1.Comparison of improvement of two kinds of preventative administration of inter-leukin-27 on airway inflammation and impaired phosphorylation of STAT1 in asthma mice

Objective:Animal models were set up to explore the best preventative intra-nasal administration of interleukin 27 (IL-27)to diminish allergic airway inflammation of asthma and the related molecular mechanisms. Methods:Ninety-six female C57/6J mice were randomly divided into four groups,a group of the control group,a group of asthma group,and two groups of the prevention group. Based on being sensitizing and challenging with Ovalbumin ( OVA ) in the asthma model, two kinds of IL-27 administration asthma animal models were set up,one of which was low-dose-multiple preventive administration before OVA sensitization,one was low-dose-multiple preventive administration after OVA sensitization but before OVA challenge. Sacrificed the mice after challenging and analyzed the IL-5 and IL-13 levels in supernatant of Broncho alveolar lavage fluid( BAL) using ELISA;and HE stain and inflammation score were done for the lungs. Sacrificed the mice before challenging and used the lungs to analyze the level of total signal transducer and activator of transcription-1(STAT1) protein and phos-STAT1 based on the method of Western blot. Results: In low-dose-multiple administration before sensitization preventions group,IL-27 inhibits the secretion of IL-5 and IL-13(P<0. 05) and inflammation around bronchial and vascular obviously, and the inflammation score was lower than asthma group ( P<0. 05 ) , while another group has no significant effects ( P>0. 05 ) . The phosphorylation of STAT1 was impaired in mice after OVA sensitization, and preventative administration of IL-27 before sensitization could reverse the impairment of STAT1, whereas another group had no obviously changes. Conclusion:Preventative administration of IL-27 before sensitization can attenuate the airway inflammation in the mouse asthma model via reversing the phosphorylation of STAT1,while the mice which has been sensitized resisting the inhibition of IL-27 due to the impairment of the phosphorylation of STAT1 and already committed Th2-CD4+T cells existed in sensitization-mice airway might be the reason for such IL-27 resistance.
2.Quality standard for Anshen Wendan Pill

AIM:To establish a quality standard for Anshen Wendan Pill(Rhizoma Pinelliae Praeparata,Pericarpium Citri reticulatae,Poria,Radix et Rhizoma Ginseng,etc.). METHODS:Pericarpium Citri Reticulatae,Fructus Schisandrae Chinensis,Radix et Rhizoma Ginseng,and Radix et Rhizoma Glycyrrhizae were identified by TLC,and the contents of schisandrin and hesperidin were determined by HPLC. The content of cinnabaris was determined by titration. RESULTS:Pericarpium Citri reticulatae,Fructus Schisandrae Chinensis,Radix et Rhizoma Ginseng,and Radix et Rhizoma Glycyrrhizae could be identified by TLC. The calibration curve of schisandrin was linear in the range of 0.017-0.52 ?g with the correlation coefficient of 1.000 0(n=5).The average recovery of schisandrin was 99.83%(RSD=0.86%,n=6). The calibration curve of hesperidin was linear in the range 0.506 -2.53 ?g,with the correlation coefficient of 1.000 0(n=5).The average recovery hesperidin was 100.43%(RSD=1.39%,n=6). The average recovery of cinnabaris was 98.30%(RSD=1.06%,n=6). CONCLUSION :The methods are simple,accurate and reproducible. They are suitable for the quality control of Anshen Wendan Pill.
3.Antimicrobial resistance of Ureaplasma urealyticum from reproductive systems of female patients at a traditional Chinese medicine hospital in three consecutive years

