1.Biological features of CD90+ tumor stem cells in ovarian cancer

BACKGROUND:Thinking from ovarian cancer stem cel theory shows that: in the tumor cels, there are a fraction of stem cels with self-renewing ability and multipotent differentiation, which are the root causes of ovarian cancer recurrence and drug resistance. Studies have shown that CD90 can be used as a surface marker of mesenchymal stem cels and stem cels of other cancers.
OBJECTIVE:To explore the biological features of CD90+ tumor cels from ovarian cancer tissues.
METHODS: Primary ovarian cancer cels were isolated from the abdominal dropsy of ovarian cancer patients to sort CD90+ and CD90- cels using flow cytometry. RT-PCR was used to detect expressions of stem cel-related genes and epithelial to mesenchymal transition-related genes. Cel invasion was observed by Transwel invasion assay, cel proliferation and differentiation observed by clone formation assay, stem cel potential observed by suspension sphere-forming assay, and tumor formation rate observed byin vivo tumorigenicity experiment.
RESULTS AND CONCLUSION:Compared with the CD90- cels, the expressions of CD44, CD133, ALDH1, N-cad and Vimentine were significantly higher in the CD90+cels (P < 0.05), but the expression of E-cad was significantly decreased in the CD90+ cels (P < 0.05). Tumor formation rates of CD90- and CD90+ cels were increased significantly with the increase of seeded cel number, which was more obvious in CD90+ cels. The number of transmembrane cels, the number of cel clones and the number of suspended spheres were significantly higher in the CD90+ cels than the CD90- cels (P < 0.05). Experimental findings from this study show that CD90+ cels highly express epithelial to mesenchymal transition-related genes and stem cel-related genes, with higher invasion, proliferation and differentiation, in vivo tumorigenicity and potential of stem cels. CD90+ cel separation may be a new method to separate ovarian cancer stem cels.
2.Determination of P-aminophenol in Pediatric Paracetamol and Amantadine Hydrochloride Granules by HPLC

Objective:To establish an HPLC method for the determination of P-aminophenol in pediatric paracetamol and amanta-dine hydrochloride granules. Methods: The column was Phenomenex Luna C8 (150 mm × 4. 6 mm, 5 μm), the mobile phase was phosphate buffer solution-methanol (93∶7), the flow rate was 0. 8 ml·min-1, the detection wavelength was 275nm, the column tem-perature was 30℃, and the injection volume was 10μl . Results: P-aminophenol showed good linearity within the range of 0. 528-42. 240 μg·ml-1(r=0. 999 9). The average recovery was 100. 03%(RSD=0. 9%, n=9). Conclusion:The method is simple with high accuracy and reproducibility, which can be used in the determination of P-aminophenol in pediatric paracetamol and amantadine hydrochloride granules.
3.Discussion on Quantitative Standard of the Pill and Granule of Liuwei Dihuang in 2005 Edition of China Pharmacopoeia

OBJECTIVE:To discuss the problems of quantitative standard and method for the pill and granule of Liuwei Dihuang in 2005 edition of China Pharmacopoeia.METHODS:The quantitative indicators,determination methods,and content standards established in 2005 edition of China Pharmacopoeia were compared between the pill and granule of Liuwei Dihuang,these indices were also compared with those in 2000 edition of China Pharmacopoeia.RESULTS &CONCLUSIONS:The quantitative indicators,determination methods,and content standards set for pill and granule of Liuwei Dihuang are not consistent between the horizontal and vertical comparisons,it is suggested that these indices should be revised when China Pharmacopoeia is reprinted.
4.Effect of lipitor on high glucose-induced HUVEC apoptosis and PI3K/AKT/eNOS signal pathway

Objectives To explore effect of lipitor on apoptosis and phosphatidyl inositol-3-kinase ( PI3K)/protein kinase B ( AKT )/endothelial nitric oxide synthase ( eNOS ) signal pathway in high glucose-induced human umbilical vein endothelial cell ( HUVEC) .Methods The cases were randomly divided into normal control group, model control group (33.3 mol/L glucose), lipitor low, medium, high-dose group (0.1,1,10 μmol/L lipitor).The viability of HUVEC was detected by MTT assay.The morphology of HUVEC was photographed by inverted microscope.The apoptosis of HUVEC was examed by Annexin V-FITC/PI flow dual-staining method.The concertration of NO in HUVEC supernatant was exmaed by Gries method.The activation of PI3K/AKT and expression of eNOS was assayed by western blot.Results HUVEC was shrinkage, rounded and brighten, the viability of HUVEC decreased, early and late apoptosis rate of HUVEC increased significantly, the level of NO, eNOS, PI3K and AKT phosphorylation also reduced in model control group ( P<0.01).1, 10 μmol/L lipitor improved HUVEC morphology, increased HUVEC' viability and expression of PI3K (P<0.05).0.1,1,10 μmol/L lipitor suppressed HUVEC' apoptosis, increased the concentration of NO, expression of eNOS and phosphorylation of AKT (P<0.05).Conclusion These results suggested lipitor exert anti-apoptosis in high glucose-induced HUVEC, which might be related to PI3K/AKT/eNOS signal pathway.

