1.in vitro Model for Targeting Tumor Vasculature Using Immunoliposome

Objective: Based on tumor angiogenesis theory, a target system of tumor endothelial cells was established to investigate the cytotoxicity against tumor vascular endothelium by an drug loaded immunoliposome.Methods: Sterically stabilized liposomes containing doxorubicin were used for conjugation to a monoclonal antibody (MAb) as a drug-vascular targeting device. The MAb M5/114 directly against MHC Ⅱ, which was expressed by vascular endothelial cells was employed. Binding of cholesteryl ether (CHE) labeled anti-MHC Ⅱ immunoliposomes to endothelial cells (SVEC ) in vitro was detected by a binding assay. Cytotoxicity of drug loaded immunoliposome to endothelial cells was determined by MTT assay.Results: At least 3-fold increase in binding for the targeted liposomes was observed. This data suggest highly selective binding of M5/114-immunoliposomes to MHC Ⅱ expressing endothelial cells. Moreover, the cytotoxicity produced by drug loaded M5/114-immunoliposome was greatly enhanced compared with that by non-targeted liposomes. Conclusion: This is the first report using immunoliposome to target endothelial cells. The results demonstrate that immunoliposome directly against tumor vascular endothelial cells might provide a general treatment for solid tumors.
2.Measured results and assessment of personal protection appliances against ionizing radiation

Objective To monitor the protective qualities of personal protective appliances and to ensure the health and safety of radiological working personnel.Methods The lead-equivalent thickness of personal protective appliances and materials was measured by means of standard lead slices.The lead equivalent thickness represents in terms of mm Pb.Results 77 pieces of products and samples were measured altogether.The results indicate that the specific lead equivalents of lead-rubber plates were between 0.20-0.39 mm Pb/mm for 37 pieces of lead-rubber plates and the values of 6 pieces of samples were less than 0.25 mm Pb/mm,which did not accord with the requirement of the relational standard.27 pieces of personal protection appliances were measured altogether.They were 12 pieces of protective clothes,4 pieces of protective headgears,5 pieces of protective neckpieces,4 pieces of protective gloves and 2 pieces of protective masks.13 pairs of lead-glass spectacles among them were measured altogether.The measured results for personal protective appliances and lead-glass spectacles showed that actually measured lead-equivalent were higher than the nominal lead-equivalent.Conclusions The protective qualities are reliable for personal protection materials and appliances to be made in home and imported abroad.But the protective qualities of interventional protection gloves should be improved and made them better.
3.Application of 4-wavelength optical intrinsic signal imaging in monitoring peri-infarct depolarizations in GFAP(⁺/⁺)Vim(⁺/⁺) mice.

OBJECTIVETo study optical intrinsic signal (OIS) imaging of peri-infarct depolarizations (PIDs) in mice and investigate the influence of knockout of glial fibrillary acidic protein and vimentin on PIDs.
METHODSGFAP(⁺/⁺)Vim(⁺/⁺) mice and GFAP(⁺/⁺)Vim(⁺/⁺) mice were subjected to MCAO by standard intraluminal filament method. The main characteristics of PIDs in 4 h were studied by 4-wavelength OIS imaging technique.
RESULTSPIDs were identified as consistent, red and blue interaction waves in the cortical reflectance that slowly propagated peripherally from the origin site. There were 5 patterns of PID propagation, namely rostro-caudal, latero-medial, caudo-rostral, contralateral and medial-lateral. No significant differences were found in PID frequency, propagation patterns, velocity or duration time between the two groups (P>0.05).
CONCLUSIONThe 4-wavelength OIS system allows acquisition of high temporal-spatial resolution color images for analyzing temporal-spatial characteristics of PIDs in detail. Knockout of GFAP and vimentin do not affect PIDs in 4 h following middle cerebral artery occlusion.

4.Influence of gender in cortical spreading depression wave in mice

Objective To observe the influence of gender in cortical spreading depression (SD) wave in mice.Methods Potassium chloride (KCl) or middle cerebral artery occlusion (MCAO) models were used to induce SD.Adult C57 mice were randomly divided into four groups:male and female KCl-induced groups,male and female MCAO-induced groups (n=8).SD waves were monitored for 2 h by electrophysiological method.Results SD waves showed lower amplitude and longer duration time in the MCAO-induced group as compared with those in the KCl-induced group.There were no obvious differences in SD amplitude,duration time,frequency and onset time of first SD between male and female KCl-induced groups (P>0.05).There were no differences in SD amplitude and duration time between male and female MCAO-induced groups (P>0.05),however,SD frequency and onset time of the first SD were signficantly different between male and female MCAO-induced group (P<0.05).Conclusion Mice gender has an effect on MCAO-induced SD,but no obvious effect on KCl-induced SD; the possible explanation for the difference is that estrogens modifying susceptibility to SD is mainly through their effect on glutamate but not on K+ metabolic pathways.
