1.The experimental study of ideal resuscitation pressure for different ages of rats in uncontrolled hemorrhagic shock model

Objective To explore the ideal resuscitation pressure for uncontrolled hemorrhagic shock in different ages of rats.Methods Ninety-six rats were divided into 3 groups by ages (6 weeks,16 weeks and 12 months representing pre-adult,adult and elderly rats,respectively).Uncontrolled hemorrhagic-shock model was established in all rats.Then every group was divided into 4 groups by the different target pressures (untreated group,40 ~ 50 mmHg group,> 50 ~ 60 mmHg group,and > 60 ~ 70 mmHg group).We recorded the survival time and 24 h survival rate of rats in each group and compared the blood gas analysis,blood lactate and hematocrit (Hct) level in each group at 90 min of establishing model.Results Different ages had different optimal recovery pressure.The optimal target resuscitation pressures for 6 weeks,16 weeks and 12 months rats were 40 ~50 mmHg,> 50 ~ 60 mmHg and > 60 ~ 70 mmHg,respectively.Under the ideal pressure they had a better survival rate and lower blood lactate level (P < 0.01 or P < 0.05).Conclusious Hemorrhagic-shock rats at different ages have different ideal target resuscitation pressures during active hemorrhage.

3.Study on the relationship between children's functional gastrointestinal disease and food allergy

Objective To study the relationship between the functional gastrointestinal disease of children and food allergies,and recent prognosis.Methods 108 children with functional gastrointestinal disease were selected as the observation group,at the same time 36 healthy cases were selected as control group.ELISA was used to detect IgG antibody concentration and total IgE level in ten kinds of allergy food.The different methods were used to guide the patients' diet,the improved quality were oberseved in the two groups after 3 ~6 months.Results In the observation group 108 cases of children with functional gastrointestinal disease,1 ~ 10 kinds of inequality of food allergen specific IgG children have increased in 90 cases,accounting for 83.3%,whereas the control group had 9 cases,only accounting for 25.0% (x2 =20.87,P < 0.05).In the observation group,the foods with highest sensitivity were milk,egg white/yellow,respectively for 84 patients (77.8%) and 60 cases (55.6%).Serum total IgE level between the observation group and control group had no significant difference (P > 0.05).17 cases (15.7%) within one month,the symptoms were improved,13 cases (12.1%) in three months,the symptoms were improved,the symptoms of 21 cases (19.4%) were not improved after 3 months.The total effective rate was 80.6%.Conclusions Children with functional gastrointestinal decease is easily to occur allergy caused by different food,and IgG antibody level can correctly reflect the relationship between them,reasonable diet can improve the symptoms of functional gastrointestinal disease.

5.Expression of mRNA for isoforms of GABA_B receptor in the spinal cord in a rat model of chronic neuropathic pain

Objective To investigate the effect of GABAB receptor in spinal cord by detecting semi-quantitatively the mRNA for isoforms of GABAB receptor in a chronic neuropathic pain model. Methods Twenty-four male SD rats weighing 200-250g were anesthetized with intraperitoneal pentobarbital. Lamina of L5 vertebra and superior articular process of L6 were removed and posterior root of left L5 spinal nerve was exposed and tightly ligated with 7/0 atraumatic suture. The animals were randomly divided into 4 groups of 6 animals in each group: group A in which animals were observed for 7 days after sham operation; group B in which animals were observed for 7 days after ligation of posterior root of L5 spinal nerve; in group C animals were observed for 14 days after sham operation and in group D animals were observed for 14 days after ligation. Pain threshold was measured by heat stimulation of bilateral hindlimbs before and on the 1st , 3rd , 7th , 10th and 14th day after operation. Lumbar spinal cord was removed on the 7th or 14th day after operation and mRNA for isoforms of GABAB receptor in the spinal cord was detected semi-quantitatively by digoxin hybridization in situ.Results Pain threshold of left hindlimb (ligated side) was significantly lower than that of right hindlimb on the 7th, 10th and 14th day after operation as measured by heat stimulation. There were less mRNA for three isoforms of GABAB receptor in group B and D as compared with group A and C respectively. Conclusion The gene expression of three isoforms of GABAB receptor in the spinal cord has plasticity and is reduced during chronic neuropathic pain.
6.The biological characteristics of FGF23 and its relationship with metabolic diseases

Fibroblast growth factor 23 (FGF23) is a new endocrine factor, mainly expressed in osteoblasts and osteoblasts. Many studies have found that FGF23 can act on kidney, parathyroid and other tissues after binding with Klotho protein, and participate in bone mineral metabolism. In addition, in recent years, the role of FGF23 in exoskeleton has been gradually discovered, such as FGF23 and thyroid diseases, diabetes and so on. Therefore, understanding the biological characteristics, regulatory mechanism of FGF23 and its relationship with related diseases is of great clinical significance for the diagnosis and treatment of diseases. This article reviews the relationship between FGF23 and metabolic diseases such as phosphorus metabolic disease, parathyroid disease, hyperthyroidism, osteoporosis, diabetes, iron metabolism and so on.
7.CXCR4 gene expression promotes K562 cells chemotaxis

