1.Analysis of 1812 Cases of Cyclosporine Therapeutic Drug Monitoring Data

OBJECTIVE:To evaluate1812cases of therapeutic drug monitoring(TDM)and the clinical drug utilization of cyclosporine.METHODS:Analysis and statistics of cyclosporine TDM data of1812cases(502subjects)in our hospital were carried out.RESULTS:Among all the1st TDM concentration,only41.4%was within the therapeutic window.Constituent ratios of patients whose drug concentration were monitored once,twice,or three times respectively was44.8%,16.3%and11.6%respectively.Among all the subjects undergoing3~4times of TDM,constituent ratio of patients whose drug con-centrations of the1st,2nd,3rd and4th TDM are within therapeutic window are40.6%,48.0%,50.0%and57.9%respec-tively.CONCLUSION:Although TDM is widely used,for some reasons,the constituent ratio of drug concentration within therapeutic window is low and the TDM times are much less than enough.Even TDM is multiple,the status of is not satisfac-tory,So clinics should pay more attention to the phenomenon that the patients’medication can’t be adjusted properly;Mean-while,it correlates with the expensive medication costs,etc.
3.Influence factors of the release rate of benorilate from sustained-release matrix tablets

Objective To study the influence factors of the release rate of benorilate from sustained release matrix tablets. Methods The matrix tablets of benorilate were prepared by using hydroxypropylmethylcellulose (HPMC) as the matrix material. The effects of the contents of HPMC, polyvinyl pyrrolidone (PVP), and microcrystaline cellulose (MCC), and the method of preparation on in vitro drug release were studied by evaluating the n value in the Peppas equation. Results The increasing HPMC content led to decrease of benorilate release. However, PVP and MCC used in this experiment accelerated the benorilate release from the tablets. The drug released from the tablet prepared by dry method was faster than that by wet method. Conclusion The contents of HPMC, PVP, and MCC, especially HPMC, have effects on the release rate of benorilate, but the two preparation methods have less effect.
4.Adenylate cyclase stimulation and ocular hypertension inhibition by forskolin analogs

ObjectiveForskolin (FSK) analogs,isoforskolin (isoF),deacetylforskolin(deaF),and 1-acetylforskolin(1-aF),extracted from Coleus forskohlii native to Yunnan,were assayed for their adenylate cyclase stimulating activities in vitro and for effects of two analogs on ocular hypertension (OHT) in water-loaded rabbits.MethodsAdenylate cyclase stimulation was determined by protein-binding method of radioimmunoassay,and intraocular pressure was monitored by pneumatonometer.ResultsIt showed that isoforskolin and forskolin stimulated adenylate cyclase in vitro with almost equal activity,deacetylforskolin with milder activity,and 1-acetylforskolin with little activity in vitro.1% deaF and 1-aF suppressed rabbit OHT induced by water-loading for at least 3h,with the maximal inhibitory rates of 6.0,10.9% respectively.ConclusionThis study suggests that two foskolin analogs (isoforskolin,deacetylforskolin) possess adenylate cyclase stimulation activities in vitro;deacetylforskolin and 1-acetylforskolin suppress OHT induced by water-loading in rabbits.
5.Effects of 1-acetylforskolin on Intraocular Pressure in Rabbits and Men

To investigate effects of l% l-acetylforskoin (l-aF) suspension eye-drops administeredtopically on intraocular pressure (IOP) in rabbits and men, and observe the loca l irritation of the drug. l% l-aF suspension eye-drops was prepared and was instilled into one of e yes while the corresponding vehicle into the contralateral eye as control. IOP was measured b y pneumatonometer before and after administration. l%1-aF suspension eye-drops s ig nificantly reduced IOP in rabbits reaching the maximum reduction of 2.2 mmHg and lasting a t least 3 hours. But it didnt show significant effects in men. There was little local irritati on neither in rabbits nor in men. l% l-aF suspension eye-drops significantly reduced IOP in normal rabbit s, but not in men. No local irritation was observed in rabbits and in men.
6.Clinical Observation on Shilin Qinghua Decoction for Treatment of Urinary Calculi

Objective To observe the therapeutic effect of Shilin Qinghua Decoction(SQD) for the treatment of urinary calculi.Methods Two hundred urinary calculi patients were equally randomized into two groups: the treatment group received SQD(mainly composed of Herba Lysimachiae,Endothelium Corneum Gigeriae Galli,Radix Paeoniae Alba,Talcum,Radix Clematidis,Poria,Semen Vaccariae,Herba Plataginis,Herba Polygoni Avicularis,Herba Dianthi,Succinum powder),and the control group received uralyt.Twenty days constituted one treatment course,and the treatment lasted 1~3 course(s) according to the illness state.The therapeutic effect was evaluated after 2 months.Results The total effective rate was 96.0% in the treatment group and 81.0% in the control group,the difference being obviously significant(P

