1.Effect of occupational injury on quality of life: 234 cases in the mining area of Tongling, Anhui

BACKGROUND: Quality of life, which is induced as a completely evaluating system to reflect new healthy viewpoints and medical patterns, plays a key role in supplying traditional therapy and evaluating rehabilitation efficacy in occupational injury.OBJECTIVE : To analyze the effects of occupational injury on quality of life in mining area of Tongling, Anhui.DESIGN: Case contrast questionnaire, Logistic single factor analysis and multiple factors analysis.SETTING: Workers General Hospital of Tongling Nonferrous Metal Group; Mining Safety Office of Tongling Nonferrous Metal Company.PARTICIPANTS: A total of 234 subjects with occupational injury were selected from four subgroups of Tongling Nonferrous Metal Company from January 2000 to December 2005 and regarded as the occupational injury group.Injured grade was evaluated by the department of safety in production. In addition, 206 employees of the same sex were regarded as the control group. All subjects provided the confirmed consent.METHODS: Questionnaires were finished in December 2006. Besides those whose educational level was lower than primary school or illness state was severe, their questionnaires were filled by somebody else through asking the feelings,and other subjects filled in the questionnaires by themselves. ① Social demography characteristics were surveyed in the two populations. ② The World Health Organization Quality of Life Assessment-100 (WHOQOL-100), which was characterized by excellent reliability, validity and sensitivity, was used to survey influencing factors (sex, age, occupation,educational level, marriage, smoking, drinking, pay, type of work in production, grade of occupational injury, injured site and type, other diseases, average income of per family member, housing area of per family member, accidental types,character styles, sleeping status, etc.) and various dimensions reflecting quality of life [physiological function (body ache,vigor, sleeping), mental state (well-being feels, confidence, and other three aspects), independence (daily living ability,work ability, and other two aspects), social relationship (interpersonal relation, social communication, and other one spect), material life (reliability, housing environment, and other eight aspects) and faith (individual faith)]. There were 24 aspects and every aspect included four items; in addition, there were 4 items in general healthy status. Therefore, there were 100 items in total. LinearitY method was used to score each item (from 1 tc 5 points). Self-scored quality of life had only one item, which was scored based on hundred-mark system. ③ Quality of life of subjects in the occupational injury group was analyzed with one-way analysis of variance. Variously dimensional indexes of quality of life in 6 fields were regarded as dependent variance, and effect factors were regarded as independent variance. The data were analyzed with multiple stepwise regression analysis.MAIN OUTCOME MEASURES: ① Characteristics of social demography; ② scores of variously dimensional indexes of quality of life; ③ correlation between quality of life and effect factors in occupational injury group.RESULTS: All 234 patients in the occupational injury group and 206 subjects in the control group were involved in the final analysis. ① There were no significant differences in sex, age, occupation, educational degree, working duty,marriage, drinking, smoking, pay, having other diseases and occupational types between the two groups (P > 0.05). ②Scores of physiological field, independence field and total quality of life were lower in the occupational injury group than those in the control group (P < 0.05). ③ Effect factors on quality of life in the occupational injury group contained intoxication and asphyxia; ankle, foot and cranial injuries; avulsed wound and task degrees. Multiple factor analysis demonstrated that quality of life of patients in the occupational injury group was affected by waist injury, pay level and relationship with colleagues and family member.CONCLUSION: Quality of life is inferior in the occupational injury group to that in the control group. The effect factors are mainly focus on injured sites, accident and injured types, economic status and relationship with society and family member.
2.Efficacy and safety of albumin-bound paclitaxel combined with epirubicin and cyclophosphamide in neoadjuvant chemotherapy for breast cancer

Objective:To investigate the efficacy and safety of albumin-bound paclitaxel combined with epirubicin and cyclophosphamide in neoadjuvant chemotherapy for breast cancer.Methods:Clinicopathological data of 48 breast cancer patients who received neoadjuvant chemotherapy and surgery from August 2018 to November 2019 in Shaanxi Provincial Cancer Hospital were retrospectively analyzed. The primary endpoints were pathological complete remission (pCR) rate, objective response rate (ORR) and clinical benefit rate (CBR). The secondary endpoint was the adverse reactions of chemotherapy.Results:The incompletion rate of the protocol-specified courses in all patients was 10.4% (5/48). The pCR rate was 31.2% (15/48), the ORR was 87.5% (42/48), and the CBR was 95.8% (46/48). No statistic difference of pCR rate was obtained in subgroups of patients' age, clinical stage, histological grade, cN stage, and human epidermal growth factor receptor 2 (HER2) status (all P > 0.05), but it was higher in the Ki-67 high-expression group (Ki-67 positive index ≥30%) [46.7% (14/30) vs. 16.7% (3/18), χ2 = 4.427, P = 0.035]. Common adverse reactions which almost were grade 1-2 included leucopenia (93.8%, 45/48), neutropenia (93.8%, 45/48), arthralgia (27.1%, 13/48), fatigue (22.9%, 11/48), peripheral neuropathy (18.8%, 9/48), and nausea and vomiting (18.8%, 9/48). The overall incidence rate of grade 3-4 adverse reactions was 18.8% (9/48). Conclusion:The albumin-bound paclitaxel combined with epirubicin and cyclophosphamide can achieve high pCR rate in neoadjuvant chemotherapy for breast cancer, and the regimen is well tolerated.
3.Effect of emulsified isoflurane preconditioning on postsynaptic density protein 95 activation in brain during focal cerebral ischemia-reperfusion injury in rats

