1.Clinical Study of Cyclosporin A for Psoriasis in China.

BACKGROUND: The efficacy of Cyclosporin A(CyA) in the treatment of psoriasis is well recongized, the dosage less than 5mg/kg per day(mg/kg/d) is recommended. OBJECTIVE: The purpose of this study is to evaluate the indication, dose, course and side-effects of CyA therapy for psoriasis, especially in China. METHODS: Forty patients with psoriasis were treated with doses of CyA 5mg/kg/d, compared with 20 patients treated with Tigason lmg/kg/d by Global PAST. RESULTS: 1. Of 40 patients 33 patients (82.5%)were cured clinically in 4-5weeks and all the patients(100%) had responses to the therapy. 2. The efficacy and the Global PASI reduction of CyA therapy were significantly better than that of Tigason therapy. 3. During the CyA therapy, no severe side-effects and obvious laboratory changes were found. CONCLUSION: CyA(5mg/kg/d) was satisfactory for psoriasis, especially for psoriasis pustulosa, arthropathica and erythroderma. CyA was found to be superior to Tigason. The therapy course and side-effects seemed to be related to the dosage.

2.Correlation between G919A polymorphism of cystathionine β synthase and breast cancer in Han nationality in Xinjiang

Objective To investigate the relationship between breast cancer and G919A polymorphisms of CBS (cystathionine β synthase) in patients with breast cancer in Han nationality in Xinjiang. Method Three hundred breast cancer patients and 300 normal controls of Han nationality were genotyped by ARMS method, and then the relative biochemical indicators were detected by the automatic biochemical analyzer. Results The Hcy level of breast cancer patients were higher than that of control group (P < 0.05). Genotypes GG, GA, AA of CBS G919A and allele G,A, in breast cancer patients were 63.7%, 24%, 12.3% and 75.7%, 24.3% respectively, which had no statistically significance when compared with those of the controls (P > 0.05). Conclusion The high level of Hcy may be a risk factor for breast cancer in Han nationality in Xinjiang and there was no relationship between CBS G919A polymorphism and breast cancer.
3.Association of serum visceral adipose tissue-derived serine protease inhibitor levels with carotid atherosclerosis in elderly patients with type 2 diabetes

The serum levels of visceral adipose tissue-derived serine protease inhibitor (vaspin)were measured in 65 elderly type 2 diabetic patients with carotid atherosclerosis( T2DM + CA group), 63 elderly type 2 diabetic patients without carotid atherosclerosis (T2DM group ) and 55 normal elderly individuals( NC group)by enzyme-linked immunosorbent assay. The results showed that the serum vaspin levels in T2DM + CA group were significantly lower than those in T2DM group[(0. 23 ± 0. 13 ) μg/L vs.(0. 41±0. 16)μg/L, P <0. 01]. The serum vaspin levels in T2DM + CA group and T2DM group were significantly lower than those in NC group respectively[(0. 63 ± 0. 11 ) μg/L, P < 0. 01 and P < 0. 05].Logistic regression analysis showed that high sensitivety C-reactive protein and Homeostasis Model Assessment insulin resistance index (HOMA-IR) were negatively correlated with serum vaspin levels,which indicates that lower serum vaspin levels might be associated with the pathogenesis of carotid atherosclerosis in elderly type 2 diabetic patients.
4.Regeneration of rat sciatic nerve inhanced by chemical acellular xenogeneous nerve grafts:a functional evaluation

[Objective]To observe the nerve regeneration and functional recovery in the adult rats.[Method]The sciatic nerves on the left side of 15 rats were exposed and 1.0 cm long segments of the nerves were removed from the mid-thigh level and replaced by rabbit nerve made acellular through chemical extraction.At 4 months after procedure,the nerve regeneration and function recovery were examined with HE staining,NF-160 immunohistochemical staining,electrophysiological tests and sciatic functional index(SFI).[Result]In the rats repaired by acellular nerves,regenerated axons re-entered into the acellular xeno-nerve segments without excessive sign of inflammation following implantation.As stimuling(1.5 mA,0.1 ms,1.0 Hz) to the proximal sciatic nerves,the implanted segment resulted in motor evoked potentials,which were recorded from posterior tibial muscles.SFI showed a partial recovery of locomotion of the limb with sciatic nerve defect.[Conclusion]Sciatic nerves defect can be repaired by chemical acellular xenogeneous nerves in rats.There was partial functional recovery,which revealed the grafting with chemical acellular xenogeneous nerves may be a promising mehod for nerve defect in clinical.
5.Establishment of detecting methods of Th1/Th2 cytokine gene

