1.Effects of transforming growth factor-β1 and transforming growth factor-α on the growth and invasive potentials of human bladder tumor cells

Objective To study the effects of transforming growth factor-β1 (TGF-β1) and transforming growth factor-α (TGF-α) on the growth and invasive potentials in human bladder tumor cells. Methods The effects of TGF-β1 and TGF-α on the growth of EJ cell line and the effects on MMPs and TIMP-2 were studied by means of MTT,Western Blot and RT-PCR methods. Results (1)TGF-β1 and TGF-α tended to inhibit the growth of EJ cells but statistically nonsigficant.(2)Higher levles of MMP-9 mRNA but lower levels of MMP-2,TIMP-2,MT1-MMP mRNA were found in EJ cells following the treatment with TGF-β1.The same was true for the expression of MMP-2,TIMP-2 mRNA in TGF-α groups.However,MMP-9 mRNA was not found in both TGF-α groups and the control groups. (3)TGF-β1 (0.1,1.0 ng/ml) enhanced MMP-2 protein but not the TIMP-2 protein,while TGF-β1 (5.0,10.0 ng/ml)decreased TIMP-2 protein but not on MMP-2.In TGF-α groups,when the concentration was 1.0,5.0,10.0ng/ml,TIMP-2 protein expression was decreased but MMP-2 did not.When the concentration reached 100.0 ng/ml,it increased MMP-2 protein level,not the TIMP-2 protein. Conclusions TGF-β1 and TGFα do not inhibit the proliferation of EJ cells whereas the enhanced-invasiveness and metastasis may be associated with regulating the expression of MMPs and TIMP-2.

4.Anatomical study of pedicle screw fixation for the cervical spine

Objective To develop an anatomically based method of accurate pedicle screw insertion for cervical spine fixation. Methods Forty adult cervical spine were mounted onto a specially designed jig. Parameters including height, width, length, and axis angulation of pedicles,depth of lateral fovea, height of superior posterior border of arcus vertebrae and angles of posterior articular prominance as well were mea sured with vernier calibre or goniometer either directly or radiographically. Results Lateral fovea did exist in all C2-C7 vertebrea. The pedicles of the cervical spine were wide and long enough to accept 28 mm long 3.5 mm screws. The abduction angle of the screw was 20? at the C1 level, 25? superiorly and 44? inferiorly at the C2 level, 45? to 47? from level C3 to C5, and 40? from level C6 to C7. The angle of posterior plane of articular mass was minus 2.30? at C3,it was 2.32? to 2.42? from level C4 to C6, and it was 15.22? at C7. The stability of cervical spine fixation and neurological improvement were satisfactory. Conclusion Transpedic ular screw fixation for the cervical spine is practicable. Anatomical symbol of the superior posterior border of the arcus vertebrae and the posterior plane of articular mass and the lateral fovea is easy to be exposed and identified in the operation. Transpedicular screw can be precisely inserted easily and safely for the cervical spine fixation with limited exposure and without using fluoroscopic guidance.
5.Therapy of fracture of femoral intercondylar and supracondylar by saddle-shaped condylar plate

[Objective]To investigate the clinical effect of saddle-shaped condylar plate in treating the fracture of the femoral intercondylar and supracondylar.[Method]Thirty-two cases with the fracture of the femoral intercondylar and sapracondylar,who were selected randomly were fixed by saddle-shaped condylar plates after reduction,of which 8 cases of patellar fractures accompanied with ipsilateral fracture of the femoral condylar.[Result]All cases were completely healed during the follow-up study of 4 months to 2 years,all cases achieved bone union in 5~9 months.[Conclusion]The design of the plate conforms to the principle of biomechanics and is good for the heal of the fracture.The saddle-shaped structure makes the plate and the cannular bone tangent as fixing so it can reduce the putrescence of the bone which is caused by pressure;The arch-bridge-shaped juncture and the mechanical bolts which are fixed across can strengthen the power of anti-bending and anti-rotation of the broken ends of the fractured bone.The stability of the fixation enables the patient do the functional exercise after the operation as soon as possible.It has definite clinical value and is worth of popularization.

Objective To investigate the role of WNK4 gene Exon8 Ala589Ser polymorphism in the pathogenesis of essential hypertension. Methods This study included 259 hypertensive patients and 235 normotensive patients as control subjects. Exon8 Ala589Ser polymorphism in WNK4 was investigated by PCR-RFLP and confirmed by direct sequencing. Genotype, genotype frequencies and allele frequencies of the polymorphism were compared among patients with essential hypertension and control subjects. Objective: To improve the physical properties of micromeritics of the extracts of Fengshilin capsule, such as anti moisture. Methods: Critical relative humidity, absoption curve of moisture, angle of repose were used as the norms of the research. The suitable excipients and formulation process of Fengshilin capsule have been sieved. Results: It was feasible that the combined excipients of microcystalline cellulose and starch (in a ratio of 1∶1) were mixed with the extract powder. The absorption of moiture of the extract powder was improved. The humidity of mass production and store was lower than 45%. Conclusion: The research has been found to be an effective in the process of Fengshilin capsule.

Objective:To study the expression of SKP2 and MRP-1/CD9 protein in glottic cancer and adjacent tissues,and to analyze its significance for a safe surgical margin.Method:Thirty-eight cases of glottic squamous cell carcinoma were studied for its cancer tissue, tissue 2 mm, 5 mm , and 10 mm away from cancer ,and 10 cases of vocal cord polyp were served as control. SKP2 and MRP-1/CD9 protein were examined by immunoh istochemical method.Result:The positive expression of SKP2 proteins decreased in sequence of polyp mucosa, those adjacent to carcinoma (10 mm, 5 mm, 2 mm ) and cancer tissue, and there was significant difference between them(P<0.05);On the contrary, the positive expression of the MRP-1/CD9 proteins increased in sequence of polypusmucosa, those adjacent to carcinoma (10 mm,5 mm, 2 mm) and cancer tissue,and there was significant difference between them (P<0.05).Conclusion:SKP2 and MRP-1/CD may act as the reference index for judging the biological speciality of LSCC. It is appropriate to regard 5 mm or above 5 mm away from tumors as a safe margin for surgical treatment of glottic carcinoma.
