1.Contrast in respiratory mechanics and blood gas between bronchial blocker and double lumen tube in one lung ventilation

Objective To compare the influences between bronchial blocker (BB) and double lumen tube (DLT) in respiratory mechanics and blood gas in one lung ventilation. Methods Thirty-six patients undergoing pulmonary lobectomy or radical operation of esophageal carcinoma were randomized into group BB (17 cases) and group DLT (19 cases). In group BB,BB was intubated under the guidance of a fibreoptic bronchoscope after intubation of single lumen tube; while in group DLT, the position of the tube was confirmed using fibreoptic bronchoscopy after intubation. The peek airway pressure (Pmax) and pulmonary dynamic compliance (CLdyn) during double lung ventilation and one lung ventilation were recorded in recumbent position. Blood samples were taken from, radial artery 20 min after one lung ventilation for blood gas analysis,partial pressure of oxygen in artery(PaO2), partial pressure of carbon dioxide in artery(PaCO2), saturation of arterial blood oxygen (SaO2) were recorded. Results No significant difference was found in Pmax and CLdyn between two groups during double lung ventilation,but Pmax was significantly lower in group BB [(21.00 ± 3.59) cm H2O,1 cm H2O = 0.098 kPa] than that in group DLT [(26.89 ±4.21) cm H2O] (P<0.05), and CLdyn was greater in group BB [(28.10 ±6.24) ml/cm H2O] than that in group DLT [(24.00±7.13) ml/cm H2O ] (P < 0.05); there were no significant differences in PaO2, PaCO2, SaO2 betweentwo groups during one lung ventilation (P> 0.05). Conclusion The influence to respiratory mechanics in one lung ventilation is smaller using BB than using DLT, BB has blocker well applicable value in clinics.
2.The clinical pathological analysis of 23 patients with renal damage caused by microscopic polyangiitis

Objective To study the clinical features and pathology of renal damage in patients with MPA. Method The clinical pathological changes of 23 MPA patients were analyzed and the patients with positive ANCA were compared with those with negative ANCA. Results Most MPA patients were senile and male with the symptoms of lung damage, pleuritis, arthritis and myalgia as well as extrarenal symptoms such as fever, weight-loss, and anorexia. 65.2% of the patients were ANCA (+). Symptoms of renal function damage were hematuria and proteinuria, which could be found in all the patients. Different degree of renal damage could be detected. Glomerular cresent formation, which were mostly fibrous, could be found in all of the 23 patients. Half of the patients have tuft necrosis and interstitial vessel vasculitis. Conclusion MPA patients often have extrarenal symptoms besides renal function damage. Patients with positive ANCA differs from patients with negative ANCA in both clinical manifestation and kidney pathology.
3.Construction of recombinant adenovirus vector carrying human miR-133a and its expression in vitro

AIM:To construct recombinant adenovirus vector carrying human miR-133a and study its expression in human mesenchymal stem cells(hMSCs).METHODS:The PCR product containing miR-133a was amplified from human genomic DNA and inserted into the adenoviral shuttle vector pAdTrack-CMV.Then the recombinant shuttle plasmid linearized by pmeⅠwas cotransformed into competent E.coli.BJ5183 with the adenoviral backbone plasmid pAdEasy-1 to generate the recombinant adenovirus vector rAd-mir-133a.rAd-mir-133a was then packaged and amplified in human embryonic kidney 293(HEK293) cells.The purified rAd-miR-133a was used to infect the hMSCs and the expression of miR-133a was detected by non-quantitative RT-PCR and real-time PCR.RESULTS:The recombinant adenovirus shuttle vector pAdTrack-CMV-miR-133a was constructed and verified by restriction endonuclease analysis and DNA sequence analysis.rAd-miR-133a was successfully packaged and amplified in HEK293 cells.The transcriptions of primary miR-133a and mature miR-133a were over-expressed in the hMSCs infected with rAd-miR-133a.CONCLUSION:The recombinant adenovirus vector carrying human miR-133a is successfully constructed,which lay a foundation for miR-133a function study.
4.Study on biomechanical properties of several scaffold materials for tissue engineering

