1.The protective effect of L-arginine on vascular endothelial cell during cardiopulmonary bypass

ve To assess the protective effect of L-arginine on vascular endothelial cell during cardiopulmonary bypass(CPB). Methods 12 adult healthy mongrel dogs of either sex weighing 10-15kg, were divided into L-arginine group (group L, n=6) and control group (group C, n=6) . Femoral artery and vein were cannulated for monitoring of MAP and CVP. Right internal jugular vein was cannulated with Swan-Ganz catheter for hemodynamic monitoring. L-arginine 100 mg?kg-1 was administered intravenously before CPB followed by continuous intravenous infusion at 10mg?kg-1.min-1 to the end of CPB in group L. Control group was treated as group L except the administration of L-arginine. Venous blood samples were taken before CPB, 5min after CPB was started, 15, 45min after aortic cross-clamping and 15, 30min after release of aortic cross-clamp for determination of the plasma levels of metabolites of nitric oxide (NO2- and NO3-), malondialdehyde (MDA) and von Willebrand Factor(vWF). Hemodynamic parameters were measured before CPB, 15 and 30min after release of aortic clamp. Specimens of tissue from femoral artery and descending aorta were taken for microscopic examination at 15,45 min after aortic cross-clamping and 30min after release of aortic cross-clamp.Results There was little difference in plasma NO2- and NO3-concentrations before CPB between the two groups. 3.Concentration Determination of Inorelbine Bitartrate in Human Plasma by LC-MS/MS

OBJECTIVE:To establish a method for determining the concentration of vinorelbine bitartrate in human plasma, and study the pharmacokinetic process of vinorelbine in human plasma. METHODS:The samples were extracted with Solid-phase extraction(SPE). LC-MS/MS was performed on the column of Agilent Extend C18 with the mobile phase of 5 mmol/L ammonium ac-etate solution(pH10.5)-acetonitrile-methano1(18∶10∶72,V/V/V)at the flow rate of 0.4 ml/min,the ion source was ESI and MRM mode was used to scan positive ion detection. The ion reaction of quantitative analysis was m/z 779.50→m/z 122.10 (vinorelbine) and m/z 811.60→m/z 224.50 (internal standard, vinblastine). 3p97 was used to calculate the pharmacokinetic parameters. RE-SULTS:The determination was not interfered by endogenous impurities,and there was no matrix effect;the lowest limit of quantita-tive analysis was 2.0 ng/ml with the linear range of 2.0-4 000.0 ng/ml(r=0.997 8);the linear range of vinorelbine in plasma was. The intra-day and inter-day precisions and accuracy results were all in line with the acceptable limit across all concentrations. t1/2γ of 30 mg/m2,40 mg/m2 Vinorelbine Emulsion for Injection groups and 30 mg/m2 Vinorelbine for Injection group were(37.958 ± 34.256)、(47.835±54.231)、(76.873±40.537)h respectively,cmax were(1 426.250±397.562)、(1 700.125±624.669)、(2 187.500± 828.040)ng/ml respectively,AUC0-48 h were(75 839±19 551)、(82 088±14 207)、(95 318±18 208)mg·h/L respectively. CONCLU-SIONS:The method is rapid,sensitive and specific,and suitable for the determination of vinorelbine and pharmacokinetic study. Metabolic processes of vinorelbine can be described as first order process of three-compartment model in cancer patients.
4.Influence of epidermal growth factor on proliferation, migration and apoptosis of adipose-derived stem cells in vitro

Objective To investigate the effects of epidermal growth factor (EGF) on the proliferation , migration and apoptosis of adipose-derived stem cells (ADSCs) in vitro. Methods Starved cell model and FasL-induced apoptosis model were established in serum-free media. The effects of 10 nmol/L and 100 nmol/L of EGF on the proliferation, migration and apoptosis of ADSCs in vitro were observed. The exrpessions of signal pathway proteins like phospholipaseC-γ(PLC-γ) , extracellular regulated kinase (ERK) and AKT were also detected. Results The proliferation, migration and anti-ap-optosis of ADSCs were promoted by 10 nmol/L or 100 nmol/L of EGF, and the expressions of PLC-'y, ERK and AKT were up-regulated. Conclusion EGF can promote the proliferation, migration and an-ti-apoptosis of adipose-derived ADSCs in vitro.
5.The impact of using a Swiss ball to exercise the lumbo-abdominal muscles

Objective To explore any changes in surface electromyogram (sEMG) signals from the lumboabdominal muscles during exercise on stable and unstable surfaces. Methods sEMG signals from the lumbo-abdominal muscles of 33 healthy young persons [18 male and 12 female; average age (26.5 ± 4.3 ) years] were measured with the FlexComp Infiniti apparatus.Each subject performed 5 exercises on and off a Swiss ball:sit,bridge,bridge with both knees flexed,reverse bridge as well as press-up. Results ①In bridging there was a significant increase in the activation of the erector spinae during exercise on the ball compared with on the stable surface.②Bridging with both knees flexed gave a significant increase in activations of the erector spinae,the external obliques and the transverses abdominus/internal obliques during exercise on the unstable surface compared with the stable surface.③During reverse bridging there was a significant increase in activation of the erector spinae and rectus abdominus during exercise on the unstable surface compared with the stable surface.④During press-ups there was a significant increase in activation of the rectus abdominus,the external obliques and the transverses abdominus/internal obliques during exercise on the ball compared with the stable surface. Conclusion The unstable surface provides better training stimulus for the activation of the lumbo-abdominal muscles.
6.Clinic significance of neutrophil-iymphocyte ratio in the early-stage diabetic nephropathy

