AIM:In order to study the effects of retinal glial cells on glaucomatous retinal ganglion cells damage,expression of TNF-? and TNF-R1 in retinal glial cells was observed in rat model with chronic elevated intraocular pressure glaucoma.METHODS:(1)Rats were rendered elevated intraocular pressure by ligating 2 episcleral veins with subconjunctival injection of 5-Fu.(2)Four weeks after operation,immuno-histological assays were carried out on cryostat sections.The co-expressions of TNF-?-GFAP,TNF-?-OX42,TNFR-1-GFAP,and TNFR-1-NeuN were observed via a confocal laser scanning microscope,respectively.RESULTS:(1)Elevated intraocular pressure was consistently maintained for up to 4 weeks in model group.(2)The co-expressions of TNF-? and GFAP,TNF-? and OX42 were detected in retina in treatment group,respectively.(3)The co-expressions of TNFR-1 and GFAP were also detected in retina,but the co-expressions of TNFR-1 and NeuN were not detected in retina in treatment group.CONCLUSION:TNF-? that activated retinal glial cells may play an important role in glaucomatous retinal ganglion cell damage.

This paper reviewed and analyzed the management and performance of 43 key clinical pmjeets in recent ten years in Fudan University(the clinical part of original Shanghai Medical University).It was found that hardware equipments of clinic,the ability of disease prevention,diagnosis and treatment obviously improved,the talent training and personnel structure received outstanding outcome,the academic level of basic researches got obvious promotion,the preponderant disciplines established gradually and consolidated,and the center radiation effect showed preliminary success under the aid of key project fund.Constructing the key clinical projects should boost actively amalgamation of medicine and sciences and pay a great attention to develop the medicine auxiliary discipline to further enlarge the clinical research achievements and fully display the guidance function for the development of our country~clinical medicine discipline under the new situation.

Objective To compare the glycemic control of different management approaches in type 1 diabetic mellitus (T1 DM) children,and to evaluate the influence factors associated with glycemic control.Methods This survey included 68 T1DM children from January 2012 to December 2014 in the Affiliated Hospital of Qingdao University of more than 1 year management.Hemoglobin A1c (HbA1c) levels were detected to assess glycemic control.Twenty-three T1DM children who participated in the World Diabetes Foundation(WDF) management program (group A) were compared with 45 T1DM children who were managed by themselves (group B).The clinical data were analyzed to explore the correlations with glycemic control.Results The HbA1c of 68 children was (8.86 ± 2.00)％,and only 21 cases (30.9％) had optimal HbAlc (HbAlc ＜ 7.5％).The level of HbA1 c of group A (7.60 ± 1.57)％ was significantly lower than that of group B[(9.40 ± 1.91) ％] (t =-4.045,P =0.001).Group A had less fluctuation in blood glucose than group B,which the duration more than 3 years.Duration (F =2.277,P =0.004),frequency of self-monitoring blood glucose (F =9.375,P =0.001),diabetic education (F =3.276,P =0.002),and diabetic diet (t =-2.046,P =0.025) were associated with glycemic control.Conclusions The glycemic control of T1 DM children at the Affiliated Hospital of Qingdao University was at the medium level,most of them haven't reached the target levels.The WDF management program could significantly and continuously monitor the glycemic level.Diabetic education and more frequent self-monitoring blood glucose and diabetic diet are associated with better glycemic control.

Aim To develop a sensitive, rapid and ac-curate LC-MSn method for determination of midazolam/1′-hydroxymidazolam and their pharmacokinetic char-acteristics in rat brain. Methods SD rats received in-travenous injection of midazolam ( 5 mg · kg-1 ) via femoral vein, a probe drug of cytochrome P450 3A. The microdialysis ( MD ) samples in situ brain were collected every 8 mins at 2. 0μl·min-1 in 2. 4 hours. Analytes in brain dialysate were quantified by the pro-posed LC-MSn method. Gradient elution was performed on an Agilent Eclipse Plus-C18 column ( 2 . 1 × 50 mm, 3. 5 μm). The mobile phase consisted of 2 mmol ·L-1 ammonium acetate and acetonitrile. The analyte was detected using electrospray ionization ( ESI ) in multiple reaction monitoring ( MRM) modes. The reac-tion selected ions were 326 . 1/291 . 1 m/z for midazo-lam, 342. 1/324. 1 m/z for 1′-hydroxymidazolam and 285. 1/154. 0 m/z for diazepam as internal standard. Result The linear ranges of midazolam and 1′-hydroxymidazolam were 0 . 78~100 and 0 . 195 ~12 . 5μg·L-1 respectively. The lower limit of quantification was 0 . 2 μg · L-1 . The RSD of intra- and inter-batch precisions was less than 7 %. The RSD of accuracy was from -1 . 34 to -8 %. Conclusion This sensi-tive and rapid LC-MSn method is suitable for determi-nation of midazolam/1′-hydroxymidazolam in rat brain dialysate. MD combined with LC-MSn method may give assistance to deep and further studies of drug metabo-lism and CYP3A enzyme in brain.

