1.Effects of 5-aza-2'-deoxycytidine and trichostatin A on the methylation and expression of TFPI-2 gene in human pancreatic cancer Panc-1 cell line

Objective To investigate effects of 5-aza-2'-deoxycytidine(5-Aza-dC)and trichostatin A(TSA)on the methylation and expression of TFPI-2 gene in pancreatic cancer Panc-1 cells.Methods We treated pancreatic cancer Panc-1 cells with 5-Aza-dC and TSA alone or combined.TFPI 2 gene's DNA,mRNA and protein were determined by MSP,RT-PCR and Western blot.Results The hypermethylation of TFPI-2 gene in Panc-1 cells was reversed after treated with 5 Aza-dC alone or in combination with TSA.The re-introduction or raised expression of TFPI-2 mRNA and protein was detected in Panc-1 cells after treated with 5 Aza dC alone or in combination.However,we did not see the hypermethylation of TFPI 2 gene reversed nor detected TFPI 2 gene mRNA and protein after treated with TSA alone.Conclusions The methylation of the promoter region for the TFPI-2 gene is a main cause for its transcriptional activation in Panc-1 cells.The hypermethylation of TFPI-2 gene in Panc-1 cell was reversed and the expression of TFPI-2 gene mRNA and protein was detected after trea ted with 5-Aza-dC alone or in combination with TSA.Treatment of TSA alone does not have significant effect on TFPI-2 gene reexpression.
2.A new function of NK cells:adaptive or memory?

Recent reports have indicated that NK cells may have the function of immunological memory in common with cells of the adaptive immune system."Memory" NK cells move through the four phases of the adaptive immune response — proliferation,contraction,memory and recall which was similar to the T-cell responses."Memory" NK cells have special phenotypes.During MCMV infection,"memory" NK cells showed a greater expression of Ly49H,KLRG1,CD43 and Ly6C,and a decreased expression of CD27.In hapten (DNFB)-induced contact hypersensitivity model,the sorted liver NK cell subsets with the phenotype of Thy-1+Ly49C-I+ had the transferable memory activity,occupying around 10% of hepaedtic NK cells.These Ly49H+"memory" NK cells might produce more IFN-gamma ex vivo in response to plate-bound antibody against NK1.1 or Ly49H.Degranulation by Ly49H+ "memory" NK cells was also enhanced,as assessed by CD107a expression after ex vivo stimulation with anti-NK1.1."Memory" NK cells may move through the same migratory routes and adhesion pathways during the priming and effector phases of DNFB-primed Rag2-/-mice as conventional T cells of normal mice.Until now,it is not clear about the requirements for the triggering,regulation and maintenance of NK cell memory,but it is absolutely clear that "Memory" NK cells must play important role in host defense against pathogens.
3.Clinical application of osseointegrated prosthetic in reconstruction of auricular defects.

Auricular deformity, a common disease of the external ear in China, can be divided into congenital deformity and acquired deformity caused by trauma or ablative origin. Ear reconstruction with autogenous rib cartilage grafts provides consistent emotional relief and psychological benefits due to the repair. However, autogenous reconstruction is complex and challenging, and can be fraught with complications, in addition sometimes it may not be an option for the patients with poor skin cover, severe craniofacial anomalies, those with microtia who have had a failed previous reconstruction and microtia patients with a very low hairline and residual lobe. As an alternative to the significant surgical morbidity of autogenous reconstruction, osseointegrated implant reconstruction has several distinct advantages. This current essay was to illustrate the superiority, indications, feasibility and complications of osseointegrated implant reconstruction, and provide alternative way to help patient who was deprived of ear(s).

4.Advances in small-for-size syndrome

The small-for-size syndrome (SFSS) is widely recognized as one of the most serious clinical complications. It substantially contributes to a poor prognosis after adult living donor liver transplantation. Currently, there is still no consensus on the exact definition and pathogenesis of SFSS. We reviewed the progress on research of pathogenesis of SFSS and put forward some relevant preventive and treatment measures, including donor selection, graft assessment, reduction of high portal vein perfusion, dual grafts technology, advanced molecular medicine and other innovative approaches. Also, we offered some relevant insights into the future research directions of SFSS.

7.Double-catheter-balloon technique for embolization of high-flow carotid cavernous fistulas.

