1.Effect of dihydroartemisinin on cell proliferation and radiotherapy sensitization of lung cancer cell H1299

Objective:To investigate the inhibitory effect of dihydroartemisinin on non-small cell lung cancer H1299 cells.Methods:Through the CCK-8 method for determining the IC 10 of dihydroartemisinin ,choose low dose IC 10 as the experimental concentration,CCK-8 to observe artemisinin in low doses of H1299 cell proliferation, cycle and the influence of radiation sensitivity.Results:IC10 of dihydroartemisinin was 14.87 μmol/L,dihydroartemisinin could inhibit proliferation of H 1299 cells,slow down the cell cycle , and increased the radiation sensitivity.Conclusion: Dihydroartemisinin can inhibit cell proliferation , cell cycle arrest in S phase ,increase the radiation sensitivity.
2.Effects of Losartan on Expression of TNF-? in Myocardium in Renovascular Hypertensive Rats

Objective To investigate the effect of angiotensin Ⅱ-1 receptor antagonist losartan on expression of tumor necrosis factor-alpha (TNF-?) in the ventricular myocardium in the renovascular hypertension rats. Methods Renovascular hypertension model was obtained by clip left renal artery in Sprague-Dawley(SD) rats. After operation the rats were divided into 3 groups: sham group, two-kidney one clip (2K1C) group, and losartan treatment group(2K1C and losartan 20 mg/kg?d by drinking). Tail blood pressure was determined every week. Animals were euthanized after treatment with losartan for four weeks. 4.Clinical observation of noninvasive ventilation in the treatment of patients with acute exacerbations of chronic obstructive pulmonary disease

Objective To investigate the therapeutic effect of non-invasive positi e pressure ventilation (NIPPV) in the treatment of patients with acute exacerbations of chronic obstructive pulmonary disease(AECOPD).Methods 68 AECOPD patients with type Ⅱ repiratory failure were randomly divided into control group and treatment group.The control group was treated by conventional therapy alone and the treatment group was given conventional therapy combined with NIPPV.The levels of power of hydrogen (pH),partial pressure of carbon dioxide in artery(PaCO2),arterial oxygen tension(PaO2),heart rate and repiratory rate were compared between the two groups.The mean length of patient stay,the frequency of tracheal intubation and the hospital mortality were analyzed after discharge from the hospital.Results Before treatment,the arterial blood gas analysis showed that the pH,PaCO2 and PaO2 had no significant differences between the two groups(all P > 0.05).After treatment,compared with the control group,the levels of pH,PaCO2 and PaO2 were obviously improved(all P <0.05).The mean length of patients stay in the treatment group was significantly shorter than the control group(P < 0.05).The frequency of tracheal intubation of the treatment group was significantly lower than the control group(P < 0.05).The hospital mortality was lower in the treatment group,but the difference between the two groups was not significant (P > 0.05).Conclusion NIPPV for treating the AECOPD can improve the arterial blood gas analysis,decrease the need of tracheal intubation and shorten hospitalization time.
5.A quantitative analysis of lower limb muscle spasticity in stroke survivors

Objective To relate the results of surface electromyography (sEMG) of the femoris among patients with hemiplegia after a brain lesion with scores on the modified Ashworth scale (MAS),and to decide whether sEMG can be used as a quantitative index for evaluating spasm.Methods The affected quadriceps femoris of sixty hemiplegic stroke survivors were assessed by the same therapist using the MAS.sEMGs were also recorded and the root mean square and average sEMG readings from the vastus medialis (VM),rectus femoris (RF) and vastus lateralis (VL) were extracted.The correlation between these sEMG indexes and the MAS grading was analyzed.Results The average sEMG signals from the VM,RF and VL were strongly correlated with the MAS grading,and they increased with increasing MAS grade.Spearman correlation analysis demonstrated highly significant correlation (P ≤ 0.001) when the quadriceps femoris was moved passively.Conclusions Surface EMG signals from the VM,RF and VL correlate well with MAS grades,rising when the affected quadriceps femoris of a stroke survivor is moved passively.sEMG can present the electrical activity during passive movements of the quadriceps femoris and can be useful in spasticity assessment and clinical treatment.

7.Clinical significance of combined detection of plasma ctDNA and BRAF V600E mutations in patients with thyroid carcinoma

Objective To detect the content of plasma ctDNA and the mutation rate of BRAF V600E in plasma of patients with thyroid carcinoma ,and to explore its clinical significance. Methods Plasma ctDNA was extracted from 16 patients with thyroid carcinoma and 59 patients with benign thyroid nodules by using the blood genomic DNA Extraction Kit. The ctDNA content was detected by fluorescence quantitative PCR ,and the mutation of circulating BRAF V600E was detected by PCR and sequencing. Then the clinical significance was analyzed by combined detection analysis. Results The content of ctDNA in thyroid cancer group was significantly higher than that in benign nodule group (P < 0.01). BRAF V600E mutation detection showed that the mutation rate was 43.75%,but benign nodules had no mutation. Parallel combined detection improved the sensitivity and the specific-ity of the combined detection was higher. Conclusion Combined detection of ctDNA and BRAF V600E in plasma is helpful for differential diagnosis of benign and malignant thyroid nodules.
8.Interstitialcells of liver cancer influence the biological function of malignant through the HGF/cMET system

