Objective To investigate whether the tyrosine phosphatase SHP-2 participate in the cell migration induced by IL-1β.Methods Two different reorganized plasmids transfected into the MCF-7,and achieved two different cell lines:SHP-2-GFP-MCF-7 cell lines.SHP-2C>S-GFP-MCF-7 cell lines.The cell migration was detected by a fluorescence microscope and the expression of E-eadhefin and MMP-9 wee detected by western blotting analysis and RT-PCR.Results The SHP-2-GFP-MCF-7 cell lines showed a higher cell migration ability,and the expression of E-cadherin in the SHP-2-GFP-MCF-7 cell lines was especially lower,neady undetectable.But in this group the expression of MMP-9 was the highest.Conclusion The tyrosine phosphatase SHP-2 participate in the cell migration induced by IL-1β,and maybe produce a marked effect by reinforcing the expression of MMP-9 and reducing the expression of E-cadherin.
2.Analysis of characteristics of cognitive function of elderly patients after total knee arthroplasty

Objective To study the influencing factors of postoperative cognitive dysfunction ( POCD) in elderly patients after knee joint replacement. Methods 165 cases of elderly patients undergo-ing knee replacement surgery were recruited in the study. According to whether there was POCD,the patients were divided into POCD group (35 cases) and non POCD group (130 cases).The gender,age,educational level,medical history,physical exercise,operation mode( cruciate-retaining total knee arthroplasty and poste-rior stabilized total knee arthroplasty) ,operation time,blood loss,preoperative depression score and postoper-ative pain score were recorded and compared.The logistic multiveariate regression analysis was conducted to analyze the risk factors of POCD in elderly patients with knee replacement. Results (1)There were statis-tically significant differences in the total score of MoCA, visual space and executive function,naming and de-laying recall between non-POCD group and POCD group after the operation ( Z=-5. 327, P=0. 000;Z=-3.140, P=0.002;Z=-2.910, P=0.004;Z=-3.333, P=0.001). (2)The results of logistic multiveariate regression analysis showed the age>70 years old ( OR=3.708,95%CI =1.037-13.261, P<0.05) ,hyperten-sion ( OR=5.250,95%CI =1.787-15.423, P<0.05),diabetes ( OR=6.560,95%CI =1.418-30.345, P<0.05) ,preoperative depression ( OR=4.648,95%CI =1.290-16.744, P<0.05) and postoperative pain score>4 ( OR=3.149,95%CI=1.072~9.251, P<0.05) were risk factors of POCD,and physical exercise ( OR=4.330,95%CI =1.294-14.488, P<0.05) and CR-TKA( OR=0.187,95%CI =0.045-0.781, P<0.05) were the protective factors of POCD. Conclusion The overall level of cognitive function in elderly patients after knee replacement surgery is low. The advanced age,hypertension,diabetes,lack of physical exercise,choice of operation method,postoperative pain and preoperative depression are important influence factors of POCD.

4.Vein Scanning Projection Instrument Based on Two-Dimensional Scanning Mirror.

With the development of science and technology, new medical equipments is toward the direction of intelligent and portable. In order to assist medical personnel to patients with blood, developing from previous devices, a new kind of vein locating projection instrument based on two-dimensional scanning mirror is put forward. It can scan and project vein image using a scanning mirror. The related algorithm is also be improved, make vein scan projection more practical. The system finally set up can perform well in vein scan projection.

5.The application of feedback debriefing in simulation-based clinical training

Objective To evaluate the educational effectiveness of feedback debriefing in clinical training.Methods Forty medical students and fifteen teachers were enrolled in the 2015 after-internship clinical training,which was newly designed to obtain the debriefing part.The test scores and single technical practices were collected and analyzed,before and after the training.DASH scoring system was used to evaluate the effect of debriefing in the team simulation.Results were statistically analyzed by SAS 9.2(SAS institute Inc).Quantitative data were described by x ± s,while qualitative data were described by number and/or constituent ratio.Student's t test and Wilcoxon rank sum test were applied as needed.A P-value < 0.05 was considered to be significant.Results The scores of the students before and after the assessment were (41.88 ± 8.54) vs.(65.06 ± 13.83),and the difference was statistically significant(P=0.000).Individual skills before and after the course had a different degree of improvement.The average DASH-scores evaluated by the students,teachers and supervisors were (39.3 ± 2.12) (very good),(35.1 ± 4.18) (good) and (37.2 ± 3.03)(very good),separately.Conclusions Potent debriefing helps to improve the effectiveness of simulationbased clinical training.
6.Inhibitory effect of methylmercury on proliferation of human small cell lung cancer cells

Objective:To investigate the inhibition of methylmercury on proliferation of human small cell lung cancer NCI-H446 and to compare with As_2O_3 and cisp latin.Methods:MTT chromatometry was adopted.Cultured NCI-H446 cells were incubated with different doses of methylmercury,As_2O_3,or cisplatin at 2.5,5,10,20,40 μmol/L.The optical density OD_(570) value were detected to calculate the inhibitory rate of proliferation.The nude mice model bearing tumor was constructed by subcutaneous injection with transplanting NCI-H446 cells.MMC was used at 10 mg/kg in administration with normal saline as the control.Tumor size was measured and tumor volume was calculated.Then the morphologic change was observed by HE stainning.Results:OD_(570) value of methylmercury groups were lower than that of the control groups at any doses and the difference was significant from 2.5 μmol/L group(P<0.05).The difference of the OD_(570) value of methylmercury groups under the same concentration among the different times were significant (P<0.05);OD_(570) value of As_2O_3 and cisplatin groups were also lower than that of the control groups.There was no significant difference with MMC and cisplatin.OD_(570) value of MMC and DDP groups were higher than that of As_2O_3 groups.The volume of the tumor of the experiment groups was smaller than that of normal saline group,and the necrocytosis was more than that of normal saline group by HE stainning.Conclusion:Methylmercury could inhibit the proliferation of human small cell lung cancer NCI-H446,and the effect is both time- and dose-dependence.At the same doses,this effect may be equal to cisplatin's and stronger than As_2O_3's.

