1.Percutaneous absorption of Rg1 in Panax notoginseng in vitro

AIM: To study the percutaneous absorption of Rg1 in Panax notoginseng and the effects of some commom enhancer and pH value on it. METHODS: Drug permeation tests were performed through excised rat skin in V-C diffusion cell.Rg1 concentration in samples was determined by HPLC. RESULTS: Rg1 could penetrate through excised rat skin with lower permeability coefficient;in the range of concentration 300 mg/mL~60 mg/mL permeability coefficient of Rg1 decreased as its concentration increasd;Among enhancers,10% ethanol,10%,20% and 30% propylene glycol,2% Azone,2% Eucalyptus Oil and 5% Oleic Acid had markedly promotive effect on transdermal penetration of Rg1. CONCLUSION: The research will provide experimental data for skin administration of Chinese medicine containing Panax notoginseng.
2.Effects of inhalation of different concentrations of sevoflurane combined anesthesia on somatosensory evoked potentials in adolescent patients undergoing scoliosis surgery

Objective To investigate the effects of inhalation of different concentrations of sevoflurane combined anesthesia on somatosensory evoked potentials (SEPs) in the adolescent patients undergoing scoliosis surgery.Methods Forty-five adolescent patients of both sexes,aged 12-17 yr,with body mass index of 15-23 kg/m2,undergoing scoliosis surgery under general anesthesia,were randomly divided into 3 groups ( n =15 each):low concentration sevoflurane group ( group S1 ),moderate concentration sevoflurane group ( group S2 ) and high concentration sevoflurane group (group S3 ).Anesthesia was induced with fentanyl 4 μg/kg,midazolam 0.03 mg/kg,propofol 2.0 mg/kg,and rocuronium 0.6 mg/kg.The patients were endotracheal intubated and mechanically ventilated.Anesthesia was maintained with propofol infusion at 8 mg· kg-1 · h-1 and remifentanil infusion at 0.25 mg· kg-1 ·min-1.After beginning of the surgery,1.8%,4.0% and 6.0% sevoflurane were inhaled for 10 min in groups S1-3 respectively,SEPs were measured by stimulating the posterior tibial nerve and significant SEP suppression was detected and recorded.The wave P40 latency,wave P40-N50 amplitude,and time for P40 waveform changes were recorded before sevoflurane inhalation (baseline) and at 10 min of sevoflurane inhalation,and the time for recovery of P40 waveform was recorded after sevoflurane inhalation was stopped.The prolonged percentage of P40 latency and decreased percentage of P40-N50 amplitude were calculated.Results The rates of significant SEP suppression were 80%,100% and 100% in groups S1,S2 and S3 respectively ( P > 0.05).Compared with group S1,the prolonged percentage of P40 latency and decreased percentage of P40-N50 amplitude were significantly increased,the time for P40 waveform changes was significantly shortened,and the time for recovery of P40 waveform was significantly prolonged in groups S2 and group S3 ( P < 0.05).Compared with group S2,the decreased percentage of P40-N50 amplitude was significantly increased,the time for P40 waveform changes was significantly shortened,and the time for recovery of P40 waveform was significantly prolonged in group S3 ( P < 0.05).Conclusion 1.8%,4.0% and 6.0% sevofiurane combined anesthesia can suppress SEPs in the adolescent patients undergoing scoliosis surgery,and they are not suitable for the surgeries requiring SEP monitoring.
3.Treatment of severe hepatitis by plasma exchange

Objective:To observe the therapeutic effects of plasma exchange in severe hepatitis. Methods:53 cases served as treatment group and 49 cases as control group. Both groups were similar in basic medical treatment, and an additional treatment of plasma exchange was carried in the treatment group. 186 times of plasmas exchange were performed in the treatment group, to observe and compare the changes in patients of the 2 groups in symptoms、liver functions and survival rates. 4.Clinical significance of detection on peripheral blood T cell subsets, sIL-2R, VEGF and LDH in patients with diffuse large B cell lymphoma

