The suppressive effects of cyclosporin A ( CsA ) made in China on immune function were studied in mice. CsA ( 25 and 50 mg/kg ig for 4 d ) caused a significant decrease of spleen plaque forming cell ( PFC ) in mice. At the dose of 25, 50 and 100 mg/kg ig for 4 d, CsA inhibited the production of hemolysin in a dose-dependent manner in mice immunized with SRBC. CsA ( 25, 50 and 100 mg/kg ig for 10d ) also markedly inhibited the delayed cutaneous hypersensitivity ( DCH ) induced by DNCB in mice. CsA 50 mg/kg ig for 14d prolongated survival time of cardiac grafts. The treatment with CsA could not reduce the clearance rate of iv charcoal particles as well as bone marrow cells in mice. There was no significant difference of immu-nosuppressive effect on PFC response between China-made CsA and Sandoz-made CsA

4.Serum concentrations of tumor markers in patients with chronic kidney diseases and the analysis of related factors

Objective Our study intended to check whether there are any changes of serum concentrations of tumor markers in patients with chronic kidney disease,and to determine the related factors.Methods Atotal of 232 hospitalized patients in Nephrology Department in Zhongshan Hospital of Fudan University from March to June in 2005 were divided into groups respectively according to their levels of Ccr,Upro and Salb.Then Kruskal-Wallis test was applied to confirm the relationships among Ccr,Upro,Salb and serum tumor markers.Furthermore,multielement logistic regression was used to analyze the independent effect of age,Ccr,effusion in serous cavity,the levels of proteinuria and serum albumin on the levels of these markers in CKD patients.Results The serum levels of CEA,CA 199,NSE and SCC in different Ccr groups,the levels of CA 199,CA125,NSE and SCC in different Salb groups,the levels of CA 125,NSE and SCC in different Upro groups,had significant statistical differences.Age was the risk factor of the increased levels of CEA and PSA;effusion in serous cavity was the risk factor of increased levels of CA 125.The decreased level of Ccr was the risk factor of CA 125 and SCC.The elevated Upro was the risk factor of SCC.The decreased Salb was the risk factor of CA 199,CA 125 and NSE.Conclusion When we diagnose some tumors according to their serum levels of tumor markers such as CEA,CA 199,CA 125,NSE,SCC and PSA,we must note that whether the patients are aged or have complications such as large proteinuria,hypoalbuminemia,effusion in serous cavity or decreased kidney function.

Objective To investigate the distribution and chemical character of nestin immunoreactive cells in the human basal forebrain. Methods We explored systematically the distribution of nestin immunoreactive cells throughout the human basal forebrain by nestin 331B, 10C2, Rat401 antibody.Furthermore, we investigated the chemical identity of these nestin immunoreactive cells by using double\|labeling immunocytochemistry with NSE, ChAT, p75NGFR, GFAP antibody or using NADPH\|d histochemistry. Results The nestin immunoreactive cells were found in the septum, diagonal band of Broca, innominate substance,amygdala,basal nucleus of Meynert of adult human brain.These nestin immunoreactive cells haven big cell body,2\|3 processes.The nestin immunoreactive cells were labeled by NSE antibody.The large majority of those were single stained,and 28% were double labeled with ChAT positive neurons,15% with NGFR positive neurons,6% with NOS positive neurons.A few nestin immunoreactive cells shared similar morphology with that of astrocyte glia which existed in the spaces between the thin septa pellucida or the midline along the septum.It could not be labeled by GFAP antibody.Conclusion\ A new cluster of nestin immunoreactive neurons that were different from the ChAT,NGFR,NOS positive neurons existed in adult human basal forebrain.\;[
7.Effect of sufentanil on analgesic effect and hemodynamics during recovery period of general anesthesia in elderly patients

Objective To study the influence of sufentanil on analgesic effect and hemodynamics during recovery period of general anesthesia in elderly patients.Methods Eighty patients with general anesthesia were randomly divided into 4 groups (20 cases in each group).Ten min before surgery ended patients in each group were intravenous injected different dose of sufentanil:group S1 with sufentanil 0.2 μg/kg,group S2 0.4 μg/kg,group S3 0.6 μg/kg,and group C (control group) with saline 5 ml.The time of spontaneous breath restores,the time of summon opening eyes and extubation,the cases occurred cough,nausea,vomiting during recovery period were recorded and the VRS pain scores and Ramsay score 10 min after extubation were also recorded.Mean arterial pressure (MAP) and the heart rate (HR) were recorded before the anesthesia induction(T0),stopped anesthetic (T1),pre-extubation (T2),5 min and 10 min after extubation (T3 or T4)Results As group S1 and S2 compared with group C,there was no significant difference on the time of spontaneous breath restores,the time of summon opening eyes,and the time of extubation and leaving the operating room (P > 0.05).However,there was significant difference on the time of spontaneous breath restores ((7.3 ± 4.1) min vs (10.2 ± 4.7) min),the time of summon opening eyes ((8.1 ± 3.1) min vs (11.2 ± 3.7)min),the time of extubation ((12.3 ± 3.1) min vs (16.3 ± 5.9) min),and the time of leaving the operating room((21.4 ±3.0)min vs (24.2 ±3.5)min) between the group S3 and group C(P<0.01 or P <0.05)Patients occurred cough in group S1,S2 and S3 were less than those in group C (P <0.05),but there was no significant difference on the occurrence of nausea and vomiting between group S1-3 and group C (P > 0.05)There was significant difference on VRS pain score and Ramsav score between group S1-3 and group C(P <0.01).MAP and HR were increased in all points of time as compared with T0 (P < 0.05 or P < 0.01).And there was significant difference on MAP and HR between group S1-3 and group C (P < 0.05 or P < 0.01).Conclusion Before the end of anesthesia sufentanil 0.2 ~ 0.4 μg/kg intravenous injection could be able to enhance analgesic effect,stabilize hemodynamics and do not affect regain consciousness in elderly patients.
8.Expression of nerve growth inhibiting factor Nogo-A in ischemic infarct brain in rats

