1.Observation of the morphological change during the adipogenesis differentiation of human bone marrow stromal cell

ObjectiveTo study the variant differentiated phase of the human bone marrow stromal cell (MSC) during induced adipogenesis course by morphological observation.MethodsAfter proliferated in vitro, MSCs isolated from bone marrow of the female adults volunteers were cultured with the adipogenetic inducers for 6—30 days. Morphological changes of the cells were observed everyday under Olympus contrast phase microscopy, and some specimens were stained with Oil-red-O.ResultsMSC showed long spindle shape before inducing, and then changed gradually to oval or round shape and the figure enlarged simultaneously. There were specifically morphological changes with lipid droplet emergence under plasmalemma and confluence gradually to lipid drop during differentiated into the phase of immature and mature adipocyte.ConclusionIt is easy to detect the MSC differentiated into the phase of immature and mature adipocyte with the specific morphological change of lipid droplet, while in the phase of preadipocyte and adipoblast, there is no special morphological change, and it may need some specific markers to detect.
2.Expression of gastrin and gastrin releasing peptide in patient's with gastric cancer by using tissue chip technique.

OBJECTIVETo study the expression of gastrin(GAS) and gastrin releasing peptide(GRP) in patients with gastric cancer and investigate the clinical significance.
METHODSThe expression of GAS and GRP in sixty patients with gastric cancer was detected by using tissue chip technique and immunohistochemical methods.
RESULTSThe positive rates of GAS and GRP were 30.0% and 11.7% respectively in 60 cases with gastric cancer. The positive rates of GAS and GRP were higher in moderately and poorly differentiated cancers than those in well differentiated cancer (P< 0.05). The positive rates of GAS and GRP were significantly higher in mucinous adenocarcinoma and signet-ring cell carcinoma than those in other types of gastric cancer (P< 0.05). The positive expression of GAS and GRP in gastric cancer was correlated with lymph node metastasis (P< 0.05).
CONCLUSIONTissue chip technique is a feasible,rapid,economic and accurate approach for screening clinical tissue specimens on a large scale.

3.Clinical monitoring of tissue oxygen pressure on acupoint Zusanli (ST36) of patients in critical condition.

OBJECTIVETo observe the local tissue oxygenation in acupoint Zusanli (ST36) of patients in critical condition, and to assess the value of applying tissue oxygen measurement via acupoint in clinical practice.
METHODSThirty-six patients with severe illness undergoing invasive positive pressure ventilation (IPPV) were divided into the infected group (n=13) and the non-infected group (n=23). Tissue oxygen pressure (PtO2) in the right acupoint Zusanli was measured with tissue oxygen tension detector when the concentration of inspired oxygen (FiO2) was 40% and 100% respectively, meanwhile, the arterial blood oxygen pressure (PaO2) was also determined to calculate PtO2 index (the ratio of PtO2/PaO2).
RESULTSThere was a positive correlation between PtO2 and PaO2 (P < 0.05); PaO2 in the infected group was lower, and PtO2 index and PtO2 were higher significantly, as compared with those in the non-infected group (all P < 0.05). As FiO2 adjusted from 40% to 100%, the increment of PtO2 in the infected group was significantly higher than that in the non-infected group (P < 0.05).
CONCLUSIONFor the severely infected patients with obstacle of systemic oxygen delivery, the local tissue oxygen demand of acupoints increases significantly, and the tissue oxygen delivery dependence is enhanced, in this time, the tissue might fulfill the oxygen demand itself through elevating PtO2 index. It is of great significance to monitor tissue oxygen pressure in acupoint to determine the state of local tissue oxygenation in critically illed patients.

4.Development of a high-throughput sequence-based typing assay for human leukocyte antigen loci.

