Objective To investigate the survival situation of thalassemia cases treated with Bushen Yisui Method. Methods The survival situation of 197 cases were investigated by telephone. Results The total survival rate of 197 cases is 87.3%, among which the thalassemia major is 65.71%, the thalassemia intermedia is 99.2%, the ?-thalassemia is 100%, the ?-thalassemia is 85.0%. 158 of 197 cases have been analyzed genotype, among which 63.16% is the survival rate of homozygote, 89.16% of the heterogenic duplex heterozygote, 100% of the heterozygote. Objective To investigate the relationship of the expressions of common β (βcR) and specific α chains of IL-5, IL-3 and GM-CSF receptors with the eosinophils(Eos) apoptosis for the roles of these receptors in asthma. Methods All of 12 guinea pigs were equally randomized into normal and ovalbumin sensitized asthmatic group. The apoptosis of hypodense and normodense Eos in bronchoalveolar lavage fluid (BALF) was detected by TUNEL method. The expression of common β and specific α chains of the 3 receptor mRNA were measured with RT-PCR and in situ hybridization. Results More Eos were observed in asthmatic group than in normal group, especially those in hypodense (P<0.01). The apoptotic rate of Eos in BALF were significantly lower in asthmatic group than in normal animals (P<0.01), but no difference was found between hypodense and normodense Eos. Compared with normal group, the expressions of the 3 receptor mRNA α chains were decreased in asthmatic group than in normal group (P<0.01), but βcR expression was increased significantly. More IL-3 Rα and GM-CSFRα mRNA were expressed in hypodense Eos than in normodense Eos. The contents of IL-5Rα and IL-3Rα mRNA were lower in asthmatic group than in normal group. Conclusion The apoptosis of Eos is inhibited in BALF from guinea pigs with asthma. The expressions of IL-5, IL-3 and GM-CSF receptors in Eos are regulated through 2 mechanisms: the reduced expressions of their specific α chains which attenuating the negative regulation on Eos activation and inhibited apoptosis and the increased expressions of their βcR which enhancing the positive regulation on the 2 aspects. These data suggest that IL-5, IL-3 and GM-CSF receptors might be involved in the pathogenesis of asthma through regulating the apoptosis of Eos.

4.Effects of Aminophylline on expression of Fas mRNA and apoptosis in eosinophils

AIM To investigate the effects of aminophylline (AM) on expression of Fas mRNA and apoptosis in eosinophils(Eos) of bronchalveolar lavage fluid(BALF) in asthmatic guinea pig, and the mechanism of AM promoting Eos apoptosis. METHODS After guinea pigs were stimulated by ovalbumin(OVA) 48 h, hypodense Eos(HEos) and normodense Eos(NEos) were purified from BALF by gradients of percoll. Purified Eos were incubated with different concentration of AM (10 -6 ~10 -3 mol?L -1 ) in RPMI 1640 medium 24 h, the mRNA of Fas in Eos was measured by hybridization and apoptosis was detected by TdT mediated dUTP nick end labeling. RESULT Eos cultured under the condition of AM administration in vitro, apoptotic Eos increased and the expression of Fas mRNA increased in a dose dependent manner. The expression of Fas mRNA was correlated with Eos apoptosis. CONCLUSION AM promote Eos apoptosis and expression of Fas in dose dependent manner in vitro. Fas might be one of mechanism that AM promoting apoptosis of Eos.
5.Protective effect of telmisartan on rats with renal failure and its mechanism

Objective:To study the protective effect of telmisartan on rats with renal failure and its mechanism. Methods:60 Wistar rats were chosen as study objective, and were divided into 4 groups randomly:15 in group A (sham operation group), 15 in group B (model group), 15 in group C (telmisartan group) and 15 in group D (telmisartan+GW9962 group). The difference of survival rate, blood-urine biochemical indexes, renal pathological change, and the expression level of PPARγ and nNOS were compared. Results:After 12 weeks, the survival rate of group A was 93.33%(14/15), that of group B was 46.67%(7/15), that of group C was 86.67%(13/15), that of group D was 60.00%(9/15), and the difference among 4 groups had statistical significance (P<0.05). After 1 week, the difference of Scr, that of BUN and that of 24h protein urine among 4 groups was not statistical significant (P>0.05);after 3 weeks, 6 weeks and 12 weeks, these difference was statistical significant (P<0.05). The difference of blood-urine biochemical indexes, that of renal pathological change, and that of the expression level of PPAR毭and nNOS was statistical significant (P<0.05). Conclusions:Telmisartan has protective effect on renal failure caused by 5/6 nephrectomy, which might be relative to the expression level of PPARγ and nNOS.