BACKGROUND:It is still a research focus on constructing substitution of the human tissues and organs, or producing the alliance for grafting by engineering methods in tissue engineering. Among these researches, it is pivotal to choose appropriate materials. The prepared scaffolds should have suitable tensile strength and mechanical toughness to withstand the various outside forces without being damaged. So, it is very necessary to evaluate the biomechanical properties of candidated materials in tissue engineering, which can supply the references for selecting materials for tissue scaffolds and their designation.OBJECTIVE: To investigate the biomechanical properties of nine kinds of scaffold materials, in order to supply a biomechanical basis for the selection and design of scaffold materials for tissue engineering.DESIGN: A repetitive measurement study.SETTING: College of Bioengineering, Chongqing University.MATERIALS: The materials involved in this study were poly (DL-lactic-co - glycolic acid) (PLGA), sodium polymannuronate, gelatine, chitosan, collagen, acellular porcine dermis (APD), acellular vascular matrix (AVM),APD-PLGA, AVM-PLGA, modified gelatine and chitosan.METHODS: All the experiments related to this study were completed in the Biorheology laboratory of the College of Bioengineering, Chongqing University from April 2006 to March 2007. The nine materials above were prepared, gelatine and chitosan were modified. Stress-strain testing was performed at 10 mm per minute by a material testing machine (INSTRON 1011, USA). The Yang's modulus was calculated in the range of 0.005 to 0.02, the ultimate strain and stress were also obtained.MAIN OUTCOME MEASURES: The ultimate strain, ultimate stress and Yang's modulus of the nine materials were analyzed.polymannuronate > AVM-PLGA > collagen > gelatine (P < 0.05). The rate of burst straining of chitosan and PLGA were greater than those of other materials, 133% and 276% respectively (P < 0.05). In addition, after being combined with ultimate stresses of APD and APD-PLGA were greater than that of other materials, i.e., their burst strengths were greater than those of other materials. The data also indicated that the burst strength of APD-PLGA was a little greater than that of APD (P > 0.05). The burst strengths of gelatin, chitosan, and collagen were similar at the range of 7.67 to 9.51 MPa (P > 0.05). The burst strengths of collagen and sodium polymannuronate were from 1.16 to 1.40 MPa, which were the least among all the materials. At the same time, being combined with PLGA, the burst strength of AVM-PLGA greatest, i.e., its rigidity was the greatest. The rigidity of APD was the least. After combined with PLGA, the rigidity of AVM and APD were increased (P < 0.05), and corresponded with PLGA (P> 0.05). Except for gelatin, the order of rigidity in the materials was AVM-PLGA > PLGA > APD-PLGA > AVM > chitosan > sodium polymannuronate > collagen > APD.CONCLUSION: AVM and APD have good biomechanical properties, which are very different from the water-soluble collagen. It is promising to improve the biomechanical properties of sodium polymannuronate, gelatin and chitosan by the complex of PLGA.
5.Effect of Tramadol Combined with Ketorolac Tromethamine in 30 Case of Patients of Patient-controlled Intravenous Analgesia after Gynecological Laparoscopic Surgery

Objective To observe the effect of combination use of tramadol and ketorolac tromethamine for patientcontrolled intravenous analgesia (PCIA) after gynecological laparoscopic surgery. Methods Ninety patients with American Society of Anesthesiologists (ASA) grade I or Ⅱ undergoing gynecological laparoscopic surgery under general anesthesia were randomly divided into three groups: group T (using tramadol for PCIA) , group TK (using tramadol and ketorolac tromethamine for PCIA) and group K (using ketorolac tromethamine for PCIA) . The visual analog scale (VAS) scores at rest and moving and Ramsay sedation scale(Ramsay) scores were recorded at 1 h (t1) ,4 h (t2) ,8 h (t3) , 12 h (t4) , 24 h (t5) , 36 h (t6) , 48 h (t7) after surgery.The total dosage of rescue analgesia drug and the times of needing rescue analgesia were recorded. Adverse reactions were also recorded. Results VAS scores at rest in t 1,t2,t3,t4,t5 and t6 after surgery were significantly lower in group T and group TK than in group K(P<0.05) . VAS scores at movement time was not significantly different. After surgery,incidence rate of nausea,vomiting,dizziness or sleepiness was significantly higher in group T than in group TK and group K(P<0.05) . The total dosage of rescue analgesia drug in group K was significantly higher than in group T and in group TK (P<0.05) . In 24 h after surgery,patients pressed the button of PCIA (2.89±0.597) times in group TK,(3.05±0.574) times in group T,(6.50±0.859) times in group K.Ramsay scores at each time points after surgery were significantly increased (P<0.05) . Total dosage amount of rescue drugs and number of patients using rescue drugs were significantly higher in group K than in group T and group TK (P< 0.05) . Conclusion Combination use of tramadol and ketorolac tromethamine for PCIA after gynecological laparoscopic surgery has better analgesia effect with reduced side effects.
6. Lingual mucosal urethroplasty for one patient of long-segment urethral stricture caused by lichen sclerosus

A 48-year-old male was suffered from slowly progressive dysuria over twenty years. Although multiple urethral dilatation was treated, the effect was unsatisfactory. He was diagnosed with penile lichen sclerosus related to urethral stricture due to the lichenification of glans penis, the stenosis of urethral meatus, and the long anterior urethral stricture shown by urethrography. Finally, the patient underwent an enlarged urethroplasty with lingual mucosal graft (17 cm in length), and obtained a good outcome. During the two-year postoperative follow-up, the patient maintained a satisfactory urination without any complication.
7.Molecular hydrogen is a promising therapeutic agent for pulmonary disease.

