2.Inhibition Effects of Nimesulide on Proliferation in Human Cholangiocarcinoma Cell Line QBC939 in Vitro

Objective To investigate the effects of nimesulide, a selective cyclooxygenase-2 (COX-2) inhibitor, on human cholangiocarcinoma QBC939 cell line in vitro. Methods The effects of nimesulide on QBC939 cells were observed with the following techniques: the influence of nimesulide on the proliferation of QBC939 cells was determined by MTT assay; the apoptosis of QBC939 cells was viewed and measured by transmission electron microscopy and flow cytometry, respectively; the expressions of proliferation cell nuclear antigen (PCNA) and COX-2 of cholangiocarcinoma cells were detected by immunocytochemistry. Results Nimesulide inhibited the expressions of PCNA and COX-2 and the proliferation of cholangiocarcinoma QBC939 cells, whose effects intensified as the dose increased and time elongated. Flow cytometry showed that the apoptotic rates of QBC939 cells increased significantly as the dose of nimesulide increased. The typical morphologic features of apoptosis were also observed by transmission electron microscopy. Conclusion Nimesulide significantly inhibits the proliferation of QBC939 cells in vitro by inducting cell apoptosis, which may be associated with the downregulation of COX-2 expression, and it also presents the features of dose and time dependents.
3.Effects of nimesulide on proliferation and apoptosis of human cholangiocarcinoma cell line QBC939

Objective To investigate the effect of a selective inhibitor of COX-2 nimesulide on growth and apoptosis of human cholangiocarcinoma cell line QBC939 in vitro. Methods MTT assay was used to determine the influence of nimesulide on the proliferation of QBC939 cells, apoptosis of QBC939 was measured by transmission electron microscopy and flow cytometry.Expression of apoptosis related genes mRNA and bcl-2 ,bax, survivin were detected by RT-PCR and immunocytochemistry. Results Nimesulide effects a dose-dependent and time-dependent growth inhibition on QBC939 cells. High concentration of nimesulide (200 ?mol/L) not only inhibits the growth of QBC939 cells but also induces apoptosis cell nuclear condensation and apoptotic bodies were seen by transmission electron microscopy. Immunocytochemistry and RT-PCR shows upregulation of bax and down regulation of bcl-2 and survivin. Conclusion Nimesulide significantly inhibits the proliferation of QBC939 in vitro by induction of apoptosis in a dose- and time- dependent manner.

Objective To study the effect of pulse pressure(PP)/mean artery pressure(MAP) ratio in evaluating arteriosclerosis.Methods Blood pressures from 200 subjects without cerebrovascular nor cardiovascular disease were obtained and analyzed by different age groups.PP/MAP ratio was calculated for each patient and compared.Results PP/MAP ratio was significantly different in different age groups.Conclusion PP/MAP ratio has clinical significance in evaluating arteriosclerosis and is worthy of further investigation.

6.Clinical Observation on Modified Bushen Huoxie Decoction for the Treatment of Vascular Dementia

