2.PROTECTIVE EFFECT OF MELATONIN ON KIDNEY IN DIABETIC RATS

To investigate the protective effect of melatonin on the kidneys of STZ-induced diabetic rats, experimental diabetes was induced by intraperitoneal injection of streptozotocin 50 mg/kg in male Sprague-Dawley rats (150~200g). Three days after streptozotocin injection, the diabetic rats were assigned to one of the following groups: (1) DM, untreated; (2) DM+Mel 1, melatonin supplement at 0.2mg?kg -1?d -1, by gavage; (3) DM+Mel 2, melatonin supplement at 5mg?kg -1?d -1, by gavage. The treatment continued for 8 weeks. Periodically, body weight, blood glucose, plasma triglyceride and cholesterol levels were measured as clinical and biochemical parameters; plasma creatinine, creatinine clearance( Ccr), urinary protein excretion were detected as renal function indexes; kidney weight, glomerular area, PAS positive area, and cell counts were evaluated as histological damage indexes. Melatonin treatment had no significant effects on blood glucose and plasma cholesteral, but lowered plasma triglyceride level in diabetic rats. Melatonin treatment reduced urinary protein excretion. Until the end of the experiment, there were no significant changes in plasma creatinine level, Ccr in diabetic rats. Melatonin treatment decreased kidney weight/body weight ratio, reduced PAS positive area, glomerular cellular number and glomerular area which were increased in DM rats. The protective effects of melatonin were more obviose in high dosage groups. Melatonin treatment can aggravate glomerular hypertrophy and mesangial cell proliferation, delay the progression of glomerulosclerosis, and improve renal function of diabetic rats.
3.THE STUDY OF OCCUPATIONAL HAZARDS OF VDT

Both monitoring of occupational hazards and assessment of healthy effects on VDT operators were made. The results showed that there were several occupational hazards in VDT,including insalubrious microclimate and air ion condition, lower illuminance, higher noise, some doses of x-radiation, and unfitting ergonomics. The healthy effects of the hazards of the VDT mainly involved the operator's nervous system, psychology and behavior, eyes, musculoskeletal system as well as the woman operator's menses and pregnancy, especially in the 18 to 25 age bracket who had more VDT work hours than that of the others, it suggested that the various preventive measures against the hazards of the VDT must be strengthened.
4.VDT OPERATORS' OCCUPATIONAL STRESS AND PSYCHOPHYSICAL STATE

VDT operators' occupational stress and psy-chophysical state were studied. The resultsshowed that the nature, environment and require-ments of VDT operation shaped the major occupa-tional stress in VDT operation. The longer VDTworking hours and more workloads gave the opera-tors more occupational strains. VDT operators hada lower satisfactory level to the job and lower psy-chological health level than contral group, espe-cially on obsession and depression. VDT operationperhaps gave the operators some kinds of psycho-physiological troubles. We did not find there wereharmful effects of VDT on the operators' autonom-ic nervous functions.

6.Relationship between colorectal carcinoma and lymph node metastasis

Objective To provide the theoretic basis for the clearance of the metastatic lymph nodes in radical operation of colorectal carcinoma(CRC) by means of exploring the relationship between mass size of both tumor and lymph nodes and the rate of lymph node metastasis. Methods A total of 702 lymph nodes from 57 cases of CRC radical operations were divided into 5 groups and each of the lymph nodes was examined. The size of lymph nodes, Dukes' phases, the pathological types of CRC and their metastasis rate were analyzed. Results ① High metastasis rate was found at Dukes' C and D phases of CRC, being 29.93% and 48.12%, respectively. ② The metastasis rates of mucous adenocarcinoma and signet ring cell carcinoma were the highest of all pathological types, being 59 52% and 59.26%, respectively. ③ Metastasis rate of lymph nodes was not correlated with the mass size of tumor, but correlated with Dukes' phase. Conclusion ① Metastasis rate of lymph nodes increases with the growth and infiltration of tumor. ② Metastasis rates of mucous adenocarcinoma and signet ring cell carcinoma are higher than those of well and mid differentiated carcinomas. ③ Metastasis may be suggested for lymph node of CRC more than 5 mm in diameter during operation. However, clearance of lymph node of CRC less than 5 mm in diameter needs further study.
7.Analysis with Pareto chart on disease component of discharged deathbed-patients from geriatric nursing organization

