ObjectiveTo compare quantitative proteomic analysis of bromotetrandrine (W198) which was a Class Ⅰ new antitumor drug in China and tetrandrine (Tet) in K562 cell line using 18O-labeling method.MethodsTo illustrate its mechanism,a shotgun quantitative proteomic strategy employing 2D LC-MS-MS and trypsin catalyzed 18O-labeling quantification was carried out in this study.Compared to normal chronic leukemia cell line K562 and K562 induced by Tet,the proteomic changes of K562 induced by W198 were investigated.In order to validate the quantitation by the 18O-labeling,the analysis was done on an equivalent sample composed of the same amount of labeled and unlabeled proteins from normally cultured cells to act as a reference to the comparative sample.ResultsA threshold of ± 2-fold change for deciding whether a protein concentration was changed was settled for the following experiments.Comparing the 105 identified soluble proteins' expression levels of the apoptosis starting up K562 cells after W198 induction with the normally cultured cells,16 proteins were found with significantly altered expression levels after W198 treatment.Eight proteins were up-expressed including HMGB2,peroxiredoxin-2,and eIF4A-I,etc.Eight proteins were down-expressed including TCP-1,GRP94,GST-π,and SFGHs,etc.Compared to K562 induced by Tet,eight proteins of K562 were found with significantly altered expression levels after W198 treatment.Five proteins were up-expressed including HSP 90-β and 40S ribosomal protein S15a,etc.Three proteins were down-expressed including phosphoglycerate kinase 1,isoform 5 of interleukin enhancer-binding factor 3,etc.ConclusionThe 18O-labeling MS-MS-based method is ideal as a discovery tool,but it is not suitable for validation using a large number of samples.Other more effective methods,such as Western blotting should be used for further validation of candidate cancer proteins discovered from 18O-labeling samples.In total,105 soluble proteins were discovered,and 16 proteins were found with significantly altered expression levels after W198 treatment.These repressed or activated proteins are the potential drug targets of W198,which may provide novel targets for future development ofbiomarkers for cancer therapy.

In recent years,the abnormal expressions of BRAF,Runx2,Runx3 and S100A4 genes in thyroid papillary carcinoma have been found in many studies.The abnomal expressions of those genes are closely related to the tumor invasion,lymph node metastasis,mulifocality of thyroid papillary carcinoma,which may be valuable in clinical diagnosis and treatment.

As an important member of the adisintegrin and metalloproteinase (ADAM) superfamily,ADAM17 can mediate a variety of membrane molecular hydrolysis off,such as adhesion molecules,cytokines,growth factors,etc,and play an important role in regulating tumor cell adhesion,apoptosis,metastasis and proliferation through the EGFR-PI3K-Akt pathway,Notch signaling pathway and other signaling pathways.The research of ADAM17 targeted drugs provides a new direction for cancer therapy.

In recent years,the abnormal expression of microRNA-21 (miR-21) in head and neck carcinoma have been found in many studies,which may be closely related to the genesis and development of thyroid carcinoma,nasopharyngeal carcinoma,laryngocarcinoma and oral carcinoma,etc.MiR-21 may also be valuable for the clinical diagnosis,treatment and prognosis of head and neck carcinoma.

Cell polarity is a common feature of many different types of cells,and it is essential to the normal differentiation and function of cells. Partitioning defective 6( PAR6)gene encodes PAR6 protein, which is crucial to asymmetric cell division and polarized growth. PAR6 protein as a member of the PAR6 polarity complex,affects the synthetic of centrosome and protein recruitment to the centrosome. The abnormal number of centrosomal and the loss of cell polarity may eventually lead to the occurrence of tumor.

A case of a fibrous histiocytoma (FH) of the larynx in a 54-year-old male is reported. Laryngeal fibrous histiocytoma is uncommon. The case recurred several times over 4-year period. Its pathology is described including arguments on potential malignancy and the way of management.

