Objective To investigate the changes of serum interleukin 8(IL-8) level in children with thyroid disfunction and its clinical significance in children with autoimmune thyroid disease(AITD).Method Serum IL-8 of 30 children with hyperthyroidism,16 children with hypotyroidism and 20 health children were detected by double antibody enzyme linked immunossorbent assay(ELISA).Results Serum IL-8 levels in hyperthyroidism and hypothyroidism children were significantly higher than that in controls(P0.05).With the remission of the diseases, IL-8 levels decreased; there was a significant difference between serum IL-8 levels in patients before and after therapy (P

Background Deficiency of neurotrophic factor is associated with the damage of optic nerve in glaucoma.Reaserches showed that ectopically applied neurotrophic factor has a transient neuroprotective effect in glaucoma model,and the viral expression of adeno-associated neurotrophie factor may provide long-term supplementation of neurotrophic factor and neuroprotection in tissues.Objective The present study Was to investigate the neuroproteetive effect of adeno-associated viral(AAV)-mediated brain-derived neurotrophic factor (BDNF)on retinal ganglion cells(RGCs)in DBA/2J mice with experimental glaucoma. Methods 10 clean DBA/2J mice were administered intravitreal injection with 1 microliter of AAV-BDNF-GFP in the left eyes at the age of 6 months,and the right eyes were injected with the same volume of saline solution as control.Intraocular pressure (IOP)was measured with Tonolab in the mice every month.Retinas were obtained after 3 months for the investigation of GFP expression in RGCs using fluorescence microscopy.Immunohistochemistry Was performed by applying TUJ1 and Cy3 antibodies to identify surviving RGCs. Results The IOP of DBA/2J mice were 11.90 mmHg and 11.40 mmHg in the right eyes and left eyes,respectively,at 4 months.The IOP of DBA/2J mice began to rise at 5 months and reached its peak in 8 month-old mice.There was no statistically significant difference in IOPs between the right eyes and the left eyes from 4 month-through 9 month-old mice(t=-1.78-0.61,P=0.11-0.90).Three months after intrlavitreal injection of AAV-BDNF-GFP,GFP was positively expressed in RGCs of retinas with the expression rate of 46.33%±8.08%.The over-expression of BDNF led to more RGCs survival than the control eyes (3168.13±1319.33/mm2 vs 2024.81±796.38/mm2,t=2.75,P=0.02). Conclusion These data suggest that BDNF can exert a protective effect in DBA/2J glaucoma mice.
3.Assessment and direction of Objective Structured Clinical Examination on clinical practice in Department of Obstetrics and Gynecology

Objective To analyze the results of the Objective Structured Clinical Examination(OSCE) of medical interns in Department of Gynecology and Obstetrics,and to assess the role of OSCE in clinical teaching and practice. MethodsAtolal of 104 medical interns of five-year education program in 2002 and seven-year education program in 2003 in School of Medicine had taken part in OSCE of gynecology and obstetrics.The OSCE consisted of six stations: gynecologic examination,obstetric examination,oral test,interrogation of standardized-patients(SP),drawing partogram,and non-stress test(NST) explanation.After gathering the data of each station,the software of SPSS 11.0 was used to make statistical analysis. Results The median scores were as follows: 92.0 for gynecologic examination,91.0 for obstetric examination,83.5 for oral test,80.0 for interrogation of SP,80.0 for drawing partogram,and 70.0 for NST explanation.There was no significant impact of familiarity to OSCE on the results of the test.The results showed that medical interns did good job in basic performance,but lack of clinical practice skills in some complicated practice of gynecology and obstetrics.Conclusion The grading system and stations setup of OSCE in Department of Gynecology and Obstetrics in Shanghai Sixth People's Hospital show objectivity and equity.The OSCE can be widely applied in the after-department examination and even graduation examination.
4.Study on knowledge,attitudes and use of the pre-packaged food nutrition labeling in Hangzhou supermarket consumers in 2010

Objective To study the prevalence rates of knowledge,attitudes and use of the nutrition labeling and related influential factors in Hangzhou supermarket consumers.Methods Using a self-developed survey questionnaire,randomly selected customers were conducted a face-toface interview program in a large supermarket of Hangzhou city.Results 586 people were interviewed,including 202 males and 384 females,with the mean age as 41.6±17.23 years.The Facts Labels'were as follows:dietary fiber(71.84%),fat(70.99%),calcium(60.75%),salt (58.36%),energy(50.85%)and sugar(39.42%).The support rates of the three attitudes indicators were"support marking the Nutrition Facts Label"(90.44%)."support marking the nutrition claims"(87.03%)and"want to know more relative knowledge"(77.13%).There were 58.36% and 80.03%first-time buyers who would read the Nutrition Facts Food Labels and the Nutrition Claims.Through logistic model analysis,male,youth,low-educated people were found to be the risk factors related to the nutrition label reading behavior.Conclusion The capacity of the reading,understanding and correct application of nutrition labeling among consumers was not satisfactory.Improvement on the readability and the authority of nutrition labeling and the development of the national nutrition knowledge and health education programs were in urgent needs.