7.Influences of dexamethasone on expression of fas, bcl-2 mRNA and apoptosis in eosinophils in vitro

AIM: To investigate the effects of dexamethasone (DM) on expression of fas and bcl-2 mRNA and apoptosis in eosinophils (Eos) of bronchoalveolar lavage fluid (BALF) in asthmatic guinea pig. METHODS: The guineas pigs were stimulated with ovalbumin (OVA) for 48 hours. Hypodense Eos (HEos) and normodense Eos (NEos) were purified from BALF by gradients of Percoll. Eos was cultured in PRMI-1640 medium and DM (10 -10-10 -5 mol/L) was added. The mRNA of fas and bcl-2 in Eos was measured by hybridization. Apoptosis was detected by TdT-mediated dUTP nick end labeling (TUNEL). RESULTS: Eos was cultured under the condition of DM administration in vitro. Apoptotic Eos increased, fas mRNA expression increased and the bcl-2 mRNA expression decreased in a dose-dependent manner. There was significant correlation between Eos apoptosis and fas mRNA expression. The expression of bcl-2 mRNA was inversely correlated with Eos apoptosis. CONCLUSION: DM promoted Eos apoptosis, increased expression of fas and inhibited expression of bcl-2 in a dose-dependent manner in vitro. fas and bcl-2 might be one of mechanisms of DM promoting apoptosis in Eos.

As an important measure to reduce casualties and prevent secondary pollution, decontamination is an impor-tant link in the process of emergency response during chemical accidents.The decontamination effect is closely related to decontamination technology and equipment.Decontamination agent selection and development are an important part of a decontamination technology.In this paper, the development and use of cleaning agents, such as alkaline, oxidation and chlorination, adsorption (degradation), metal oxide and oxygen acid salt, chemical compounds, biological (enzymatic), and individual disinfection package, light decontamination equipment, multifunctional integrated large-scale decontamination equipment at home or abroad, are reviewed.By laying bare the gap between China and advanced countries in the related field, we hope to raise the concern of relevant professional counterparts and promote the development of domestic decontami-nation technology and equipment with decontaminant agents at the core.

Taking the bull by the horns,pooling the wisdom and efforts,the experts and rescue teams carried out and implemented the decision spirit of ″it should not hurt one man,and should not appear serious secondary disasters in the late treat?ment″which put forward by the Party Central Committee and State Council. In order to better learn and sum up experience,the cause of the disaster,the disaster rescue and disposal process,the enlighten?ment brought by the disaster and the recommendations deal with unexpected chemical incidents in the future was discussed in this paper.
10.Effect of Vit E on myosin light chain kinase activity and expression in the liver of atherosclerosis model rabbit

AIM To study the effect of Vit E on the MLCK activity and expression in the liver of atherosclerosis model rabbits. METHODS The MLCK activity of rabbit liver was measured by the method of ?- 32P incorporated and its expression was detected by immunofluorescent. RESULTS The model of atherosclerosis was estabilished. After rabbits were fed with cholesterol for four weeks and twelve weeks, the activity of MLCK increased markedly, and there was significantly statistical difference compared with the normal control (P0.05). MLCK expression increased after the rabbits was fed with cholesterol for four weeks, and this increase became more obvious had been the rabbits was fed with cholesterol for twelve weeks. The expression decreased when vitamin E had been added into the cholesterol fed. CONCLUSION The pathology of liver may be associated with the increase of the activity of MLCK. Vit E may reduce MLCK activity and protect hepatocyte from injury.