Objective To investigate the changes of peripheral blood T cell subsets and serum soluble interleukin-2 receptor (sIL-2R),vascular endothelial growth factor (VEGF) and LDH in patients with diffuse large B cell lymphoma,and to investigate their relationship of the cellular immune function,clinical stage and disease progression.Methods 35 cases of DLBCL patients were selected as observe group,20 cases of healthy individuals as normal control group.Peripheral blood T cell subsets were detected by flow cytometry.Serum soluble interleukin-2 receptor (sIL-2R) and vascular endothelial growth factor (VEGF) were detected by ELISA assay.Serum levels of LDH were detected by the rate method for measuring.Results The expression levels of CD4,NK cells and CD4/CD8 ratios in all patients with DLBCL were lower than healthy individuals [(68.62±12.82) % vs(73.00±6.12) %,(14.28±7.93) % vs (39.49±12.01) %,(1.03±0.84) vs (1.45±0.28),all P < 0.05].Serum sIL-2R in patients with DLBCL was significantly higher than healthy individuals [(501.50±157.70).μg/L vs (128.30±82.40) μg/L,P< 0.001].Serum levels of sIL-2R with Ⅲ-Ⅳ stage in patients with DLBCL was higher than that in ones with Ⅰ-Ⅱ stage [(618.38±192.22) μg/L vs (432.45±75.48)μg/L,P < 0.05].Serum expression levels of sIL-2R in patients with DLBCL with B symptoms was higher than that in ones with A symptoms [(616.50±183.03) μg/L vs (441.52±103.48) μg/L,P < 0.01].The expression levels of VEGF,LDH in patients with DLBCL were higher than healthy individuals [(304.03±163.05) pg/ml vs (156.25±47.83) pg/ml,(300.74±300.91) U/L vs (177.00±32.14) U/L,both P < 0.05].The high expression level of serum sIL-2R was correlated with low CD4/CD8 ratio (r =0.4635,P < 0.05).The high expression levels of serum sIL-2R was correlated with the high expression levels of serum VEGF,LDH (r =0.447,P < 0.05; r =0.448,P < 0.05).Conclusion High expression levels of serum sIL-2R in DLBCL are closely associated to low cellular immune function,tumor burden in vivo,disease progression and invasion.
5.Clinical value of transthyretin from patients with early rheumatoid arthritis

Objective To investigate the clinical value of transthyretin (TTR) from patients with early rheumatoid arthritis (ERA). Methods 58 patients with ERA , 34 patients with later RA (LRA) and 34 healthy control (HC) were included in the research. TTR was analyzed by ELISA, whose variance was analyzed. TTR density, disease activity score28 (DAS28) score and rheumatoid factor (RF) were tested, and their correlation with TTR was analyzed. Results Serum level of TTR with ERA significantly increased compared with that with LRA and HC (P < 0.05), no statistical significance with LRA group and HC. TTR level was no correlation with the number of swelling and tender joints, disease activity score 28, RF, ESR, CRP, anti-cyclic citrylinated peptide antibody and anti-keratin antibodies, hemoglobin, thrombocyte and albumin. Conclusion Serum level of TTR significantly increased with ERA patients, contributing to early diagnosis for RA.
6.The immune responses to hepatitis B gene vaccine in mice and the immune adjuvant effect of cytokines