Objective To investigate neutrophil lymphocyte ratio (NLR) in early-stage diabetic nephropathy and its clinic significance. Methods The 145 subjects were divided into two groups:the healthy control group (n=54) and early stage diabetic nephropathy group (n=91). The numbers of neutrophils (N) and lymphocytes (L) as well as the NLR values of peripheral blood and other biochemistry index were examined. Factors of early stage diabetic nephropathy were calculated us?ing variance and logistic regression analysis. Results Creatinine(Cr), total cholesterol(TC), triglyceride(TG), LDL-C, neu?trophils number and CRP in DN group were significantly higher than those of the control group and lymphocytes numbers of DN group were significantly lower than that of the control group (P<0.05 respectively);NLR values were significantly higher in diabetic nephropathy group compared with those of healthy control group(2.52±0.57 vs 1.82±0.60,t=6.997, P<0.01). Lo?gistic regression analysis showed that the risk factors of DN include NLR, TG and total cholesterol. NLR ( P <0.001, OR=8.951, OR 95%CI:3.595-22.287) was significantly associated with DN. Conclusion High NLR values may be a predic?tive and reliable marker ofearly-stage DN.
7.Expression and purification of SARS coronavirus membrane protein.

To construct a recombinant plasmid Pet23a-M, the gene encoding severe acute respiratory syndrome (SARS) coronavirus membrane protein was amplified by RT-PCR and cloned into the expression plasmid Pet23a. Results of restriction endonuclease analysis, PCR detection and DNA sequencing analysis revealed that the cloned DNA sequence was the same as that reported. The recombinants were transformed into Escherichia coli (E. Coli) BL21 (DE3) and induced by Isopropyl-beta-D-thiogalactopyranoside (IPTG). The expression of 27 kD (1 kD=0.992 1 ku) protein was detected by SDS-PAGE and pured by metal chelated chromatography. Results of Western-blot showed that this expressed protein could react with antibodies in sera of SARS patients during convalescence. This provided the basis for the further study on SARS virus vaccine and diagnostic agents.

8.Efficacy and safety of ciprofol in combined intravenous and inhalation anesthesia in elderly patients undergoing thoracoscopic surgery

Objective To research the efficacy and safety of ciprofol in combined intravenous and inhalation anesthesia in elderly patients undergoing thoracoscopic surgery.Methods A retrospective analysis was conducted on 806 elderly patients who underwent thoracoscopic surgery from January 2021 to July 2022,416 males and 390 females,aged 65-85 years,BMI 16-30 kg/m2,ASA physical status Ⅰ-Ⅲ.Patients were divided into two groups according to different intravenous anesthesia drugs during the sur-gery:ciprofol group(group C,n=229)and propofol group(group P,n=577).The preoperative and intraoperative indexes of the two groups were balanced by 1∶1 propensity score matching(PSM).Multiva-riate logistic regression was used to analyze the correlation between intravenous anesthetics and the incidence of adverse reactions(hypotension,bradycardia,hypoxemia during recovery,and postoperative nausea and vomiting).Multivariate linear regression was used to analyze the correlation between intravenous anesthetics and short-term recovery indicators after anesthesia(extubation time,postanesthesia care unit residence time,and length of postoperative hospitalization).Results Compared with group P,the total dosage of va-soactive drugs in group C were significantly reduced(P<0.05),extubation time and PACU residence time were significantly shortened(P<0.05).Multivariate logistic regression analysis showed that the use of cip-rofol was associated with a reduced incidence of intraoperative hypotension(OR=0.378,95%CI 0.236-0.603,P<0.001)and hypoxemia during recovery(OR=0.542,95%CI 0.336-0.874,P=0.012).Multivariate linear regression analysis showed that the use of ciprofol was associated with the reduction of ex-tubation time and PACU residence time(P<0.001).Conclusion Compared with propofol,combined in-travenous and inhalation anesthesia with ciprofol can significantly reduce the incidence of anesthetic adverse reactions in elderly patients,and is conducive to short-term recovery after anesthesia.
9.Primary study on quality standard of carbonizing drug characteristic of ginger carbon.

OBJECTIVETo establish the quality standard for carbonizing drug characteristic of ginger carbon.
METHODGingers and different carbonized gingers were compared by the absorption of pigment, tannin content, pH, mouth's coagulation time and bleeding time.
RESULTThe study resulted in the recommended carbonizing standard that the absorption capacity shall not be less than 7.50 mg x g(-1) for methylene blue, the tannin content shall not be less than 2.103 mg x g(-1), the pH shall be (5.50 +/- 0.10), and coagulation time and bleeding time shall be the shorter the better.
CONCLUSIONThe established assessment standard for carbonizing drug characteristic of ginger carbon is reasonable, easily operated and feasible.

10. Effect of cigarette smoke exposure during pregnancy on offspring of ovarian development and oocyte DNA methylation in female mice

Objective:
To explore the effects of cigarette smoke exposure during pregnancy on the quality of oocytes and the whole genome DNA methylation in the offspring of female mice during the period of germinal vesicle (GV) .
Methods:
The pregnant 7 d mice were divided into 3 groups, exposure on the 0, 0.5, 1 lit cigarettes cabinet (volume 18 L) , at 9 am and 3 pm for 1 h twice daily, until delivery. When the mice were 6 weeks old, the organ index and the number of follicles in the ovary were detected by weighing and making HE stained sections. GV stage oocytes were obtained by Hoechst 33342 staining and indirect immunofluorescence to detect the quality of oocytes, chromatin configuration and whole genome DNA methylation level.
Results:
Compared with the control group and low dose group, the offspring ovarian organ index of female mice in the high dose group decreased, the difference was statistically significant (
