 3D-MRI demonstrated infarctions in the left ventral mesial occipitotemporal area and in the left side of the splenium of the corpus callosum. The left lateral mid-fusiform cortex, which has been identified as the visual word form area(VWFA), was almost intact. DTT indicated the major forceps fibers running through the splenium were all disconnected due to the infarction of the left splenium. Conclusion As a result of disruption of the splemium-major forceps pathway, visual character information in the left visual field which is initially projected to the right occipital cortex cannot be transferred from the right visual cortex to the left VWFA. This mechanism of left hemiparalexia and left hemialexia in reading Chinese characters is similar to that in reading English words.
9.The effect of ropivacaine on proliferation and cell cycle of human breast cancer MCF-7 cells

Objective To observe the different behavior of proliferation and cell cycle of MCF-7 cells when exposured to ropivacaine of different concentrations and further explore its underlying mechanism.Methods Human breast cancer cells MCF-7 were inoculated into culture medium for 24 h,then were randomly divided into four groups:Control group(group C),Ropivacaine 100 μg/ml group(group R1 ),Ropivacaine 200 μg/ml group(group R2),Ropivacaine 400 μg/ml group(group R3).We medicated each group and incubated for 48 h,then detected the cell proliferation and cell cy-cle immediately.The level of protein TCF-4 and beta-catein of groups R3 and C were measured at the same time.Results MCF-7 cell viability of groups R2 and R3 was significantly lowed (P <0.05 ), MCF-7 cell viability of group R1 had no significant difference when compared to group C.G0/G1 phase cells of groups R1,R2 and R3 were significantly less than those of group C,S phase cells of groups R1,R2 and R3 were significantly more than group C,G2/M phase cells of groups R1,R2 and R3 were significantly more than group C (P <0.05).The expression level of TCF-4 and beta-catenin in group R3 was significantly lower than that in group C (P <0.05).Conclusion Ropivacaine inhibits the proliferation of breast cancer cells MCF-7 by down-regulating TCF-4 and beta-cateni.
10.Application Research on Medical Equipment Management Information System in CAPF

Objective To develop a medical equipment management information system in CAPF so as to improve current medical equipment management and maintenance level.Methods Based on a thorough research for current medical equipment management model in CAPF,the platform of NET was utilized to develop the software.The connection,combination and operation among all medical equipment management and maintenance work were realized by hardware construction at last.Results The management information system not only met the requirement in management of medical equipment,but also supplied a data basis to directors.Conclusion The management information system provides a powerful tool for the scientific and standard management of medical equipment in CAPF.
