1.New evidences of α7nAChR participating in 100 Hz EA regulating NK cells activity

Objective On the basis of previous experiments ,we try further to seek the new evidences of α7nAChR participating in 100 Hz electroacupuncture (EA ) regulating natural killer (NK ) cells activity .Methods α7nAChR antibody and the specificα7nAChR antagonist α-bungarotoxin (α-Bgt) were used to study the effect of α7nAChR on NK cell activity in 100 Hz EA mice .Re-sults Compared with 100 Hz EA group ,100 Hz EA plus α7nAChR-Ab group showed a significant enhanced in NK cell activity (P<0 .05) .100 Hz EA treated with 1 .5 or 3 .0 μg α-Bgt groups had higher NK cell activity than 100 Hz EA group(P<0 .01) . Conclusion α7nAChR is involved in 100 Hz EA modulation for immune function ,which provide the theoretical and practical basis for further clarifying and expanding the clinical application of EA .
2.Celastrol induces caspase-dependent apoptosis through ROS/JNK path-way in Saos-2 cells

AIM:ToexploretheeffectofcelastrolonapoptosisofSaos-2cellsanditsmechanism.METH-ODS:Saos-2 cells were treated with various concentrations of celastrol , and the cell viability was measured by MTT assay . Apoptosis and reactive oxygen species ( ROS) production were determined by flow cytometry .The protein levels of cleaved caspase-9, cleaved caspase-3 and phosphorylated JNK were evaluated by Western blot .RESULTS:The viability of Saos-2 cells was significantly inhibited by celastrol .Celastrol significantly induced apoptosis of Saos-2 cells.Celastrol signifi-cantly induced ROS production in the Saos-2 cells.Western blot analysis demonstrated that celastrol significantly increased the protein levels of cleaved caspase-9, cleaved caspase-3 and phosphorylated JNK in the Saos-2 cells.CONCLUSION:Celastrol induces caspase-dependent apoptosis through ROS/JNK pathway in Saos-2 cells.
3.Reconstruction of unilateral cleft lip with vermilion musculomucosal flap: a clinical study

Objective To design a new vermilion musculomucosal flap to reconstruct the normal appearance of unilateral cleft lip,and to analyze the clinical effects.Methods All patients were repaired with Millard method,and vermilion musculomucosal flap was formed with fixed point method to repair vermilion.Normal function and morphology were recovered by reconstructing lip beads and forming a peak of cupid's bow.Results 78 cases were enrolled in this study.They were followed-up for 6 to 36 months after operation.Except for 6 cases of nasal deformity and 5 mild lift of vermilion of affected side,67 cases obtained satisfied nasolabial form and function,with full decline and symmetry of cupid's bow peak,and well form of nasolabial fold,and natural transition of vermilion,and significance of lip beads,and inconspicuous scars.Conclusions Appling vermilion musculomucosal flap for reconstructing vermilion of unilateral cleft lip has good effect,and it is simple and feasible.It can be a regular surgery method,and is worthy of clinical promotion.
4.EFFECTS OF SELENIUM-AND ZINC-ENRICHED PROBIOTICS ON BLOOD CONTENT OF SELENIUM AND ZINC,ANTIOXIDATION FUNCTION AND INTESTINAL MICROFLORA IN CANINES

Objective To investigate the effects of selenium-and zinc-enriched probiotics on the content of selenium and zinc in blood,antioxidation function and intestinal microflora in canine.Method Eight 18-month native canines,female and male in half,were randomly divided into the control and treatment groups on average.The control group received basal diet,the treatment group received basal diet supplemented with 2.0g selenium-and zinc-enriched probiotics everyday.To determine the experimental indices,the samples were collected on D0,D15 and D30,respectively.Results Compared with the control group,on D15,the content of selenium and zinc in blood,blood GPX,serum SOD,T-AOC and the amount of Lactobacillus in the experimental group were significantly increased,while the amount of Escherichia coli significantly decreased,but the serum MDA and the amount of Bifidobacterium,Staphylococcus and Enterococcus had no significant change.On D30,the content of selenium in blood,serum SOD,T-AOC and the amount of Lactobacillus were very significantly increased,while the content of zinc in blood,blood GPX and the amount of Bifidobacterium significantly increased;but serum MDA and the amount of Escherichia coli,Staphylococcus and Enterococcus very significantly decreased.Conclusion Selenium-and zinc-enriched probiotics could increase content of selenium and zinc in blood,enhance antioxidation function,improve and regulate the intestinal microflora.

