1.Value of echocardiography in the diagnosis of non-compaction of ventricular myocardium

Objective To explore the echocardiography characteristics of non-compaction of ventricular myocardium(NVM),and assess its value in the diagnosis of NVM.Methods Philips SONOS 5500 color Doppler ultrasound system with 2-4 MHz transducers was used in 9 patients with NVM.Results In all 9 patients,the presence of multiple trabeculations and multiple deep intertrabecular Space communicating with the ventricular cavity was demonstrated by color Doppler imaging.Both ventricles were involved in 1 patient,only the left ventricle in 7 patients,and right ventricle in 1 patient.Conclusions NVM has typical ultrasonic appearance on echocardiography.It Can find out the NVM manifestations even if patients do not have any symptoms.It is a reliable and convenient way for diagnosing NVM.
2.Effect of wearing of soft corneal contact lens for long term on apoptosis of corneal epithelial cell

Background Wearing of soft contact lens,as one of the important methods to correct ametropia,has been widely applied at present.The clinically deleterious effects of soft contact lens have been reported,but researches about influence of apoptosis of corneal epithelial cells are scant.Objective This study was to detect and compare the ratio of apoptosis of corneal epithelial cells between the patients with contact lens wearing and without contact lens wearing,and explore the influence of soft contact lens wearing for long term on growth of corneal epithelial cells after injury.Methods A retrospective nest case-controlled study was designed.Forty eyes of 20 myopic patients with wearing of soft contact lens for ≥5 years and 40 eyes of 20 myopic patients without wearing of soft contact lens were included in Affiliated Hospital of Guiyang Medical College.All the patients received the off-flap epipolis laser in situ keratomileusis (Epi-LASIK) from 2010 October to 2011 June.The corneal flaps were collected during the surgery.The apoptosis of corneal epithelial cells was detected by flow cytometry(FCM),and the difference of apoptotic proportion between the two groups was compared using independent sample t test.Results The FCM scatter plot showed that compared to the without contact lens wearing group,the early stage of apoptotic cells with high staining annexin V and low staining proaium iodide (PI) were much more,and survival cells with low staining annexin V and PI were less,and necrotic and late stage of apoptotic cells with high staining of annexin V/PI were more in the with contact lens wearing group.The early stage of apoptosis proportion of corneal epithelial cells was (11.23 ± 5.31)% in the with contact lens wearing group,and that in the without contact lens wearing group was (7.31± 5.43)%,showing a significant difference between them (t=2.754,P=0.008).Conclusions A long term wearing of contact lens induce more apoptosis of corneal epithelial cells,which can result in a disorder of structure and function of ocular surface during the wound healing stage.
3.Clinical observation of diclofenac sodium eye drops for postoperative recurrence of pterygium

To observe the clinical efficacy of diclofenac sodium eye drops for postoperative recurrence of pterygium, and to provide safer drug regimens to prevent the recurrence of pterygium.
● METHODS: From November 2011 to September 2013, 124 patients 134 eyes who underwent surgery because of pterygium in our hospital were randomly divided into trial group ( diclofenac sodium eye drops and sodium hyaluronate eye drops) and control group ( tobramycin dexamethasone eye drops and sodium hyaluronate eye drops), all patients were followed up for 3mo ( once a week ) and the cornea, intraocular pressure and recurrence of pterygium were observed and compared between the two groups.
●RESULTS: Followed up for 3mo, 43 cases (45 eyes) in the trial group and 38 cases (39 eyes) in the control group had done the follow up as planned, in which 14 cases (15 eyes) showed the signs of recurrence, 2 cases (3 eyes) recurred eventually in the trial group, and in the meantime there were 12 cases (13 eyes) with signs of recurrence and 2 cases ( 2 eyes ) with a recurrence eventually in the control group, but no statistical difference was found between the two groups (P>0. 05);there were 6 cases (9 eyes) dropped out of the study with a higher intraocular pressure in the control group, compared to trial group (0 cases of 0 eye), the difference was statistically significant ( P < 0. 05); cornea conditions between the two groups had no statistical significance (P>0. 05).
●CONCLUSlON:Diclofenac sodium eye drops is effective and safe in preventing the recurrence of pterygium.
4.Entry of antibiotic ointment into the anterior chamber following sutureless clear corneal incisions phacoemulsification:a case report

