1.Studies on the relationship of apoptosis and multidrug resistance in 5-fluorouracil resistant human hepatocellular carcinoma cell line Bel7402/5-FU

AIM To elucidate whether apoptosis is involved in the development of multidrug resistance in 5-fluorouracil resistant cells Bel7402/5-FU. METHODS Multidrug resistance of Bel7402/5-FU cells was determined by SRB assay, apoptosis was measured by Annexin V-FITC/PI double labeled assay, cell cycle phases were analysed by flow cytometry, and expression of genes related to apoptosis bcl-2, bcl-xl, bcl-xs, bax, fas, p53, and cpp32 was detected by RT-PCR assay. RESULTS Bel7402/5-FU cells were resistant not only to 5-FU but also to multiple anticancer drugs. When treated with the same concentration of 5-FU (30 ?mol?L -1 or 150 ?mol?L -1 ), the proportion of apoptosis was significantly increased and G_0/G_1 phase was increased and S and G_2/M phases were reduced in Bel7402 cells, but the proportion of apoptosis and cell cycle was not changed in Bel7402/5-FU cells. The expression of bcl-xl, bcl-xs, and bax mRNA were significantly increased and the expression of p53 and cpp32 mRNA were significantly decreased in resistant Bel7402/5-FU cells, as compared with Bel7402 cells. CONCLUSION Decrease of apoptosis may be related to multidrug resistance of human hepatocellular carcinoma cells Bel7402/5-FU. The up-regulation of anti-apoptotic genes and down-regulation of pro-apoptotic genes are involved in the development of multidrug resistance in Bel7402/5-FU cells.
2.Influence of different storage ways on the testing results of AD7C-NTP in urine specimen

Objective To explore the influence of different preservation conditions and time period on the testing results of Alzheimer's disease(AD) associated neuronal thread protein(AD7C-NTP) in urine specimen.Methods From Oct.2015 to Jan.2016,urine specimen were collected from 50 AD patients,and divided into three groups,according to the different storage temperature,including room temperature group,4 ℃ group and-20 ℃ group.Preservatives were added into specimen of 4 ℃ preservation group and-20 ℃ preservation group.AD7C-NTP level was detected at different preservation time of all specimen.Results The testing results of AD7C-NTP in specimen of room temperature group and 4 ℃ groups,detected within three days,were not significantly different with initial detection value(P>0.05).After seven days,the testing results in specimen of 4 ℃ group were not significantly different with initial detection value(P>0.05).However,after one day,the testing results in specimen of-20 ℃ group were significantly different with initial detection value(P<0.05).Contrast with 4 ℃ without preservative group,the adding of preservative could not increase the stability of AD7C-NTP.The adding of preservative in specimen of-20 ℃ could obviously increase stability,but the deviation of testing results was beyond acceptable limits.Conclusion 4 ℃ without preservatives could be the optimal storage condition for detection of AD7C-NTP in the urine.
3.Early treatment and Ⅰ-stage repair of acute facial and brain injuries

Objective To study the method for early treatment and Ⅰ-stage repair of the acute facial and brain injuries and its significance. Methods A retrospective study was performed in 38 cases with acute cranial injury,who underwent early treatment and surgical repair of the cranial bone and/or dura defects with self-tissues,glue material and titanium net depending on individual conditions. Of all,33 cases (87%) received Ⅰ-stage repair. Antibiotic therapy was performed in all the cases after operation. Results All patients recovered with satisfactory regional appearance. No intracranial infection or rhinorrhea of cerebral spinal fluid occurred among them during the follow-up for from six months to 11 years. Conclusion For the acute facial and brain injuries,early treatment and Ⅰ-stage repair based on the injury site and injury severity can attain safe and effective results.
4.The Relationship of VASP Signaling Molecule and Colorectal Cancer

GCC/PKG/VASP,PKA/VASP et.al are associated with the occurrence and development of colorectal cancer.As one of the important signaling molecules of above signal pathways,VASP has constributed to the invasion,metastasis,apoptosis,angiogenesis and other events of colorectal cancer.The role of VASP signaling pathways in the development of CRC and its potential clinlic significance are reviewed in this article.
5.The clinical analysis of fungus ball sphenoid sinusitis

OBJECTIVE To improve the knowledge on the early diagnosis and treatment of the isolated fungus ball sphenoid sinusitis.METHODS A retrospe ctive study was made on10cases of isolated fungus ball sphenoid sinusitis to discuss the clinical manifestation,radiographic features and treatment with endoscopic sinus surgery.RESULTS The common symptoms in10cases were headache and/or bloody nasal discharge.Radiographic features of the disease were a soft tissue image occupying the sphenoid sinus with bones wall thickening.A soft tissue image with calcification in the sphenoid sinus was the feature of CT image.All the patients were treated by nasal endoscopic sinus surgery and cured after followed up for3to18.CONCLUSION Isolated fungus ball sphenoid sinusitis was difficult to diagnose in early stage because of its non-specif ic symptoms.The nasal endoscopy and CT scan make the early diagnosis possible.The diagnosis can be def initely confi rmed by pathological studies.Treatment with endoscopic sinus surgery is an effective method for fungus ball sphenoid sinusitis.

