Objective To observe the phenotypic changes between renal tubular epithelial cells and mesenchymal cells in rat tubulointerstitial fibrosis(TIF) .Methods The renal tubulointerstitial fibrosis models of Wistar rats were established by gavage with excessive adenine. The morphological changes of model kidney were observed by light microscopy, electron microscopy, polarizing microscopy. The?-smooth muscle actin(a-SMA), vimentin, and keratin were examined by immunohistochemistry staining. While the proliferating cell nuclear antigen (PCNA) was also assessed. Results There were many 2, 8-dioxyadenine crystals derived from the metabolite of adenine to deposit in the renal tubules, which induced the injury. Tubular epithelial cells degeneration and regeneration, loss of renal tubules, mononuclear cells infiltration in renal interstitium and the accumulation of extracellular matrix protein were observed. Both ?-SMA and vimentin were positive in some of renal tubules. And consecutive sections of immunohistochemical staining showed that the area of distribution of a-SMA positive cells was almost the same area as that of vimentin positive cells. Polarizing microscopy results showed that there was a mass of interstitial collagen ( Ⅲ and Ⅰ) accumulation. And electron microscopy examination proved that epithelial cells invaded into the renal interstitium. Conclusion There is tubular epithelial cells-myofibroblasts transdifferentiation in the process of TIF, and which plays a key role in the accumulation of extracellular matrix.

Objective: To compare the advantages and disadvantages of three staining methods of HBV. Methods: Normal Liver tissue and HBV-infected, HCV-infected ,or dually infected (HBV and HCV) liver tissues were selected for this study. Formalin- fixed, paraffin-embedded sections(4 ?m) were prepared. Each of the liver tissue specimens was detected by three staining methods, including immunohistochemical methods ,Shikata’s orcein stain and Victoria blue stain, respectively. Results: In the three methods , all of six HBV -infected cases showed intense staining, and three cases with dual infection (HBV and HCV) were weakly positive. However, both normal and HCV-infected liver tissues showed no staining. HBsAg stained dark brown with Immunohistochemical stain; HBsAg containing ground-glass hepatocytes stained magenta with Shikata’s orcein stain; HBsAg stained blue with Victoria blue. Conclusion: Each of three methods has its own advantages and disadvantages: high specificity and sensitivity, but high cost for immunohistochemical methods;complicated and overelabrate procedure for preparation of solutions, lower specificity and sensitivity, but low cost, for special staining methods.

Objective To evaluate the radiation dose from enhanced dual-source computed Tomography(DSCT) scan on children with congenital heart disease(CHD). Methods Seventy children with CHD, age from 1 month to 8 years old, were scanned with enhanced DSCT. Children were divided by age into < 1 year old group, 1-5 years old group, > 5 years old group. The differences among three groups were tested by F test. Then, the SNK test was used to compare the difference between each group. Multiple linear regression analysis was used to test the relationship of dose length product (DLP) with the age, weight, voltage, current, pitch and scan sheet. Results The average value of DLP was (144.46 ±test showed statistical differences of DLP between < 1 year old group and 1-5 years old group, and between <1 year old group and >5 years old group(q =5.21,6.52,P =0.009,0.004). The difference of DLP between 1-5 years old group and > 5 years old group did not reach significant ( q = 0.28, P = 0.48 ). The differences of ED was not statistically significant among the three groups [< 1 year 3.20 mSv, 1-5 years (5.17 ± 1.98 ) mSv, > 5 years ( 3.74 ± 1.33 ) mSv, F = 0.54, P = 0.59]. DLP was positively correlated with age (4.3 years,r = 0.541 86, P = 0.0008 ), weight ( 12.1 kg, r = 0.563 71, P = 0.0004 ), voltage [(95.48±6.99) kV,r =0.632 69, P<0.01], current [(138.55±40.67) mA,r =0.796 08, P<0.0001] and scan sheet (236.10 ±46.51 ,r = 0.721 92, P < 0.01 ). DLP was negative correlated with pitch (0.48 ± 0.03,r = - 0.466 93, P = 0.0047 ). Conclusion Higher DLP was observed in children over 1 year old under enhanced DSCT scan, but ED was not statistically significant among the three groups due to the higher K value in the children under 1 year old.

A method was developed for the determination of fumonisin B1 ( FB1 ) and fumonisin B2 ( FB2 ) in livestock and poultry formula feeds by high performance liquid chromatography-tandem mass spectrometry (LC-MS/MS. After extracted by acetonitrile-water (50: 50, V/V) and purified with MAX solid phase extraction column, the fumonisins were separated by Thermo C18(100 mm×2. 1 mm, 5 μm) column with 0. 1% formic acid in water and methanol as the mobile phase. Multiple reaction monitoring (MRM) was used to acquire mass spectrometric data under electrospray positive ionization mode ( ESI+) . The results showed that the linear correlation coefficients (R2) of fumonisin FB1 and FB2 were all greater than 0. 999 in the range of 1-500 μg/L. The limits of quantitation (LOQ) were 0. 098 and 0. 197 μg/L, and the limits of detection ( LOD) were 0. 328 and 0. 656 μg/L, respectively. At different spiked levels, the recoveries of FB1 and FB2 were ranged from 89. 7% to 95. 1%, and the relative standard deviation ( RSD) was ranged from 3. 2% to 8. 6%. Additionally, the detection rate reached 98. 11% screening through the established method in the 106 livestock and poultry formula feeds collected from markets. This result indicates that the method is suitable for accurate quantitative analysis of FB1 and FB2 in different complicated livestock and poultry formula feeds.