·AIM:To report a case of entry of antibiotic ointment into the anterior chamber following sutureless clear corneal incisions phacoemulsification.·METHODS:A 62-year-old man had uneventful phaco-emulsification with clear corneal incisions with intraocular lens(IOL) implantation. Postoperative medication included antibiotic/steroid ointment; the eye was firmly patched at the end of the procedure. On the first postoperative day, the patient was seen an oily droplet floating in the anterior chamber. Immediate surgical removal of the ointment without IOL exchange was performed.·RESULTS:During follow-up for six months after irrigation/aspiration of the anterior chamber and removal of the oily droplet, visual acuity and intraocular pressure (IOP) were unchanged and anterior segment was formed and quiet.·CONCLUSION:Ophthalmic ointment may ingress into the anterior chamber through unstable clear corneal incisions. The case highlights the importance of appropriate wound construction and integrity, as well as the risk of tight eye patching following placement of ointment.
5.Effect of complex prescription of Chinese medicine on ischemic brain of rat

ObjectiveTo study the protection of Chinese herbs on the ischemic brain of rats. Methods 75 Wistar rats were divided into 5 groups, Group 1 for false operation. For the other four groups, the common carotid artery was exposed then was ligatured and cut off, Group 2 for model. From the 20th hour after operation on,Group 3, 4, 5 were fed with complex prescription of Chinese medicine named Prescription 1, 2 and 3 once a day respectively. 3 hours after taking medicine, Groups 2-5 were put in the hypoxia environment for 1 hours, then taking the medicine for 7 days. On the 7th day after operation,the blood was taken from R. atria then the rats were killed and the whole right brains were cut off. Malonaldehyde (MDA), notric oxide synthetase (NOS) and superoxide dismutase (SOD) in the brain tissue and serum, and calcium in the brain were measured respectively. ResultsThe three prescriptions can decrease the quantity of MDA both in brain tissue and serum and the calcium in brain tissue(P<0.05-0.001).Prescription 1 can enhance the activity of SOD in brain tissue while the others can decrease the activity of NOS. The hippocampus cells show tidy, and the number of the necrotic cells decrease greatly among them, Groups 4 and 5 are better than Group 3.Conclusions Prescriptions 1-3 can protect the brain tissue of the rat form ischemic brain injury.
6.Comparison of Rhizosphere Bacteria Diversity Between Fusarium Wilt Resistant and Susceptible Watermelon

The traditional culture methods and the molecular biology methods were used to study the rhizosphere bacterial diversity between fusarium wilt resistant and susceptible watermelon. The results showed that the diversity and the equality of cultured rhizosphere bacteria of resistant watermelon were higher than those of the susceptible watermelon. The reason was that the cultured rhizosphere bacterial di- versity index H′ and 1/D of the resistant watermelon were higher than those of the susceptible watermelon and that the cultured rhizosphere bacterial equality index E of the resistant watermelon were higher than those of the susceptible watermelon. The dominant cultured bacterial genotypes were different between re- sistant and susceptible watermelon. The genotype 1 is the dominant genotype of resistant watermelon, con- sists 51.1%. The genotype 7 is the dominant genotype of susceptible watermelon, consists 58.7%.
7.Human Umbilical Venous Endothelial Cells as Feeder Layer to Support the Growth of Embryonic Stem Cells

Objective To explore whether human umbilical venous endothelial cells could be used as feeder layer to support the growth of embryonic stem cells (ESC) and keep ESC undifferentiated.Methods The venous vessels of umbilical cord obtained from healthy puerperal were perfused with collagenase.The isolated endothelial cells went through primary culture and passages for expansion.Factor Ⅷ antigens determination was implemented.Endothelial cells with good growth and 3 or above passages were treated with mitomycin-C(10 mg/L) and prepared as feeder layer,on which E14.1 ESC was transplanted for subculturing to observe the morphological characterization and determine ESC alkaline psphatase (AKP) activity and the expression of stem cell marker Oct-4.Severe combined immune deficiency(SCID) mouse in vivo terotoma formation experiment was performed to identify its pluripotent properties.Results Human umbilical vein-derived endothelial cells grew well in culturing in vitro and regenerate in large numbers.The endothelial cells maintained normal cellular morphological and biological characterization after 10 passages.The cells stopped proliferating after being treated with mitomycin-C,but its activity and morphological properties were well-maintained with 24 hours,which was a fundamental property of serving as feeder layer.E14.1 ESC remained undifferentiated in human umbilical venous endothelial cells after 3-8 passages,the cells grew in colony and showed high expression of AKP and stem cell Oct-4.In vivo pluripotency experiment showed that 6 weeks after being transplanted to SCID mice E14.1 ESC of 6 and 10 passages in endothelial cells both could form teratoma containing 3 layers of tissue cells.Conclusions Human umbilical venous endothelial cell serve as a convenient feeder layer cell with rich sources.It can effectively support ESC growth and heterogenous and prevent the heterogeneous protein pollution and pathogenic microorganisms caused by animal cell feeder layers,thus solve the problem of biological safety of ESC clinical application.
8.Effect of bone morphogenetic protein-7 on aristolochic acid induced renal tubular epithelial cells transdifferentiation.

