1.Suppression of Delayed Type Hypersensitivity Response to DNP_6OVA in Mouse by UV --Irradiated Urocanic Acid

In order to investigate the effects of cis-UCA on cell -mediated immunity, we established a murine model of DTH response to DNP6OVA which is indicated by the increasing in the ear thickness of the mouse after resensitization, compared with those of the mouse injected with 200 g cis - UCA and trans -UCA intraperitoneally (i. p. ). The average of the increasing in ear thickness of each group was 9. 1 1. 13 10-2 mm (n=8, the number of ears) for the group administrated with cis-UCA; 17. 45 0. 88 10 - 2 mm (n= 8) for the group administrated with trans-UCA ; 16. 43 + 1. 83 10 - 2 mm(n= 7) for control group. It is suggested that there are immunosuppression of DTH response to DNP6OVA in mouse administrated with ets-UCA and no effecs of trans-UCA on this response.
2.CXCL12/CXCR4 and breast cancer

CXCL12 and its receptor CXCR4 can express in breast cancer,and are regulated by several factors in the genesis and development of breast cancer. Lots of researches have proved that CXCL12 and CXCR4 can be used as new independent prognostic makers of breast cancer.Treatment targeted for CXCR4 can be seen as a new kind of combination therapy,which may provides patients a more ideal treatment.
3.Situation survey of use of the antipsychotic in schizophrenic inpatients

Objective To investigate the present situation of use of the antipsychotic in schizophrenic inpatients,To find out factors of using the antipsychotic.Methods The prescriptions of schizophrenic inpatients was investigated in cross-sectional survey.Results The 47 remedy in 465 schizophrenic inpatients were investigated.The remedy of use of single antipsychotic had 175 patients(37.6%),the remedy of use of combined 2 kinds of antipsychotic had 262 patients(56.3%),the remedy of use of combination 3 kinds of drugs had 28 patients(6.0%),combined more than two drugs therapy was very common;Using rate and constituting rate of clozapine in prescription were No.1,non-classical antipsychotics was playing a leading role in the antipsychotic;Inpatients with public health services used non-classical antipsychotics more than inpatients of non-public health services.Conclusion Non-classical antipsychotics is replacing classical antipsychotics in curing schizophrenic.Paying way of patient influences the antipsychotic choice.
4.Pretreatment with a low dose of concanavalin A attenuated the development of concanavalin A-induced hepatitis

Objective:To investigate the influence of concanavalin A(Con A) pretreatment on the development of the fulminant murine hepatitis induced by an adequate dose of Con A.Methods:An adequate dose of Con A(15 ?g/g.body.wt) was injected intravenously into C57BL/6 mice to provoke severe liver damage,an extensively-used murine hepatitis model.A low dose of Con A(3 ?g/g.body.wt) was administrated intravenously 12 h before injection with an adequate dose of Con A.Liver injury was evaluated by serum transaminase assay and H&E staining.Hepatic lymphocytes were analyzed by flow cytometry and the absolute amount of lymphocytes per liver was calculated.Results:Pretreatment with a low dose of Con A significantly inhibited Con A-induced liver injury.Both the percentage and the absolute number of liver CD3~+T cells in Con A-pretreated mice were lower than those of PBS-pretreated mice.The activation of liver CD3~+T cells in Con A-pretreated mice was also prominently inhibited.Conclusion:Con A pretreatment exerted the negative effect on the development of Con A-induced hepatitis,which may be result from the decreased recruitment of T cells into the liver.The underlying mechanisms are under investigation.

Cao reported that melagenine extracted from human placenta was effective in the clinical treatment of vitiligo, to elucidate the possible mechanism involved, melagenine and placenta peptide were added to melanocytes in the culture, and the culture without melagenine as control. 6.Expression of CXCR4 in breast cancer and the relationship between CXCR4 and Hedgehog signaling pathway