7.Clinical analyses of spontaneous cerebrospinal fluid rhinorrhea

OBJECTIVE To investigate the best approach of diagnosis and treatment of spontaneous cerebrospinal fluid rhinorrhea.METHODS The clinical data of 13 cases with spontaneous cerebrospinal fluid rhinorrhea performed in our hospital from 1998 to 2007 were analyzed retrospectively.RESULTS The diagnosis of spontaneous cerebrospinal fluid rhinorrhea was established for these 13 cases on a set of diagnostic program,including qualitative determination of sugar in the nasal leakage,nasal endoscopy, CT and MRI scan.All the patients were failed with conservative treatment.All of the 13 patients were cured by surgical repairing under nasal endoscope for follow up of 6 months to 6 years.CONCLUSION The diagnosis program for spontaneous cerebrospinal fluid rhinorrhea includes qualitative determination of sugar in the nasal leakage,nasal endoscopy,CT and MRI scan.Transnasal endoscopic management is the best method for repairing spontaneous cerebrospinal fluid rhinorrhea.
8.Clinical analysis of orbital apex syndrome caused by sinus diseases

Objective:To investigate the clinical characters and therapeutic methods of orbital apex syndrome caused by sinus diseases.Method:Six cases of orbital apex syndrome originated from sinus diseases were retrospective analyzed in our hospital from August 2003 to February 2009. Different therapeutic methods were taken according to different causes of disease.Result:Four cases of sinus infection,one cases of sphenoethmoid mucocele , one cases of sinus squamous cell carcinoma. The results of patients with orbital apex syndrome included cure and effectiveness in 2 cases respectively, death from myocardial infarction in one case, and one case is still following up.Conclusion:Orbital apex syndrome originated from sinus diseases was rare, which was mainly caused by sinus infection . The key point is the correct etiology diagnosis in early phase . Treatment with endoscopic sinus surgery early is a good effective method for orbital apex syndrome caused by sinus diseases, in the same time with enough dosage of antibiotic and glucocorticoid therapy intravenously.

Ain. To study the effect of kanglemycin C (KC) on production and gene transcription of lymphokins. Methods. Cell proliferation and lymphokin activities were quantified with MTT colorimetry and ELISA, and genetranscripdons of lymphokins semi-quantified with RT-PCR. Restlts. Suppression of KC on proliferation of enriched T-and B-cell respectively mediated by Con A and LPS was declined by addition of exogenous IL-1, IL-2, and IL-6. KC 80 nmol/L markedly inhibited IL-2 and IL-6 pro-duction and mRNA transcription of incubated mouse splenocytes induced by Con A. Additionally, KC had some sup-pression on IL-1β and IL-6 productions of peritoneal macrophage stimulated by LPS (5 μg/mL), whereas cyclosporine(CS) had not. Condusion. Immunosuppression of KC came true partially through the decrease of IL-1β, -2 and -6 productions,especially of IL-2. However, CS's immunosuppression was mainly through the decrease of IL-2 procduction.
10.Biocompatibility of beta-tricalcium phosphate/alpha-calcium sulfate hemihydrate artificial bone and its application in a spinal fusion model

BACKGROUND: Beta-tricalcium phosphate (β-TCP)/alpha-calcium sulfate hemihydrate (α-CSH) artificial composite bone has a porous morphology and good biocompatibility, and it is helpful to improve the fusion rate in a spinal fusion model, which however has not yet been confirmed.OBJECTIVE: To investigate the preparation methods, biocompatibility and application effect of β-TCP/α-CSH composite bone in the spinal fusion model.METHODS: (1) Calcium sulfate dihydrate under certain conditions and at a proper temperature can be dehydrated to prepare α-CSH. Healthy bovine cancellous bone was decellularized, degreased and sintered under the certain condition and at the certain temperature to prepare β-TCP particles. Then, the β-TCP particles were dissolved in anhydrous ethanol, suspended, dried, and then used to prepare the β-TCP/α-CSH composite bone. Osteoblasts from the rabbit periosteum were co-cultured with the composite bone, and then cell morphology, adhesion and proliferation were observed. (2) Twenty New Zealand white rabbits were selected to make bilaterally posterolateral spinal fusion models of the multiple thoracic vertebrae, in which β-TCP/α-CSH composite bone was implanted into the left side (experimental group) and autogenous bone implanted into the right side (control group). The spinal fusion rate was compared between the two groups.RESULTS AND CONCLUSION: (1) Under the phase contrast microscope, a relatively small amount of L929 cells adhered to the composite bone after 3 days of co-culture, while the number of adherent cells became relatively dense. Under the scanning electron microscope, there were many crystalline particles on the surface of the composite bone,indicating a higher number of adherent cells on the composite bone surface. (2) The spinal fusion rate was increased at 4 weeks after implantation of β-TCP/α-CSH composite bone, which was significantly higher than that after implantation of autogenous bone (P < 0.05). (3) At 4 weeks after autogenous bone implantation, the bone trabecular bone was scarce and clumped, and newborn bone tissues and the boneless autograft bone were dominant. After 8 weeks after spinal fusion, the newborn bone tissues around the autograft were further increased. At 4 weeks after β-TCP/α-CSH composite bone implantation, there was no degradation of debris, but existed a few new bone tissues; at 8 weeks after implantation,the composite bone was surrounded by newborn bone tissues, and thickened trabecular bone and degradation of the composite bone were found. To conclude, the prepared β-TCP/α-CSH composite bone can achieve a higher fusion rate in the spinal fusion model.
