1.The clinical diagnosis of acute coronary syndrome and the analysis of coronary arteriograghy

Objective\ To evaluate the importance of coronary arteriography with acute coronary syndromes (ACS).Methods\ The clinical data of 271 ACS patients with coronary arteriography and electrocardiogram were compared.Results\ The incidence of normal coronary arteriography with ACS patients occurred in 21 03%,220 of 271 patients had ischemic ST-T change or abnornal Q wave (81 1%).The change of anterior wall was system frequently accompanied by LAD or three branch lesions,inferior wall and inferior wall system frequently accompanied by RCA,Lcx and RCA or three branch lesions,anterior wall and inferior wall system freguently by LAD and RCA or three branch lesions,respectively.The lesions were more than one branch and severe in patients with unstable angina.Q-wave myocardal infarction usually occurred in single branch and severe lesione,non-Q wave myocardial infarction usually occurred in other branches and severe lesion.Conclusion\ Patients with ACS should be performed coronary arteriography,it is very significant in correct diagnosis of coronary heart disease,fixed position and severe degree of lesion vessel.
2.Advance of growth factor-independent 1B(GFI1B) in common hematologic diseases

Recent studies have demonstrated that GFI1B is most prominently expressed in cells of the fetal and adult erythroid and megakaryocytic lineages. In 2006,it is first reported that GFI1B is highly expressed in acute myeloid leukemia, especially in erythroleukaemia and megakaryocytic leukaemia.This report provides theoretical basis for diagnosis and differential diagnosis of subtypes of acute myeloid leukaemia.In this review,we covered the expression of GFI1B in leukemia,T-cell lymphoma,myelodysplastic syndrome and so on.
3.RNA interference in mediated Bcl-2 gene silence affects growth of human gallbladder carcinoma xenograft in nude mice

Objective To investigate whether RNA interference in mediated Bcl-2 gene silence can decrease the gallbladder carcinoma xenograft formation rate and inhibit growth of xenograft in nude mice.Methods In the xenograft formation ability group,the Bcl-2-siRNAs were transfected into the gallbladder carcinoma cell.Transfected gallbladder carcinoma and gallbladder carcinoma were injected into nude mice hypodermically to form a gallbladder carcinoma tumor.The formation rate and volume of the tumors were measured and the tumor growth rates were calculated.In the therapy group,the animal models of human gallbladder carcinoma in nude mouse body were constructed.The nude mice were randomly divided into pSilencerTM-EGFP sh515 group,pSilencerTM-EGFP shCon group,and normal control group.After the formation of tumor knot,siRNA expressing plasmids pSilencerTM-EGFP sh515 and pSilencerTM-EGFP shCon were injected into the knots of pSilencerTM-EGFP sh515 group and pSilencerTM-EGFP shCon group.The tumor volume was measured and tumor growth rate was calculated.All mice were sacrificed after 6 weeks.The distribution of Bcl-2 in the tumors was detected by immunohistochemistry.Results The nude mouse model of human gallbladder cancer xenograft tumor was constructed successfully.Xenograft formation ability:The xenograft formation rate of gallbladder carcinoma and Bcl-2 siRNA transfected gallbladder carcinoma was 100% and 60%,respectively;the average tumor volume in gallbladder carcinoma group and transfected gallbladder carcinoma group was(1 914.6?125.0)mm3 and(629.7?78.9)mm3;the growth rate was 45.58% and 14.99%,and the average tumor weight was(2.24?0.33)g and(0.77?0.12)g,respectively.The tumor volume,weight and growth rate in the transfected gallbladder carcinoma group decreased significantly when compared with those in control group(P

Silent information adjustment factor(Sirtuins)are Ⅲ class histone deacetylase on the nicotinamide adenine dinucleotide(NAD +), and regulate various enzyme activities and biological processes.Recent studies have found that members of the Sirtuins family are closely related to the occurrence of bronchial asthma.This review summarizes the role of the Sirtuins family in the pathogenesis of bronchial asthma and provides a basis for the Sirtuins family as a potential therapeutic target for bronchial asthma.
5.Effect of platelet derived growth factor on inducing hepatic stellate cell activation in tumor microenvironment of hepatocellular carcinoma

The function of hepatic stellate cell (HSC)in liver fibrosis has been well recognized.In recent years,the role of HSC in the liver tumor microenvironment have been paid increasing attention.Platelet derived growth factor (PDGF) is the most important mitogen of hepatic stellate cell,and plays an important role in the activation of hepatic stellate cell.Hepatic stellate cell is not only the target cells of PDGF,but also PDGF-secreting cells.Thus a bi-directional cycle mode of PDGF activating hepatic stellate cell has been established.The signaling pathways of HSC activation include MAPK,PI-3K,Ca2+ and JAK pathways.To explore PDGF activating hepatic stellate cell in liver tumor microenvironment and to find the new methods of targeting PDGF and hepatic stellate cell,may help us find a new direction for the treatment of hepatocellular carcinoma.
6.Expression and significance of breast cancer suscepterbility gene 1 in leukemia