OBJECTIVETo observe the effect of bone morphogenetic protein-7 (BMP-7) on aristolchic acid induced renal tubular epithelial cell trans-differentiation to look for new therapeutic approach for aristolchic acid nephropathy (AAN).
METHODSIn vitro cultured human proximal renal tubular epithelial cell line HK-2 cells were treated with different concentrations of BMP-7 (75 ng/mL, 150 ng/mL and 300 ng/mL) after trans-differentiation of the cells was induced by AA (10 microg/mL). Levels of alpha-SMA mRNA and protein expressions were detected by semi-quantitative reverse transcription polymerase chain reaction (RT-PCR) and Western blotting respectively.
RESULTSBMP-7 reversed the AA inducing alpha-SMA expressions in HK-2 cells in a dose-dependent manner.
CONCLUSIONBMP-7 can inhibit the trans-differentiation of human renal tubular epithelial cell induced by AA, thereby might be a new potential drug for AAN prevention and treatment.

9.Experimental study on the pathological rat modelof paraquat-induced acute lung injury and pulmonary fibrosis

Objective:To determine the half lethal dose(LD50) of paraquat in rats and to establish a relatively safe and stable pathological animals model of pulmonary fibrosis.Methods: Ninety-six SD rats totally in half genders.Fifty SD rats in half genders were randomly divided into 5 groups,each had 10 rats.Feed the rats with different doses of liquor of paraquat intraperitoneally one time and definite the half lethal dose of one and two weeks.After that,prepare another forty-six SD rats,also in half genders,as intonication group,twenty-eight rats were treated with the liquor of paraquat in dosage of 18 mg/kg intraperitoneally.As control group,sixteen rats were treated with equivalent volume of normal saline.Observe the toxic symptom daily and rats were sacrificed on day 1,3,5,7,14,21,28,35 and 42 respectively for the histological examination.Results: The half lethal doses of intraperitoneal paraquat of 1 and 2 weeks were 18.27 and 17.29,with 95% confidence intervals of 16.61-20.09 and 15.99-18.67,respectively.After intraperitoneal paraquat injection at the dose of 18 mg/kg,typical toxic symptoms were observed at different times in the rats.The whole process of acute lung injury and fibrosis induced by paraquat intoxication could be seen with the naked eyes or under the light microscope.Conclusion: Paraquat has a strong toxicity to rats.A proper dose of paraquat solution can not only reduce the number of experimental rats,but also induce typical pulmonary fibrosis in rats.
10.Study on in vivo pharmacokinetics of cucurbitacin injection in rats.

To establish a method for the determination of cucurbitacin in plasma samples, in order to study the in vivo pharmacokinetic characteristics of cucurbitacin in rats. Rats were intravenously injected with cucurbitacin. With diphenhydramine as the internal standard (IS), the plasma concentrations of cucurbitacin in rat plasma at different time points were determined by liquid chromatography tandem mass spectrometry (LC-MS/MS). With electrospray ionization source, the positive ion detection in the multiple reaction monitoring mode was conducted to determine the ion-pairs for target compound and IS were m/z 503.2/113.1 and m/z 256.0/167.2, respectively. Agilent ZOBAX SB-C18 column (2.1 mm x 50 mm, 1.8 microm) was adopted and eluted with methanol and 0.1% formic acid (55:45), and the flow rate was 0.2 mL x min(-1). DAS 2.0 software was applied to fit the blood concentration and calculate corresponding pharmacokinetic parameters. The rats were intravenously injected with cucurbitacin at the concentration of 3.0 mg x kg(-1). The target blood quality concentration show good linear relations within the range of 10.5-3 150 microg x L(-1) (R2 = 0.996), the lower limit of the standard curve was 10.5 microg x L(-1), and the signal to noise ratio S/N = 12. Intra- and inter-day precisions RSD was less than 6.9% and 14%, respectively; The accuracy RE ranged between 0.20% and 3.7%; The extraction recoveries ranged between 92.7% and 97.1%. Regarding the pharmacokinetic parameters of tail intravenous injection of cucurbitacin, AUC (0-t) was (811.615 +/- 111.578) microg x h x L(-1), (t1/2) was (1.285 +/- 1.390) h, CL was (3.627 +/- 0.487) L x h x kg(-1), and V(d) was (6.721 +/- 7.429) L x kg(-1). In this study, researchers established a simple, accurate, sensitive and highly specific method for determining the blood concentration of cucurbitacin, and reported the in vivo pharmacokinetic characteristics of cucurbitacin in rats for the first time.