Objective To detect the expressions of CXCR4,Smoothened (Smo) and Patched (Ptch) in breast cancer,to analyze the clinical significance of the expression of CXCR4,and to evaluate the relationships among CXCR4,Smo and Ptch.Methods The expressions of CXCR4,Smo and Ptch from 121 cases of breast cancer specimens were detected by immunohistochemistry,and the results were analyzed.Results The positive expression rate of CXCR4 in breast cancer was 66.9%,which was positively associated with the lymph node metastasis (r =0.181,P =0.044).The positive expression rates of Smo and Ptch in breast cancer were respectively 58.9% and 64.5%.The expressions of Smo and Ptch were positively associated with the expression of CXCR4 in breast cancer (r =0.189,P =0.036 ; r =0.230,P =0.010).Conclusion The expression of CXCR4 in breast cancer is associated with the lymph node metastasis,and it is positiviely associated with the expressions of Smo and Ptch.Patients with breast cancer who express both CXCR4 and Hedgehog signaling pathway may have a higher risk of recurrence.
7.Improvement of the turnaround times of STAT biochemistry test in clinical laboratory

Objective To shorten the stat test turnaround time (TAT) and improve the quality of service in clinical laboratory.Method Stat test TAT related data of clinical laboratory in Tongji hospital in Wuhan from August 1st to September 30th were collected by laboratory information system and hospital information system.EXCEL and SPSS were used for data analysis.Median and the 90th percentile were calculated for TATs from order to collection, collection to transfer, transfer to reception, reception to inspection and inspection to report.Outlier rates were calculated for TAT from collection to reception, reception to report, and collection to report using 2 h, 2 h, and 4h as target TAT value, respectively.Meanwhile descriptive statistics were calculated for TAT from order to collection in different clinical department, TAT from collection to reception during different collection time frames, and TAT from reception to report during different reception time frames.Results 32 235 stat biochemistry test data were included in this survey.Among three periods cut by collection and reception time, TAT from order to collection were the longest (P50: 681 min,P90:1261 min), followed by TAT from collection to reception(P50:94 min,P90:169 min) and TAT from reception to report(P50:68 min,P90:111 min).TATs from order to collection were longer in gynecological tumor department and organ transplantation department while shorter in infection department and cardiac vascular department.The TATs from collection to reception of specimen collected during 2:00 to 3:59 were longer than other collection time.While the TATs from reception to report of specimen received during 6:00 to 7:29 were longer than other reception time.There was no significant correlation between the amount of emergency specimens collected and TAT from collection to reception in different collection time period.However, the amount of emergency specimens collected was significantly correlated with the TAT from reception to report in different reception time period.Conclusions Analysis of TAT data can be used to identify existing problems and provide improved directions to shorten TAT in clinical laboratories.
8.Preliminary observations of differentially expressed genes in cholangiocarcinoma

Objective To analyze differentially expressed genes between hilar and distal cholangiocarcinoma and to clarify the molecular mechanism of tumorigenesis. Methods Gene-expression profiles of 3 samples of hilar cholangiocarcinoma and 4 samples of distal cholangiocarcinoma were analyzed using oligo microarray containing 21 329 genes. The differentially expressed genes between the two groups were analyzed using the Significance Analysis of Microarrays (SAM) in order to get the specifically differentially expressed genes. The gene expression presence was verified by real-time PCR (RT-PCR). Results A total of 725 genes of cholangiocarcinoma was regulated significantly compared to normal bile duct. Of them, 244 genes were upregulated and 399 genes were downregualted in both groups; on the other hand, 82 gene expressions between hilar and distal cholangiocarcinoma had significant differences (ratio ≥ 2.0 or ≤ 0.5). The SAM analysis showed that 40 genes, including AREG, EPHA2, SPP1, PACE4, and so on, were identified as differentially regulated between the two groups (q=0). Conclusions These data are helpful for a better understanding of the tumorigenesis of cholangiocarcinoma and contribute to the development of diagnostic and therapeutic strategies. The gene expressions between hilar and distal cholangiocarcinoma are significantly different, which suggests that the two tumors have different molecular mechanisms of tumorigenesis.
10.Long term outcomes of antebrachial flap with a pedicle of the posterior interosseous artery

Objective To analyze the long term outcomes of antebrachial flap with a pedicle of the posterior interosseous artery Methods Ninety cases were followed up for 1~16 years, the indication and long term outcomes and dorsal aspect of the forearm were analyzed and evaluated retrospectively Results Eighty nine cases survived, except one case, the conteur of forearm and hand were satisfied Conclusion Antebrachial flap with a pedicle of the posterior interosseous artery is an optimal method to repair soft tissue defects of hand.