Objective To investigate the effect of emulsified isoflurane preconditioning on postsynaptic density protein 95 (PSD95) activation in brain during focal cerebral ischemia-reperfusion (I/R)injury in rats.Methods Thirty-two pathogen-free male SD rats weighing 280-300 g were randomly divided into 4 groups (n =8each): sham operation group(group S),group I/R,emulsified isoflurane group(group EI) and lipid emulsion group(group LE).Focal cerebral I/R was induced by middle cerebral artery occlusion for 2 h followed by reperfusion.8% emulsified isoflurane 10.5 ml/mg or 30% lipid emulsion 10.5 ml/mg was injected intraperitoneally at 24h before I/R in groups EI and LE respectively.The neurologic deficit score (NDS) was evaluated at 6 h of reperfusion and then 4 rats were sacrificed,and brains were removed for determination of phosphorylatied PSD95 (pPSD95) expression in ischemia cortex and hippocampus by Western blot.Four rats were sacrificed at 24 h of reperfusion and then their brains were removed for determination of infarct volume percentage.Results NDS,infarct volume percentage and pPSD95 expression in ischemia cortex and hippocampus were higher in groups I/R,EI and LE than in group S( P < 0.01 ).NDS,infarct volume percentage and pPSD95 expression in ischemia cortex and hippocampus were lower in group EI than in group I/R( P < 0.05).There was no significant difference in NDS,infarct volume percentage and pPSD95 expression between groups I/R and LE(P > 0.05).Conclusion Emulsified isoflurane preconditioning can attenuate focal cerebral I/R injury in rats by inhibiting PSD95 activation in brain.
4.Effects of emulsified isoflurane preconditioning on expression of platelet-activating factor and its receptor in brain during focal cerebral ischemia-reperfusion in rats

ObjectiveTo investigate the effects of emulsified isoflurane preconditioning on the expression of platelet-activating factor (PAF) and PAF receptor during focal cerebral ischemia-reperfusion (I/R) in rats.MethodsThirty-two healthy adult male SD rats weighing 250-300 g were randomly divided into 4 groups ( n =8each):group sham operation (group S); group I/R; group emulsified isoflurane preconditioning( group EI) and group lipid emulsion (group LE).Focal cerebral I/R was induced by 2 h occlusion of middle cerebral artery followed by reperfusion in groups I/R,EI and LE.8% emulsified isoflurane 10.5 ml/kg and 30% lipid emulsion 10.5 ml/kg were injected intraperitoneally at 24 h before cerebral ischemia in groups EI and LE respectively.The neurologic deficit score (NDS) (0 =no deficit,4 =unable to control) was evaluated at 12 h of reperfusion.Venous blood samples were collected for measurement of plasma PAF concentration.The animals were then sacrificed and their brains removed for determination of infarct size (by TTC staining) and PAF receptor expression in hippocampus and cerebral cortex (by Western blot).ResultsFocal cerebral I/R significantly increased NDS,the infarct size,plasma PAF concentration and PAF receptor expression in cerebral cortex and hippocampus in group I/R as compared with group S.Emulsified isoflurane preconditioning significantly attenuated the focal cerebral I/R induced above changes in group EI as compared with group I/R,but there was no significant difference between groups I/R and LE.ConclusionEmulsified isoflurane preconditioning can attenuate focal cerebral I/R injury by inhibiting PAF and PAF receptor expression.
6.Investigation on Clonorchis sinensis infection and its risk factors in Futian District,Shenzhen City

Objective To understand the status of Clonorchis sinensis infection and its risk factors in Futian District Shen?zhen City so as to provide the evidence for formulating the strategy of prevention and control. Methods Two monitoring points were randomly selected and the permanent population aged≥3 years were investigated and their stool samples were detected for the eggs of C. sinensis by using Kato?Katz technique. Meanwhile the risk factors of infection were also investigated. Results A total of 743 subjects were examined and 7 persons were infected with C. sinensis and the infection rate was 0.94%. Most of the infected cases were focused on 20?40 and 40?60 age groups. The risk factors were eating raw or semi?cooked fish and the protec?tive factor was the use of separated cutting board for cooked or uncooked food. Conclusion The infection rate of C. sinensis is low in the population of Futian District. However the health education especially for diet health education still should be strengthened.
7.Unreported Hospital Infection: Analysis and Countermeasures

