1.Microglia mediated inflammatory signaling pathways after ischemic stroke
International Journal of Cerebrovascular Diseases 2017;25(2):174-178
Microglia is the most important immune cell in the central nervous system. It plays a key role in mediating the immune response in the central nervous system.Sterile inflammation is the key factor in the pathophysiological process following ischemic stroke.Microglia is activated and induces a series of inflammatory signals through binding of injuring related ligands to their corresponding receptors. This article introduces the activation of microglia and inflammatory response after ischemic stroke from Toll-like receptors, inflammasome, cytokine receptors,Notch signaling and other signaling pathways.

Objective To improve the diagnosis and treatment of hermaphroditism. Methods The data of 138 cases of hermaphroditism who were hospitalized in our department between March 2005 and April 2008 were reviewed and and analyzed to summarize our experience in the diagnosis and treatment. Results Among the 138 cases with a age ranging from 2 to 36(mean 17 years old),42 were socially male and 96 were socially female. With the help of medical examination,,laboratory examination, ultrasonic examination, radiological examination and pathologic diagnosis,54 patients were diagnosed as female pseudohermaphroditism,66 as male pseudohermaphroditism,10 patients as true hermaphroditism,2 patients as pure gonadal dysgenesis,2 as testicle degeneration, and the left 4 patients as Klinefelter syndrome.Among the 138 patients,132 patients were received surgery operation, including 76 of them receiving laparoscopy. After operation,121 patients were maintained female sex,11 patients were maintained male sex.Female pseudohermaphroditism were all maintained female sex.Fifty-nine patients of male pseudohermaphroditism were maintained female sex. There were 4 patients being maintained male sex,and 3 patients having not received operation.Of the cases of the true hermaphroditism,8 patients were maintained female sex and 2 patients were maintained male sex. Conclusion Early accurate diagnosis is very important to the treatment of hermaphroditism.Etiologic diagnosis is useful in the course.Although it is good for the hermaphroditism to maintain female sex,the decision of the patients and the family should be considered.

Aim To study the changes of the biological behavior of dermal fibroblasts in Ⅲ skin burns wound in rats using chitosan. Methods Wistar rats were randomly divided into 5 groups as follows:1% chitosan(W/V)group,2% chitosan(W/V)group,4% chitosan(W/V)group,bFGF(basic fibroblast growth factor) group and the control group.Rats were made for Ⅲ skin burns. The wound healing time was recorded, and the wound healing rate was calculated. Then the cell cycle and apoptotic dermal fibroblasts were determined and the amount of Hydroxyproline(HOP) in the skin tissue was analyzed. Results The wound healing rate of 4% chitosan(W/V) group was higher and the wound healing time of 4% chitosan(W/V) group was shorter than that of the control group. On the 7th,14th day post-injury,the content of protein of 4% chitosan(W/V) group was higher than that of the control group. The content of HOP of 2% and 4% chitosan(W/V) group was highest on the 7th day post-injury. Compared with that in control group, the percentage of cells of S stage in 4% chitosan(W/V) group was aboundant, and was reduced in apoptotic dermal fibroblasts. Conclusion The changes of cellular biological behaviors might be one of the mechanisms of that Chitosan could promote the wound healing of Ⅲ skin burns in rats significantly.

AIM To investigate the effect of puerarin (Pue) on tumor necrosis factor (TNF) ?, interleukin (IL) 6 secreted by neonatal rat cardiomyocytes during hypoxia/reoxygenation injury. METHOD The activities of TNF ? and IL 6 in the supernatants of cultured myocytes, which were sampled from different groups (control, model, and therapeutic groups with 1 g?L -1 Pue, 0 1 g?L -1 Pue, 0 01 g?L -1 Pue) at different time, were assayed by bioassay method. RESULTS TNF ? and IL 6 activity increased compared with that of control ( P

AIM: To investigate antioxidative and antinitrosative effects of volatile oil from A.longiligulare T.L.Wu on dextran sulfate sodium(DSS)-induced ulcerative colitis mice. METHODS: Balb/c mice were fed with 4% DSS solution for 7 d to induce ulcerative colitis.Using biochemical method, the activity of antioxidative enzyme SOD,MDA and NO were determined in normal, model,SASP and three mouse's groups with low, moderate and high volatile oil from A.longiligulare T.L.Wu respectively. At the same time, the activity of iNOS was also measured by immunohistopathology. RESULTS: The concentration of MDA and NO were reduced and SOD increased significantly in high-and moderate-volatile oil groups compared with those in the model group. The activity of iNOS was reduced significantly in high-and moderate-volatile oil groups compared with those in the model group. The results demonstrated that the expression of iNOS was significantly inhibited in DSSinduced ulcerative colitis mice after being treated with high or moderate-dosage volatile oil. CONCLUSION: Volatile oil from A.longiligulare T.L.Wu has antioxidative and antinitrosative effects which may be one of the mechanism for treating UC.

AIM To investigate the therapeutic effect of ethyl acetate extract from Pongamia pinnata roots (PREA) on ethanol-induced gastric lesions. METHODS The experimental gastric mucosal injuries were prepared by ig ethanol to rats, and the protective effect of PREA was evaluated by calculating lesion index, observing pathological changes, and measuring the contents of nitric oxide (NO) and malondialdehyde (MDA), and the activity of superoxide dismutase (SOD) from gastric mucosal tissue. In addition, gastric secretary and gastric wall adherent mucus were studied with the pylorus-ligation rat model. RESULTSCompared with the model control group, PREA (50, 150 and 450 mg·kg-1, ig) dose-dependently prevented the gastric mucosal damages induced by ethanol, its inhibition rates were 28.7%, 57.7% and 78.7 %, respectively. The pathomorphology lesions of mucosal tissue were obviously ameliorated. PREA obviously antagonized the ethanol-induced elevation of MDA content, and reduction of NO level and SOD activity of gastric mucosa. PREA significantly reduced gastric juice volume, free acidity, total acidity and total acid output, but didn′t affect the pepsin activity. Moreover, PREA obviously increased adherent mucus quantity of stomach wall, as well as free mucus quantity dissolved in gastric juice of pylorus-ligation rat. CONCLUSIONPREA has protective effect on ethanol-induced gastric mucosal injuries, which suggests that PREA may be used for protection or treatment of human ethanolinduced gastric lesions.

AIM: To study the effect of extract from Pongamia pinnata roots on anti-inflammation and analgesia and acute toxicity. METHODS: The models of mice ear edema induced by xylene and Cotton pellet granuloma in rats to observe the anti-inflammation effect of PRE via oral administration. The effect of PRE on analgesia was tested by measuring the latent period licking hind foot with the hot plate method and counting body twisting induced by acetic acid in mice. The acute toxicity of PRE was measured by the method of Bliss. RESULTS: PRE could significantly inhibit the ear edema caused by xylene in mice, granuloma hyperplasia caused by cotton in rats. It could significantly prolong the pain threshold on hot-plate in mice, reduce the writhing times in mice. The LD50 of PRE was 6. 371 8 g/kg, its 95% confident limit was 5. 408 4-7. 723 2 g/kg. CONCLUSION: PRE has obvious effect on anti-inflammation and analgesia and the lower acute toxicity.
