AIM: To investigate the effect of low intensity white light irradiation on the retinas of mice.
METHODS: Thirty C57BL/6J mice were randomly divided into two groups. The number of the mice in each group was 15. The mice in experimental group received dark adaptation from 5:00p. m. to 6:00p. m. , and then exposed to LED white light from 6:00p. m. to 7:00p. m. everyday for a month. At 1, 3, 7, 14 and 30d after the beginning, we examed the histology of mice retinas, calculated the thickness of outer nuclear layer ( ONL ) , inner nuclear layer ( INL ) and analyzed electrophysiology of mice.
RESULTS:One month after experiment, compared to the control group, the latency of Rod-R a wave of the mice in experimental group significantly prolonged, the amplitude of Cone-R b wave of the mice in experimental group significantly decreased and the latency of b wave of the mice in experimental group significantly prolonged ( P<0. 05). There are no significant difference in the histology of retina, ONL and INL thicknesses.
CONCLUSION: 100lux low intensity white light could give rise to the impairment of the retinal functions in dark-adapted mice.

OBJECTIVETo observe the curative effects of combined therapy with Kangyanling (KYL, a Chinese herbal preparation) and Omeprazole on post-burn digestive dysfunction.
METHODSPatients with post-burn digestive dysfunction were assigned to two groups, the 32 in the treated group, including 18 with acute stress gastrointestinal mucosal hemorrhagic lesion and 14 with toxic enteroparalysis, were treated by KYL plus Omeprazole, and the 20 patients in the control group, 11 with acute stress gastrointestinal mucosal hemorrhagic lesion and 9 with toxic enteroparalysis were treated with Omeprazole alone. The pH value in gastric mucosa was determined before and 12 h after treatment, the hemostasis effects in 48 h, and the anti-paralysis effects in 72 h were observed as well.
RESULTSThe pH value in gastric mucosa of both groups before therapy were all lower than the normal range, it raised after treatment in the treated group (P < 0.05), approaching to the normal range, but with no significant change in the control group. The total hemostatic rate and the anti-paralysis rate was 77.8% and 85.7% respectively in the treated group, and 45.5% and 0% in the control group, all shown statistical significance between groups (P < 0.05).
CONCLUSIONCombined therapy with Kangyanling and Omeprazole has obvious curative effects on post-burn gastric dysfunction.

Objective To find out the cut-off value of rose bengal plate agglutination test(RBPT) in rapid diagnosis of brucellosis. Methods From May to June 2009,398 people who came to the outpatient department of Inner Mongolia Center for Endemic Disease Control and Research were diagnosed brucellosis by RBPT and tube agglutination test (SAT).Tube agglutination test as a gold standard, rose bengal plate agglutination test, its authenticity and reliability were evaluated. Results Taking positive predictive value 100.0% as the selection standard, the cut-off value of RBPT was ++ which could be used to diagnose brucellosis. The sensitivity was 83.3%; the specificity was 100.0%; the Youden index was 0.832; and the compliance rote was 89.9%.Conclusion The cut-off value ++ of RBPT to diagnosis brucellosis is worthy of clinical promotion.

Objective To evaluate the effect of multiplex polymerase chain reaction(Multiple-PCR) in identification of Brucella strains. Methods Six standard Brucella strains (Brucella abortus,Brucella melitensis,Brucella suis,Brucella canis,Brucella ovis,Brucella neotomae) were used as positive controls and Escherichia coli O∶157 and Yersinia enterocolitica O∶9 were used as negative controls.A total of 29 Brucella strains were tested.Brucella strains were amplified by BCSP31-PCR and the species of Brucella with positive results were identified with Multiple-PCR method. Results The results of all 29 amplified Brucella strains were positive with BCSP31-PCR.The identified results of all Brucella strains were positive with Multiple-PCR,including 20 strains of Brucella melitensis,5 isolates of Brucella suis,3 strains of Brucella abortus and 1 strain of Brucella canis. Conclusion Multiple-PCR method is a rapid, specific, simple and low risk method for identification of Brucella species.

Mesaconitine was incubated with rat liver microsomes in vitro. The metabolites of mesaconitine in rat liver microsomes were identified by ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) method with high resolution power. A typical reaction mixture of 100 mol L-1 Tris-HCI buffer (pH 7.4) containing 0.5 gL-1 microsomal protein and 50 micro molL-1 mesaconitine was prepared. The above reaction mixture was divided into six groups, and the volume of each group was 200 micro L. The incubation mixture was pre-incubated at 37 degrees C for 2 min and the reactions were initiated by adding NADPH generating system. After 90 min incubation at 37 degrees C, 200 micro L of acetonitrile was added to each group to stop the reaction. The metabolites of mesaconitine were investigated by UPLC-MS/MS method. Mesaconitine and 6 metabolites M1-M6 were found in the incubation system. The structures were characterized according to the data from MS/MS spectra and literatures. The metabolic reactions of mesaconitine in rat liver microsomes included the demethylation, deacetylation, dehydrogenation and hydroxylation. The major metabolic pathways of mesaconitine in rat liver microsomes were determined by UPLC-MS/MS on multiple reaction monitoring (MRM) mode combined with specific inhibitors of cytochrome P450 (CYP) isoforms, including alpha-naphthoflavone (CYP1A2), quinine (CYP2D), diethyldithiocarbamate (CYP2E1), ketoconazole (CYP3A) and sulfaphenazole (CYP2C), separately. Mesaconitine was mainly metabolized by CYP3A. CYP2C and CYP2D were also more important CYP isoforms for the metabolism reactions of mesaconitine, but CYP1A2 and CYP2E1 haven't any contribution to MA metabolism in rat liver microsomes.

Objective To investigate the epidemiological characteristics of human brucellosis in Ulanqab of Inner Mongolia. Methods Three hundred and twenty patients with suspected brucellosis were selected, who had registered in the Ulanqab Center for Endemic Disease Control of Inner Mongolia from April to June 2011.The investigation covered general situation, such as gender, age, occupation and main clinical symptoms and so on.Blood samples were collected, and Rose Bengal plate agglutination test(RBPT) was used for serum screening. Those who were tested positive in RBPT were confirmed with tube agglutination test (SAT).Brucellosis was diagnosed according to Diagnostic criteria for Brucellosis (WS 269-2007).Data were analyzed with statistical software(SPSS 17.0).Results One hundred and thirty-four cases were positive in RBPT of the 320 people surveyed,of which 93 cases were positive in SAT; antibody titers were higher than 1 ∶ 100(++),therefore they were diagnosed as brucellosis, and the ratio was 29.1%(93/320).The number of patients with suspected brucellosis who were negative in SAT test was 41,and the ratio was 12.8% (41/320).Among the 93 people who were infected, the constituent ratio of farmers and herdsmen who engaged in livestock was the highest, accounted for 63.4%(59/93) and 24.7% (23/93) of the total number of patients ; infection rate of male (30.9%,55/178) was higher than that of females (26.7%,38/142) ; the number(39) of brucellosis patients who were over the age of 51 was the highest, and the ratio is 42.0%.The onset season mainly in May and August; main route of exposure was bare hands lambing, midwifery and contact with infected sheep pollutants. Conclusions Sheep is the main source of human Brucella infection in Ulanqab. It is the key to control the spreading of brucellosis through improving awareness of disease prevention among farmers and herdsmen as well intensifying the prevention and control of Brucella infection between livestock.
