1.Study on the weaning method of mechanical ventilation in the treatment of 51 patients with acute severe respiratory failure caused by organophosphate poisoning

ObjectiveTo explore the weaning method of mechanical ventilation in the treatment of acute severe respiratory failure caused by organophosphate poisoning.MethodsThe clinical features and weaning situation of 51 patients with acute severe respiratory failure caused by organophosphate poisoning by the treatment of mechanical ventilation were retrospectively analyzed.ResultsIn 51 cases,45 cases were cured,the survival rate was 88.2% and 6 cases died,the fatality rate 11.8%.Conclusion Early ventilator to rescue and choose opportunity for weaning could directly impact on the qualitv of life of patients,and improve outcomes,and reduce mortality.

3.Effects of arsenic trioxode and adriamycin on survivin expression in (hepatocellular) carcinoma cell line HepG2

Objective To investigate potential mechanism of arsenic trioxide (As2O3) and/or adriamycin (ADM) induces apoptosis in hepatocellular carcinoma (HCC) cells. Methods Human hepatocellular (carcinoma) cells, HepG2, were treated with As2O3 combined with/without ADM in various concentrations for various time, and then the expression of survivin was detected by immunocytochemistry and RT-PCR (respectively). Results (1)Before the cells were interfered with drugs, survivin was highly expressed in HepG2 cells. (2)Expression of survivin was reduced in cells treated with various concentrations of As2O3 or ADM , and the reduction was dependent on durg concentration and time, these two factors were interrelated. (3)Expression of survivin was reduced more markedly in cells treated with ADM than in those treated with As2O3 and the reduction was additionally increased in cells treated with a combination of the two compounds. Conclusions (1)Either As2O3 or ADM can downregulate the expression of survivin in HepG2 cells in a concentration-or time-dependent manner; (2)At the same concentration, ADM is more effective than As2O3 on the downregulation of survivin expression in HepG2 cells, but the effect can be enhanced by combination of these two drugs. (3)As2O3 and ADM may induce apoptosis in HepG2 cells through the downregulation of survivin expression and thus exert their anti-cancer effect. Their combination may enhance their anti-tumor effect, so that in clinical use, the dosage of each durg may be reduced.

6.Preliminary study of apoptosis in human ovarian cell line COC1 induced by ultrasound and its mechanism

Objective:To investigate the antitumor effect of Low-Frequency Pulse Forcusd Ultrasound(LFPFU) exposure in human cell line COC1,and to clarify its probable mechanism.Methods:The growth inhibitory effect of ultrasound exposure[0.8MHz,83.6W/cm 2,pulse duration(PD) 50?s;sonication time of 10s] on COC1 cell was observed by MTT colormetric assay.Apoptosis was analyzed using flow cytometry(FCM) and terminal deoxynucleotidyl transferase-mediated dUTP-digoxin nick end labeling(TUNEL),and fluorescent microscope and electron microscope were applied to detect the morphological changes of COC1 cell.The protein expression of bcl-2 and bax affected by LFPFU were determined by FCM and immunofluorescent techniques.Results:Sonication could obviously suppress proliferation of COC1 cells surviving ultrasound exposure.The absorbance value(A 570 )of MTT decreased significantly at 12h,24h,36h and 48h after sonication as compared with control group.The hightest apoptosis rate was found to reach 23.57?4.62% by FCM,and brown-colored positive apoptotic cells were observed with TUNEL method at 6h after sonication.Typical morphological changes of apoptosis such as apoptotic body was found under fluorescent microscope and transmission electron microscope.Fluorescence index(FI) of bcl-2 decreased,and Fl of bax increased after LFPFU exposure.Conclusion:LFPFU can induce apoptosis in vitro in human ovarian cell line COC1 surviving ultrasound exposure.The bcl-2 and bax protein may be involved in the apoptotic process.
7.Experimental study on hydroxychloroquine raised insulin sensitivity in insulin resistance rat models

Objective:To observe the effects of hydroxychloroquine on insulin sensitivity in high-fat feeding rats.Methods:After administrating hydroxychloroquine,the glucose infusion rate (GIR) determined by euglycemic clamp was used to measure the insulin sensitivity,and determining fasting plasma glucose,lipid and insulin.Results:(1)In high-fat feeding rats,the GIR60-120 was lower and the fasting plasma glucose,insulin,lipid levels were higher than those in normal feeding rats.(2)Hydroxychloroquine,rosiglitazone and metformin increased insulin sensitivity (the value of GIR60-120 was 9.3?1.2,11.6?1.4 and 9.8?1.3 mg.kg/min,respectively) in insulin resistance (IR) models.Moreover,the fasting glucose and some plasma lipid indices were improved in the IR rats with hydroxychloroquine administration.Conclusion:It is indicated that hydroxychloroquine could raise insulin sensitivity of IR rats with high-fat feeding,which was similar to metfomin but lower than rosiglitazone.In addition,hydroxychloroquine also could improve the glucose and blood lipid of the IR rats in some extent.

9.Establishment of hyperinsulinemic euglycemic clamp technique

Objective:To establish a hyperinsulinemic euglycemic clamp technique for the study of insulin sensitivity in Chinese.Methods:A hyperinsulinemic euglycemic clamp technique was used to the study of methodology in 8 normal weight subjects with normal glucose tolerance.Results:(1)When a higher level of insulin was created during maintaining fasting euglycemia,the secretion of endogenous insulin and hepatic glucose production were completely inhibited.The secretion of counter-regulatory hormones(cortisol,growth hormone and glucagon) was not significantly stimulated..(2)During the steady-state of hyperinsulinemic euglycemic clamp,the glucose disposal rate was 9.25?0.25mg/(kg.min)Conclusion:The hyperinsulinemic euglycemic clamp technique was established successfully in Chinese.During the steady-state of exogenous insulin and glucose metabolism,glucose utilization increased.
10.Antitumor components from sponge-derived fungus Penicillium auratiogriseum Sp-19

Objective To isolate and identify the antitumor bioactive components from the fermentation of the fungus Penicillium auratiogriseum Sp-19 derived from sponge Mycale plumose.Methods The antitumor components were isolated by bioassay-guided fractionation and using solvent extraction,silica gel column chromatography and preparative HPLC.Their structures were established by physicochemical properties and spectral analyses.The cytotoxicities of compounds were evaluated by SRB method.Results and Conclusion Five alkaloid compounds were isolated and identified as fructigenines A(1),deacetyl fructigenines A(2),fructigenines B(3),1,4-benzodiazepine-2,5-diones(4) and cyclopenin(5),respectively.Compound 4 and 5 showed cytotoxicity at the concentration of 3 ?g?mL~(-1)against tsFT210 cell line.