OBJECTIVETo develop a reliable assay for simultaneous sequence-based typing (SBT) of HLA-DPA1 and HLA-DPB1, and to apply it for the study of allelic polymorphisms in southern Chinese Han population.
METHODSBased on full-length HLA-DPA1 and HLA-DPB1 allelic sequences, locus-specific PCR primers were designed and applied to amplify the target sequence encompassing the entire exon 2 of HLA-DPA1 and HLA-DPB1. PCR products were purified with magnetic beads, and run through an ABI 3730 DNA sequencer. Genotypes were assigned with an Assign 3.5 SBT software.
RESULTSThe target sequences of HLA-DPA1 and HLA-DPB1 were both amplified with the PCR procedure. Little background and noise was observed in the derived sequences. Among 176 non-related healthy individuals, 4 HLA-DPA1 alleles with the frequencies of DPA1*02:02 (0.589) > DPA1*01:03 (0.284) > DPA1*02:01 (0.096) > DPA1*04:01 (0.031) were identified. In addition, 14 HLA-DPB1 alleles, including 4 common alleles (with a frequency of more than 5%, namely DPB1*05:01, DPB1*02:01, DPB1*04:01 and DPB1*02:02), 7 alleles with a frequency ranging from 1%-5% and 3 alleles with a frequency of less than 1% were identified. The results of HLA-DPB1 genotyping were all in accordance with the typing results derived from an Atria AlleleSEQR HLA-DPB1 kit.
CONCLUSIONA reliable technique has been established for simultaneous genotyping of HLA-DPA1 and HLA-DPB1, which may have a broad application in population and disease association studies.

5.Clinical Application Status of TCM for Prevention and Treatment of Osteonecrosis of Femoral Head

Osteonecrosis of the femoral head (ONFH) is a common refractory disease in orthopedics, and the rate of disability is very high, and its pathogenesis is unknown. TCM has unique advantages in the prevention and treatment of ONFH, and it has gradually received attention. This article reviewed the clinical application status of TCM for prevention and treatment of ONFH in recent years from ONFH's TCM name, etiology, pathogenesis, syndrome differentiation, and clinical treatment, with a purpose to provide a reference for the clinical treatment of ONFH.
6.Precision medicine of osteonecrosis of the femoral head: concepts and patterns

BACKGROUND: In recent years, precision medicine has become a hot topic in the medical field. The application of precision medicine to osteonecrosis of the femoral head is wider than before. OBJECTIVE: To summarize the current status and progress of precision medicine to osteonecrosis of the femoral head. METHODS: A computer-based search of PubMed and CNKI databases was performed for literatures about application and prospect of precision medicine for osteonecrosis of the femoral head in and outside China published within the past 10 years using the keywords of "precision medicine, osteonecrosis of the femoral head, three dimensional (3D) printing, computer assisted navigation, minimal invasive, gene" in English and Chinese. RESULTS AND CONCLUSION: (1) In the study on the prevention and treatment of osteonecrosis of the femoral head, 3D printing technology, computer assisted navigation technology, precision minimally invasive orthopedic technology, research and treatment of gene are four effective ways to achieve precise and personalized healthcare. (2) The appearance of precision medicine, a novel medicinal model, enriches the treatment of osteonecrosis of the femoral head and enhances the curative efficacy of the disease. It brings the new dawn to the patient and provides some new clues for medical workers and researchers engaged in the clinical and basic researches of osteonecrosis of the femoral head.

8.Biological evaluation of ~(18)F-FDTP as a potential dopamine D_4 receptor PET imaging agent

