The fused gene (PTH-HSA) of parathyroid hormone (PTH) gene and Human Serum Albumin(HSA) gene was amplified without linker by Overlapping PCR technology. The spliced gene was clone into Pichia pastoris secretory vector pPIC9K. With the help of promoter AOX1 and mat ? signal peptide, the PTH-HSA gene was designed to secretory expression.Linearized by restriction enzyme SalI, The recombinant plasmid pPIC9K/PTH-HSA was transformed into Pichia pastoris KM71 by electroporation. The recombinant strains which were identified by G418 and PCR analysis were induced by methanol to express protein PTH-HSA. The target protein was expressed in fermentation supernatant. Western blot analysis of the fusion protein showed that the expressed fusion protein PTH-HSA had the antigenicity of HSA.adenylate cyclase assay proved that the fused protein exhibited the bioactivity to stimulate cAMP synthesis The specific activity of broth was about 318IU/ml.

OBJECTIVETo estimate the feasibility of 16-multidetector spiral computed tomography (16-MDCT) on detecting coronary plaques in comparison with intravascular ultrasound (IVUS).
METHODSSixty-eight patients suspected of coronary heart diseases were examined by 16-MDCT, quantitative coronary angiography (QCA) and IVUS. Coronary stenosis was defined as lumen diameter reduction (DS) >or= 50%. Hounsfield units (HU) were used to determine different types of plaques: soft plaque (
RESULTSCompared to QCA, the sensitivity and the specificity for patients with DS >or= 50% were 91.8% (112/122) and 97.8% (556/568) respectively, the positive and negative predictive value were 90.3% (112/124) and 98.2% (556/566) respectively. In 96 plaques evaluated both by 16-MDCT and IVUS, 20 and 21 soft plaques, 37 and 36 fibrous plaques, 39 and 38 calcified plaques were identified by 16-MDCT and IVUS respectively. HU value of soft (11 +/- 36), fibrous (83 +/- 20), and calcified (292 +/- 80) plaques were significantly different (P < 0.05).
CONCLUSIONSNoninvasive 16-MDCT could correctly estimate coronary stenosis and coronary plaques compositions.

Journal of Preventive Medicine 2015;(1):14-16,20
Objective To explore the heterosexual intercourse characteristics and its influencing factors among MSM population. Methods Voluntary counseling/testing and questionnaire survey were carried out at the bathhouses where MSM usually assemble. Results Among 342 MSM surveyed,52. 05% were married. 76 people( 22. 22%) were completely homosexual, while 266 people(77. 78%)were bisexual. 24(31. 57%)MSM who answered″ totally gay″ in self-assessment of sex orientation were married, significantly lower than those answered bisexual(57. 89%)(p<0. 05). 197 of participants had sex with women ,and 123 MSM had sex with women during the last six months,49 MSM of which used condoms at each heterosexual activity. MSM who were married or bisexual were easy to have heterosexual intercourse. Among the MSM surveyed, the HIV and syphilis infection rates were 11. 40% and 16. 67% respectively. There was no significant difference between the use of condoms and infection( both p>0. 05). In the recent six months, there was no significant difference in the use of condoms between infected( 43. 34%)and non-infected persons( 43. 34%)( p >0. 05). Conclusion Among MSM population, the prevalence of heterosexual intercourse is high, while condom use rate is relatively low. It is necessary to strengthen the behavioral intervention and promote condom use.

Objective To explore causal relationship between atopic diseases(asthma and atopic dermatitis)and os-teoarthritis(OA)by using mendelian randomization(MR).Methods Asthma and atopic dermatitis as instrumental variables were selected, searched them through IEU database, and selected the latest data with a large number of cases and single nu-cleotide polymorphism(SNP).Data were collected and processed using R language, inverse varianceweighted(IVW)method was adopted as main MR Evaluation method.Single linear regression was performed to estimate causality based on pooled knee and hip data from genome-wide association studies(GWAS).The forest map was drawn to visualize the results, and gene pleiotropy and sensitivity were analyzed by scatter plot and funnel plot.At the same time, asthma, atopic dermatitis, body mass index(BMI),osteoporosis and OA were selected for multivariate MR Analysis to exclude the effect of horizontal pleiotropy on the results in GWAS data. Results Analysis of MR-IVW results showed asthma was positively correlated with causal effect of OA[OR=1.41,95%CI(1.07,1.85),P=0.02],multivariate Mendelian randomization(MVMR)adjusted for BMI and osteo-porosis and a direct causal effect on OA was observed[OR=1.57,95%CI(1.03,2.39),P=0.03)].MR Results of two samples of atopic dermatitis and OA were[OR=1.01,95%CI(0.97,1.04),P=0.76],and MVMR results were[OR=1.02,95%CI(0.99,1.05),P=0.25],indicating no clear causal relationship between two samples. Conclusion Asthma could increase risk of OA,atopic dermatitis has no obvious relationship with OA,and the relationship between atopic diseases and OA still needs to be discussed.
