Objective To explore serum liver and kidney function,blood lipid,complement indicators in patients with hepatitis B-associated nephropathy and the relationship between HBV infection and kidney function.Methods 141 cases with hepatitis B-associated nephropathy were randomly selected from January 2015 to July 2016 from Renmin Hospital of Wuhan University including 96 male,female 45 eases.65 cases with nephropathy included 31 male,female 34 eases.68 cases with healthy people included 36 male and female 32 cases.Liver and kidney function parameters were detected by the automatic biochemical analyzer Siemens ADVIA 2400.Complement C3 and C4 were detected by the automated immunoassay analyzer Beckman 1MMAGE 800.HBV DNA load was detected by fluorescence quantitative PCR instrument ROCHE LightCycler(R)480.Results ALT,Cr,UA,Urea,β2-M,LDL,TCh,TG,C3 and C4 were significant difference among hepatitis B complicated with nephropathy,nephropathy group and healthy groups (FALT =6.50,FCr=46.02,FUA =32.89,FUrea =37.60,Fβ2 M =44.98,FLDL=4.13,FTch =5.20,FTG =26.90,FC3 =14.54,Fc4 =11.01,all P＜0.05).ALT,AST,β2-M,C3 and C4 were significant difference among hepatitis B activity period complicated with nephropathy,hepatitis B non-activity period complicated with nephropathy and nephropathy group (FALT =5.96,FAST =7.45,Fβ2-M =18.70,FC3 =5.32,Fc4 =4.16;all P＜0.05).Conclusion Concentrations of liver and kidney function,blood lipid abnormalities and complement had a certain relationship with the hepatitis B complicated with nephropathy and HBV had a certain effect on serum concentrations of β2-M and complement.

Objective: To examine the emotional states of asthmatic children with different degrees of severity, as well as the effects of emotion on change of cytokines in airway. Methods: Asthmatic children were divided into two groups according to the degrees of severity: moderate and mild. Their emotional states were measured and results were compared. Correlation analysis was conducted between scores on emotional scales and sputum levels of IL-8.Results: Total scores on anxiety and depression were higher in the moderate group than in the mild group. Negative correlation was found between levels of anxiety and IL-8 during acute exacerbation of asthmatic condition. Conclusion: Emotional distress was found to be increased with severity of asthmatic condition in children. Anxiety contributed to the decreased concentration of IL-8 in asthmatic children's airway.

Objective Investigating the expression of TLR3 and TLR7 mRNA in peripheral blood mononuclear cells (PBMC) of patients with chronic hepatitis C infection (CHC),to explore the effects of gender and age on Toll-like receptor expres-sion.Methods Peripheral blood was collected from 1 1 5 patients of chronic hepatitis C infection,1 1 3 healthy individuals.Ex-pression levels of TLR3 and TLR7 mRNA were detected by real-time quantitative PCR.Results The expression of TLR3 and TLR7 mRNA were significant difference between patients with CHC infection and healthy individuals (t=38.73,6.16, P<0.05),respectively.The expression of TLR3 and TLR7 mRNA were significant difference between premenopausal fe-males with CHC infection,postmenopausal females and young males of CHC infection (t=61.210,6.464,P<0.05;t=24.166,26.266,P<0.05),respectively.The expression of TLR3 and TLR7 mRNA were significant differences between old males and young males of chronic hepatitis CHC infection (t=86.349,19.583,P<0.05).The expression of TLR3 mRNA was significant differences between old males and postmenopausal females of CHC infection (t=122.941,P<0.05).There was no correlation between the expressions of TLR3 and TLR7 and HCV-RNA load of CHC patents of Gender and age dis-crepancies (|r|<0.40,P>0.05).The HCV-RNA load of premenopausal females was significant lower than young males and old females (t=3.49,2.51,P<0.05),the load of old males was lower than old females (t=2.35,P<0.05),however, there was no significant differences between old males and old females (t=1.20,P<0.05).Conclusion Gender and age dis-crepancies have a relationship with the expression of Toll-like receptors of patients with CHC infection,which may provide a theoretical basis and a new method for CHC.

Objective To detect the expression of retinoic acid-inducible gene Ⅰ ( RIG-Ⅰ) in peripheral blood mononuclear cells (PBMCs) of patients with chronic hepatitis C (CHC), and to explore its correlations with type Ⅰinterferon gene expression and serum level of hepatitis C virus .Methods A total of 101 na?ve patients with CHC were enrolled from Renmin Hospital of Wuhan University during July 2014 and July 2015, and 69 healthy subjects served as controls .The expressions of RIG-Ⅰ, IFNαand IFNβmRNA in PBMCs and serum level of HCV RNA were detected by real-time quantitative polymerase chain reaction.Pearson correlation analysis was performed to investigate the correlations of RIG-Ⅰ mRNA expression with IFNα/βmRNA expression and serum level of HCV RNA .Results The relative expressions of RIG-Ⅰ, IFNα, IFNβmRNA to healthy controls in CHC group were 0.082, 0.022 and 0.156, respectively (t=7.83, 3.65 and 2.13, all P<0.05).RIG-Ⅰ mRNA expression in patients with HCV loads <(1 ×106 ) IU/mL was 1.79 times higher than that in patients with HCV loads≥(1 ×106 ) IU/mL (t=2.60, P <0.05).Pearson correlation analysis showed that the expression of RIG-Ⅰ mRNA was positively correlated with that of IFNαmRNA and IFNβmRNA (r=0.247 and 0.489, P<0.05), but not correlated with HCV load (r=0.187, P>0.05).Conclusion Expression of RIG-ⅠmRNA decreases in patients with CHC , and is positively correlated with that of IFNαand IFNβmRNA.