Objectives:To observe the effect of eukaryotic expression vectors coding IL-2 and IL-12 on immune responses induced by DNA immunization of HBV surface antigen(pCR3.1-S)in BABL/c(H-2d) and the protection against P815 mastocytoma cells stable expressing HBV surface antigen in mice after immunized with HBV gene vaccine.Methods:The immunization was performed by intramuscular injection,three weeks later,we directly inoculated P815-HBV-S into mice by subcutaneous injection .Tumor growth was measured every five days.Anti-HBs in serum was detected by ELISA and HBsAg specific cytotoxic T lymphocytes (CTLs) activity was measured by 51 Chromium release assay.Results:Eight weeks after immunization,the A value of mice serum in 450 nm and CTLs activity of mice codiog IL-2 and IL-12 eukaryotic expression vectors were significant higher(P<0.05) than that of mice intramuscular injected HBV-S DNA vaccine,these values are significant higher than that of mice injected pCR3.1(P<0.05).The spleen cells CTLs activity have decreased obviously after treated with anti-CD8+ monoclonal antibody and have no significant change after treated with anti-CD4+ monoclonal antibody.The HBV-S gene vaccine could evidently inhibit the tumor growth,prolong the survival period (>38.2 days) and improve the survival rate in mice.Conclusions:The DNA vaccine of HBV ( pCR3.1-S) had strong antigenicity in cellular and humoral immunity and had marked killing effect on HBV infected cells in vivo,which could be promoted by vector coding murine IL-2 or IL-12.CTLs activity was performed by CD8+ cells.
7.Arsenic trioxide (As(2)O(3)) induced apoptosis and its mechanisms in a human esophageal squamous carcinoma cell line.

OBJECTIVETo study whether As(2)O(3) has an apoptotic effect on human solid tumor cells, and the possible cellular and molecular mechanisms of this treatment using human esophageal squamous carcinoma cells (EC8712) as a model.
METHODSDNA microarray, biochemical and cytological analyses were used.
RESULTSThe growth and survival of EC8712 cells were markedly inhibited by As(2)O(3) treatment at a concentration of 1, 2 and 4 micromol/L. EC8712 cells were obviously arrested at G2/M phase with As(2)O(3) treatment and apoptosis induced at micromolar As(2)O(3) concentrations, as shown by morphology, histogram related nuclear DNA contents, and DNA gel electrophoresis. As(2)O(3) activated caspase-3, which might be involved in the process of As(2)O(3), induced apoptosis in EC8712 cells.
CONCLUSIONSAs(2)O(3) changes the expression of many genes at transcription level. The regulation of expression of many genes might be involved in the process of As(2)O(3) inducing apoptosis. These results suggest that As(2)O(3) can be clinically useful for solid tumor treatment.

8.Clinical and genetic analysis of five patients with familial male-limited precocious puberty

Objective:To analyze the clinical and genetic characteristics of five patients with familial male-limited precocious puberty(FMPP).Methods:The clinical data, laboratory and imaging results of the five patients with FMPP were collected. Whole exome sequencing was carried out to identify the potential variants. Suspected variants were verified by Sanger sequencing of family numbers.Results:Of the five patients, four were children and one was an adult. All the four children presented to hospital with premature sexual development at age less than 4 years. Serum testosterone was elevated, luteinizing hormone(LH) and follicle stimulating hormone(FSH) basal values were at prepubertal levels, and gonadotropin-releasing hormone(GnRH) stimulation test suggested peripheral precocious puberty. Genetic analysis revealed the mutations of LHCGR genes in all the five patients. Patients 1, 2, 3, and 4 carried the same heterozygous mutation c. 1713G>C(p.M571I), and the patient 5 carried the c. 1741T>C(p.C581R)variation. The four children were treated with anti-androgen preparations and the third-generation aromatase inhibitors, all of which were effective.Conclusion:The c. 1713G>C mutation of LHCGR gene is a novel one which expands the mutation spectrum of LHCGR gene. Combined treatment with bicaluamide and the third generation aromatase inhibitors can improve clinical symptoms and delay epiphyseal closure in children with FMPP.
9.Exploration of cognitive inhibitory function in negation and color-word Stroop task of patients with schizophrenia