Objective To investigate the expressions of Stat3, CyclinD1 and proliferating cell nuclear antigen (PCNA) in non-small cell lung cancer (NSCLC), and analyze their relationships with carcinogenesis and progression of NSCLC. Objective To investigate the clinical effects of the posterior atlantoaxial pedicle screw fixation combined with unilateral axial spinous process and lamina screws and allogeneic bone graft for atlantoaxial instability.Methods From March 2010 to April 2014,data of 10 patients with atlantoaxial instability who had undergone atlanto-axial vertebral pedicle fixation and interbody fusion combined with posterior atlanto-axial spines lamina nails were retrospectively analyzed.There were 6 males and 4 females with an average age of 39 (range,16-62) years old.The inclusion criteria was atlantoaxial instability together with unilateral vertebral artery segment high cross deformity (7 patients) or atlantoaxial joint break bad involving unilateral pedicle (7 patients).3 patients have spinal cord disease symptoms and physical signs,both suffered limited neck mobility and pain.The VAS scores were 1-8,with an average of 3.70±2.11.Preoperative X-ray,CT three-dimensional reconstruction and MRI were collected.X-ray and CT examination were performed 7 d and 1,3,6,12 months after operation to evaluate the internal fixation and bone grafting fusion.Results There's no occurrence of cervical spinal cord and vertebral artery injury.Screws were successfully implanted.1 case's incision occurred infiltration liquid 3 days after operation.For allograft rejection consideration,we changed the plan,and the infiltrating stopped 5 days later.Other incisions were all primary healing.VAS scores were 1.01±0.89 after operation,which was significantly reduced.X-ray showed good recovery of cervical sequence after the operation,and CT revealed 1 patient with atlas pedicle screw in the medial cortex damage.Spinal canal was without infringing,and the other screw positions were normal.6 months after operation,X-ray or CT examination both showed bony fusion.Conclusion Atlantoaxial pedicle screw fixation combined with unilateral axial spinous process and lamina screws and allograft bone graft fusion for the treatment of atlantoaxial instability can observe good clinical effects.
9.Role of topical tranexamic acid plus cocktail analgesic in reducing blood loss during total knee arthroplasty

Objective To investigate the effect and safety of topical tranexamic acid (TXA) plus cocktail analgesic for reducing blood loss during total knee arthroplasty (TKA).Methods A prospective case control study was made on 60 patients scheduled to undergo TKA because of knee injuries between August 2015 to June 2016.There were 13 males and 47 females,with the mean age of 65.5 years (range,51-80 years).Traumatic arthritis occurred in 44 patients and degenerative arthritis in 16 patients.The patients were assigned to separate cocktail analgesic group (Group A,n =30) and topical TXA plus cocktail analgesic group(Group B,n =30),according to the random number table.Patients in Group A received multiple-point intra-articular cocktail analgesic injection before implantation of the prosthesis in TKA.While patients in Group B received multiple-point intra-articular TXA plus cocktail analgesic injection before implantation of the prosthesis.Between-group differences were compared with respect to intraoperative blood loss,hemoglobin change (Hb),haematocrit (Hct),postoperative drainage,total blood loss,hidden blood loss,blood transfusion rate,Hospital for Special Surgery (HSS) score,incidence of deep venous thrombosis (DVT) and other complications.Results All patients were followed up for 3 months.Perioperative Hb reduction in Group B was 18.5 (13.0,26.0) g/L,less than 23.0 (21.0,35.5) g/L in Group A (P < 0.05).Hct was reduced by 5.6 (4.1,7.8) % in Group B,while 7.2 (6.1,10.7) % in Group A (P < 0.05).Postoperative drainage volume,total blood loss and occult blood loss in Group B were 105.0(60.0,223.8) ml,596.0(426.1,795.3) ml,422.3 (228.9,624.0) ml respectively,decreased compared to Group A [162.5 (118.8,245.0) ml,788.3 (583.0,1 082.4) ml,603.2 (435.2,884.7)ml respectively] (P <0.05).There were no significant differences in intraoperative blood loss,blood transfusion rate,HSS score and DVT incidence between the two groups (P >0.05).Conclusion Topical TXA plus cocktail analgesic can reduce blood loss during perioperative period in TKA,without increasing the risk of DVT.
10.Culture of Blastocystis hominis in vitro in Medium 199

Objective To establish the culture of Blastocystis hominis in vitro in Medium 199 for further research on the biological characteristics of this intestinal parasite.Methods The growth, reproduction and relevant factors of B.hominis under different culture conditions including pHs, sorts and concentrations of serum, and the number of inoculation were compared. Results The optimal conditions for the continuously anaerobic culture of B.hominis in Medium 199 were as follows: concentrations of new born bovine serum (or human serum and horse serum) ranged from 10%-20%, the number of inoculation was no less than 1?10~5 organisms per tube, pHs ranged from 7.0-8.0, penicillin, streptomycin should be added, anaerobic cultured at 37 ℃.The reproductive peaking-days were 4, 8 at pH 7.0; 3, 6, 9 at pH 7.5 and 2, 4, 7, 9 at pH 8.0. Conclusion B.hominis could be continuously cultured in vitro in Medium 199.