Objective To study the possibility and value of double-catheter-balloon technique for embolization of high-flow carotid cavernous fistula (CCF).Methods Three high-flow CCFs, which can't be completely occluded by 1 detachable balloon, were treated with the technique. By the way of bilateral femoral arteries approach, 2 detachable balloons were advanced to the internal carotid artery (ICA). After passing through the fistula, both balloons were inflated on the venous side in order to occlude the fistula. Results All CCFs were occluded completely and the ICAs remained patent. This procedure was easy and safe without increasing the risk of interventional therapy.Conclusions The application of double-catheter-balloon technique for high-flow CCF should be an effective method of choice during failure of 1 balloon procedure and simultaneously maintainning the patency of ICAs completely.
8.Auricular prostheses by osseointegration technique for auricular defect after trauma

OBJECTIVE To investigate the clinical results and feasibility of auricular prostheses by osseointegration technique for total auricular defect.METHODS Nineteen patients with traumatic total auricular defect who had underwent operation of bone-anchored auricular prosthesis were studied retrospectively.There were 12 cases with congenital microtia after an unsuccessful total auricular reconstruction,1 case with sulfuric acid burn,1 case with high temperature aluminum liquid burn,4 cases with traffic accident and 1 case with knife trauma.All the cases were followed-up for 3 to 5 years.RESULTS All the patients were cured after operation once.They all had vivid appearances with natural color and luster.The auricular prosthesis by osseointegration technique was stable and had no harm for daily life.CONCLUSION Auricular prosthesis by osseointegration technique is a convenient method with less trauma,short curing time,no complication and good result.It is a salvage treatment for patients after unsuccessful total auricular reconstruction.It is also a priority method for patients with scar formation after total auricular defects.
9.Assessment of left ventricular function in patients with type 2 diabetes mellitus by quantitative tissue velocity imaging

ObjectiveTo assess the left ventricular systolic and diastolic function in patients with type 2 diabetes mellitus(DM) using quantitative tissue velocity imaging(QTVI) ,and to provide reliable evidence for early diagnosis and prevention in diabetic cardiomyopathy.MethodsOne hundred and twenty one type 2 DM patients were divided into two DM 1 groups (61 patients without microangiopathy) and DM2 group (60 patientswith microangiopathy).Fifty normal subjects were enrolled as control group.The index of echocardiography,including the LVEF, FS and mitral peak flow velocity during early and late diastole (E/A) were measured by conventional echocardiography,and the ratio of E/A was calculated.The average peak velocities of six LV wall sites at mitral annuluses during systole, early and late diastole(MEm, MEm, MAm) were measured by QTVI, and the ratio of MEm/MAm was calculated.Results Compared with the normal group, MSm ([7.13 ± 1.42])cm/s vs.([6.49 ± 1.29]cm/s), MEm ([6.22 ± 1.39]cm/s) vs.([4.53 ± 0.94]cm/s) and MEm/MAm ([0.79 ±0.17]vs.[0.59 ± 0.19]) in DM patients were significantly decreased(F = 5.32,8.01 ,4.89 ; Ps <0.05).There was no significant differences among three groups in the comparisons of LVEF ([67.45 ±5.47]%),([65.91 ±4.83]%),([68.01 ±6.16]%) and FS([38.84±4.23]%,[37.82±5.43]%),([40.17 ± 4.53]%)(F = 1.89 and 2.46 respectively, P > 0.05) .In addition, E/A of DM2 group (0.71 ±0.21)decreased more dramatically than DM1 and normal (0.91 ± 0.18,1.02 ± 0.24)(F = 4.71, P < 0.05)ConclusionCompared with EF,FS and E/A obtained by conventional echocardiography,QTVI-derived MSm,MEm and MEm/MAm are more sensitive indexes to defect early LV dysfunction.The functional disorder appears early than microangiopathy, and the left ventricular systolic and diastolic function gets worse along with the microangiopathy.
10.Assessment of left ventricular hypertrophy and impaired diastolic function in elderly hypertensive patients by brain natriuretic peptide level in combination with tissue doppler imaging

Objective To investigate the clinical values of brain natriuretic peptide (BNP) in combination with TDI in diagnosing left ventricular hypertrophy (LVH) and impaired diastolic function in elderly hypertensive patients. Methods The 140 elderly hypertensive patients were divided into LVH group (n=69) and NLVH group (n=71). Control group consisted of 50 normal subjects. Plasma BNP level and index of echocardiography, including mitral peak flow velocity during early and late diastole (E, A), ratio of E/A, average peak velocities of six LV wall sites at mitral annuluses during early and late diastole (MEm, MAm), ratios of MEm/MAm and E/MEm were measured in all patients. The correlation of plasma BNP level with cardiac ultrasonographic findings was also examined. Results The level of BNP [(61.64±37.18)ng/L, (138. 65±30. 23)ng/L] and the ratio of E/MEm (11.3±1.83, 15.7±1.45) were significantly higher in NLVH group and LVH group than in normal group (P<0. 05 or P<0. 01). MEm [(6.32±0. 94)cm/s, (4.29±0. 91)cm/s]and MEm/MAm (0.76±0.19, 0.51±0. 11) were significantly lower in NLVH and LVH group than in normal group (P<0.05 or P<0. 01). The BNP level was negatively correlated with E/A, MEm and MEm/MAm (r=- 0. 294, r= 0. 387 and r= 0. 422, all P<0. 01), and was positively correlated with LVMI and E/MEm (r=0.342, r=0.501, all P<0.01). Conclusions Left ventricular diastolic function is impaired in elderly hypertension patients regardless of LVH or NLVH.Plasma BNP level in combination with echocardiography parameter is accurate to evaluate the LVHand impaired diastolic function in elderly hypertensive patients.