Objective To understand the main and multiple kinds of diseases and probe into key points of diseases for hospice care in geriatric nursing organization in Beijing, Tianjin and Shanghai city. Methods Pareto chart wag used to analyze situation on disease component of deathbed-patients discharged from 22 geriatric nursing organizations from years 2005 to 2007 in Beijing, Tianjin and Shanghai. Results Components of cardiovascular disease, cerebrovascular disease, respiratory system disease, finale malignancy and senile decrepitude were accounted more than 95%.Conclusions Kinds of diseases in deathbed-patients discharged from nursing organizations in city were relatively concentrated, especially cardiovascular disease, cerebrovascular disease, respiratory system disease, finale malignancy and senile decrepitude, etc should be focal point of hospice care service in geriatric nursing organizations.

9.Effects of rosiglitazone on the expression of TNF-? in visceral adipose tissue of insulin-resistant rats

Objective To explore the effects of rosiglitazone on the expression of TNF-? in visceral adipose tissue of insulin-resistant rats induced by high-glucose diet. Methods Sprague-Dawley (SD) rats were fed with high-glucose diet for 6 months to reproduce the non-obesity insulin-resistant rat model (abbreviated as IR rats). The model rats were then randomized into insulin resistan (IR) group and rosiglitazone (ROS) group (10 each). Ten rats fed with normal diet were enrolled as controls (CN group). Rats in ROS group were administered with 10?mol/(kg?d) of rosiglitazone for 6 weeks from the 7th week of experiment,and those in CN and IR groups were administered with same amount of distilled water. At the 6th and 12th week,the systolic pressure,body weight,and biochemical parameters (fasting and 2-hour post-glucose load plasma glucose,insulin,free fatty acid,triglyceride,high density lipoprotein cholesterol) were measured,and insulin resistance index (HOMA-IR) was calculated. At the end of the 12th week,the visceral adipose tissue was collected to calculate the ratio of visceral fat to body weight (VF/BW),and to determine the protein and mRNA expressions of tumor necrosis factor-? (TNF-?) with inmmunohistochemical assay and RT-PCR. Results As compared with CN group,the systolic pressure,HOMA-IR,plasma insulin,triglyceride and free fatty acid increased,whereas high density lipoprotein cholesterol decreased (P0.05). Inmmunohistochemical examination showed positive expression of TNF-? in intercellular substance,but not in visceral adipocytes. The expressions of TNF-? protein and mRNA in visceral adipose tissue were dramatically higher in IR group than in CN and ROS groups (P0.05). Conclusion Rosiglitazone may significantly abate insulin resistance,reduce the abnormality of plasma lipid and blood pressure,and inhibit the over expression of TNF-? in visceral adipose tissue in the non-obesity insulin-resistant rats induced by high-glucose diet.
10.Study on differentially expressed genes in papillary thyroid carcinoma by cDNA microarray

Objective To screen the differentially expressed genes between papillary thyroid carcinoma (PTC) and normal thyroid tissues using cDNA microarray. Methods mRNA from both PTC and normal thyroid tissues were reversely transcribed to cDNAs with the incorporation of fluorescent dUTP (Cy-5 or Cy-3) to prepare the hybridization probes. The mixed probes were hybridized to the cDNA microarray, which was scanned for the fluorescent signals and showed difference between two tissues. Two among the differentially expressed genes were verified by RT-PCR and immunohistochemistry. Results In PTC 48 genes were upregulated while 17 were downregulated. The results of RT-PCR and immunohistochemistry were consistent with that of the gene chips. Conclusion cDNA microarray technique is effective in screening the differentially expressed genes between 2 different kinds of tissue. The obtained genes are mainly related with extracellular matrix, cytokine, signal transduction and so on.
