1.Adaptive Behavior Assessment of Mental Retardation(review)

Adaptive behavior is attracting more and more attention and have become necessary basis of diagnosis and intervention of mental retardation. The definition of adaptive behavior have not unified yet, AAMR(2002) suggests that adaptive behavior is the skill of concept, society and use, the constraint of adaptive behavior will influence individual's daily life, and then life adaption. Approachs of mental retardation assessment include interviews, behavioral observations, functional assessment, quality of life assessment and sociometric techniques. No matter which approach is used in assessment, the goal is to improve lives of individuals with mental retardation, the limits of each approach should be considered, and, the assessment of individuals with mental retardation must contain all aspects.
2.Polysaccharides activate signaling pathways of macrophage.

Polysaccharides extracted from various sources are natural active substances, which may lead to the activation of macrophage via multiple pathways and mechanisms. This article intends to illustrate the signaling pathways of polysaccharides from plants, fungi, algae and other sources, to identify the mechanisms on the molecular level, and to explore the novel target immunomodulatory agents.

3.Design, synthesis and antiproliferative activities of artemisinin derivatives substituted by N-heterocycles.

Increasing attention has been focused on the antitumor activity of artemisinin derivatives in recent years, for artemisinin had been reported to have cytotoxic effects against HL-60, P388 and MCF-7 tumor cells. We report here the synthesis and evaluation for antitumor activity of a series of artemisinin-ether derivatives bearing tetrahydropyrrole, morpholine, piperidine, substituted piperidines and azoles with various linkers. Sixteen 10-O-substituted dihydroartemisinin derivatives were designed and synthesized, all of which have never been reported in literatures and whose antiproliferative effects on human breast cancer MCF-7, MCF-7/Adr and HL-60 cells were determined by MTT assay or direct cell counting. Each of these artemisinin derivatives possessed better effects than dihydroartemisinin evidently against HL-60 and MCF-7 cells growth, while less potent than doxorubicin. All target compounds exhibited significantly improved potency compared to DHA and doxorubicin on the doxorubicin-resistant MCF-7/Adr cells, so did they in their sensitive counterparts MCF-7 cells. Among them, compounds GF02, GH04 and ZH04 showed strong activity against these three cell lines growth. Further research is undergoing.

4.Stimulation by Lycium bararum polysaccharides of the maturation of dendritic cells in murine bone marrow.

OBJECTIVETo study the effect of Lycium bararum polysaccharides (LBPs) stimulation on the maturation of murine bone marrow derived dendritic cells (BMDCs).
METHODSMurine bone marrow cells were cultured in GM-CSF and IL-4 for 5 days, then were purified with a MACS column. Respectively, BMDCs were stimulated with LBPs, LPS and RPMI1640 for 2 days. Cell phenotypes and antigens uptake by BMDCs were analyzed by flow cytometry. Cytokines released by BMDCs were detected. The antigen presenting by BMDCs was evaluated by mixed lymphocyte responses.
RESULTCompared with to the BMDCs that only subjected to RPMI 1640, the expression of I-A/I-E, CD11c and secretion of IL-12 by BMDCs stimulated with LBPs were increased, the phagocytosis of FITC-dextran by BMDCs stimulated with LBPs was impaired but the activation of proliferation of allogenic lymphocytes by BMDCs was strengthened.
CONCLUSIONLBPs promote not only the maturation of cultured murine BMDCs in vitro, but also the immune response initiation induced by BMDCs.

5.Based on supramolecular chemistry to explore the scientific connotation of predecocting gypsum in Maxingshigan decoction preliminarily

It has gradually become a consensus in the industry that the traditional Chinese medicine gypsum should be decocted first, but the understanding of decocting method is not completely unified in the works of doctors since ancient times, and there are occasional disputes about whether it is necessary to decocting first. In this study, the phase determination, physical and chemical characterization, qualitative and quantitative analysis of inorganic and organic components of the decoctions of herbal pairs and the whole prescription Maxingshigan decoction with gypsum as the center, and the pre-decoctions and co-decoctions of them were carried out to explore the scientific connotation of the pre-decoctions of gypsum. Results show that decoction phases were different between the co-decoctions and pre-decoctions of licorice-gypsum (Gancao-Shigao, GC-SG), ephedra-gypsum (Mahuang-Shigao, MH-SG) and almond-gypsum (Xingren-Shigao, XR-SG). The results of the micromorphology, particle size and zeta potential of herbal pairs and prescription (Quanfang, QF) showed that the supramolecular particles in pre-decoctions were smaller, more uniform and more stable than the co-decoctions. The results of organic components analysis showed that different cooking methods did not change the organic composition and content. ICP-OES results showed that the content of inorganic components in pre-decoctions was higher than in co-decoctions for the same boiling time of gypsum. The IR results showed that the pre-decoctions had stronger chemical functional group effect than the co-decoctions. To sum up, compared with the co-decoction, the pre-decoction of gypsum has different phase state and chemical composition interaction, and the difference of inorganic composition is an important material basis affecting the change of phase state compared with the co-decoction. It indicates that the material basis of traditional Chinese medicine decoction is indeed different whether gypsum is decocted first or not, which can provide a basis for the clinical application of decocted gypsum.
6.Molecular characterization of hantavirus Shandong isolate JNL virus strain.