Objective To investigate the value of multi-slice spiral computed tomography (MSCT) in diagnosing gastric cancer. Methods The MSCT signs of 48 gastric cancers proved histologically were evaluated retrospectively. Results The main CT signs included thickening of gastric wall and mass.The lesion showed marked enhancement in the arterial and portal venous phases, and 35 cases showed marked enhancement in the portal venous phase compared with the arterial phase. 12 cases had tumor invasion in the adjacent organs and lymph mode metastasis, and 11 cases had tumor distant metastasis. The accurate rate of enhancement staging was 87 5%. Conclusions MSCT enhancement scan can help us evaluate the lesion more perfectly, and also is helpful in diagnosing and staging of gastric cancer.

Ischemic stroke, gravely affecting human health with its increasing incidence rate , is a common disease of old age . JAK/STAT way , a recently discovered signaling pathway , is not only widely involved in processes of cell growth ,differentiation, and apoptosis ,but also closely related to the pathophysiology of stroke.However,the pathway function and mechanism in ische-mic cerebral stroke has not yet been fully elucidated .We will review the role and mechanism of JAK/STAT signal transduction pathway in ischemic stroke ,and scientifically draw network chart between various related signal molecules in the process of ische-mic stroke neuropathy combining both domestic and foreign re-search reported in this paper , in order to better understand the pathological mechanism process of brain stroke , find new drugs for the treatment of cerebral ischemic diseases ,then provide more systematic scientific literature support .

Aim To develop a sensitive,rapid and ac-curate LC-MS /MS method for the simultaneous deter-mination of cytochrome P450 probe substrates,inclu-ding caffeine and its metabolite paraxanthine for CYP1 A2,omeprazole and its metabolite 5-hydroxyome-prazole for CYP2C1 9,dextromethorphan and its metab-olite dextrorphan for CYP2D6,midazolam and its me-tabolite 1 ′-hydroxymidazolam for CYP3A4.Methods Probe drugs with the IS diazepam were extracted using ethyl acetate.Gradient elution was performed on an Agilent Eclipse Plus-C1 8 column (50 mm ×2.1 mm, 3.5 μm).The mobile phase consisted of 0.01 % for-mic acid(1 mmol·L -1 ammonium formate)and aceto-nitrile.The flow rate was 0.3 mL·min -1 ,and the in-jection volume was 1 0 μL.The analyte was detected u-sing electrospray ionization(ESI)in positive multiple reaction monitoring(MRM+)mode.The reaction se-lected ions were 1 95.0 /1 38.1 m /z for caffeine, 1 81 .1 /1 24.1 m /z for paraxanthine,346.1 /1 98.1 m /z for omeprazole,362.1 /21 4.1 m /z for 5-hydroxyome-prazole, 272.2 /1 47.1 m /z for dextromethorphan, 258.1 /1 57.1 m /z for dextrorphan,326.1 /291 .1 m /z for midazolam,342.1 /324.1 m /z for 1 ′-hydroxymid-azolam and 285.1 /1 54.0 m /z for diazepam as internal standard.Results The linear ranges of caffeine,pa-raxanthine,omeprazole,5-hydroxyomeprazole,dextro-methorphan, dextrophan, midazolam and 1 ′-hydroxymidazolam were 1 .95 ~2 000,0.98 ~250, 0.48 ~2 000,0.98 ~250,0.98 ~2 000,0.48 ~1 25,1 .95 ~2 000 and 1 .95 ~250 μg·L -1 respec-tively.The RSD of all probe drugs was less than 1 5%and matrix effects in plasma on the ionization of probe drugs were negligible.Conclusion This sensitive and rapid LC-MS /MS method is suitable for determination of the drug/metabolite concentrations in plasma,so as to study the metabolism of CYP1 A2, CYP2C1 9, CYP2D6 and CYP3A4 in depth.

AIM To study the pharmacokinetics of arbidol capsule in Chinese healthy volunteers.METHODS A single oral dose of arbidol capsule 200 mg was given to 20 healthy volunteers respectively.Plasma samples were prepared based on a simple liquid-liquid extraction.The extracted samples were analyzed by HPLC equipped with UV detection.Pharmacokinetic parameters were calculated by 3P87 software. Objective To compare the clinical outcomes of anterior cruciate ligament(ACL)reconstruction by using allograft and autograft of patella tendon.Methods From August 2003 to December 2005,82 ACL reconstruction patients were studied retrospectively,including 59 cases using autograft of patellar bone-tendon-bone(BPTB)and 23 cases using BPTB allograft.Physical examination,scores of IKDC,Lysholm-Tegner and VAS as well as KT-2000 measurements were used for evaluation pre-and post-operation.Results 41 autograft and 17 allograft patients were finally followed up.One allograft patient suffered re-rupture of the reconstructed ligament.Positive ADT test were found in 5 autograft and 1 allograft patients and positive Lachman test were found in 3 autograft and 1 allograft patients.No statistically significant differences were revealed in ROM.IKDC,Lysholm-Tegner Score and KT-2000 measurements showed no significant differences between two groups,but VAS for autograft group were significantly higher than allograft group(P