H4IIE hepatoma cells transfected by recombinant adiponectin adeno-associated virus vectors were effectively mediated adiponectin gene expression and enhanced the ability of suppressing glucose production of H4IIE cells at low concentration of insulin.Improvement of the insulin sensitivity in hepatocytes may contribute to the glucose-lowering effect of adiponectin.

OBJECTIVETo investigate the myocardial electrophysiological effect and its underlying mechanisms of atorvastatin (Ator) on isolated rat hearts injured by ischemia/reperfusion (I/R).
METHODSIsolated SD rat hearts were mounted on Langendorff system, and a local I/R was induced by ligation (30 min) and release (15 min) of the left anterior descending artery. During the reperfusion period, the effect of Ator on diastolic excitation threshold (DET), effective refractory period (ERP) and ventricular fibrillation threshold (VFT) on rat heart were measured.
RESULTCompared with the control group, medium concentration of Ator prolonged the ERP in normal rat hearts; low, medium and high concentration of Ator significantly inhibited the decrease of DET, ERP and VFT induced by I/R. However, pretreatment with L-NAME cancelled these cardiac electrophysiological effects of Ator.
CONCLUSIONAtor reduced electrophysiological alteration induced by I/R in isolated rat hearts, which may be mediated by activating nitric oxide pathway to enhance the myocardial electrophysiological stability.

8.Safety and efficacy of endovascular stenting for treatment of ruptured vertebral artery dissecting aneurysms

Objective To evaluate the safety and efficacy of endovascular stent placement in the management of ruptured vertebral artery dissecting ancurysms. Methods A retrospective analysis was conducted in 9 patients with ruptured vertebral artery dissecting aneurysms treated in our department between July, 2004 and January, 2007 by means of endovascular stenting. Of these cases, 3 were treated with exclusive stent placement and 6 with stent-assisted coiling. Results Stent deployment and coil embolization were carried out successfully in all the cases. The vertebral arteries with stent placement remained patent after the surgery, and stent thrombosis or aneurysmal rupture occurred in none of the cases. During the follow-up for 4 to 39 months, no clinical evidence of rebleeding was found in these patients. Follow-up angiography was performed in 6 of the patients 3-18 months after the surgery, and two patients treated with stent-assisted coiling showed aneurysm recurrence or enlargement and underwent secondary internal trapping or proximal occlusion; two patients receiving endovascular stenting showed complete embolization of the anenrysm, and one patient showed partial embolization. Conclusion Endovascular stent placement is safe and effective for management of raptured vertebral artery dissecting aneurysms and allows the involved artery to remain patent after the surgery. Close follow-up is necessitated in these patients, and the long-term effect of this treatment remains to be further observed.

10.Abnormal abundances of calcium cycling regulatory proteins in rabbit myocytes with heart failure.

OBJECTIVETo investigate the abnormal abundances of calcium regulatory proteins in rabbit myocytes with failing hearts.
METHODSSixteen rabbits were divided into two groups: 8 rabbits with heart failure induced by volume plus pressure overload and 8 sham-operated animals. The hemodynamic parameters and cardiac structure and function were detected via catheterization and echocardiography respectively. L-type calcium channel (LTCC), Ryanodine receptor 2 (RyR2), Sarcoplasmic reticulum Ca(2+)-ATPase (SERCA2a), and Na(+)-Ca(2+) exchanger (NCX) protein abundances were determined by Western blot analysis.
RESULTSThe ratio of left ventricular mass to body weight, heart rate and left ventricular end diastolic pressure in heart failure rabbits were significantly increased compared with sham-operated rabbits (P < 0.01), but their left ventricular shorten fraction [(21.3 +/- 4.00)% vs. (36.5 +/- 1.36)%] and ejection fraction (0.45 +/- 0.07 vs. 0.70 +/- 0.02) were decreased (P < 0.01). In heart failure rabbits, the abundances of LTCC and RyR2 were significantly decreased (R(LTCC/actin): 0.287 +/- 0.029 vs. 0.624 +/- 0.009; R(RyR2/actin): 0.106 +/- 0.001 vs. 0.203 +/- 0.011; P < 0.01), whereas the expressions of SERCA2a and NCX were markedly increased (R(NCX/actin): 0.497 +/- 0.015 vs. 0.221 +/- 0.014; R(SERCA2a/actin): 0.611 +/- 0.036 vs. 0.433 +/- 0.008; P < 0.01).
CONCLUSIONSReductions of LTCC and RyR2 might contribute to risk factors of systolic dysfunction in failing hearts. In early stage of heart failure, upregulated SERCA2a and NCX protein levels may be helpful for maintaining cardiac performance.
