1.Effect of Asi-antidiarrheal capsule on gastrointestinal goblet cell of thyroid hormone-induced diarrheic rats

Objective To study the effect of Asi-antidiarrheal capsule on gastrointestinal goblet cell of thyroid hormone-induced diarrhea.Methods Total of 120 SD male rats aged about 8 weeks were randomly divided into 2 groups:control group(10 rats)and thyroid hormone-induced diarrheic group(110 rats).Rats in control group were lavaged with normal saline 1 ml/d.Thyroid tablets were partly desolved into normal saline forming a 40 mg/ml suspension.Rats in thyroid hormone-induced diarrheic group were given the thyroid suspension 1 ml/d to make thyroid hormone-induced diarrheic model.Serum FT3 and FT4 were tested.Fourty thyroid hormone-induced diarrheic rats were screened out according to serum FT3 and FT4 levels,body weight and wet stool.The fourty rats were randomly divided into 5 groups,8 rats in each group:positive control group,berberine group,low-dose,mediandose and high-dose groups.Normal saline of 1 ml/d was admnistered to diarrhea control group,1.94 g·kg-1·d-1 Berberine capsule was given to positive control group,and 0.63,1.26,2.52 g·kg-1·d-1 Asi-antidiarrheal capsule to low-dose,mediandose and high-dose groups,respectively.After sever days treatment,rats are executed.Duodenum,jejunum,ileum and colon were dissected,respectively.Histology observation and cell counting were carried out under light micmscopo on HE coloration.Cell counting unit was defined as:cell/high power field of vision (cells/hpf).Results In jejunum,the number of goblet cells in berberine group,mediandose group and high-dose group[(15.32±2.53),(20.24±1.24),(14.98±1.10)cells/hpf,respectively],were all lower than that of the diarrhea control group[(25.73±4.55)cells/hpf,all P<0.05]with an exception of low-dose group[(23.98±2.28)cells/hpf].The numbers of goblet cells in berberine control group,low-dose group,mediandose group and highdose group[(18.29±1.33),(20.61±2.12),(19.38±2.01),(16.34±1.55)cells/hpf,respectively]were all less than that of the control group[(23.36±3.10)cells/hpf,all P<0.05].The numbers of goblet cells of diarrhea control group and high-dose group were obviously lower than that of the low-dose group(all P<0.05)in jejunum and colon.The numbers of goblet cells of Duodenum and ileum were not significantly different between groups(F=2.81,2.67,all P>0.05).The numbers of goblet cells in the diarrhea control group increased markedly observed under microscope,but decreased following therapeutic treatment.Conclusions The numbers of goblet cells from jejunum and colon in thyroid hormone-induced diarrheic rats are increased significantly.Asi-antidiarrheal capsule can remarkably decrease the number of goblet cells in jejunum and colon,and reduce mucus secretion.

Objective To explore the clinical significance of the blood lactic dehydrogenase(LDH), ?_2-microglobulin(?_2-MG),D-dimer measuring in the diagnosis and treatment of Non-Hodgkin lymphoma. Methods In 40 cases with NHL,LDH was measured by L-P continuous monitoring method,?_2-MG was measured by luminescent immunoassay,D-dimer was measured by immunoturbidimettic assay.Results The levels of the blood LDH,?_2-MG and D-dimer in patients with NHL were higher than those of in the controls(P 0.05).Con- clusion The levels of blood LDH,?_2-MG,D-dimer can be taken as an auxiliary clinical index to diagnose, classify the phase,evaluate the effectiveness of treatment and prognosis in the NHL patients,and have impor- tant clinical significance.