Objective To investigate antimicrobial resistance and change trends ofUreaplasma urealyticum (Uu) from reproductive system of female patients at a traditional Chinese medicine hospital in three consecutive years. Methods Antimicrobial resistance of Uu isolated from gynaecology leucorrhea and cervical secretion specimens of patients of dermatology & sexually transmitted diseases department and gynaecology department from January 2011 to December 2013 were analyzed retrospectively.Results A total of 327 Uu strains were isolated,except resistance rates of Uu to doxycycline(5.23%-6.12%)and josamycin(0-1 .96%)remained at a relatively low level in three consecutive years,resistance rate to the other antimicrobials showed an upward trend year by year,but the difference was not significant (P >0.05).The resistance of Uu to ofloxacin,clarithromycin and sparfloxacin were relatively high(42.11 %-61 .40%),to josamycin,doxycycline and minocycline were lower(0-13.07%),and multidrug re-sistance was serious.Conclusion Antimicrobial resistanc of Uu isolated from reproductive system of female patients in this hospital is high.It is necessary to realize the status and change trends of antimicrobial resistance of Uu for the treatment of disease and control of the emergence of multidrug resistance pathogens.
4.Epidemiological characteristics and therapeutic strategies of hepatitis C virus genotype 6 in Guangxi area

Objective To investigate the epidemiological characteristics and therapeutic strategies of patients infected with hepatitis C virus (HCV)genotype 6 in Guangxi area.Methods Serum samples were collected from 150 patients with serologic HCV RNA positive in Guangxi, China. Reverse transcription nested polymerase chain reaction (PCR)was employed to amplify HCV NS5B fragments and the DNA products were sequenced.The sequences obtained were compared with the sequences deposited in GenBank to construct a phylogenetic tree.Among the patients who accomplished 48-week treatment of interferon plus ribavirin and 24-week follow-up after stopping medication,10 cases were infected with genotype 6a and 28 cases with genotype 1 HCV.The virological responses were evaluated at week 4,week 12,week 24 of treatment and week 24 after the end of the treatment.Results Among all recruited 150 cases,21 (14.0%)cases were HCV genotype 6 including two subtypes 6a (n = 20 )and 6d (n = 1 ). Genotype 6 HCV mainly affected intravenous drug users, especially with age of ≤ 40 years old. Phylogenetic tree showed that there was very close evolutionary distance between HCV 6 strains of Guangxi and Hongkong,China strains (Y12083,DQ 480515)and Vietnam strain (EU246930).All of 10 HCV genotype 6a patients who completed 48 weeks of antiviral therapy achieved sustained virological response (SVR).The rate of SVR was higher than that of genotype 1 patients,but without statistically different significance (10/10 vs 75 .0%,P >0.05).Conclusion HCV genotype 6 in Guangxi area mainly affects young intravenous drug users with age of ≤ 40 years old,which has high homology with Hongkong,China and Vietnam standard strains.Patients with HCV 6 genotype infection treated with interferon plus ribavirin for 48 weeks usually achieve favorable SVR.
5.The B lymphocytes D8/17 antigen expression and characteristics of patients with rheumatic heart disease in Guangdong

ObjectiveTo investigate the D8/17 antigen expression of patients with rheumatic heart disease (RHD) in Guangdong province and study the antigen's characteristics.Methods The level of D8/17 antigen expression on B lymphocytes was determined with flow cytometry assay in 96 RHD patients and 83 unaffected controls.The percentage of B-cells expressing the D8/17 antigen having more than 10% was considered to be positive. D8/17 antigen was extracted by immunopreeipitation,and the antigen characteristics was analyzed by tandem mass spectrometry. Results The mean percentage of B-cells expressing the D8/17 antigen was (85.36 ± 15.15)% in the RHD patients and (82.89 ±4.55)% in the controls,with no significant difference between the two groups ( P =0.436).Moreover,the positive rate of the D8/17 cxprcssion was 100% in either the RHD patients or the controls.The molecular weight of D8/17 antigen was found to be 40 000-67 000,and the purified protein was most likely to match moesin or β-actin.ConclusionsB-cell antigen D8/17 is not associated with RHD in Guangdong province of China.Moesin or β-actin is the most likely protein to match D8/17 antigen.
6.Recent technical research hot spots and development progresses in medical whole-body positron emission tomography.