Hepatitis C virus (HCV)was easily developed into hepatitis,cirrhosis and liver cancer after infecting human.Cur-rently,the traditional method of treatment of HCV was pegylated interferon and ribavirin program,which was ineffective and had poor side effects,while the new direct antiviral drugs were expensive.Therefore,looking for cheap and non-toxic side effects of treatment was the focus of current research.More and more evidence indicate that micro-Ribose Nucleic Acid (miRNAs)play an important role in the development of liver disease,regeneration and functional regulation.This review studies the relationship between miR-122 and Hepatitis C virus and hepatocellular carcinoma.

Objective To investigate mRNA expressions of Toll-like receptors (TLR3 and TLR7)and type Ⅰ interferons (IFN-α and IFN-β) in peripheral blood mononuclear cells (PBMCs) of patients with chronic hepatitis C (CHC).Methods Thirty patients with CHC and 30 healthy controls were collected from Renmin Hospital of Wuhan University during September 2013 and May 2014.Liver function was detected using enzyme-linked immunosorbent assay (ELISA).mRNA expressions of TLR3,TLR7,IFN-α and IFN-β were detected by real-time quantitative PCR,and their relative expression values were obtained by 2-△△Ctmethod.The t test was performed for the comparison of mean differences between CHC patients and healthy controls,and Pearson analysis was used to analyze the correlations between TLR mRNA and IFN mRNA,liver function,HCV RNA load.Results Taking mRNA expressions in healthy control group as 1,the relative mRNA expressions of TLR3,TLR7,IFN-α and IFN-β in CHC group were 0.086,0.633,0.145 and 0.423 respectively (t =4.25,2.39,2.37 and 2.80,all P ＜ 0.01).Pearson correlation analysis showed that mRNA expressions of TLR3 and TLR7 were positively correlated with that of IFN-β (r =0.381 and 0.487,all P ＜ 0.05),but not correlated with IFN-α mRNA expression,HCV RNA load,ALT and AST (all P ＞ 0.05).Conclusion The mRNA expressions of TLR3,TLR7 and IFN-α,IFN-β decrease in CHC patients,and the mRNA expressions of TLR3 and TLR7 are positively correlated with that of IFN-β,indicating that increasing the expression of TLRs might be a new strategy in CHC treatment.

Objective To detect mRNA expressions of STING and type Ⅰ interferons(IFN-α and IFN-β) in peripheral blood mononuclear ceils (PBMCs) of patients with chronic hepatitis C(CHC).Methods 113 patients with CHC and 94 healthy controls were collected from Renmin Hospital of Wuhan University during January 2015 and January 2016.Expressions of mRNA of STING,RIG-1,IFN-α and IFN-β were detected by real-time quantitative PCR,and their relative expression values were obtained by 2-△△Ct method and the results were statistically analyzed.Results The expression of mRNA of STING,RIG-1,IFN-α and IFN-β in CHC were 0.018,0.361,0.578 and 0.573 times than its in healthy controls respectively and the differences were statistically significant (t-8.28,4.26,2.18 and 2.07,all P ＜ 0.05).Pearson correlation analysis showed that mRNA expressions of STING in healthy controls were positively correlated with that of IFN-α mRNA and IFN-β mRNA (r=0.487 and 0.207,all P＜0.05),which had no correlation in CHC (r=0.174 and 0.091,all P＞0.05).The expressions of STING mRNA was positively correlated with that of RIG-1 mRNA in healthycontrols (r=0.222,P＜0.05),but there was no correlation of expression in CHC between STING mRNA and RIG-1 mRNA (r=-0.029,P＞0.05).Conclusion Compared with healthy controls,the expression of STING,RIG-1,IFN-α and IFN-β mRNA were all reduced.There was positive correlation between STING and RIG-1,IFN-α and IFN-β in healthy controls,but no correlation in CHC.