[ Objective ] To observe the therapeutic effect of Bushen Huoxie Decoction (mainly composed of Radix Rehmanniae Preparata, Fructus Corni, Rhizoma Dioscoreae, Carapax et Plastrum Testudinis, Radix Polygoni Multiflori, Fructus Lycii, Radix Morindae Officinalis, Herba Cistanches, Rhizoma Acori Tatarinowii, Radix Polygalae, etc. ) for the treatment of vascular dementia (VD) . [Methods] Eighty cases of VD were randomized into two groups by using random number table: group A ( n = 40) was treated with modified Bushen Huoxie Decoction and group B (n = 40) with duxil and nimodipine. After a 3-month treatment, the therapeutic effect of the two groups was evaluated, and mini-mental state examination (MMSE), activities of daily life (ADL) and blood rheology before and after treatment were observed. Objective To investigate the value of ionic-contrast esophagogram in diagnosis of intrathoracic anastomotic leakage after oesophagectomy.Methods The data of 728 patients suffered esophagectomy were retrospectively collected.723 patients without clinical manifestations of anastomotic leakage were divided into group A(esophagogram before eating,n=465) and group B(non-esophagogram before eating,n=258).The incidence of anastomotic leakage was compared between the two groups.Results The discovery rates of anastomotic leakage in group A and B were 0.43%(2/465) and 0.78%(2/258),respectively,the difference was no statistically significant between the two group(P=0.55).The total incidence rates of anastomotic leakage in group A and B were 0.65%(3/465) and 0.78%(2/258),respectively,the difference was no statistically significant between the two groups(P=0.84).5 patients with clinically suspected anastomotic leakage,all had loculated pleural effusion,4 cases had fever,and 2 cases had abnormal chest drainage.Finally,4 cases(80%) were confirmed to be anastomotic leak by esophagogram.Conclusion Routine ionic-contrast esophagogram before eating do not improve intrathoracic anastomotic leakage detection rate after esophagectomy,but when patients have fever,loculated pleural effusion and abnormal chest drainage,esophagogram is necessary and can improve the detection rate of esophageal anastomotic leak.CT radiography may detect smaller anastomotic leakage than conventional esophagogram.

The demand of hospital car in first-aid and patients carrying is urgent in paroxysmal epidemic,disaster and wartime.According to types and characteristics of civil-train,many methods are used to rapidly refit civil-train to hospital car,such as rapid taking down devices from civil-train,installing medical appliances and unloading the device in the civil-train and auxiliaries.Based on analysis and comparison,YZ25G hard-seat train is the best one to refit from civil-train to hospital car.
9.Relative Bioavailability of Ibuprofen Sustained Release Capsules in Healthy Volunteers

OBJECTIVE:To study the pharmacokinetics and relative bioavailability of ibuprofen sustained release capsules in healthy volunteers for evaluating the bioequi valency .METHODS: 20 healthy volunteers took a single dose(600mg) of ibuprofen sustained release capsules and fenbid capsules(as reference drug) alternatively, then the plasma concentrations of ibuprofen were determined by HPLC.The pharmacokinetic parameters were calculated with 3p87 pharmacokinetic program and the bioequivalence was calculated .RESULTS: The pharmacokinetic parameters of ibuprofen sustained release capsule and fenbid capsule wereT1/2= (2.88+ 0.25)h, (2.91 + 0.33)h;Cmax = (30.66+ 4.21)mg/L, (29.09+3.54)mg/L;Tmax = (4.4 + 0.52)h, (4.3+0.48)h;AUCo-1, = (174.26+13.95)mg/(h . L), (165.80+ 11.89)mg/(h ?L) ;C = (16.25+ 1.47)mg/L, (15.63+1.83) mg/L; Cmin= (1.36 + 0.56) mg/L, (1.24 + 0.46) mg/ L; FI = (1.83 + 0.07), (1.84 + 0.06) .The relative bioavailability of ibuprofen sustained release capsule was( 105.10 + 7.77) % .CONCLUSION: Ibuprofen sustained release capsule and fenbid (reference drug) were bioequivalent.
10.Determination of the Concentration of Argine Asprine in Human Plasma With HPLC

OBJECTIVE:To develop a HPLC method for determining the concentration of argine asprine in plasma MET_HODS:The analytical column was YWG C18(4 6mm?250mm,10?m) and kept analyzing at 33℃ The mobile phase consisted of a mixture of purified water-acetonitrile(96∶4,v/v)with 20?l of butylamine and 5?l of acetic acid in per 100ml mobile phase And the plasma sample was extracted with iso-propyol alcohol-chloroform(95∶5,v/v) RESULTS:The calibration curve was linear within the range of 0 97~125 0mg/L plasma concentration and the detective limit was 0 97mg/L The RSDs for inter-day and intra-day in sample of 125 0,31 25,1 95mg/L were less than 10% This method has been used for determining plasma drug concentrations in 8 healthy volunteers and the pharmacokinetic parameters of the drug were obtained CONCLUSION:The method was sensitive,accurate and stable,which is suitable for the study of bioavailibility and pharmacokinetics of argine asprine
