1.The study of the possible correlation between the MDA and adenylate cyclase or cAMP of Ehrlich's ascites cancer
Chinese Journal of Pathophysiology 1986;0(03):-
Ehrlich's ascites cancer (EAC) in BALB/C mice by ip transplantable EAC cell is reproduced by us. The change of serum malondialdehyde(MDA), plasma cellular cAMP and adenylate cyclase (AC) are examined in the early, middle and late phase of mice bearing EAC respectively in comparison with that of control group. Results indicate that serum MDA in mice bearing EAC is continuously increased with the progress of the disease in the remarked ways in comparison with the control group. Whereas the plasma cAMP in the mice bearing EAC is markedly decreased in that order. Relation between the change of serum MDA and the cellular cAMP, AC or plasma cAMP shows a significant negative correlation efficiency (P
2.The study of plasma MDA, cAMP/cGMP in nude mice bearing human NPC and effects of PSP
Chinese Journal of Pathophysiology 1989;0(05):-
Nasopharyngeal carcinoma (NPC) in BALB/c nude mice (NM) was re-produced by hypodermic implanting human CNE-2 cell to the right-side of neck-backposi-tion. The changes of plasma malondialdehyde (MDA) and cAMP/cGMP ratio in NM bea-ring human NPC were examined. Meanwhile, we observed that tumor's size, plasma MDAand cAMP/cGMP ratio in NM bearing human NPC were affected by polysacchoridopetide(PSP). Results indicated that in comparison with the control group, NM bearing humanNPC showed a marked increase in plasma MDA(18.25?1.37 nmol/ml),cAMP/cGMP ratio(4.55?0.35), a marked decrease in plasma, cAMP (71.83?8.51 pM/ml) and cGMP(15.77?0.65 pM/ml), both high or low-concentration PSP inhibited NPC growth.Rate of inhibition was between 59.58 to 95.53 percent. PSP decreased plasma MDA,elevated cAMP, cGMP's levels and recovered cAMP/cGMP ratio. Our results suggestedthat the elevated level of MDA in NM bearing human NPC was closely associated withthe presence of NPC. PSP had antitumor effect, probably by means of decreasing freeradicals and recovering cAMP/cGMP ratio.

Objective To develop and test a stressor scale for in-patients with spinal cord injury (SSIPSCI). Methods Based on in-depth interviews and the second-level International Classification of Functioning, Disability, and Health (ICF) categories, a 39-item SSIPSCI was generated and pilot tested. It was then used with 284 inpatients with spinal cord injury. Principal components factor analysis and correlation analysis were performed. Results The exploratory components factor analysis revealed 7 significant stress factors: social activity, economy security, individual activity, relationship with others, physical disorders, bowel and bladder problems and medical fees. Internal consistency was high, with a Cronbach's alpha coefficient of 0. 950 overall and 0. 741 ~ 0. 920 for the individual factors. Test-retest reliability was 0.848. The correlation between SSIPSCI scores and Beck depression inventory (BDI) scores was strong ( r = 0. 665 ). Conclusions The reliability and validity of the SSIPSCI are both satisfactory enough to meet psychometric requirements.

Objective:To explore the in vitro anticancer effect of bleomycin (Ble) combined with cisplatin (DDP) on human gastric cancer cells.Methods:CCK-8 experiment was conducted to detect the effect of Ble and DDP alone or in combination on the proliferation of gastric cancer cells; Flow cytometry was used to detect changes in cell apoptosis and changes in reactive oxygen species. Cell scratch test was employed to detect cell migration; Western blot experiment was conducted to detect protein expression. Results:The results of CCK-8 showed that 20 μM DDP could inhibit the proliferation of six types of gastric cancer cells by 40%, while the effect of 20 μM Ble was only about 20%. When used in combination, the inhibitory rate on six types of gastric cancer could reach over 60%, with the strongest inhibitory effect on AGS cells. When 10 μM Ble and 10 μM DDP were combined, the proportion of AGS apoptotic cells reached 75.68%, higher than that of the two drugs treated with 20%, respectively 20 μM processing effect. When 10 μM Ble and 10 μM DDP were used in combination, the number of migrating and invading cells was significantly reduced, and the inhibition rate reached 50%. Compared with single use, the difference was statistically significant ( P<0.05). Flow cytometry results showed that the combination of Ble and DDP promoted cancer cell apoptosis by up-regulating the level of reactive oxygen species in cells; Western blot results showed that the expression of p-JAK2 and p-STAT3 decreased after Ble and DDP were combined, indicating that the JAK2/STAT3 signal pathway was inhibited. Conclusions:The combination of Ble and DDP can inhibit the proliferation and migration of gastric cancer cells, promote the apoptosis of gastric cancer cells, and play a synergistic anti-cancer effect. Its mechanism may be related to the up-regulation of ROS levels in cells, thereby inhibiting the activation of the JAK2/STAT3 pathway.

Objective:This study aimed to compare the audiological characteristics between children with unilateral auditory neuropathy (UAN) and single-sided deafness (SSD) to establish a valid basis for the differential diagnosis of children with UAN.Methods:A retrospective analysis was conducted on audiological and imaging evaluations of children with UAN and SSD who were treated at Beijing Children′s Hospital of Capital Medical University between May 2015 and June 2023. There were 17 children with UAN, comprising 10 males and 7 females, with an average age of 4.7 years. Additionally, there were 43 children with SSD, consisting of 27 males and 16 females, with an average age of 6.5 years. Audiological assessments included Auditory brainstem response (ABR), Steady-state auditory evoked potential (ASSR), Behavioural audiometry, Cochlear microphonic potential (CM), Distortino-product otoacoustic emission (DPOAE), and acoustic immittance test. The results of the audiological assessment and imaging phenotypic between the two groups of children were compared and analyzed by applying SPSS 27.0 statistical software.Results:(1) The UAN group (77.8%) had a significantly higher rate of ABR wave III L than the SSD group (20.9%) ( P<0.01). The PA thresholds at 500 Hz and 1 000 Hz of children with SSD were higher than those of children with UAN, while the ASSR thresholds at 500 Hz, 1000 Hz, 2 000 Hz, and 4 000 Hz of children with SSD were significantly higher than those of children with UAN ( P<0.05). (2) The degree of hearing loss in both UAN and SSD children was predominantly complete hearing loss. The percentage of complete hearing loss was significantly higher (χ2=4.353, P=0.037) in the SSD group (93.0%, 40/43) than in the UAN group (63.6%, 7/11). However, the percentage of profound hearing loss was significantly higher in the UAN group (27.3%, 3/11) than in the SSD group (2.3%, 1/43) ( Fisher′s exact test, P=0.023). In terms of hearing curve configuration, the percentage of flat type was significantly higher in the SSD group (76.7%, 33/43) than in the UAN group (36.4%, 4/11). The proportion of the UAN group (27.3%, 3/11) was significantly higher than that in the SSD group (2.3%, 1/43) in ascending type ( P<0.05). There were no statistically significant differences in the hearing curves of the declining type and other types between the two groups ( P>0.05). (3) The proportion of imaging assessment without abnormality was significantly more common in the UAN group (81.8%) than in the SSD group (37.1%) (χ2=6.695, P=0.015). Conclusions:Compared to children with SSD, the occurrence of wave III L on the ABR test was significantly more common in children with UAN. The percentage of ascending hearing curves was significantly higher in children with UAN than in children with SSD. ASSR thresholds were significantly lower in children with UAN. The normal imaging phenotype was significantly more common in children with UAN than in children with SSD.