Objective: TO establish methods for sensitive, rapid, economical detection of Th1/Th2 Cytokine gene expression. Method: sixpairs of Primers specific for the IL-2, IL-4, IL-10, IL-13, IFNr and B-actin were designed. A Sensitive and rapid method for detection of Th1/Th2cytokine genes by RT-RCR was established. Meanwhile another method: the RNA hybridization with sir cDNA probes were established. Results:The RT-PCR reaction and RNA hybridization was used to test five kinds of Cytokine mRNAs expressed 20 tumor cell lines on mRNAs were expressed in eleven Cell lines;The mRNAs in eight cell lines. Conclusion:The preliminary results suggested that RT-PCR and RNA hybridizationnight be used to test Th1/Th2 cytokine gene expression.
6.The gene expression of cytokines in human tumor cell lines and specimens

Objective:To study the role of IL-2,IL-4,IL-6,IL-10,IL-13 and IFN? in pathogenesis of human tumor.Methods:The gene expression of 6 cytokines in human tumor cell lines and specimens were detected by RT-PCR or RNA dot hybridization.Results: There were predominant expression of Th2 type cytokines in 183 different human tumor cell lines and specimens.The positive rate of IL-4,IL-6,IL-10,IL-13 .IFN? and IL-2 in 183 tumor cell lines and specimens were 65%,70.5%,83.6%,61.7%, 12.6% and 22.9%,respectively.Conclusion: Predominant expression of Th2 type cytokines play an important role in the pathogenesis and development of tumor, may be related to the evasion of tumor cells from immune surveillance.
7.Pretreatment with a low dose of concanavalin A attenuated the development of concanavalin A-induced hepatitis

Objective:To investigate the influence of concanavalin A(Con A) pretreatment on the development of the fulminant murine hepatitis induced by an adequate dose of Con A.Methods:An adequate dose of Con A(15 ?g/g.body.wt) was injected intravenously into C57BL/6 mice to provoke severe liver damage,an extensively-used murine hepatitis model.A low dose of Con A(3 ?g/g.body.wt) was administrated intravenously 12 h before injection with an adequate dose of Con A.Liver injury was evaluated by serum transaminase assay and H&E staining.Hepatic lymphocytes were analyzed by flow cytometry and the absolute amount of lymphocytes per liver was calculated.Results:Pretreatment with a low dose of Con A significantly inhibited Con A-induced liver injury.Both the percentage and the absolute number of liver CD3~+T cells in Con A-pretreated mice were lower than those of PBS-pretreated mice.The activation of liver CD3~+T cells in Con A-pretreated mice was also prominently inhibited.Conclusion:Con A pretreatment exerted the negative effect on the development of Con A-induced hepatitis,which may be result from the decreased recruitment of T cells into the liver.The underlying mechanisms are under investigation.

AIM: To investigate the phenomenon that miR-let-7d regulates the proliferation and invasion abilities of the lung cancer cells through nuclear receptor peroxisome proliferator-activated receptors γ (PPARγ).METHODS: The relation between PPARγ and microRNA was analyzed by bioinformatics.The plasmid reporter assay was used to verify that PPARγ was the target of miR-let-7d.The lung cancer cell line with low expression of PPARγ was selected from different lung cancer cell lines by Western blot.The regulatory role of miR-let-7d in the lung cancer cells was determined by dual luciferase labeling and Western blot.The effect of miR-let-7d on the proliferation ability of lung cancer cells was detected by colony formation assay, the effect of miR-let-7d on the invasive ability of lung cancer cells was detected by Transwell invasion assay.RESULTS: The results of bioinformatic analysis showed that miR-let-7d regulated the expression of PPARγ, and the 3'UTR of PPARγ contained 2 functional miR-let-7d binding sites, indicating that PPARγ is a direct target of miR-let-7d.miR-let-7d was able to directly regulate the expression of PPARγ at mRNA and protein levels.Transfection of miR-let-7d inhibitor promoted the proliferation and invasion abilities of lung cancer cells by increasing the expression of PPARγ.CONCLUSION: miR-let-7d increases the expression of tumor suppressor PPARγ to inhibit the proliferation and invasive abilities of lung cancer cells.