Molecular hydrogen exerts biological effects on nearly all organs. It has anti-oxidative, anti-inflammatory, and anti-aging effects and contributes to the regulation of autophagy and cell death. As the primary organ for gas exchange, the lungs are constantly exposed to various harmful environmental irritants. Short- or long-term exposure to these harmful substances often results in lung injury, causing respiratory and lung diseases. Acute and chronic respiratory diseases have high rates of morbidity and mortality and have become a major public health concern worldwide. For example, coronavirus disease 2019 (COVID-19) caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has become a global pandemic. An increasing number of studies have revealed that hydrogen may protect the lungs from diverse diseases, including acute lung injury, chronic obstructive pulmonary disease, asthma, lung cancer, pulmonary arterial hypertension, and pulmonary fibrosis. In this review, we highlight the multiple functions of hydrogen and the mechanisms underlying its protective effects in various lung diseases, with a focus on its roles in disease pathogenesis and clinical significance.

8.Dynamic changes of tear fluid matrix metralloproteinase-9 within 1 year after laser in situ keratomileusis.

OBJECTIVETo investigate the dynamic changes of matrix metalloproteinase-9 (MMP-9) level in tear fluid within 12 months after laser in situ keratomileusis (LASIK).
METHODSTwenty-two myopic patients undergoing uneventful LASIK were enrolled in this study. Tear fluid samples were collected from the patients for measurements of MMP-9 level using Western blotting preoperatively, at 7 and 14 days, and at 1, 2, 3, 6, and 12 months after the surgery.
RESULTSMMP-9 concentrations in the tear fluid of post-LASIK patients showed a time-dependent variation pattern. MMP-9 reached its peak level in the tear fluid at 14 days postoperatively, which was 2.70 times the preoperative level; it gradually decreased thereafter but was still 1.38 times the preoperative level at 12 months after the surgery.
CONCLUSIONSMMP-9 concentrations in the tear fluid of post-LASIK patients show a time-dependent variation pattern and remains higher than the preoperative level even at 12 months after the surgery, suggesting that corneal wound healing after LASIK lasts for more than 12 months.

9.Effect of processing on metabolism of amygdalin from bitter almond in rat.

OBJECTIVETo study the influence of processing on metabolism of the main component of bitter almond-amygdalin in rat.
METHODThe blood was collected at different times after amygdalin given by injection and oral, bitter almond and its processed production given by oral respectively, and then detected by both HPLC and HPLC-MS(n) methods after extraction pretreatment.
RESULTAfter injection, amygdalin was absorbed in prototype to blood rapidly, while the other three kinds of medicine given by oral were all not detected the prototype of amygdalin, but two metabolites were detected which were isomers of prunasin confirmed by mass spectrometry. The metabolic pathway of prunasin in processed bitter almond group was markedly different from the bitter almond group.
CONCLUSIONProcessing has a significant effect on bitter almond metabolic processes in rats.

10.Germplasm resources of Polygala tenuifolia and determination of presenegenin in processing products.

OBJECTIVETo research the germplasm resources and the contents of senegenin in processing products of Polygala tenuifolia.
METHODThe contents of senegenin in wild Polygala tenuifolia and cultivated samples of Polygala tenuifolia were determined by RP-HPLC, and compared.
RESULTThe contents of senegenin in wild reduce gradually along Shaanxi, Shanxi, Hebei to Dongbei. The contents of senegenin in cultivated three-year samples of three year Polygala tenuifolia from five main place was similar, 0.44%-0.49%. The content of senegenin were 0.44%-0.64% in the wand and 0.03%-0.09% in the residual part of stem, and the content of senegenin in Polygala tenuifolia was more than that in processing products.
CONCLUSIONThere is a correlation between the content of senegenin in Polygala tenuifolia and ecology environment that show a is inverse proportion with the quality grade, and the contents in the processing products were decreased. Senegenin can be used as a characteristic marker in range. This research provides a reference for search a index for quality control of Radix polygala and its processing products.
