Chinese Pharmaceutical Journal 2016;51(24):2169-2174
OBJECTIVE: To study the chemical constituents of the total flavonoids from Sophora flavescens and establish a method for simultaneous determination of seven compounds. METHODS: The compounds were isolated by chromatography on silica gel and ODS column and their structures were elucidated by spectroscopic analysis. The samples were analyzed on a Dikma C18 column (4.6 mm × 250 mm, 5 μm) ; gradient elution was performed using mobile phase composed of methanol (A) and water (B); the detection was carried out using a photodiode array detector at 280 nm. RESULTS: Seven compounds were isolated and their structures were identified as kuratidine (1), sophoraflavanone G (2), kurarinone (3), isoanhydroicaritin (4), isoxanthohumol (5), formononetin (6), and trifolirhizin (7). The calibration curve was linear within 8.70-87.00, 44.25-442.50, 128.10-1 281.00, 9.40-94.00, 48.40-484.00, 14.20-142.00, and 25.70-257.00 μg·mL-1 for kuraridine, sophoraflavanone G, kurarinone, isoanhydroicaritin, isoxanthohumol, formononetin, and trifolirhizin, respectively (r > 0.999 0), and the extraction recoveries varied from 95% to 105%. CONCLUSION: The main chemical components contributing to antibacterial activity of total flavonoids may be sophoraflavanone G, kurarinone, and isoxanthohumol. The method is simple, rapid, accurate, and can be used simultaneously to determine the contents of the seven active ingredients.

Asparaginyl endopeptidases (AEPs) in plants belong to the family of cysteine protease that undergo self-activation in the form of zymogen in acidic vacuole and play important physiological roles in maturation of seed storage proteins, protein degradation, programmed cell death and host defense. Bioprocessing enzymes (peptidyl Asx-specific ligases, PALs) that promote the maturation of cyclotides have recently been isolated and identified from several cyclotide-rich plants. PALs derived from AEPs can site-specifically catalyze the formation of asparagine or aspartate peptide bonds. Due to the advantages of relatively traceless peptide bonds and broad substrate spectrum and high catalytic efficiency, they have been playing important roles in the cyclization and modification of peptides and proteins, and are powerful tools for improving the stability of peptide drugs. This review describes the physiological functions of AEPs in plants and summarizes the discoveries, structural characteristics, catalytic mechanism and protein engineering of PALs, as well as the limitation of their applications and future trends. In addition, the applications of PALs in cyclotides biosynthesis and the development of macrocyclic peptides are highlighted, with the aim of providing a new idea for the biocatalytic synthesis of cyclic peptides.

How to remove the well fixed cement or cementless prosthesis and get a completely distal cement removal in the rTHR are critical to the outcome of revision. Because of higher rate of union, excellent intraoperative exposure, and adjustment of abductor tension, ETO has been widely applied to rTHR and complicated primary THR by foreign scholars. Furthermore, this technology has wide indications, very few contraindications, high cure rates, and low complications rate. ETO turns out to be a safe and effective revision technology. In the article, the indication, contraindication, complications and advantages of this technique were reviewed.