6.Observation of effects of zinc on the injury induced by oxygen free radical in cultured heart cells

In present study, the injury induced by oxygen free radical in cultured cardiac cells was observed by adding xanthine and xanthine oxidase into the medium The effects of zinc on the injury of cells were also observed, taking the electro physiological changes and threshold concentration of BaCl_2 which induces cell stop beating as index. The results showed that the electric parameters of action potential (the amplitude, overshoot, maximum diastolic potential, threshold, maximum rate of depolarization and duration of action potential ) were decreased in XOD group, compared with the control. The threshold concentration of BaCl_2 was decreased too. However, there were no significant differences between Zn group and control group except the TP value. The electric parameters of action potential was increased notab(?)y in Zn group, compared with XOD group. Furthermore, the threshold concentration of BaCl_2 in Zn group was increased too. It is suggested that Zn possessed the protective effects on cultured cardiac cells from the injury induced by oxygen free radical.
7.Study on Cultivation Model for Characteristics and High-end Interdisciplinary TCM Informatization Talents

8.Effect of extrinsic vascular endothelial growth factor (VEGF) on the viability of rat skin flap received radiotherapy

Objective:To observe the relationship between the vascular endothelial growth factor (VEGF) and the survival of flap in the rat back which received radiotherapy. Methods: Forty Wistar rats were divided into four groups. A 3×4 cm~2 full thickness dorsal flap with pedicle was prepared at the posterior end. The skin flaps were given~(60)Co radiotherapy in the 2, 4, 6 day postoperation. Group A was given of VEGF in the third, fifth, seventh day. Group B was given a high quantity of VEGF. Group C was the control of Group A,Group D was the control of Group B. Control groups received normal saline. Results: Pathological observation revealed that the survival of flap and blood vessel lumen diameter of group A were higher than that of group B(P<0.05). Group A showed remarkable difference compared with group C(P<0.01). Group B showed remarkable difference compared with group D(P<0.05). Results of fluorescence staining and SDH examination showed that three times injection had better results than one time injection. Conclusion: Three times injection of VEGF has better effect on the viability of rat skin flap than one time injection.

ObjectiveToinvestigatethecorrelationsofmethylenetetrahydrofolatereductase(MTHFR) gene C677T polymorphism w ith ischemic stroke and hyperuricemia in Chinese Han population in Shandong, China.Methods The patients w ith acute ischemic stroke and age-and sex-matched controls in Chinese Han population in Shandong, China w ere enrol ed. Polymerase chain reaction amplification and microarray hybridization w ere used to detect the MTHFR gene C677T polymorphism, and the serum uric acid concentration w as measured. Results A total of 145 patients w ith acute ischemic stroke and 145 age-and sex-matched controls in Chinese Han population in Shandong, China were enroled. The proportion of diabetes ( 26.90%vs. 6.89%; χ2 = 20.653, P< 0.001 ) and fasting blood glucose ( 5.56 ± 1.57 mmol/L vs.5.01 ±1.11 mmol/L;t= -3.390, P=0.001), homocysteine (median, interquartile range:18.2[16.30-22.55 μmol/L] vs.15.20[12.10-17.85 μmol/L]; Z= -6.323, P<0.001), and uric acid (43.0[361.60-490.45 μmol/L] vs.285.9[267.00-346.25 μmol/L]; Z= -10.360, P<0.001) levels in the ischemia stroke group w ere significantly higher than those in the control group. The distribution frequencies of TT genotype (42.07%vs.15.17%; χ2 =25.673, P<0.001) and T alele (58.28%vs. 34.48%; χ2 =33.008, P<0.001) in the ischemia stroke group w ere significantly higher than those in the control group. Multivariate logistic regression analysis showed that the uric acid (odds ratio [OR] 1.018, 95%confidence interval [ CI] 1.013-1.024; P<0.001), TT genotype ( vs.CT genotype OR 6.774, 95%CI 1.779-25.507; P=0.005), hypertension ( OR 1.919, 95%CI 1.013-3.636; P=0.045), and homocysteine (OR 1.153, 95%CI 1.059-1.258;P=0.001) w ere the independent risk factors for ischemic stroke. The ischemic stroke group w as combined w ith the control group, a total of 101 patients had hyperuricemia, and 189 had normal uric acid. The proportion in patients w ith diabetes ( 32.67%vs. 11.64%; χ2 = 23.749, P< 0.001), as wel as total cholesterol ( 5.67 ±1.56 mmol/L vs.5.10 ± 1.33 mmol/L; t= -3.255, P< 0.001 ) and homocysteine ( 19.50[17.10-24.70 μmol/L] vs. 15.40[12.60-18.05 μmol/L ]; Z= -7.236, P< 0.001 ) levels in the hyperuricemia group w ere significantly higher than those in the normal uric acid group. The distribution frequencies of the TT genotype (55.45%vs.13.76%;χ2 =56.409, P<0.001) and T alele (71.9%vs.32.54%;χ2 =79.561, P<0.001) w ere significantly higher than those in the normal uric acid group. Multivariate logistic regression analysis show ed that the TT genotype ( vs.CC genotype, OR 6.434, 95%CI 2.334-17.736; P<0.001 ), CT genotype ( vs.CC genotype, OR 2.234, 95%CI 1.019-4.898; P= 0.045 ), homocysteine ( OR1.081, 95%CI 1.010-1.157;P=0.024), and total cholesterin ( OR 1.363, 95%CI 1.123-1.653;P=0.002) w ere the independent risk factors for high hyperuricemia. Conclusions MTHFR gene C677T TT genotype and serum uric acid level are the independent risk factors for ischemic stroke in Chinese Han population in Shandong, China. MTHFR gene C677T TT genotype is also an independent risk factor for hyperuricemia in this population. Adjusting dietary habit may have a positive significance for the prevention of ischemic stroke in Chinese Han population in Shandong, China.