OBJECTIVETo understand the molecular epidemiologic characteristics of hantavirus Shandong isolate JNL virus strain.
METHODSThe complete M and S gene of the JNL virus isolated from Shandong Province was amplified by RT- PCR, and the purified PCR product was cloned into T vector for sequencing.
RESULTSThe results revealed that the JNL M segment was 3615 bp in length, encoding 1135 amino acids, and the S segment was 1698 bp encoding 429 amino acids, JNL belongs to HTN virus. The comparison of homology with HTN and SEO types showed that the difference of M and S complete sequences between JNL and all other HTN virus strains reached 20.0%-20.6%, and 15.5%-16.0%, respectively. Phylogenetic tree also showed that the position of JNL is located at a different clade.
CONCLUSIONSHTN virus Shandong local isolate JNL strain is a new specific HTN subtype virus.

7.Construction and development of a training system for military pharmacy professionals

Amid continuous development of new military revolutions,the cultivation of high-quality professionals practical ability is becoming increasingly important.By learning from the successful experience and practices from foreign systems for training practical skills of graduate students majoring in military pharmacy,the education mode and methods for training military pharmacy postgraduates were discussed in this paper in order to optimize the training mode of practical skills for students of military pharmacy,explore a suitable training mode in China,improve teaching methods and formulate a set of scientific and optimized strategies.It is hoped that the practical ability of military pharmacy graduate students will be improved considerably.
8.Establishment and application of a novel method for detecting MLL fusion genes of acute leukemia.

This study was aimed to establish an efficient method to detect 10 common MLL fusion genes in patients with acute leukemia. Firstly, the relevant references and databases were searched to thoroughly investigate all fusion breakpoints; the primers and probes were designed according to nearly all the involved fusion types of gene. Then the multiplex real-time PCR system was established and optimized by using the established 16 positive plasmids and negative cell lines. Finally, the detection system was clinically evaluated by means of collected 54 samples of leukemia. The results indicated that the established detection system could efficiently detect all positive plasmids with sensibility to 10 copies. Four kinds of fusion gene types such as MLL-AF4, MLL-AF9, MLL-AF10, MLL-ELL could be detected in 54 samples, the sequencing of positive samples showed consistency of sequencing results with detection results. It is concluded that a novel multiplex real-time PCR detection method is established which can detect 10 common MLL fusion genes covering about 90% of the cases harboring MLL fusions. This method is fast, sensitive, specific and reliable, and should be an useful clinical tool for identification and management of leukemia patients with MLL fusions.

9.Study on TCM Physical Personality and Constitution Based on Diabetes Mellitus Complicated with Coronary Heart Disease Risks Predicted by Decision Tree Model

OBJECTIVE To study the prediction effect of TCM physical personality and constitution on the risks of MD complicated with CHD.METHODS TCM physical personality and constitution of 110 T2MD patients were tested by five pat-tern personality inventor and five-five constitution.CHAID model of decision tree in EXCEL and SPSS was used to establish disease risk model of MD complicated with CHD predicted by TCM personality and constitution types.And the prediction ac-curacy of the model was evaluated.RESULTS Tested by cross validation,the prediction accuracy of MD disease model estab-lished with the Shao-yin personality,Yin-cold constitution and blood-stasis constitution as the predictor variables was relatively higher(93.6%),and three rules for predicting CHD risk were obtained.CONCLUSIONS The prediction rules of the MD complicated with CHD by TCM personality and constitution are consistent with the traditional understanding of Chinese medi-cine,which provides a reference for high-risk population's early screening,CHD prevention and treatment,clinical differentia-tion and treatment.
10.Effect of hepatocyte growth factor gene transfection on biological features of lymphoma cells.

OBJECTIVETo investigate the biological effect of hepatocyte growth factor (HGF) on HGF gene-transfected Raji cells.
METHODSTotal RNA was extracted from human hepatic tissue, HGF gene cDNA was amplified by RT-PCR, and then cloned into vector pVITRO2-mcs to construct recombinant eukaryotic expression vector pVITRO2-mcs-HGF. The recombinant vector was transfected to Raji cells, and the stably transfected cells were selected by homomycin B in serial passages, and confirmed by real-time fluorescent quantitative PCR, ELISA, immunocytohistochemistry. The biological features of transfected Raji cells were evaluated by semisolid culture.
RESULTSRT-PCR results showed that Raji cells were transfected successfully with recombinant eukaryotic expression vector pVITRO2-mcs-HGF. HGF mRNA and protein were expressed successfully in Raji cells. Expression of HGF gene enhanced proliferation, metastasis and invasion of Raji cells.
CONCLUSIONHGF gene has been cloned and recombined to construct recombinant eukaryotic expression vector pVITRO2-mcs-HGF successfully. Transfected HGF may change the biological features of Raji cells.
