Receptor activity modifying proteins (RAMPs) is a kind protein with transmembrane functionality.RAMPs can interact with TypeⅡ G protein coupled receptors (GPCRs) such as calcitonin gene-related peptide receptor (CGRPR),calcitonin receptor (CTR) and calcitonin receptor-like receptor (CRLR),to form a stable heterodimer expression on the cell membrane.Different RAMPs can combine with CRLR or CTR to produce different receptor phenotypes with ligand-specific affinity and thus can decide biological effect of the receptors. In addition,RAMPs can also interact with other GPCRs,which promise broader application in function regulation of G-proteincoupled receptor of RAMPs'.RAMPs' regulation of GPCRs depends on its molecular basis. Our studies of RAMPs provides a new perspective to further researches on GPCRs functionality and CGRP signal transduction.

[Objective]To provide a precise individualized design proposal for locating acetabular prosthesis by means of 3D reconstruction, reverse engineering software and rapid prototyping technology.[Method]3D CT scan pelvis image data were obtained from six patients, which displayed osteonecrosis of femoral head on one side. The data were transferred via a DICOM network into a computer workstation.A 3D model of hip was reconstructed using Amira 3.1 software, and saved in STL format. Then the 3D model was imported into Imageware 12.0 software. The normal hip joint centre of abnormal side was determined and the best orientation of the prosthesis(which was central axial ray of acetabula)was defined using reverse engineering.Locating template was designed according to the anatomic features of the acetabula and the central axial ray.Entity model was produced by rapid prototyping technology. The orientation of the prosthesis was located by matching the model to acetabular concavity at operation.[Result]By measuring postoperative standard hip anteroposterior film, the abduction angle of acetabular prosthesis was(46.00?2.49)? and the anteversion angle was(15.68?2.85)?.The expected result was obtained.[Conclusion]The navigation template designed and constructed preoperatively can provide precise location for acetabular prosthesis and avoid procedural errors at operation.

METHODS: Pericarpium Citri Reticulatae,Fructus Psoraleae,Rhizoma Atractylodis macrocephalae and Radix Codonopsis in Wenweishu Tablets were identified by TLC at two thin-layer chromatographic plates. The contents for hesperidin, psoralen and isopsoralen in this preparation were determined simultaneously by HPLC. RESULTS: The methodological study showd that a good linear correlation existed in the range of 0.039 6-0.495 ?g(r=(0.999 99)) of hesperidin,0.018 5-0.231 ?g(r=0.999 98) of psoralen and 0.019 7-0.246 2 ?g(r=0.999 91) of isopsoralen applied respectively. The average recovery of hesperidin was 98.42%(n=9),RSD was 2.69%;The average recovery of psoralen was 98.29%(n=9),RSD was 1.08%.The average recovery of isopsoralen was(98.93%)(n=9),RSD was 2.41%. CONCLUSION: The method is simple、rapid and practicle.

Objective To investigate the effects of co-exposure to LPS and heat on plasma tumor necrosis factor-? (TNF-?), interleukin-6 (IL-6), malondialdehyde (MDA) and activity of superoxide dismutase (SOD) in rats. Methods Male pathogen-free Wistar rats were randomly assigned to the following groups: saline-injected normothermic control (Group C), saline-injected heat exposure (Group H), LPS-injected normothermic control (Group L), LPS-injected heat exposure (Group HL). Mean arterial pressure (MAP) was continually monitored. Plasma levels of TNF-?, IL-6, MDA and SOD were determined at 0, 40, 80, 120 min. Results The rats in Group HL displayed much lower MAP, higher MDA and lower SOD than the other 3 groups. The rats in Group HL displayed an early rise in plasma TNF-?, IL-6 at 40 min, the peak values of both at 80 min significantly higher than the other three groups at the same time piont. Conclusion Co-exposure to LPS and heat primes the rats to advance and augment systemic inflammatory response syndrome.

Objective To investigate the genotype distribution of hepatitis C virus (HCV) in Guangdong Province by linear probe assay (LiPA) and core,non-structural protein 5B (NS5B) sequence analysis, and to evaluate the accuracy of LiPA for genotyping.MethodsOne hundred and ten HCV specimens from chronic hepatitis C (CHC) patients in Guangdong Province were genotyped by both core,NS5B sequence analysis and INNO-LiPA 2.0.The data were then analyzed with SPSS 10.0 software.ResultsAmong the 110 specimens, core and NS5B fragment could be amplified from 97 and 62 specimens, respectively, while both fragments were obtained from 57 specimens. The results of genotyping by core sequence analysis were completely consistent with the results of NS5B sequencing. One hundred and two specimens were classified into five subtypes,i.e.genotype 1a,2a,3a,3b,6a,with frequencies of 61.8% (64),9.8% (10),3.9% (4),3.9% (4),20.6% (21),respectively.All but genotype 6 strains can be genotyped correctly by using INNO-LiPA 2.0.However,only subtype 1b and 3b could be genotyped accurately.81.5% genotype 6a strains were genotyped as 1b by mistake. Conclusions Genotype 6a has become the second most prevalent genotype in Guangdong Province. The LiPA cannot distinguish genotype 6a from 1b strains accurately which needs further investigation.

Objective To study the surgical treatment of multilevel arterial occlusion of lower extremities. Methods Single or jumping arterial bypass using grafting and percuteneous transluminal angioplasty(PTA)and stenting combined with infrainguinal revascularization were used for the multilevel arterial occlusion in 25 lower extremities. Results 25 cases showed satisfactory results in 6 months to 3 years follow-up. Postoperatively, the ischemia symptoms of limbs were improved or disappeared,4 toes were amputared because of preoperative dry gangrene, the wounds received eurement in a 2 to 4 months,2 graftings became occluded in 6 months postoperation. There were no procedural or postoperative morbidity or mortality. The cumulative patency rate of grafting was 92%(23/25),The rate of curement was 100%.Conclusion According to the individual principle,it is an effective therapy method to choose single or jumping arterial bypass and PTA and stenting combined with infrainguinal revascularization for high risk patients with multilevel arterial occlusion.

Objective: To investigate the inhibitory effect of survivin antisense oligonucleotide (ASODN) on the expression of survivin gene and proliferation of human hepatocellular carcinoma cell line SMMC-7721. Methods: The 20 mer antisense oligonucleotide (ASODN) targeted to the promoter region of survivin mRNA was designed and synthesized. The expression of survivin gene in hepatocellular carcinoma cell line SMMC-7721 was blocked by means of ASODN transfection mediated by DOTAP liposomal reagent. The changes of survivin protein and mRNA expression after transfection were as-sessd by Western blot and in situ hybridization, respectively. The apoptotic rate was detected by flow cytometer. The changes of cell adherent rate, cell growth activity, and the inhibitory rate of cell growth were also studied. Results: The expression of survivin protein and mRNA were decreased markedly after survivin ASODN transfection. Meanwhile, the cell adherent rate also decreased markedly while the apoptotic rate increased markedly. Conclusions: Transfection of ASODN targeted to the promotor region of survivin mRNA by DOTAP liposomal transfection reagent could down-regulated the expression of survivin protein and mRNA significantly in 7721 cell line and inhibit the proliferation of cancer cells. Survivin could be an important target in the therapy of hepatocellular carcinoma.