Objective To construct the tranfected cell expressing the human CXCR4 gene and to identify the effect on its immigration. Methods Total RNA was isolated from peripheral blood monouclear cell (PBMC),the full-length CXCR4 gene was amplified by RT-PCR and was inserted into plasmids PBudCE4.1 which have two promtors, after the identification by digestion and sequencing,the recombinant was transfected into K562 cell by lipofectamineTM2000. After screening culture by zeocin, stable transfected K562 cell line was established, and transcription and exression of CXCR4 were checked by flow cytometry; the chemotactic activity of K562 cell transfected and untrandfected CXCR4 was analyzed by Transell plate. Results The eukaryotic expression plasmid PBudCE4.1/ CXCR4 was constructed successfully. The stable trasfected K562/CXCR4 cell lines which highly express CXCR4 was established,the chemotactic activity of K562/CXCR4 was increased significiantly than K562. ConclusionCXCR4 transfected K562 cell line was successfully established, and it can make the basis for the further research on mechanism of extramedullary infiltration in leukemia.
8.Comparison of 18F-FDG PET/CT and contrast enhanced ultrasound for detection of hepatocellular carcinoma recurrence after therapy

Objective To compare the efficiency of 18F-FDG PET/CT and contrast enhanced ultrasound (CEUS) in detection of HCC recurrence after surgery or radiofrequency ablation (RFA).Methods Twenty-seven HCC patients (22 males,5 females; average age:47 years) were included in this study,of which 15 patients underwent surgery and 12 patients underwent RFA therapy.18F-FDG PET/CT and CEUS were performed within 2 weeks in all patients to detect recurrence.Final diagnosis was made by pathology or clinical follow-up (>6 months),and the sensitivity,specificity and accuracy of 18F-FDG PET/CT and CEUS were calculated and compared.McNemar test was used for data analysis.Results Twenty-five patients were diagnosed with HCC recurrence,including 11 cases with extrahepatic metastases.Another 2 patients had extrahepatic metastases without HCC recurrences.The sensitivity,specificity and accuracy in detection of liver lesion with 18F-FDG PET/CT were 92.0% (23/25),2/2 and 92.6% (25/27),respectively.While with CEUS,the corresponding values were 60.0% (15/25),2/2 and 63.0% (17/27),respectively.In CEUS positive group,the sensitivity and accuracy were 100%(15/15) for both methods.In CEUS negative group,the sensitivity,specificity and accuracy of 18 F-FDG PET/CT in detection of liver lesion were 8/10,2/2 and 83.3%(10/12),respectively.The sensitivity and accuracy of 18F-FDG PET/CT were significantly higher than those of CEUS (0,16.7%(2/12) ; x2 =5.373,5.250,both P<0.05).Moreover,the sensitivity of 18 F-FDG PET/CT in detecting extrahepatic metastasis was 100% (13/13).Conclusions 18 F-FDG PET/CT is more sensitive and accurate than CEUS (especially in CEUS negative cases) in detecting recurrence of HCC after surgery or RFA.18F-FDG PET/CT also could be useful for detection of extrahepatic metastasis.

Objective To evaluate the measurement uncertainty for the determination of ethanol in human blood by auto-headspace GC/MS.Methods Each source of uncertainty,arising from the procedure of testing,was analyzed and confirmed according to the guidelines of the uncertainty in measurement.After each uncertainty component was evaluated,the combined standard uncertainty and the expanded uncertainty of the result were calculated.Results The expanded uncertainty was 0.084mg/mL when the concentration of ethanol in blood sample was 0.738 mg/mL.Conclusion The measurement uncertainty of the concentration of ethanol was came primarily from the sample determination,standard solution of the ethanol and the calibration curve.

SDF-1α, a ligand for the chemokine receptor CXCR4, is well known for mediating the migration of breast cancer cells. In a previous study we demonstrated that a synthetic 21-mer peptide antagonist of CXCR4 (NT21MP) derived from the viral macrophage inflammatory protein II could antagonize tumor growth in vivo by inhibiting cellular proliferation and inducing apoptosis in breast cancer cells. However, the role of SDF-1α in the signaling pathways underlying the proliferation of human breast cancer cells and associated signaling pathways and inhibiting signal pathways of NT21MP remained unclear. The present study investigated the mechanism of NT21MP on anti-tumor in breast cancer in vitro. The effect of NT21MP on the viability of cells was determined by the MTT assay. Annexin V-FITC and PI staining was performed to detect early stage apoptosis in SKBR3 cells treated with SDF-1α and AMD3100 or NT21MP. Western blotting techniques were used to assay the composition of phosphoproteomics and total proteins present in the SKBR3 breast cancer cells. RT-PCR and Western blotting technique were used to detect the effect of NT21MP and AMD3100 on Bcl-2 and Bax expression. The results indicated that SDF-1α prevented apoptosis and promoted the proliferation of SKBR3 human breast cancer cells. As compared with untreated SKBR3 cells, Treatment with SDF-1α significantly increased cell viability, and NT21MP abolished the protective effects of SDF-1α dose-dependently (P<0.05). There was a significant decrease in the percentage of apoptotic cells after SDF-1α treatment as compared with control group (2.7%±0.2% vs. 5.7%±0.4%, P<0.05). But pretreatment of SKBR3 cells with NT21MP significantly attenuated the antiapoptotic effects of SDF-1α as compared with SKBR3 cells without NT21MP pretreatment. The proliferative and anti-apoptotic effects of SDF-1α in SKBR3 cells were associated with an increase in AKT and ERK1/2 phosphorylation as well as a decrease in Bax expression and an increase in Bcl-2 expression. These changes in intracellular processes were blocked by NT21MP in a dose-dependent manner(P<0.05). In conclusion, NT21MP efficiently inhibits SDF-1α-induced proliferation and antiapoptosis in SKBR3 cells by reducing the levels of phosphorylated AKT and ERK1/2, as well as decreasing the ratio of expression of Bcl-2 relative to Bax.