Objective To investigate antithrombotic and antiplatelet effects of Dragon's Blood,?-cyclodextrin inclusion of Dragon's Blood.Methods The methods of injection of adenosin diphosphate(ADP) into mouse tail vein,electrically stimulated arterial thrombosis in rats were used to evaluate the anti-thrombotic effects of Dragon's Blood and?-cyclodextrin inclusion of Dragon's Blood,respectively.Platelet aggregation(PRP or washed platelets) was tested according to Born's method.Results ?CYDB significantly protected against thrombosis caused by ADP on mice,and electrical stimulation on rats.In vitro?-cyclodextrin inclusion of Dragon's Blood inhibited AA,ADP,PAF-induced platelet aggregation.Conclusions The results suggest that compared with Dragon's Blood,?-cyclodextrin inclusion of Dragon's Blood has more potent anti-thrombotic effects on the thrombotic models,its mechanisms may be closely related to the inhibition of anti-platelet aggregation.?-cyclodextrin inclusion of Dragon's Blood can improve the bioavailability of Dragon's Blood.
8.Establishment of normal value of posterior arch length of adolescence in Chengdu.

OBJECTIVETo investigate the development of third molar embryo and establish the normal value of posterior arch length of adolescence in Chengdu.
METHODSThe samples consisted of 40 males and 41 females with Class 1 dentitions, normal second molar occlusion, no history of orthodontic treatment, and good facial balance. Cephalometry and statistic analysis were conducted for all the subject.
RESULTSThe normal value of posterior arch length of maxilla in male was(16.52 +/- 2.35) mm and in female was(16.42 +/- 2.55) mm. The normal length of mandible was(13.00 +/- 2.44) mm in male and(12.43 +/- 2.18) mm in female. But statistic analysis showed no gender difference. Most of the third molar embryo had been formed and their tooth crowns had been calcified, the occurrence ratio was 84.0% in maxilla and 85.2% in mandible. Most of them located in the middle and inclined mesially.
CONCLUSIONThe data obtained in this study will be valuable for our clinical practice and be helpful for the study concerning dento-facial growth and development.

9.An experimental study on the detection of Porphyromonas gingivalis with multifunctional nanospheres

Objective:To development a new method for sensitive detection of Porphyromanus gingivalis (P.gingivalis) based on magnetic encoding nanospheres and upconversion fluorescent encoding nanospheres.Methods:Magnetic and upconversion fluorescent encoding nanospheres were prepared by sol-gel method respectively,combined the monoclonal antibodies specific to P.gingivalis after modifying the surface of nanospheres.The system was used to detect P.gingivalis from mixed bacteria solution of P.gingivalis,F.nucleatum and S.mutans.Fluorescent microscopy with an external 980 nm near-infrared hght pulse laser,scanning and transmission microscope were used to evaluate the effectiveness of the detection system.Results:Magnetic and upconversion encoding nanospheres had better dispersion,particle size uniformity and homogeneous morphology.Besides,the magnetic encoding nanospheres had good magnetic properties and strong fluorescence intensity.P.gingivalis was captured by magnetic and upconversion encoding nanospheres in a mixed solution of the 3 bacteria with a detection limit of 10 CFU/ml.Conclusion:The method designed in this study can capture P.gingivalis sensitively in a mixed bacteria liquid.
10.Effect of sex on reperfusion time delay in ST-segment elevation myocardial infarction patients after diagnosis

Objective To study the effect of sex on reperfusion time delay in ST-segment elevation myocardial infarction (STEMI) patients after diagnosis.Methods A total of 1647 STEMI patients admitted to our hospital were included in this study.Their clinical data and treatment time points were recorded.The reperfusion time delay after diagnosis refers the time from ECG-based diagnosis to PCI.The reperfusion time delay after diagnosis was divided into 0-3 h,>3-6 h,>6-12 h,>12-24 h and >24 h.The effect of sex and other risk factors on reperfusion time delay after diagnosis was assessed according to the established logistic regression model.Results The age of female STEMI patients was older than that of male STEMI patients (65±10 years vs 60±11 years,P<0.05).The rate of past CABG and PCI was significantly higher and the reperfusion time delay was significantly longer in female STEMI patients than in male STEMI patients (4.3 % vs 1.5%,20.7% vs 17.4%,P<0.05;404±34 min vs 280±14 min,P<0.01).Multivariate logistic regression analysis showed that female and visiting form were related with the reperfusion time delay for >3-6 h and >12-24 h (95%CI:1.052-264.306,P=0.046;95%CI:1.089-2.751,P=0.013).Conclusion Female is related with reperfusion time delay,visiting form and call for emergency treatment in STEMI patients.