Objective Investigate the Asi-antidiarrheal capsule's effect on gastrointestinal horrnones in blood plasma of thyroid hormone-induced diarrheic(Abbreviation:Hyperthyroid Diarrhea.,D)rats.Methods One hundred and twenty SD male rats about 8 weeks old were randomly divided according to their constitution into control group of 10 rats and thyroid hormone-induced diarrheic group of 110 rats.The control group rats welle hvaged with isotonic Na chloride 1 ml/d.Thyroid tablets were made with isotonic Na chloride into 40 g/L susnl. The such solution with 1 ml/d was intragastrically administered to each rat in thyroid hormone-induced diardleic group.After three weeks,blood was sampled from vena caudalis of each rat.FT4 were then detected in blood semm.Fourty-two thyroid hormone-induced diarrheic rats were screened based on FT3 and FT4 level in blood serum, wet stool and body weisht.Fourty thyroid hormone-induced diarrheic rats were stochastically re-divided into 5 groups with 8 in each.The physiological saline with 1 ml/d was given to blank group,1.94 g·-kg-1·d-1 Berberine capsule to positive control group,and 0.63,1.26,2.52 g·kg-1·d-1 to low-dose,moderate-dose and high-dose groups respectivelv. Intragastric administration of each group continued for 7 days.Venous blood was centrifuged before and after administration and underwent radioimmunoassay to observe the effect of Ashi-antidiarrheal capsule on motillty (MTL),gastrin(GAS),somatostatin(SS),vasoactive intestinal polypeptide(VlP)in blood plasma of thyroid homlone- induced diarrheic rats.Results①Weight of thyroid hormone-induced diarrheic rats decreased[(344.0±12.9)g], FT3[(4,58 ±0.70)mol/L]and FT4[(23.44±4.40)mol/L]increased,and weight of wet stool[(17.4±3.2)g] increased.Compared to control group[(386.0±1.8)g,(2.08±0.10)mol/L,(10.18±2.00)mol/L,(9.1±0.6)g], there was a statistical significance(t=6.85,9.80,7.66,7.18,P<0.01).②After treatment,high-dose Ashi-antidiarrheal group[(80.54 ±3.80)ng/L]and positive control group[(90.63 ±9.99)ng/L]blood plasma MTL, compared to pre-therapy[(204.27±17.69),(187.79±13.32)ng/L]was decreased,there wag a statistical significance (t=8.60,4.57,P<0.01)③GAS contentshad respectively decreased comparedtopre-therapy[(192.75±11.80), (193.09±3.81),(190.60±9.31),(196.33±18.13)ng/L]in positive control group[(56.06 ±6.36)ng/L],low- dose group[(90.88±4.18)ng/L],midst-dose group[(75.64±7.09)ng/L]and hish-dose group[(44.32±3.72) ng/L],except for blank group.There Wag a statistical significance(t=15.27,7.62,13.43,13.22,all P<0.01).The intm-group difference of MTL,GAS and VIP level had statistic signifieances before and after the treatment(F= 166.68,1503.53,216.68,P<0.01).Conclusion Ashi-antidiarrheal capsule Can significantly lower the level of MTL and GAS in blood plasma。And raise the level of VIP.
4.Implementing humanistic concern in nursing work to build the harmonious relationship between nurses and patients

Objective To improve the patients' satisfaction to the nursing work through implementing the humanistic concern in nursing work. Methods We held some activities to provide the humanistic concern in nursing work, developed the educations to improve the professional quality of the nurses, constructed the criterion of civilized srvice, and strengthened the construction of nursing culture in the service demonstration ward. Meanwhile, we intensified the service ideas to create the harmonious patient-nurse relationships, the harmonious working settings, the harmonious treatment surroundings and the harmonious humanistic environment. Results The whole quality of the nurses was improved. especially the consciousness of active service. The relationship between nurses and patients was more harmonious, and the nursing quality and the degree of patients' satisfaction were elevated. Conclusions to implement the humanistic concern in nursing work is not only the base of building the harmonious relationships between patients and nurses. but also the guarantee of elevating the nursing quality and the degree of patients' satisfaction.
5.Optimization of short tandem repeats and their application in prenatal diagnosis of spinal muscular atrophy