Objectives To establish the two-dimensional electrophoresis(2-DE)profile of cell. Secreted proteins.Difierential expression profiling of fibroblast cell secreted proteins between nasopharyngeal carcinoma and normal nasopharyngeal tissue was analyzed.Methods Five tissue specimens each from patients with nasopharyngeal carcinoma and nasal polyp were collected individually.Fibroblast eells from above-mentioned tissue were cultured in serum-free medium,and cell-secreted proteins from the cultured medium were harvested by uhrafihration concentration and desalination.Samples were analyzed by 2-DE,and the differentially expressed proteins were analyzed and identified by matrix-assisted laser desorption/ionization time of flight mass spectrometry.Galectin-1 wa8 analyzed by EUSA test.Results 2-DE diagram of fibroblast cell-secreted proteins Was constructed.1 8 protein spots displayed quantitative changes in expression,and 11 protein spots among them were identified by mass speetrometrv.3 proteins including cystatin C,complement subcomponent C1S precursor,heterogeneous nuclear ribonueleoprotein A1 were down-regulated in the cultured medium of nasopharyngeal carcinoma associated fibroblast cells(CAFs). Nevertheless,the rest cell-secreted proteins including galectin-1,14-3-3 protein sigma,eathepsin L and etc,were up-regulated.Meanwhile,the expression of galectin-1 in the cultured medium was also analyzed and Its results were compared between CAFs and the normal fibroblast cells by ELISA.There Was statistical significance difference between them,and galectin-1 was up-regulated in the cIlltured medium of CAFs.Conclusions The changes of fibroblast cell-secreted proteins during nasopharyngeal carcinogenesi8 are analyzed by 2-DE analysis.The variation of pattern of secreted proteins is involved in signal transduction,protein synthesis,degradation and other pathways.CAFs may regulate tumor microenvironment by the abeve-mentioned pathways,and influence nasopharyngeal carcinogenesis,progress,invasion and metastasis.This study provided experimental basis for the eell secreted proteomics studv in future.

Objective:To investigate the dynamic changes of glycine (Gly) and taurine (Tau) in corpus striatum of rats with ischemia-reperfusion injury (IRI),and the regulation effect of electroacupuncture on them.Methods:Thirty SD rats were randomly divided into sham surgery group,model group,and treatment group.Rats in the sham surgery group were operated without ischemia.Rats in the model group and treatment group were operated to make model of cerebral IRI.The ischemia status lasts 1.5 h,and reperfusion was performed for 3.75 h.Rats in the treatment group were treated with electroacupuncture at Fengchi (GB 20).Neurologic deficit score (NDS) was used to evaluate the rats functions.Microdialysis and High Performance Liquid Chromatography technique were used to detect the changes of Gly and Tau in corpus striatum of rats.Results:Contents of Gly and Tau increased at 1.5 h after ischemia.Them decreased to level as same as those rats in the sham surgery group after reperfusion.The content of Gly in the model and treatment group increased again at 2-2.5 h after reperfusion,and then decreased to the same level of the sham surgery group.There was a third increase of Gly content in the treatment group at 2.75 h after reperfusion.Electroacupuncture treatment could delay the decrease of Tau content after reperfusion,and make it increase at 1.5,2 and 2.75 h after reperfusion.The peak appeared in the last time.NDS in the model and treatment group were more than that in the sham surgery group,and lowered at 5.25 h after surgery.Effects in treatment group was better than that in the model group (P＜0.05).Conclusion:IRI makes contents of Gly and Tau in corpus striatum increase at a special time.Electroacupuncture treatment could increase contents of Gly and Tau at a special time after reperfusion,which maybe an important mechanism of protecting effects of electroacupuncture on the brain.

Objective To explore the effect of self-confidence training on social anxiety and self-esteem of patients with social anxiety disorder.Methods 80 patients with social anxiety disorder were randomly divided into the study group and the control group.The study group was given the Self-confidence Training twice a week for five weeks and the control group was not given any training.Social Anxiety Subscale of the Self-Consciousness Scale and Self-Esteem Scale were used to evaluate the effect before the intervention and after the intervention for three months,six months.Results The social anxiety and self-esteem had significant differences between the two groups (F=20.422,P＜0.001 ; F=11.239,P=0.001),they had a tendency to change over time (F=120.080,P＜0.001 ; F=143.503,P＜0.001),and the role of the time factor was different between two groups (F=18.777,P ＜0.001 ; F =24.864,P＜0.001).Further pairwise comparisons revealed that the study group showed significantly decrease in social anxiety ((10.00 ± 3.05) vs (13.42 ± 2.76),(9.53 ± 3.72) vs (13.21± 2.48)) and improvement in score of self-esteem((28.75 ± 4.19) vs (24.27 ± 3.84),(29.50 ± 4.85) vs (24.15 ±3.55)) than control group on post-test and delayed-test (P＜0.001).Conclusion Self-confidence training can reduce social anxiety and improve self-esteem for patients with social anxiety disorder.