AIM: To establish the hyperlipemic rat model by long-term gastric perfusion of intralipid and investigate the effects of hyperlipidemia on bone. METHODS: Eighteen SD rats were randomly divided into control group (C) and high-fat-diet group (HDF), with 9 rats per group. Rats of C group were treated orally with normal saline, and rats in HDF group were oral gavage of intralipid at dose of 5.0 mL/kg, once a day for 20 weeks. At the end of experiment, their serum levels of lipid were measured. 7.Expression of smad2/3 protein and the effects of flunarizine on its expression in brain tissue following transient cerebral ischemic reperfusion in gerbils

Objective To study the expression of smad2/3 protein and the effects of flunarizine on the its expression in braln tissue following transient cerebral ischemie reperfusion in gerbils. Methods A cerebral ischemia-reperfusion model in gerbils was established by clamping both common carotids. Thirty-five gerbils were randomly divided into three groups, sham operation group, cerebral ischemia-reperfusion group and flunarizine treatment group. The expression of smad2/3 protein in brain tissue was detected by immunohistochemistry technique. Results Experimental results revealed that smad2/3 protein was expressed in neuroeyte in 35 gerbil brain. Compared with sham operation group, the expression of smad2/3 protein in neurecytes of cerebral isehemia-reperfusion group was evidently increased at the lst day, 3rd day and 7th day (P <0. 01). Compared with cerebral ischemia-reperfusion group, the expression of smad2/3 protein in neurecytes of gerbils in flunarizine treatment group was evidently decreased at these time point (P < 0.05). Condusions Smad2/3 protein was expressed in nettrcvytes of gerbils. Expression of smad2/3 protein in neuroeytes of gerbils was evidently increased following cerebral ischemic reperfusion, and its expression in flunarizine treatment group was evidently decreased.
8.The platelet characteristics of 22q11 microdeletion and its clinical application

Objective 22q11 microdeletion is featured with hemotological dysfunction,among others:hypocalcemia,thrombocytopenia and megathrombocyte.This study was trying to work out the platelet characteristics of 22q1 1 microdeletion patients and its potential clinical application.Methods A total of 80 cases who had undergone open-heart surgery were selected and confirmed with fluorescence in situ hybridization (FISH) to have 22q1 1 microdeletion among 40 cases while the others were negative.The relevant data in full blood routine of all the 80 cases in hospital information system (HIS) then were collected and analyzed with suitable statistical methods.Results The mean platelet volume in microdeletion group was statistically higher than that in the control group [(11.20 ± 1.94)fL vs (8.95 ± 1.58) fL,P <0.01].The area under the receiver operating characteristic (ROC) curve was 0.82,meaning significant predictive values.The corresponding sensitivity and specificity for mean platelet volume (MPV) =10 fL were 70.0 % and 80.0 %,respectively.Conclusions MPV in congenital heart defect patients with 22q11 is significantly higher than those without 22q11.It is an effective method for preliminary screening 22q11.Being obtained from full blood routine data,it is economic and quick.MPV =10 fL can be used as a cutoff for guidance for irradiated blood transfusion postoperatively.
9.Short-and long -term outcomes of pneumonectomy for non -small cell lung cancer

Objective To assess the mortality,complications and major morbidity of pneumonectomy for non -small cell lung cancer(NSCLC)and to establish the importance of various prognostic factors.Methods The data of 64 consecutive patients who underwent pneumonectomy for NSCLC were retrospectively reviewed.Results The 30 -day mortality rate was 7.8%(5 /64).Complications developed in 29.7%(1 9 /64)and overall 5 -year survival was 1 9.0%.Pathological stage Ⅲ(P =0.030)and right pneumonectomy(P =0.01 0)were independent risk factors of an adverse outcome.Survival was not significantly influenced by histological types (P =0.1 25)or curability (P =0.587).Conclusion Pneumonectomy is associated with acceptable overall morbidity and mortality.However,the patients with pathological stage Ⅲ or right pneumonectomy require special consideration.Pneumonectomy should be performed only in selected patients.