Objective To optimize the short tandem repeats(STR)which link closely to survival motor neuron(SMN)and have redundant polymorphism information contents,and to use these STR in the prenatal diagnosis of spinal muscular atrophy(SMA).Methods Eleven STR loci(D5S435,D5F153, DSF151,D5S637,D5S1413,D5S125,D5S464,D5S1556,DSF149,D5S351,MAP1B-5')were amplified by PCR.Then the PCR products were detected by polyacrylamide gel electrophoresis(PAGE)and analyzed by silver staining.STR loci were evaluated and optimized by their PIC values.PCR-PAGE and gene scan were combined to make genetic link analysis for SMA families based on the optimized STR.Results Three STR loci(D5S435,DSF149 and D5S351)were selected with 8,19 and 18 polymorphic fragments detected respectively in 100 normal individuals.Their PIC values were 0.84,0.91 and 0.92 respectively.Four carriers and 2 normal individuals were detected from 6 SMA families with linkage analysis by using the 3 STR.Conclusion This genetic diagnosis system based on the 3 STR loci can provide rapid prenatal diagnosis for SMA families,can eliminate maternal blood contamination,and also can discriminate carriers from normal individuals in the fetuses,which makes the prenatal diagnosis system of SMA perfect.
6.Effects of lemon peel extracts on lactate dehydrogenase and sucrase activity of Streptococcus mutans

Objective To investigate the effect of lemon peel extracts(LPE) on the activity of lactate dehydrogenase and sucrase of Streptococcus mutans (Sm). Methods After serial dilution with trypticase soy broth (TSB) medium containing 2% glucose, LPE was used as the experimental group, and TSB without LPE as the control group. Sm was added to each group,which was then cultured for 6,18,24 and 48 hours in the anaerobic tank. The activity of lactate dehydrogenase (LDH) was measured with the method of oxidation of reduction coenzyme I and the pH value of the culture solution was also detected. The activity of the sucrose was determined with the method of coloration of 3,5-dinitrosalicylic acid. Results The activity of LDH, sucraae and the changes of solution pH were decreased with the increase of the concentration of LPE (P < 0. 01 ). The activity of LDH were declined from (0. 8025 ± 0. 0913 ) × 103 U/L to (0. 2099 ±0. 0283) × 103 U/L; the activity of sucrase were declined from ( -0. 0107 ±0. 0003) × 103 U/L to ( -0.0078 ±0.0002) × 103 U/L; the△pH were declined from(2.8067 ±0.0404) to (2.5033 ±0. 0416) (24 h results). The differences were significant between experimental groups and the control group (P < 0. 01 ), and there were also significant differences among experimental groups with different LPE concentration( P <0. 01 ). The inhibitory effect of acid generation and lactate dehydrogenas' activity of Sm were positively correlated ( P < 0. 01 ). Conclusions LPE can inhibit the activity of lactate dehydrogenase,sucrase and the acid production capacity of the Sm in a dose dependent manner. The inhibitory effects in logarithmic phase is stronger than that in other phases of growth cycle.
7.An animal experiment for the regeneration of periodontal defect by application of the dual-release chitosan thermosensitive hydrogel system

Objective To observe the effect of the self-made chitosan thermosensitive hydrogel system with dual-release bone morphogenetic protein and chlorhexidine on periodontal defects repair.Methods The furcation defect model was established on dog premolar.The models were divided into five groups,including three experimental groups,one control group and one blank control group.The hydrogel with the chlorhexidine/β-cyclodextrin inclusion complexes(IC)/rhBMP-2,hydrogel with rhBMP-2,hydrogel with IC,the pure hydrogel were applied to the defects of the four groups,respectively,and the blank control group did not receive any agent.The dogs were sacrificed 8 weeks later and the periodontal regeneration and gingival condition were observed by histological examination.Results Obvious periodontal tissue regeneration was found in group one and two.The heights of new bone reached 99.2% of the defects in group one,87.8%,63.6%,37.0% and 34.3% in group two,three,four and blank control groups,respectively.The inflammation of the affected gingiva showed less significant in group one and group three than in the other groups.Condusions rhBMP-2 and chlorhexidine played their independent role in repairing periodontal defects and the dual-release chitosan thermosensitive hydrogel system is effective and convenient to use.
8.Transcranial Sonographic Alterations of Substantia Nigra and Third Ventricle in Parkinson's Disease with or without Dementia