Objective To explore the effects of interstitial cells of liver cancer and normal liver cells co‐cultured on the biological function of liver cancer malignancy so as to understand the signal pathway involved by the interaction between these cells and confirm the role of interstitial cells in cancer progression in tumor microenvironment .Methods We co‐cultured interstitial cells or hepatocyte growth factor (HGF ) and human normal liver cell L‐02 ,and then detected the expressions of the tumor‐suppressing gene PTEN and the oncogene K‐RAS and changes of cell proliferation .The downstream signaling pathways were detected by Real‐time PCR and Western blot .Results The expression of PTEN was downregulated at the transcription level by 1 .15 times and translation level by 10 times (P<0 .05) ,while the transcription level and translation level of K‐RAS increased by 1 .4 times and more than 9 times , respectively ( P< 0 .05 ) in normal liver cells co‐cultured with liver cancer mesenchymal cells .The proliferation ability was increased by more than 2 times .ELISA experiment results showed that the medium from co‐culture contained HGF over 3 times more than the control group ( P<0 .05 ,1 085+108 vs .387+23) .At the same time ,cells in the experimental group expressed more than four times of cMET than the control group cells (P< 0 .05) .Exogenous HGF consistently promoted liver cell proliferation and viability (P<0 .05) .Conclusion Our study shows that liver cancer interstitial cells activate the HGF/cMET signaling pathway by secreting HGF and promote the proliferation of normal liver cells ,suggesting a new way to explore the molecular mechanism of tumor microenvironment in tumor development and treatment of hepatocellular carcinoma .

Objective To estimate the value of applying left heart bypass technique in esophageal carcinoma resection and gastroesophageal reconstruction. Methods The operation was performed under lef t he art bypass. Results Applying left heart bypass during esophagea l carcinoma resection and gastroesophageal reconstruction increased the probabil ity of the resection. The patient lived better and without operative complicatio n. Conclusion In case of esophageal carcinoma with invasion of descending aorta by carcinoma, left heart bypass may increase the probability of the resection and enhance the safety of surgical treatment.
10.Effect of intra-and post-operative mild hypothermia with ice blanket in treatment of severe traumatic brain injury

Objective To observe effect of intra-and post-operative mild hypothermia using an ice blanket on patients with severe traumatic brain injury (sTBI).Methods Twenty sTBI patients with Glasgow Coma Scale (GCS) of 3-8 points were included and were assigned to either ice bag cooling (Bag group) or ice blanket cooling (Blanket group) (n =10 each) according to random number table.Patients in Bag group had temperature reduction by placing ice bag over great vessels,whereas in Blanket group an ice blanket (temperature was set as the nasopharyngeal temperature of 33℃-34℃) was employed to have temperature reduction.Hypothermia therapy in the two group groups was initiated from the beginning of operation and continued for 48 hours after operation.Intracranial pressure,cerebral perfusion pressure (CPP) and GCS in both groups were recorded respectively at 10 minutes before operation (T0) and at 8,12,24,48 and 72 hours after operation (T1,T2,T3,T4 and T5).Venous blood of the two groups was harvested to assay the serum concentration of neuronspecific enolase (NSE),myelin basic protein (MBP)and S-100β at T0,T3,T4,-Ts and at 96 hours after operation (T6) by ELISA method.Glasgow Outcome Scale (GOS) was evaluated at postoperative six months.Results In Bag group,body temperature (T1-T5) of the patients had no significant decrease (P > 0.05) and NSE (T3-T6),S-100β (T3-T6) and MBP (T4-T6) were increased (P < 0.05 or 0.01) when compared with those in T0 ; intracranial pressure (T2-T5) was increased (P < 0.05) and CPP (T3-T5) was lowered (P < 0.05) when compared with those in T1.In Blanket group,body temperature (T1-T6) of the patients presented was decreased significantly (P < 0.01) and NSE (T3-T6),MBP (T5-T6) and S-100β (T4-T6) were increased (P < 0.05 or 0.01) when compared those in T0 ; intracranial pressure (T2-T6) was increased (P < 0.05) and CPP had no significant changes (P >0.05) when compared with those in T1.By contrast with those in the same time points in Bag group,lower body temperature (T1-T5) (P < 0.001),lower intracranial pressure (T2-T5),higher CPP (T3-T5) as well as lower NSE (T4-T6),MBP (T4-T6) and S-100β(T6)were observed in Blanket group (P <0.05 or 0.01).Changes of GCS and GOS in the two groups were no significance (P >0.05).Conclusion Intraoperative and postoperative mild hypothermia therapy using an ice blanket may alleviate the degree of brain injury in sTBI patients.
