Objective To identify the miR-122 which regulateing GATA4 expression during the induction of rat bone marrow mesenchymal stem cells (bMSCs) differentiating into cardiomyocyte-like cells. Methods BMSCs were isolat-ed from bone marrow and induced to differentiate into cardiomyocyte-like cells using 5-azacytidine. The miR-122 which may regulate expression of GATA4 were predicted using miRanda and TargetScan softwares and identified by dual luciferase report system. The expressions of miR-122 and GATA4 were determined using q-PCR during the differentiation of bMSCs into cardiomyocyte-like cells. Results The induced cells were completely in contacted with adjoining cells and uniform in shape and aligned parallelly. Cardiac troponin I (cTnI) expression was detected by immunofluorescence cytochemistry. Using dual luciferase reporter system in vitro, miR-122 were proved to be able to effectively inhibit GATA4 expression by binding the 3′UTR of GATA4 mRNA. q-PCR results showed that the expression of miR-122 is negatively correlated with that of GATA4 mRNA transcription. Conclusion These results indicated that miR-122 regulate the expression of GATA4 during the induction of cardiomyocyte-like cells.

Chinese Journal of Epidemiology 2008;29(12):1230-1234
Objective To explore the molecular types of methicillin-resistant Staphylococcus aureus (MRSA) strains present in major hospitals in Qingdao area, using pulsed-field gel electrophoresis (PFGE) and multilocus sequence typing (MLST) methods, trying to find out the epidemiological characteristics of these MRSA isolates. Correlation of the PFGE types with microbiological phenotypes and clinical data was also studied. Methods 360 isolates of MRSA were procured during 2003 to 2007 from major hospitals in Qingdao. PFGE technology was applied to comparatively analyze the chromosomal DNA digested with endonuclease Sma Ⅰ . Comparison of DNA fragments patterns from each MRSA strain and cluster analysis were performed with the Bionumericus version ' 2.0' software. A dendogram was generated using PFGE macrorestriction fragments on gel images. Data was used to predict the possibility of each PFGE type via SPSS software version 11.0, using the variables as predictors including groups on patient's age, gender, source and the site where MRSA was isolated. Antibiotic sensitivity patterns of these MRSA isolates were determined by K-B tests, and a correlation between these patterns and PFGE types was investigated. Housekeeping genes were amplified with PCR and sequenced in representative strains of variant PFGE types to identify their allelic profile. Results 5 types of PFGE patterns (M0-M4) were identified with MI being the predominant and M2 next to it which was significantly correlated to the isolates from wounds. M3 type strains were mainly isolated from ICU wards and there were a few cases complied with M4 type with no correlated variant factors found in this study. A unique pattern of MRSA isolates with its M0 distinct from other types had not been reported. No significant association was found between PFGE individual types, gender or age groups. M1 and M2 types were the major proportional PFGE patterns among different hospitals. No vancomycin-resistant isolates were detected among 360 MRSA strains. No significant association was found between individual antibiotic resistance and specific PFGE types. Data from MLST analysis showed that the aUelic profiles of M1 and M3 type strain had the same ST239 linage which was commonly present in China. For M2 and M4 representative strains, the allelic profiles were ST5 and ST240, respectively. ST45 and ST398 were corresponding to two PFGE patterns clustered as M0 type. Conclusion Nosocomial infection due to MRSA was evenly distributed among different age groups and no gender bias was observed. The PFGE types of MRSA strains isolated in major hospitals in Qingdao were highly correlated with the sources of isolates and ST239 isolate seemed the prevalent and widespread one. Strategies should be designed to further monitor and prevent or minimize the spread of ST5 MRSA isolates and the like, in Qingdao area.

Objective To investigate fracture resistance and fracture patterns of maxillary anterior endodontically treated teeth restored with two different shapes of glass fiber post systems. Methods Twenty-four human sound maxillary anterior teeth of similar size were collected, and randomly divided into 3 groups,8 teeth each. After endodontical treatment, they were given the following treatments:Group A:parallel glass fiber posts (coltene parapost fiber lux) and composite core;Group B:taper glass fiber posts (coltene parapost taper lux) and composite core; Group C (control group):intact endodontically treated teeth.Teeth in Groups A and B reserved 2 mm dentin ferrule. Then the teeth were prepared and restored with IPS e.max Press all ceramic crowns. All the teeth were embedded in acrylic resin 2 mm below the cementoenamel junction (CEJ),with the silicon impression material stimulating periodontal membrane. All the specimen were loaded under a universal testing machine,at the palatal junction of incisor third and middle third, with an angle of 135 degree to the longitudinal axis of the tooth until fracture occurred,at a cross-head speed of 1.0 mm/min.Fracture loads and patterns were recorded.Statistical analysis was performed using SPSS software. Results The mean fracture loads in the three groups were as follows:(468.8±142.5) N,(440.2±202.7) N,and (459.0±147.6) N,respectively. There was no significant difference in fracture loads among groups (P>0.05).The incidence of unfavorable fracture in Group A was higher than Groups B and C.Conclusions There is no significant difference in fracture strength between parallel and taper fiber post groups. However, the group with parallel fiber posts demonstrates a higher risk of unfavorable fracture than the group with taper fiber posts, which indicates that taper fiber posts are more favorable to preserve the remaining tooth structure.