Objective:To explore the impact of negation effect on color judgment of the color-word pairs and the impairment of cognitive inhibition function of the patients with schizophrenia (SCH) through embedding negation into the Stroop task.Methods:From January to December 2021, " Affirmative/ negative + color words" as the experimental materials, classical Stroop paradigm as the experimental paradigm, a mixed experimental design of 2 (groups: the SCH group vs the normal control group)×2(phrase polarity: affirmative phrases vs negative phrases)×2 (word-color congruence: consistency vs inconsistency) was designed to explore the inhibition function of negative phrases of the SCHs under bipolar negation conditions (Experiment 1) and multi-polar negation conditions (Experiment 2) respectively. The schizophrenic patients in Experiment 1(28 patients and 28 normal controls) and Experiment 2(30 patients and 30 normal controls) were sampled from the inpatients of Tangshan Kailuan Mental Health Center and Suzhou Guangji Hospital respectively, and the healthy controls were recruited from the community. SPSS 25.0 software was adopted as statistical analysis, and accuracy, response time of Stroop and the Stroop effect were analyzed by repeated measurement analysis of variance. Results:(1) The group main effects of response time were both significant in bipolar negation conditions(Experiment 1) ( F=49.22, P<0.001, ηp2=0.48) and multi-polar negation conditions (Experiment 2) ( F=37.58, P<0.001, ηp2=0.39). In bipolar negation (Experiment 1) conditions, as for the Stroop effect of response time, there was a significant interaction between the groups and polarities( F=4.42, P<0.05, ηp2=0.08), and there was a statistically significant difference between the affirmative Stroop effect (-0.137±0.522)and the negative Stroop effect(0.082±0.169) in the schizophrenia group( F=7.15, P<0.05, ηp2=0.12). In multi-polar negation conditions (Experiment 2), there was a marginal significant of the interaction between Stroop conditions and polarities in accuracy( F=3.81, P=0.056, ηp2=0.06). The accuracy under the word color consistency condition was higher than that under inconsistency condition((96.51±1.55)%, (90.00±2.54)%) ( F=5.15, P<0.05, ηp2=0.08). (3)In bipolar negation (Experiment 1) conditions, as for the accuracy, the interaction among groups, word color consistencies, and polarities was significant( F=6.66, P<0.05, ηp2=0.11). The interaction between word color consistencies and polarities was significant in the schizophrenia group( F=9.16, P<0.05, ηp2=0.15). In the normal group, the interaction between Stroop conditions and polarities was not significant( P>0.05). Conclusion:The impairment of cognitive inhibition function in schizophrenia is severe, which is reflected in negative processing; negation functions as cognitive inhibition, which is particularly prominent in bipolar negation.
10.Research of herb components on scavenging harmful components and reducing cytotoxicity of cigarette smoke.

OBJECTIVETo study the in vitro effect of herb components on scavenging harmful components of cigarette smoke such as radicals, polycyclic aromatic hydrocarbons, nitrosamines in vitro, and its reducing effect on cytotoxicity of cigarette smoke.
METHODspectrophotometry was used to examine the scavenging effect of herb components on DPPH free radicals, superoxide anion radical, and hydroxyl radical, and the results were compared with the anti-oxidation of ascorbic acid. Fluorescence spectroscopy was used to examine the scavenging effect of herb components on polycyclic aromatic hydrocarbons. UV spectrophotometry was used to examine the scavenging effect of herb components on volatile nitrosamines. MTT assay was used to examine cytotoxicity of cigarette smoke.
RESULTAll the herb components showed a certain scavenging effect on DPPH free radicals, superoxide anion radical, hydroxyl radical, polycyclic aromatic hydrocarbons and volatile nitrosamines, espacially the ginkgo biloba extract (GBE), flavonoids of glycyrrhiza, procyanidine, total saponins in ophiopogonin, total saponins of astragalus and epimediun flavonoids. After these six herb components were added to cigarette, the cytotoxicity of cigarette smoke on BEP2D cells was remarkably reduced, by increasing cell survival fraction (SF, %) and mean lethal dose (DML).
CONCLUSIONThe herb components can scavenge harmful components of cigarette smoke such as radicals, polycyclic aromatic hydrocarbons and nitrosamines, which reduce the damage of cigarette smoke on human being.