Objective To investigate the expression and significance of breast cancer suscepterbility gene 1 (BRCA1) in leukemia.Methods Fluorescent quantitative reverse transcription-polymerase PCR was used to investigate the expression of BRCA1 in 18 patients with ALL-L2 (13 denovo ALL patients,5 relapsed ALL patients),20 patients with CML-CP and 15 normal controls.Results The mRNA expression of BRCA1 in denovo patients with ALL was lower than that in normal control,with statistical significance (P < 0.05).The mRNA level of BRCA1 in ALL patients in CR was higher compared with before to cure, with statistical significance (P < 0.05),but lower than that in normal control,without statistical significance(P > 0.05).The mRNA expression of BRCA1 in patients with relapsed ALL was lower than that in normal control with statistical significance (P < 0.05), and lower than that in denovo patients with ALL, without statistical significance (P > 0.05).The mRNA level of BRCA1 showed no difference in CML-CP patients compared with normal control (P > 0.05). Conclusion The different expression of BRCA1 in leukemia indicates that he has closely relationship with the prognosis of leukemia and guides the clinical diagnosis and treatment.
7.Treatment of Fraser type Ⅰ floating knee injuries with double intramedullary nail under minimally invasive surgery

Objective To discuss role of double intramedullary nail in the treatment of floating knee injuries under minimally invasive surgery.Methods From July 2006 to July 2010,23 patients with floating knee injuries ( Fraser type Ⅰ ) were fixed with tibial intramedullary nail and femoral retrograde intramedullary nail assisted by minimally invasive method such as portable bracket,AO large distractor and Schanz screw.The postoperative continuous passive motion (CPM) was performed for functional exercises of the knee.Results The operation lasted for average 95 minutes ( range,70-160 minutes),with average blood loss of 260 ml ( range,100-600 ml).All patients were followed up for average 11 months (range,7-35 months).According to assessment systems of Karlstrom and Olerud,the clinical results were excellent in 17 patients,good in 4 and fair in 2,with excellence rate of 91%.Conclusions Combined with the methods of portable bracket of lower limb,AO large distractor and Schanz screw,the intramedullary nail is much convenient and minimally invasive for the floating knee injuries.
8.Construction and identification of recombinant adenovirus vector of human neuropilin-1 gene

Objective To construct a adenovirus vector containing human NRP-1 gene and 3Flag gene to interaction between tumor and interstitial cell.Methods Plasmid containing NRP-1 gene was digested by AgeⅠand NheⅠ restriction endonuclease.Then the DNA was ligated into linearized pDC315-3Flag vector.After having been constructed,the pDC3 1 5-NRP-1-3 Flag plasmid was co-transfected with framework plasmid pBHGlox△E1 , 3Cre into HEK 293 cells to obtain the homologous recombinant adenovirus,which was then amplified and purified its titer tested.Expression of NRP-1 protein was detected using Western blot.Results Polymerase chain reaction and sequencing analysis confirmed that the shuttle plasmid pDC3 1 5-NRP-1-3 Flag and design were consistent. Cytopathic effect was observed by inverted phase contrast after transfecting HEK2 9 3 cells with shuttle plasmid pDC315-NRP-1-3Flag.95-130 ku was detected by Western lot after transfecting HEK293 cells with shuttle plasmid pDC315-NRP-1-3Flag and recombinant adenovirus,the size being consistent with the NRP-1-3Flag fusion protein (104 ku),with a titer of 2.00E+11PFU/mL.Conclusion The recombinant adenovirus vector for human NRP-1 gene was successfully constructed expressed in HEK 2 9 3 cells.
9.The effect of sacral nerve stimulation (SNS) on spastic pelvic floor syndrome

Objective To evaluate sacral nerve stimulation (SNS) in the treatment of spastic pelvic floor syndrome (SPFS).Method In this study,36 patients of spastic pelvic floor syndrome who received SNS treatment from 2011.3 to 2011.12,were reviewed in terms of clinical curative effect,changes of anal pressure and defecography.Result After a course of SNS treatment,patients were followed up for 3 months,12 cases were cured,22 cases improved and 2 cases were ineffective,the total effective rate was 94.4%.The symptoms such as endless defecate feeling,difficulty in defecation,anal pain and anal obstruction feeling improved significantly (P <0.01 ).After the therapy,rectal anal reflex( RAR)threshold value volume rose,anal maximum contraction pressure (AMCP),anal rest perssure (ARP) decreased (P < 0.01 ).While the anal longest contraction time (ALCT)and rectal rest pressure (RRP) did not change significantly (P > 0.05 ).After treatment,when patient defecate the anorectal angle (ARA) increases,the puborectal muscle spasm notch (PMSN)attenuates (P < 0.01 ).Conclusions SNS is effective and minimally invasive in treating spastic pelvic floor syndrome.