AIM: To investigate the dynamic changes of Nogo-A expression in ischemic infarct brain in rats. METHODS: The model of middle cerebral artery occlusion (MCAO) in 80 rats was established and expression of Nogo-A mRNA was measured by immunohistochemistry and hybridization. RESULTS: In the brain of MCAO rats, Nogo-A mRNA expression was decreased at the third day and increased significantly at the 7th day, and reached high level at the 21th day, then remained the high level to the 28th day. Nogo-A protein expression showed the same results. CONCLUSION: Expression of Nogo-A did not change in the early stage of MCAO, but increased significantly in the late stage of MCAO, suggesting that Nogo-A expression may play an important role in the nerve regeneration of brain ischemic injury. [
9.Specific humoral immune responses in adult rhesus monkeys vac cinated with A?_(1-15) peptide vaccine

AIM: To observe the humoral immune response in adult rhesus monkey induced by A? 1-15 vaccine. METHODS: 5 adult male rhesus monkeys were injected intramuscularly with A? 1-15 vac cine at baseline and at week 2, 6, 10, 14, 18, 22. The titer and IgG isotypes of the antibody against A? 1-42 in the serum were measured with ELISA. The specificity of the antibody against A? 1-42 was determined by Wester n blotting. The A? plaques in Tg2576 transgenic mouse brain were stained with t he antisera using immunohistochemistry method. RESULTS: At the eighth week after the vaccination, antibody against A? 1-42 bega n to develop significantly i n the serum. The titers of the antibody increased following vaccine boosted and reached 1: 3 840 at the twenty-fourth week, then decreased after the terminat ion o f inocunation. The IgG1 was accounted for the highest level in the antisera pool . The antibody against A? 1-42 showed high specificity. The A? plaques in Tg2576 transgenic mouse brain were labeled with the antisera. CONCLUSION: A? 1-15 vacci ne could induce vigorously specific humoral immune responses in adult rhesus mon key.
10.Effects of Intensive Induction Chemotherapy on Consolidation Radiotherapy in Young Children with High-risk Hodgkin Lymphoma

OBJECTIVE:To investigate the safety of induction chemotherapy and the implementation method of consolidation radiotherapy in young children with high-risk Hodgkin lymphoma(HL),by evaluating the effects of intensive induction chemother-apy on consolidation radiotherapy. METHODS:Six pediatric patients with high-risk HL received low-dose involved field radiation therapy (IFRT) via volumetric modulated arc therapy (VMAT) following 6 cycles of intensive chemotherapy [Ara-C/VP16 (cyto-sine arabinoside+etoposide),ABVE-PC(adriamycin+bleomycin+leurocristine+etoposide+metacortandracin+cyclophosphamide)and CHOP(cyclophosphamide+leurocristine+adriamycin+ prednisone)in turn]. Therapeutic efficacy and toxic effects were evaluated af-ter treatment. RESULTS:Intensive induction chemotherapy and consolidation radiotherapy were acceptable by young children,and mild acute or subacute toxic reaction were observed. Intensive induction chemotherapy didn’t affect toxic effects of consolidation ra-diotherapy. In this regimen,the cumulative doses of bleomycin and adriamycin were 20 mg/m2 and 270 mg/m2,respectively. VMAT optimized plan to ensure that the dose that involved organs received was safe. In the patient with mediastional radiation therapy,the mean lung and heart doses were 525.6 cGy and 503.9 cGy,respectively. CONCLUSIONS:It is safe to give high-risk HL young children 6 cycles of intensive induction chemotherapy(Ara-C/VP16,ABVE-PC and CHOP in turn)before radiotherapy. It is feasi-ble to conduct IFRT with 18-20 Gy and an additional 20-25 Gy boost,1.5-1.8 Gy/fraction. VMAT deserves to be advised.