OBJECTIVE To ascertain the causes of unreported hospital infection in our hospital and apply countermeasures. METHODS To adopt investigation way to review and analyze 34 694 hospital files in our hospital from Aug 2002 to Jul 2004. RESULTS From them 1164 cases had experienced hospital infection,264 were unreported,the monthly unreported rate decreased from 52.50% to 0;the yearly unreported rate decreased from 36.17% to 10.67%. CONCLUSIONS The unreported reason is because the relevant staff lack infection knowledge,the control and supporting system isn′t so efficient.So the key to reduce the missing report rate is to enhance the awareness of the hospital infection control among the staff in hospital,to strengthen communication and cooperation,and to implement the administrative regulation.
8.Experimental study of bellidifolin enhancing rehabilitation of injuried sciatic nerve

Objective To study whether bellidifolin has rehabilitation effect on injured sciatic nerve rats,and whether ciliary neurotrophic factor (CNTF) is involved in this mechanism.Methods 225 male wistar rats were made to be sciatic nerve injured models,with right sciatic nerve being cut and sewed under microscopy,left sciatic being sham.Rats were randomly divided into control group,bellidifolin 25 mg,50 mg,75 mg groups and mecobalamin group,with 45 rats in each group.Rats in control group were just injected sodium chloride,others were intra-peritonealiy injected different doses of bellidifolin and mecobalamin after operation.Results Three weeks after operation,SFl of control group,mecobalaming group,bellidifolin 25 mg,50 mg and 75 mg group were-84.35± 4.87,-45.20±2.30,-70.42±4.21,-57.73±3.46 and-64.38±4.38 respectively.Compared with control group,others showed significant differences (P<0.05).There were statistically differences between bellidifolin groups and mecobalaming group(P<0.05).Within bellidifolin groups,50 mg group showed difference compared with 25 mg and 75 rg groups(P=0.031).TSW results also showed differences among bellidifolin groups,control group and mecobalaming group.There were statistical differences among bellidifolin groups(P<0.05).Each groups with immunohistochemistry analysis,CNTF expression showed statistically differences among bellidifolin 50 mg group and 25 mg,70 rg groups,Bellidifolin 50 mg group was higher than others(P<0.05).Conclusion Bellidifolin can promote the recovery of injured sciatic nerve,especially the concentration of 50 mg bellidifolin,and CNTF is involved in the rehablitation process.
9.Molecular mechanism of oncogenic B-RafV600E induces chromosome instability in melanoma Sbcl2 and SK-MEL31 cells

Objective To explore the molecular mechanism of BRAFV600E inducing chromosome instability in Sbcl2 and SK-MEL31 melanoma cells.Methods The endogenous Mps1 in stable Sbcl2-and SK-MEL31-B-RafV600E expression cells were depleted by siRNA approach.To test the effect of B-RafV600E on the centrosome amplification and the formation of multipolar spindles,cells at S-phase with HU-treatment were arrested and then the centrosomes and mitotic spindles structure were detected through immunofluoresence.Results The percentage of B-RafV600E expressing Sbcl2 and SK-MEL31 cells (Sbcl2-B-RafV600E and SKMEL31-B-RafV600E) with centrosome amplification and multipolar spindle was reduced from 36 % to 6 % when Mps1 was absent.Conclusion B-RafV600E leads to centrosome amplification and multipolar spindle through Mps1,thus results in chromosome instability in Sbcl2 and SK-MEL31 melanoma cells.
10.Neurofunctional behavior and recognition influence of rat bone marrow mesenchymal stem cells transplant from lateral cistern to cerebral vasospasm rat

Objective To observe whether bone marrow mesenchymal stem cell transplantation can improve vasospastic rats sense and motor function.Methods Rats grouped with randomized number method as Control group,Subarachnoid hemorrhage group.Stem cell culture media group and Stem cell transplantation group.Subarachnoid hemorrhage model were made with tail artery blood twice injection,2 days after 2' nd injection.Bone marrow mesenchymal stem cell were transplanted to lateral cistern.Subarachnoid hemorrhage(SAH) group didn' t transplant stem cell.Stem cell culture media group injected DMEM media as DMEM group.Stem cell transplantation group injected 30μl Bone Marrow mesenchymal stem cell suspension,so called BMSCs group.Neurofunctional score and learning memory expression were detected with morris mazer and Neurofunctional Score Scale in each group.Results After transplantation for 7 d,functional score of Control,SAH,DMEM and Stem cell group were 3.95 ±2.51,7.20 ± 1.03,7.23 ± 1.79 and 5.81 ± 1.11 respectively.Compared with others groups,Stem cell group score was significantly decrease(P=0.017).After transplanting stem cell for 14 d,the mean spanning plate time in Control group,SAH group,DMEM group and Stem cell group were 7.38 ± 1.73,4.52 ± 0.90,5.11 ± 1.93 and 7.32 ± 2.16 respectively,SAH and DMEM group vs other 2 groups,there were clearly statistically differences (P =0.009),while between control group and stem cell group,there were no statistically differences (P =0.14).Conclusion SAH rat transplant stem cell can improve sense,motor and learning expression in certain level.
