BACKGROUND:Currently, bioactive glass and polylactic acid have been used in clinical dentistry and plastic surgery; however, their therapeutic outcomes are not satisfactory, because the material properties have some limitations. OBJECTIVE:To explore the cytocompatibility of oxygen plasma-treated polylactic acid and bioactive glass guided bone regeneration membrane. METHODS:Bioactive glass and polylactic acid were used as the basic materials to prepare polylactic acid membrane, polylactic acid and bioactive glass composite membrane and oxygen plasme-treated polylactic acid and bioactive glass composite membrane, al of which were used to culture MG63 cels. Cel adhesion rate, cel proliferation rate and alkaline phosphatase activity of MG63 cels on these three kinds of membranes were observed. RESULTS AND CONCLUSION: With the growth of time, in these three groups of membranes, the cel adhesion rate and cel proliferation rate were al significantly increased. Alkaline phosphatase activity showed a decreasing trend after the first increase, and reached its peak at the 7thday of culture. The cel adhesion rate and cel proliferation rate in oxygen plasma-treated polylactic acid and bioactive glass group were significantly higher than those in the other two groups, while the cel adhesion and proliferation rates in polylactic acid and polylactic acid and bioactive glass groups were similar. At the 3rd day of culture, the alkaline phosphatase activity in the polylactic acid and bioactive glass group and oxygen plasma-treated polylactic acid and bioactive glass group was significantly higher than that in the polylactic acid group. At the 7th and 14th days, there was no significant difference in the alkaline phosphatase activity among these three groups. These results show that oxygen plasma-treated polylactic acid and bioactive glass composite membrane has good biocompatibility, which can better promote cel adhesion, proliferation and matrix secretion from osteogenic cels.

Objective To estimate any influence of strong pulsed magnetic fields on the expression of growth-related genes in human bladder cancer BIU-87 cells. Methods Human BIU-87 cells were cultured in vitro and randomly divided into a magnetic field group and a control group. Each group was further divided into 24 h, 48 h and 72 h sub-groups. The magnetic field group cells were exposed to an 8 T magnetic field pulsed at 15 Hz for 2 h every day. The control group cells also placed on the same environment, but not exposed to any strong, pulsed magnetic field. The expression of B cell lymphoma/leukemia gene-2 (Bcl-2) mRNA, Bax mRNA and caspase-3 mRNA was measured with RT-PCR, and flow cytometry was used to evaluate the expression of the Bcl-2, Bax and caspase-3 genes of the tumor cells in vitro. Results The expression of Bax mRNA and protein was significantly higher in the cells exposed to the magnetic field than in the control groups. The expression of Bcl-2 mRNA and protein was significantly less. The expression of caspase-3 mRNA and protein in the two groups showed no significant differences. Conclusions A strong, pulsed magnetic field can inhibit the growth of bladder tumor BIU-87 cells and promote their apoptosis. The mechanism is probably related with the magnetic field promoting Bax mRNA and protein expression and inhibiting Bcl-2 mRNA and protein expression.

Objective To investigate the effects of heparanase and E-cadherin on the invasion and metastasis of gastric cancer. Methods Fifty specimens of gastric cancer which had been resected at Cancer Hospital of Liaoning Province from February 2005 to May 2007 were collected. The expression of heparanase mRNA and E-cadherin mRNA in these gastric cancer specimens was detected by RT-PCR, and the expression of E-cadherin in these gastric cancer specimens was detected by immunohistochemistry. Data were analyzed by t-test and variance analysis, and the enumeration data analyzed by chi-square test. Results There were significant differences in the expression of heparanase and E-cadherin between gastric cancer cells with high and low differentiation, presence and absence of metastasis, and TNM stages Ⅰ and Ⅱ versus Ⅲ and Ⅳ (t = 1.999, 4.258, 1.735 ; 1.286, 6.794, 3.091; χ~2 =6.273, 9.397, 5.640, P <0.05). The co-expression of heparanase (+) and E-cadherin (-) was correlated with tumor undifferentiation, lymph node metastasis and advanced TNM staging (χ~2 =11.306, 10.208, 8.420, P <0.05). Conclusions Heparanasc shows high expression while E-cadherin shows low expression in gastric cancer tissue. There is a synergistic effect between the abnormal expression of heparanase and E-cadherin, and the gastric cancer cells with coexpression of heparanase and E-cadherin have more malignant potential.
