3.Intervening and therapeutic effect of cordyceps mycelia extract on liver cirrhosis induced by dimethylnitrosamine in rats.

OBJECTIVETo explore the intervening and therapeutic effect of Cordyceps mycelia extract (CME) on liver cirrhosis induced by dimethylnitrosamine (DMN) in rats.
METHODSRat liver cirrhosis model was established by peritoneal injection of DMN at a dose of 10 microg/kg, once daily in the first 3 days of every week for 4 successive weeks. Experimental study on CME-intervention was conducted from the beginning of modeling to the end of the 4th week, while the CME-treatment experiment was carried out from the 4th week of modeling, when terminating the modeling factor, to the end of the 8th week, by administering CME at a dose of 0. 74 g/( kg d) once a day. Animals were killed in batches on the 3rd day, the 2nd (T1), 4th (T2), 6th (T3) and 8th (T4) week after modeling, to observe the histopathologic change in liver and the immunohistochemical staining of alpha-smooth muscle actin (alpha-SMA) and collagen type I (Col I), determine the content of hydroxyproline (Hyp) in liver, and the liver function was tested as well.
RESULTSCME-intervention experiment showed that as compared to those in the modeled rats at corresponding time points, in rats at T1 and T2, serum alanine aminotransferase (ALT), aspartate aminotransferase (AST) activity and total bilirubin (TBIL) content were significantly lower, and albumin (Alb) obviously higher; while at T2, Hyp content, ct-SMA and Col I positive expression were significantly lower (P < 0.05), the proliferation of collagen fibre attenuated. CME-treatment experiment showed that as compared to those in the modeled rats at corresponding time points, lower serum ALT, AST activity and TBIL content, and higher serum level of Alb were shown in rats at T1; and lower Hyp content, liver collagen fibre, and alpha-SMA positive expression were shown at T1 and T2; while less Col I positive expression at T2 was also shown in them (all P < 0.05).
CONCLUSIONCME could not only prevent the development of liver cirrhosis induced by DMN in rats, but also effectively promote the reversion of already formed liver cirrhosis, having a favourable prospect of clinical application.

The strain of DG7 isolated from the oilwater sample in DAGANG oilfield could produce surfactant ?acids and gas in the culture medium in which the diesel was sole carbon source .The strain was Gram-negative, motive, rod and growed facultatively in the 0 to 18.5% NaCl. Based on its characters, the strain was identified as a member of the genus Aeromonas, but there were some differences between this strain and the described species of this genus in some biochemical features, suggesting that it could be a new species of the genus.
5.Comparative Study of Mesenchymal Stem Cells Derived From Human Umbilical Cord Vein and Adult Bone Marrow

Mesenchymal stem cells (MSCs) are promising candidates to develop new cell-based therapeutic strategies and bone marrow represents the main source of MSCs for experimental and clinical application currently. How to obtain enough MSCs is the biggest challenge faced by the experimental and clinical application. The difference of morphology, growth pattern, immunophenotype and multilineage differentiation of MSCs derived from human umbilical cord vein and their multiple differentiation capacity were investigated. The results showed that MSCs derived from human umbilical cord vein and normal adult bone marrow were similar in biological characteristics. They not only grew in fibroblast-like cells mode but had great expansion potency and multilineage differentiation ability in vitro. The MSCs derived from human umbilical cord vein may be an excellent alternative source for experimental and clinical application.
6.Meta analysis on the association between parental 5, 10-methylenetrahydrofolate reeducates C677T polymorphism and the neural tube defects of their offspring

Objective To explore the relationship between 5, 10-methylenetrahydrofolate reeducates gene polymorphism of the parents and the susceptibility to their offspring having neural tube defects. To forecast and evaluate the onset risk of neural tube defects (NTDs) on the basis of 5, 10-methylenetrahydrofolate reeducates (MTHFR) C677T polymorphism on parents of the patients.Methods Electronic search strategy was carried out among the five databases from home and abroad to collect qualified research papers, according to the inclusion and exclusion criteria. Case-control studies on association between MTHFR polymorphism and susceptibility to NTDs were collected and divided into two groups as mothers and fathers. The combined OR values and their 95% CI were calculated with Review Manager 5.0. Results 33 eligible studies were included. Statistics of the combined data showed a significant difference between the mothers with NTDs offspring carrying TT/CC, CT/CC of MTHFR gene C677T and controls. The pooled OR (95%CI) were 1.68 (1.32-2.14)and 1.20 (1.07-1.35) respectively. These was a significant difference between the fathers of the offspring with NTDs that carrying of TT/CC, CT/CC of MTHFR gene C677T and controls. The pooled ORs (95%CI) were 1.38 (1.08-1.76) and 1.32 (1.13-1.55) respectively. Conclusion The results suggested that the paternal and maternal MTHFR gene C677T polymorphism were risk factors to NTDs.
7.Inhibitory Effects of Suramin in Combination with PG-Rg3 on the Growth of Lung Adencarcinoma in Mice

Objective To observe the effect of suramin combinated with PG-Rg3 on xenograft growth of lung adeno-carcinoma in mice, and the related mechanism thereof. Methods Forty C57BL/6J mice bearing Lewis cells were random-ized into five groups:control group, cisplatin (DDP) group, suramin group, PG-Rg3 group and combination group. Appropri-ate interventions were given in five groups of mice. Mice were sacrificed at day 24 after tumor inoculation. The subcutaneous tumors were stripped for histological examination. The tumor inhibitory rate was measured. The expressions of erythropoietin-producing hepatoma amplified sequences (Eph) B4 protein, Bcl-2 and tumors microvessel density (MVD) were determined by immunohistochemistry method with image analyze system. The apoptosis of tumor cells was measured by biotinyated dUTP nick and labeling (TUNEL) method. Results There were significantly lower values in subcutaneous tumor volume and weight in drug-treated groups than those in control group (P<0.05). The inhibitory rates were 39.20%, 49.11%, 54.86%and 62.49%in cisplatin group, suramin group, PG-Rg3 group and combined group (P<0.05). The values of EphB4, MVD and Bcl-2 grey values were significantly decreased, the apoptotic index was significantly increased, in suramin group, PG-Rg3 group and combined group than those of control group and DDP group (P<0.05). The values of EphB4, MVD and Bcl-2 grey values were significantly increased, the apoptotic index was significantly decreased, in suramin group and PG-Rg 3 group than those of combined group (P<0.05). Conclusion Suramin combinated with PG-Rg3 can produce a synergetic inhibitory activity against tumor growth of lung adenocarcinoma, which may be associated with the effect of suppressing the expression of EphB4 and angiogenesis, and the promotion of tumor cell apoptosis.