Objective To evaluate changes in cerebral blood flow before and after cranioplasty by 256-slice Spiral CT perfusion imaging, and evaluate the effect of cranioplasty on the cerebral blood flow in patients with skull defect. Methods 256-slice spiral CT scan was performed in 20 cases with early cranioplasty surgery,CTP check time points were 1 to 2 days before and 10 to 14 days after cranioplasty surgery. We recorded the the CBF and CBV of the cortex, basal ganglia, and thalamus and other parts,MTT on rCBV,parameter values rCBF,MTT and 1TrP etc.and analyzed and compared.(RCBF,rCBV,MTT and TTP) Results The CBF of cortex after cranioplasty at injured side had statistically significant increase (P<0.05).The CBF of cortex, basic nuclei, thalamus on contrateral had no statistically significant difference. The cerebral blood flow on both sides of the basal ganglia and the thalamus was increased after surgery, but there was no significant difference between before and after surgery (P>0.05) Conclusion Cranioplasty can significantly improve the ipsilateral cortex cerebral blood flow, and CT brain perfusion can accurately assess changes in brain tissue blood flow before and after cranioplasty.
6. Study on trifolirhizin phospholipid complex self-microemulsifying drug delivery system and its transmembrane transport
Chinese Traditional and Herbal Drugs 2016;47(4):573-579
Objective: To improve the absorption of trifolirhizin (Tri) isolated from Sophora flavescens through preparing it to be phospholipid complex self-microemulsion. Methods: In this study, we used the silica gel column chromatography and recrystallization technology to separate and prepare trifolirhizin; And we used physical and chemical properties and spectral data to identificate the structure. We used the orthogonal experiment and the central composite design response surface method (CCD-RSM) to optimize the Tri phospholipid complex (TPC), Tri phospholipid composite material self micro-emulsifying drug delivery system (TPC-SMEDDS), and used the Caco-2 model to investigate the transmembrane transport of Tri, TPC, and TPC-SMEDDS, respectively. Results: The concentration of the phospholipid complex was 4 mg/mL, the ratio of drug to lecithin was 1:1.5, the reaction time was 3 h, the composite rate was (93.20±2.01)%. The emulsifier was polyoxyethylene castor oil (Cremphor EL40), the auxiliary emulsifier was diethylene glycol ethyl ether (Transcutol HP), and the weight ratio (Km) of phospholipid complex was 7.58. The experiments of Caco-2 cell showed that the Papp of Tri was 2.45×10-7 cm/s; The Papp values of TPC and TPC-SMEDDS were 5.13×10-6 and 1.847×10-5 cm/s, respectively. Conclusion: The permeability coefficient of Tri can be obviously improved by using phospholipid complex and self-microemulsion technique, which also can improve the efficiency of drug delivery.

OBJECTIVETo observe the clinical efficacy on spastic pelvic floor syndrome (SPFS) treated with electroacupuncture (EA) at Shangliao (BL 31), Ciliao (BL 32), Zhongliao (BL 33) and Xialiao (BL 34).
METHODSThirty-six cases of SPFS were treated with EA at Shangliao (BL 31), Ciliao (BL 32), Zhongliao (BL 33) and Xialiao (BL 34), intermittent wave, 60 times/min in frequency, retained for 20 min. In general, the acupoints on one side were stimulated in each treatment. The bilateral acupoints stimulation was applied in serious cases. The treatment was given once every two days, five treatments made one session and totally three sessions were required. Before and after treatment, the clinical symptoms, anal kinetic indices defecation radiographic changes were observed. The follow-up observation was done in three months after the end of treatment.
RESULTSIn three-months follow-up after treatment, 14 cases were cured, 18 cases improved and 4 cases failed. The total effective rate was 88. 9%. After treatment, the cases of incomplete defecation, difficult defecation, anal pain and anal obstruction were reduced apparently as compared with those before treatment, indicating the statistically significant differences (all P<0.01). After treatment, rectal anal reflex threshold (ARA) was increased, anal maximal contraction pressure (AMCP) was reduced to (16.62±1.54) kPa and anal rest pressure (ARP) was significantly reduced to (7.22±0.36) kPa, indicating the statistical differences as compared with those before treatment (all P<0.01). After treatment, anorectal angle (ARA) in forceful defecation was increased to (116.55±9.42)°, the distance between the anorectal junction and the pubococcygeal line was decreased, and the impression of puborectal muscle was alleviated apparently as compared with that before treatment (P<0.01).
CONCLUSIONEA at Shangliao (BL 31), Ciliao (BL 32), Zhongliao (BL 33) and Xialiao (BL 34) achieves definite efficacy on SPFS and this therapeutic method obviously relieves the symptoms and deserves to be promoted in clinic.