Background It is thought in recently that photodynamic therapy (PDT) is an effective treatment method for chronic central serous chorioretinopathy (CSC), but the dosage of verteporfin and its long-term efficacy and complications is rarely elucidated ever before.Objective This study was to observe the long-term efficacy and safety of 60％ dose verteporfin PDT for chronic CSC.Methods This is a retrospective study and a self-controlled design was used.The clinical data of 25 eyes of 21 chronic CSC patients who received 60％-dose verteporfin PDT in Henan Eye Institute from January 2009 to May 2010 were reviewed, with the male 18 (85.71％) and female 3 (14.29％) as well as monocular CSC 17 patients and binocular CSC 4 patients.The average ages of the patients were (43±5) years.Fundus fluorescein angiography (FFA) , indocyanine green angiography (ICGA), optical coherence tomography(OCT) and best corrected visual acuity (BCVA) were examined in all the patients before and after treatment.PDT with the 60％-dose verteporfin (3.6 mg/m2) was carried out on the CSC eyes.The treated eyes were examined 2 weeks, 1 month and 3 months after PDT.The BCVA,subfoveal choroid thickness,FFA and ICGA findings before and after PDT were compared.The following-up duration was 5 years or more.Results The BCVA before and 3 months after PDT were 0.5 ±0.1 and 0.9±0.2, respectively, with a statistically significant difference between them (t =19.17,P =0.00).The subfoveal choroidal thickness value 3 months after PDT was (326.56±39.47) μm,which was significantly reduced in comparison with (486.24 ±47.53) μm before PDT (t =25.17, P =0.00).FFA and ICGA showed that the leakage of fluorescein (hyperfluorescence) was disappeared in all the treated eyes.No systemic or local adverse effects and recurrence were observed during the follow-up period.Conclusions On the basis of the results of this study and available information,60％-dose verteporfin PDT seems to have a better long-term efficacy and safety than full-dose verteporfin in treating chronic CSC.

Objective To evaluate the efficacy of preoperative biliary drainage (PBD) in the pancreaticoduodenectomy for malignant obstructive jaundice.Methods Database including PubMed,EMBASE,Cochrane Central Register of Controlled Trials,Academic Degree Dissertation Database and Conference Database were searched with malignant obstructive jaundice,pancreaticoduodenectomy,preoperative biliary drainage,comparative study.Literatures about the randomized controlled trials of PBD (PBD group) and efficacy of early surgery (ES group) in the pancreaticoduodenectomy were retrieved from January 2001 to December 2013,and then a Meta analysis was carried out based on the data.The count data were analyzed using the odds ratio (OR),relative risk (RR) and 95％ confidence interval (95％ CI),and the measurement data were analyzed using mean difference (MD) and 95％ CI.The heterogeneity of the data was analyzed using the I2 test.Data were integrated by fixed or random effect model.Results Twelve literatures including 1 982 patients were selected.There were 1 029 patients in the PBD group and 953 in the ES group.The results of Meta analysis showed that the operation time,volume of blood loss and rate of postoperative wound infection in the PBD group were significantly different from those in the ES group (MD =10.50,107.92,95％ CI:6.34-14.66,16.43-199.42;RR =1.62,95％CI:1.19-2.21,P ＜0.05).There were no significant differences in the postoperative mortality,incidence of pancreatic fistula,incidence of bile leakage,incidence of delayed gastric emptying and duration of hospital stay between the 2 groups (RR=0.69,95％CI:0.52-0.92;OR =0.68,1.35,95％CI:0.38-1.21,0.93-1.95;MD =0.69,95％CI:-0.67-2.05;RR =0.00,95％ CI:-0.02-0.01,P ＞0.05).Conclusion PBD in the pancreaticoduodenectomy for malignant obstructive jaundice cannot reduce postoperative mortality and incidence of complications in patients,and should not be used as the conventional management in the perioperative period.

Objective:To investigate the mRNA expression of interferon‐induced protein with tetratricopeptide repeats 1 (IFIT1) and IFIT3 in the peripheral blood mononuclear cells (PBMC) in patients with chronic hepatitis C (CHC) with different genders and ages .Methods :Anticoagulant whole blood was collected from 215 patients with CHC and 79 healthy individuals . The mRNA expression levels of IFIT1 and IFIT3 were detected by real‐time quantitative PCR .Alanine aminotransferase(ALT) and aspartate aminotransferase (AST ) were detected by automatic biochemical analyzer . HCV‐RNA was detected with fluorescent probe .Results:Expression levels of IFIT1 and IFIT3 in PBMC of patients with CHC were significantly different from those in healthy people (t=22 .35 , P< 0 .05 ;t= 22 .75 , P< 0 .05) .There were statistically significant differences in ALT and AST between patients with CHC and healthy people (t=34 .38 , P<0 .05 ;t=21 .38 , P<0 .05) .The correlation analysis of gene expression levels of IFIT 1 and IFIT3 in PBMC with age ,gender ,liver function and HCV load in CHC patients revealed that the mRNA expression levels of IFIT 1 and IFIT3 were positively correlated with age (r=0 .395 , P<0 .05 ;r=0 .482 ,P<0 .05) and gender(r=0 .453 ,P<0 .05 ;r=0 .358 ,P<0 .05) .Conclusions :The expressions of IFIT1 and IFIT3 in PBMC are correlated with gender and age in CHC patients .
