1.ISOLATON AND DETERMINATION OF VITAMIN B_(12) IN FERMENTED SOYBEAN FOODS

Foods derived from plants are thought to be devoid of vitamin B12. We have found that some fermented foods contained microbiologically active vitamin B12 components.In order to ascertain the amount of physiological active vitamin B12, we have established a bioautographic area quantitative method of determination of vitamin B12. The procedure is described briefly as follows:The extract of fermented soybean food sample is spotted on a filter paper strip. The Paper is developed with a mixture of secbutanol-water-ammonia water- 5 % potassium cyanide (100: 50: 1: 0.5) using descending technique. After developing for 24-72 hours, the strip is taken out and dried in air. It is then put on the surface of nutrient agar containing E. coli 44110 and 2,3,5,-triphenyl tetrazolium chloride. After incubation, the area of spot of growth is measured and the amount of vitamin B12 in the sample is calculated.Two types of fermented soybean food samples were assayed by the above procedure, mean values of vitamin B12 content are: fermented soybean curd with strong smell contains 0.97 ?g/100g sample, fermented soybean curd contains 0.41 ?g/100g sample. Compared with animal foods, both types of fermented soybean foods may be considered as good source of vitamin B12.
2.Effect of Polysaccharides-2b from Mudan Cortex on High Glucose Induced Proliferation of Rat Mesangial Cells and Reactive Oxygen Species in Culture

Objective To investigate the effect of high glucose on proliferation of rat mesangial cells and reactive oxygen species (ROS) in culture and the intervention of polysaccharides-2b from Mudan Cortex (PSM2b). Methods Cell proliferation was assessed by MTT colorimetric assay. Colorimetric assay was used to detect SOD and MDA in the supernatant of high glucose culture of rat mesangial cells. Results PSM2b could significantly inhibit the proliferation of mesangial cells, the inhibiting effect showed a dose-dependent manner. And SOD increased with PSM2b compared with that only with high glucose, while MDA decreased. Conclusion High glucose contributes to an increase in rat mesangial cells and ROS produced by rat mesangial cells and PSM2b can better control high glucose-induced proliferation of rat mesangial cells and oxidative stress.
3.Study on improving the critical relative humidity of Qubi granule

Objective:To increase the critical relative humidity(CRH) of Qubi granule,thus to improve its humidity resistance. Methods:The best prescription of Qubi granule was optimized by comparative method,and to determine its critical relative humidity. Results:The old prescription of the Qubi granule's CRH was 50%.With new prescription,the Qubi granule’ s CRH was 61%. the latter has increased by 11% as compared with the former . Conclusion:Considering above result,there is a hope to improve the hygroscopicity of Qubi granule.
4.The cardioprotective effect of the inhibitor of JAK/STAT on simulating ischemia-reperfusion injury in cultured immature rat cardiacmyocytes

Objective To investigate the cardioprotective effects of the inhibitor of JAK/STAT pathway on ischemia-reperfusion injury in cultured immature rat cardiacmyocytes.Methods The cultured immature rat cardiacmyocytes were randomly divided into normal control group,ischemia-reperfusion group,AG490 treatment group and RMP treatment group.The cell viability was measured by MTT method.The contents of lactate dehydrogenase(LDH) and malondialdehyde(MDA),the superoxide dismutase(SOD) activity and the rate of cell apoptosis were detected.Results Compared with the normal group,the cell viability decreased significantly,the contents of LDH and MDA in the supernatant of the cells increased significantly,the SOD activity decreased and the rate of apoptosis of cells increased significantly in ischemia-reperfusion group(all P
5.The experience in one case of adult-to-adult combined liver-kidney transplantation from the same living donor

Objective To summarize the experience in one case of adult-to-adult combined liver-kidney transplantation.Method In Sep.2007,one case of adult-to-adult liver-kidney transplantation from the same living donor was performed on a patient with liver cirrhosis (liver failure decompensation) and chronic renal failure (uremia).There was a donation of the right liver with the middle hepatic vein and right kidney in the same time from the living donor.The piggyback liver transplantation and ectopic kidney transplantation were performed for the recipient.Basiliximab and methylprednisolone were given for immune induction therapy in operation.Tacrolimus,MMF and prednisone were given for anti-rejection.There were hepatoprotective treatment,anti-infection treatment and nutritional support for the donor and recipient after operation.The follow-up period has now been more than five years.Result The donor and the patient were smooth in the perioperative period.The liver and kidney function of the donor is well so far.There was no significant influence on quality of life of the donor.The transplanted liver and kidney function of the recipient is well so far.There were no significant complications for the recipient.Conclusion The living liver-kidney transplantation is an effective means for the treatment of liver and kidney failure.The safety can be ensured for the donors that donate the right liver and one kidney simultaneously.
6.Determination of relative molecular weight in polysaccharide-2b of moutan(PSM_(2b))

