Objective To explore the effect of hyperoxia through mechanical ventilation induced lung injury(VILI) in newborn rabbit. Methods One hundred newborn rabbits aged 1-5 days were randomly divided into 4 groups: with 900 mL/L O2 and mechanical ventilation(MV),group A received high peak inspiratory pressure(HPIP),group B received moderate peak inspiratory pressure(MPIP),group C received low peak inspiratory pressure(LPIP),and group D with no mechanical ventilation with room air. There were 30 rabbits in group A,B,C,10 rabbits in D group. All rabbits were killed at 1,3,6 h after trial respectively.Wet-to-dry weight ratio(W/D) of left lung, and white blood cell(WBC)counts in broncho alveolar lavage fluid(BALF) were measured. The changes of lung histopathology were assessed by hematoxylin-eosin(HE) staining and observed under light microscope. Results Compared with group B and C,the group A demonstrated more pulmonary hemorrhage and pulmonary bullae formation, and the WBC and neutrophils counts in BALF increased and the W/D was higher compared with group B and group C.Meanwhile, alveolar epithelial cellⅡ(AECⅡ) hyperplasy transformed to AECⅠ.In group C,AECⅡswelled and lung tissue edema showed obviously, after 6 hours collagen fibers hyperplasia appeared.Part pulmonary atelectasis was obvious in group C.Conclusions HPIP ventilation can increase lung injury induced by hyperoxia in newborn rabbits, but has minimal effects on MPIP and LPIP ventilation.Pulmonary histopathologic changes participate in the newborn rabbit machine ventilation induced lung injury.

Objective:To observe the changes of visual evoked potential(VEP)in diabetic rats and the influence of nerve growth factor(NGF)and aminoguanidine(AG)on VEP.Methods:Diabetes was induced in adult male Wistar rats with streptozotocin(STZ).Rats were divided into normal control group(CON),diabetes model group(DM).NGF-treated group(D +N)and AG-treated group(D+A).VEP was measured during the 3~(rd)month,6~(th)month,9~(th)month,and 12~(th)month.Results: Compared with the CON group, all rest groups had longer latencies and lower amplitudes(P

Objective To investigate the developmental characters of neural stem cells(NSCs) in occipital of cortex from human fetal brain at different age. Methods Ninety cases of embryoes at gestational age 16-32 weeks and by induction of labor with water bag were collected for determining distribution, shapes, growth modes and the number of NSCs in the occipital of cortex with immunohisto- chemical method under light microscope. Results It was noted that NSCs existed in the occipital of cortex from human fetal brain at different ages.NSCs mainly distributed in layers of cone cells and inner granule cells.NSCs existed in the occipital of cortex of different fetal age included middling round cells,NSCs had enations from 0 to 1.Nucli were larger than plasm. Each NSC had nucleoli from 2-4 and rarefaction chromatin. Most of NSCs distributed in three growth modes including crowd, cluster and clone, occasionally with a single growth mode among other nerve cells. There were no differences including distribution, shapes, growth modes and the number of NSCs in the occipital of cortex between groups, but,NSCs gradually decreased with increasing of age. Conclusion NSCs exists in the occipital of cortex from different gestational age,and the number of NSCs decreases with increasing of age.

Organic anion transporting polypeptide 1B1 (OATP1B1) is an important liver-specific uptake transporter, which mediates transport of numerous endogenous substances and drugs from blood into hepatocytes. To identify and investigate potential modulators of OATP1B1 from natural products, the effect of 21 frequently used natural compounds and extracts on OATP1B1-mediated fluorescein methotrexate transport was studied by using Chinese hamster ovary cells stably expressing OATP1B1 (CHO-OATP1B1) in 96-well plates. This method could be used for the screening of large compound libraries. Our studies showed that some flavonoids (e.g., quercetin, quercitrin, rutin, chrysanthemum flavonoids and mulberrin) and triterpenoids (e.g., glycyrrhetinic acid and glycyrrhizic acid) were inhibitors of OATP1B1 with IC50 values less than 16 µmol · L(-1). The IC50 value of glycyrrhetinic acid on OATP1B1 was comparable to its blood concentration in clinics, indicating an OATPlB1-mediated drug-drug interaction could occur. Structure-activity relationship analysis showed that flavonoids had much higher inhibitory activity than their glycosides. Furthermore, the type and length of saccharides had a significant effect on their activity. In addition, we used OATP1B1 substrates fluvastatin and rosuvastatin as probe drugs to investigate the substrate-dependent effect of several natural compounds on the function of OATP1B1 in vitro. Our results demonstrated that the effect of these natural products on the function of OATPlB1 was substrate-dependent. In summary, this study would be conducive to predicting and avoiding potential OATP1B1-mediated drug-drug and drug-food interactions and thus provide the experimental basis and guidance for rational drug use.

Objective To investigate whether leukotriene D4 (LTD4) regulates eotaxin-3 (Eot-3) expression in bronchial epithelial cells, and study effect of pranlukst on the regulation. Methods BEAS-2B cells and normal human bronchial epithelia cells were pre- treated with LTD4 for 1 hour, stimulated with interleukin-4, the cells were incubated for 24 hours. Eot-3 protein in supernatant were measured by enzyme linked immunosorbent assay(ELISA). The cells were pretreated with pranlukast in different concentration, then the above procedure was repeated. Results The untreated bronchial epithelial cell expressed Eot-3 protein on a very low level. After stimulating with IL-4 and incubating for 24 hours, Eot-3 production increased significantly. Pretreating the cells with LTD4 enhanced the inducing effect of IL-4. Pranlukast inverted the upregulation of LTD4. Conclusions Upregulating the expression of Eot-3 induced by IL-4 on bronchial epithelial cells may explain partially the mechanism of leukotrienes involving airway allergic inflammation of asthma. The invertion impact on upregulation of LTD4 by pranlukast may be one of mechanisms that leukotrienes receptor antagonist cure asthma.

Objective To investigate the changes of insulin-like growth factor-1(IGF-1) and its associated protein in Balb/C mice with viral myocarditis. Methods Sixty Balb/C mice were randomly divided into 2 groups:infected group(n=40) and control group(n=20).Mice in infected group were inoculated intraperitoneally with coxsackievirus B3(CVB3) while control mice with 0.1mL of Eagle′s minimal essential medium(EMEM).Blood and myocardial musculature of all mice were collected on the day of 3,7,15 and 30 after inoculation. The expression of IGF-1 and its associated protein in plasma or myocardium were detected respectively by enzyme-labeled immunosorbent assay(ELISA),immunohistochemistry and reverse transcription polymerase chain reaction(RT-PCR).Results The expression of IGF-1 and its associated protein of mice with viral myocarditis was higher than those in control group and gradual ascent after mice being infected with virus. The expression reached summit on d15,then decreased on d30,but still sustain in a high level. The difference was significant(F=19.53 P