Medical whole-body positron emission tomography (PET), one of the most successful molecular imaging technologies, has been widely used in the fields of cancer diagnosis, cardiovascular disease diagnosis and cranial nerve study. But, on the other hand, the sensitivity, spatial resolution and signal-noise-ratio of the commercial medical whole-body PET systems still have some shortcomings and a great room for improvement. The sensitivity, spatial resolution and signal-noise-ratio of PET system are largely affected by the performances of the scintillators and the photo detectors. The design of a PET system is usually a trade-off in cost and performance. A better image quality can be achieved by optimizing and balancing the key components which affect the system performance the most without dramatically increases in cost. With the development of the scintillator, photo-detector and high speed electronic system, the performance of medical whole-body PET system would be dramatically improved. In this paper, we report current progresses and discuss future directions of the developments of technologies in medical whole-body PET system.

8.Effect of early rehabilitation intervention on acute cerebral infarction

Objective To investigate the effect of early rehabilitation intervention on acute cerebral infarction.Methods 132 patients with acute cerebral infarction were evenly divided into the observation group and the control group: the former received early rehabilitation intervention and the latter routine nursing intervention.The two groups were assessed and compared with Fugl-Meyer Assessment(FMA),Bathel Index(BI)and Self-rating Depression Scale(SDS).Results The scores on FMA and BI in the observation group were higher than those in the control group(P<0.05),while the scores on depression in the observation group were lower than those in the control group(P<0.05).Conclusion The early rehabilitation intervene may improve the motor function and activities of daily living and reduce the degree of depression.
9.Changes of plasma nesfatin-1 levels in patients with impaired glucose regulation and type 2 diabetes mellitus

Plasma nesfatin-1 levels were assayed by ELISA in patients with impaired glucose regulation (IGR),type 2 diabetes mellitus( T2DM ),and healthy subjects with normal glucose tolerance( control group).The results showed that plasma nesfatin-1 levels in IGR and T2DM groups were significantly higher than that in control group [ ( 1.91 ±0.79 and 1.80±0.80 vs 1.41 ±0.58) μg/L,P<0.01 ].Plasma nesfatin-1 levels were positively correlated with body mass index(BMI),HbA1c,waist hip ratio,fasting plasma glucose,fasting insulin,and homoestasis model assessment insulin resistant index( HOMA-IR,P<0.05 or P<0.01 ).Multiple regression analysis showed that BMI and HOMA-IR were independent related factors in influencing plasma nesfatin-1 levels (both P<0.01 ).These results suggest that nesfatin-1 may partially contribute to the pathogenesis of insulin resistance and T2DM.
10.High resolution melting analysis for the rapid and sensitive detection of KRAS codon 12 and 13 mutations in colorectal cancer

Objective To establish a HRM assay to screen for KRAS mutations in clinical colorectal cancer patients.Methods The sensitivity of HRM was analyzed by detecting somatic mutations in exon 2,notably codons 12 and 13 of the KRAS gene in the serial plasmid mixture samples which were mixed using the different proportions mutation plasmid and wide type plasmid of KRAS.HRM analysis was performed for KRAS on DNA insolated from a panel of 60 colorectal cancer samples derived from fresh tissues.The results were compared with the direct sequencing data.Results After the PCR amplification,the mutation results could be available by performing HRM analysis in the same tube on a real time PCR machine with HRM capability.HRM detection could identify KRAS mutation in a proportion of 10% of mutation plasmid DNA.All 60 samples identified the KRAS mutation by HRM and sequencing.17 samples were positive(28.3%) by HRM for KRAS exon 2 mutations,and 15 samples were confirmed the presence of codon 12 or 13 mutations(25.0%) and the other 2 samples were wild type by sequencing.The 60 samples detected by HRM were given 100% sensitivity with 96% specificity.Conclusions HRM is a sensitive intube methodology to screen for mutations in clinical samples.HRM will enable high-throughput screening to gene mutations to allow appropriate therapeutic choices for patients and accelerate research aimed at identifying novel mutations in human cancer.