Objective To determine the relative molecular weight(M_r) of polysaccharide-2b of moutan(PSM_(2b)) to establish the standard of reference substance.Methods PSM_(2b) was separated by a Sepharose 4B column and then PSM_(2b-A) and PSM_(2b-B) were obtained after cryodesiccation.Then HPLC was used to determine their M_r.Ultrapure water was used as mobile phase to compare with 0.7% Na_2SO_4 solution.Results PSM_(2b-A) was pure.The M_r and polydispersity(D) of PSM_(2b-A) obtained by use of dextran as standard were 3.596 2?10~(5) and 4.15 in 0.7% Na_2SO_4 solution phase,respectively,and in ultrapure water phase the M_r and D were 4.758 5?10~(5) and 1.03,respectively.Conclusion Different results are detected by different mobile phases in determination of M_r and D of PSM_(2b-A) by HPLC.And the ultrapure water as mobile phase is more suitable than 0.7% Na_2SO_4 solution.
7.A SURVEY ON POTASSIUM INTAKES AND SERUM POTASSIUM CONTENTS OF POPULATION IN THE AREAS WITH DIFFERENT OCCURRENCE RATES OF GOSSYPOL RELATED HYPOKALEMIA

The result of a nutritional survey on potassium intakes of population and serum potassium contents of healthy adults in Shanghai (high occurrence of hypokalemia), Sichun (moderate occurrence of hypokalemia) and Shandong (null occurrence of hypokalemia) revealed that the average daily potassium intakes for the three areas were 60.1?16.6, 54.6? 13.4 and 58.7? 11.1 mmol per capita per day respectively; The serum potassium contents of the three areas were 4.2?0.3,4.0 ?0.3 and 4.2?0.4 mmol/L respectively. These results indicated that the daily potassium intakes and serum potassium contents between Shanghai and Shandong were not significantly different and might suggest that the occurrence of gossypol related hypokalemia in Shanghai and Sichun could not be attributed to the low potassium intakes and low serum potassium contents of the population.
8.DIETARY FIBER CONTENTS AND ITS COMPONENTS IN FOODS

Dietary fiber contents in 34 common foods perchased in Beijing markets have been determined with detergent method, including neutral detergent fiber, acid detergent fiber, cellulose, hemicellulose and lignin. On dry basis, the dietary fiber contents of vegetables varied with kinds of food: the dietary fiber contents were higher in tender stems, leaves, stalks and flowers; lower in starchy tubers and roots; medium in non-starchy tubers and roots. The propotions of dietary fiber components were different in various categories of foods. Cellulose was the major component in vegetables and legumes, but in most cereals, hemicellulose content was higher than cellulose. All foods had the lowest lignin content with the exception of garlic bolt.
9.Study on the index framework of medium-developed health level in Zhejiang Province

Objective To formulate the index framework of medium developed health level in accordance with Zhejiang Provinces development goal of primarily realizing modernization by 2020. Methods Such research methods as information retrieval, cluster analysis, and Delphi expert consultation were used to select index items of medium developed health level. Results 14 indexes were selected, falling respectively into 4 major categories, viz. health level of the residents, allocation of health resources, provision of and access to health services, and environment and form of the residents health and life. Conclusion The index framework is capable of reflecting in a comprehensive way the overall health level of a certain region.

AIM To study the effect of polysaccharides 2b from Mudan cortex (PSM 2b) on type 2 diabetes mellitus (T2DM) rats and explore its mechanism. METHODS T2DM rat model was established by low dosage streptozotocin and high sucrose fat diet. The drugs were administrated by ig for 5 weeks. The serum glucose was assayed with GOD POD method. Insulin was determined by radioimmunoassay. Insulin receptors of the plasma membrane from rat liver were determined with radioligand binding assay of receptors (RBAR). RESULTS PSM 2b could significantly lower fasting blood glucose (FBG), improve impaired glucose telarance (IGT) and dyslipidemia. It could also remarkably raise receptor total (RT2) of the low affinity insulin receptor (InsR) and insulin sensitivity index (ISI) in T2DM rats. CONCLUSION PSM 2b has an obvious therapeatic effect on T2DM in rats. Its mechamsm is relatively improve insulin resistance (IR) in T2DM rats by raising the number of InsR.