Background: Numerous studies have demonstrated that patients with Parkinson's disease (PD) have a higher prevalence of substantia nigra (SN) hyperechogenicity compared with controls. Our aim was to explore the neuroimaging characteristics of transcranial sonography (TCS) of patients with PD and those with PD with dementia (PDD). The correlation between the echogenicity of the SN and clinical symptoms in Chinese patients with PDD was also assessed. Methods: The ratios of SN hyperechogenicity (SN+), maximum sizes of SN+, and widths of third ventricle (TV) were measured using TCS for all the recruited patients. Data were analyzed using one?way analysis of variance, rank?sum test, Chi?square test, and receiver?operating characteristic (ROC) curve analysis. Results: The final statistical analysis included 46 PDD patients, 52 PD patients, and 40 controls. There were no significant differences in ratios of SN+ and maximum sizes of SN+ between PDD and PD groups (P > 0.05). TV widths were significantly larger in PDD group (7.1 ± 1.9 mm) than in PD group (6.0 ± 2.0 mm) and controls (5.9 ± 1.5 mm, P < 0.05); however, the ratios of enlarged TV did not differ among the three groups (P = 0.059). When cutoff value was set at 6.8 mm, the TV width had a relatively high sensitivity and specificity in discriminating between PDD and PD groups (P = 0.030) and between PDD group and controls (P = 0.003), based on ROC curve analysis. In PDD patients, SN+ was more frequently detected in akinetic?rigid subgroup, and patients with SN+ showed significantly higher Hoehn and Yahr stage and Nonmotor Symptoms Questionnaire scores (P < 0.05). Conclusions: Compared to Chinese patients with PD, patients with PDD had a wider TV, altered SN sonographic features, and more severe clinical symptoms. Our findings suggest that TCS can be used to assess brain atrophy in PD and may be useful in discriminating between PD with and without dementia.

10.Successful transcatheter ablation of fascicular potential in pediatric patients with left posterior fascicular tachycardia.

OBJECTIVETo simplify the methods of transcatheter mapping and ablation in the pediatric patients with left posterior fascicular tachycardia.
METHODWhile in sinus rhythm, the fascicular potential can be mapped at the posterior septal region (1 - 2 cm below inferior margin of orifice of coronary sinus vein), which display a biphasic wave before ventricular wave, and exist equipotential lines between them. When the fascicular potential occurs 20 ms later than the bundle of His' potential, radiofrequency was applied. Before applying radiofrequency, catheter position must be observed using double angle viewing (LAO 45°RAO 30°), and it should be made sure that the catheter is not at His' bundle. If the electrocardiogram displays left posterior fascicular block, the correct region is identified and ablation can continue for 60 s. Electrocardiogram monitoring should continue for 24 - 48 hours after operation, and notice abnormal repolarization after termination of ventricular tachycardia. Aspirin [2 - 3 mg/(kg·d)] was used for 3 months, and antiarrhythmic drug was discontinued. Surface electrocardiogram, chest X-ray and ultrasound cardiography were rechecked 1 d after operation. Follow-up was made at 1 month and 3 months post-discharge. Recheck was made half-yearly or follow-up was done by phone from then on.
RESULTFifteen pediatric patients were ablated successfully, and their electrocardiograms all displayed left posterior fascicular block after ablation. None of the patients had recurrences during the 3 to 12 months follow-up period. In one case, the electrocardiogram did not change after applying radiofrequency ablation and the ventricular tachycardia remained; however, on second attempt after remapping, the electrocardiogram did change. The radiofrequency lasted for 90 seconds and ablation was successful. This case had no recurrences at 6 months follow-up.
CONCLUSIONTranscatheter ablation of the fascicular potential in pediatric patients with left posterior fascicular tachycardia can simplify mapping, reduce operative difficulty and produce a distinct endpoint for ablation.