Objective To evaluate the feasibility of 3-(4-~(18)F-fluorobenzyl)-8,9-dimethoxy-1,2,3,4-tetrahydrochromeno [3,4-c]pyridin-5-one ( is F-FDTP) as a potential dopamine D4 receptor PET imaging agent.Methods ~(18)F-FDTP solution in ethanol-physiological saline was incubated with calf serum to test its in vitro stability through the determination of radiochemical purity.Normal rats were injected intravenously with ~(18)F-FDTP and then sacrificed at 2,5,10,15,30,60 and 120 min after anesthesia.Blood,organs and brain tissue samples were collected.All samples were weighed and measured for radioactivity.The uptake of samples was expressed as percentage activity of injection dose per gram of tissue ( % ID/g).Results The stability of ~(18)F-FDTP was satisfactory and its radiochemical purity was above 95% after incubation 120 min at 37℃ in calf serum.The biodistribution showed that ~(18)F-FDTP could penetrate through the blood-brain barrier and selectively accumulate in striatum,hypothalamus,frontal certex,hippocampus,cerebellum,where the D_4 receptor was reportedly located.The radioactivities in hippocampus,hypothalamus,striatum,frontal cortex,cerebellum,pons were (0.42±0.03),(0.46±0.05),(0.54±0.04),(0.39±0.04),(0.45±0.06),(0.35±0.04) %ID/g,respectively,2 min post injection.And there was difference between the normal biodistribution results and the blocking experimental results:(0.36 ±0.05),( 0.33±0.05 ),(0.55±0.05 ),(0.30±0.07 ),(0.34±0.07 ) and (0.32±0.04) % ID/g in hippocampus,hypothalamus,striatum,frontal cortex,cerebellum and pons,respectively.Conclusions ~(18)F-FDTP can penetrate through the blood-brain barrier and selectively accumulate in striatum,hypothalamus,frontal cortex,hippocampus,cerebellum,where the D_4 receptor was known to concentrate.These preliminary results suggest that ~(18)F-FDTP is a potential dopamine D_4 receptor imaging agent and further studies are needed.
9.Clinical features and experience of diagnosis and treatment of thyroid neoplasm in children.

OBJECTIVETo study the clinical features, diagnosis and therapy of thyroid neoplasm in children.
METHODSA retrospective study was performed on 32 children with thyroid nodular who were underwent operation in Xiangya Hospital between January 2002 and December 2010.
RESULTSOf the 32 cases, there were 23 girls and 9 boys. Six cases were diagnosed as nodular Goiter adenoma and 26 cases were diagnosed as thyroid papillary carcinoma. B-ultrasonic examination showed a 100% accurate rate for the diagnosis of thyroid carcinoma. Fourteen children (44%) were proven to have concurrent Hashimoto's thyroiditis. Twenty-two (69%) children with thyroid carcinoma were found to have lymph metastasis in the lateral neck. The children younger than 10 years showed a high rate of metastasis than those older one (94% vs 56%, P<0.05). All 32 children received a surgical therapy. Subtotal thyroidectomy was performed on the 6 children with nodular Goiter adenoma. Total thyroidectomy (17 cases) or ipsilateral thyroidectomy (9 cases) was performed according to the stage of thyroid carcinoma. The surgical outcomes were followed up for 3 months to 9 years and no recurrence or death occurred. The development and growth were normal in the children.
CONCLUSIONSChildhood thyroid nodular attacks girls more than boys, and the frequency of malignancy is high. Hashimoto's thyroiditis is a common concurrent disease. The incidence of local lymph metastasis is high in those younger than 10 years. The surgical therapy for thyroid neoplasm may lead satisfactory outcomes in children.

10.Postoperative detection of AFP mRNA in the peripheral blood of hepatic cellular carcinoma patients and its correlation with recurrence.

OBJECTIVEThe postoperative presence of alpha-fetoprotien (AFP) mRNA in the peripheral blood of patient with hepatic cellular carcinoma (HCC) may suggest that carcinomatous cells still exist in the peripheral blood which may someday develop into metastasis. Therefore, the expression of AFP mRNA in postoperative peripheral blood is detected and its correlation with recurrence of HCC is investingated.
METHODSAFP mRNA in peripheral blood of HCC patients and non-hepatic cellular carcinoma patients treated by partial hepatectomy on the first, 7th, 14th postoperative day was examined by means of RT-PCR/ Southern blot. All HCC patients were followed up for 36 months.
RESULTSOf the 29 HCC patients, 18 (62.1%) had recurrence, and 17 (58.6%) was found to have AFP mRNA expression in peripheral blood during postoperative period. Fourteen of these 17 patients (82.4%) with positive AFP mRNA expression developed recurrence, but only 4 patients (33.3%) with negative AFP mRNA expression had recurrence. The difference between two groups was statistically significant (P < 0.05).
CONCLUSIONOur preliminary results suggest that detection of AFP mRNA during postoperative period may be helpful in predicting recurrence for hepatic cellular carcinoma.
