Cytokine storm plays an important role in the pathogenesis of liver failure.It is closely related to hepatocyte necrosis in the clini-cal course and the prognosis of this disease.With Kupffer cells as a clue,it is elucidated that related cells and cytokines influence each other and jointly function in the development and progression of liver failure,leading to serious liver tissue damage and necrosis.Cytokine -based research can help improve early diagnosis,disease assessment,and individualized treatment for liver failure.
2.Comparative Studies on Kinetics of Mitochondrial LDH and Solubilized Mitochondrial LDH in Rat Liver

In this paper, we studied the kinetics of nitochondrial lactate dehydrogenase (LDH) and solubilized mitochondrial LDH in the rat liver. The apparent Km values of mitochondrial LDH and solubilized mitochondrial LDH for substrate pyruvate were 50.0 ?mol/L and 33.8 ?mol/L, and those for NADH were 35.3?mol/L and 21.4 ?mol/L, respectively. The apparent Km values of mitochondrial LDH were greater than those of solubilized nitochondrial LDH. The mitochondrial LDH in the rat liver was mainly LDH-5, which could be solubilized by 0.15 mol/L NaQ solution.
3.Interaction of Rabbit Skeletal Muscle Lactate Dehydrogenase with Asolectin Liposome

In this paper, the effect of asolectin liposome on the activity of rabbit skeletal muscle lactate dehydrogenase (LDH) is reported. The results suggested that asolectin liposome could inhibit LDH activity, KC1 could restore the enzyme activity, and NAD+ and NADH could protect the enzyme from being inhibited by asolectin liposome.
4.Integrated“U”Shaped Cutting Edge Rongeurs of Pruning Finger-Toe and Bending-Truncating Pin

Objective In order to overcome the problems that bone rongeur and Kirschner forcep's less function and easy damage, trivial and inefficient, bulky volume, bone and needle broken edge is not neat,needle tail easy spatter wounding and other defects, integrated"U"shaped cutting edge rongeurs of pruning finger-toe and bending-truncating pin is desigened. Methods Pruning-truncating rongeurs set the trimming, bending and shearing,straight and twisting, loading and unloading, filing and stripping, string devices, aintenance functions and other functions in one, compare with bone rongeurs in clinical application. Results Pruning-truncating rongeurs are molding once, manipulation convenient, light and safe, anti damage and maintenance free, cost-effective. Conclusion Pruning-truncating rongeurs are highly integrated and portable anti-lost, preparation instrument swift, man-machine coordination, sharp instrument injury prevention, to improve the operation efficiency.
5.Effects of Interleukin?17A on Acute Paraquat?intoxication?induced Kidney Injury in Mice

Objective To investigate the effects of interleukin?17A on kidney injury induced by paraquat(PQ). Methods Seventy?two ICR mice were randomly divided into 3 groups:NS,PQ,and PQ+Ab (n=24 for each). The PQ?poisoning model was established by administering a gavage of PQ solution;mice in the PQ+Ab group were then administereda dose of anti?IL?17A antibody 2 hours later by i.p. injection,whereas the NS group were administered a corresponding volume of normal saline instead.The mice were killed at 8,24,48,or 72 h to obtain renal tissues and serum. An enzyme?linked immunosorbent assay(ELISA)was used to determine serum IL?17A,serum creatinine(SCr),and blood urea nitrogen (BUN)levels.Chemical colorimetry was used to detect the viability of myeloperoxidase(MPO )in renal tissue,and hematoxylin?eosin(HE)stain?ing was used to observe the renal pathologic changes. Immunohistochemistry(IHC)and PCR were used to examine IL?17A expression in renal tis?sues. Results Serum IL?17A,renal tissue MPO viabilities,BUN,and SCr were increased in the PQ and PQ+Ab groups,compared to those in the NS group(P<0.01). However,the above?mentioned parameters were lower in the PQ+Ab group than in the PQ group(P<0.01). Conclusion IL?17A promotes mouse kidney injury induced by acute PQ?intoxication through activating and/or recruiting neutrophils;therefore,blockade IL?17A,with antibody can attenuate the injury.
6.Application of combination transfer pump to continuous blood purification

Objective To evaluate the effect of combination transfer pump and single blood pump to 32 cases of renal failure patient who need blood purification therapy.Methods The combination transfer pump and single blood pump were used.According to the clinical requirement,the parameters of transfer pump were set such as therapy time,replacement liquid flow rate,dialysate flow rate,temperature of warmer and filtrate flow rate,etc.Results 32 patients with varying degrees of illness have been alleviated.Patients' acute left heart failure,water and electrolyte turbulences,acid intoxication and azotemia caused by liquid retention were corrected,which ensured a role for further treatment.Conclusion It is the functions of adjustable time and transfusion volume that are used in the combination transfer pump,the task of which is accomplished by several transfer pumps together.This method is easy to implement and can meet the clinical requirement largely without special CBP instrument.
7.Experimental study of the oriented differentiation of bone marrow derived mesenchymal stem cells into chondrogenic phenotype in a specific culture fluid

BACKGROUND:To construct tissue engineering cartilage would open up a novel way for the repair of cartilage damage in avoidance of the disadvantages of traditional therapeutic method.OBJECTIVE: To probe the techniques for the isolation of mesenchymal stem cells (MSCs) from bone marrow, as well as the in vitro differentiation into chondrocytic phenotype in a specific culture fluid.DESIGN:A complete randomized experimentSETTING:The Department of Traumatic Orthopedics and Hand Surgery of the First Affiliated Hospital of Guangxi Medical University, and Teaching and Research Faculty of Histology and Embryology of Guangxi Medical University.METHODS: The experiment was carried out at Guangxi Medical University between August 2002 and April 2003. Twenty SD neonatal weaning rats were selected. Bone marrow was aspirated from the bones of rat limbs and was isolated by gradient centrifugation in Percoll, and MSCs could be obtained in combination with adherent screening method, which were then cultured in DMEM-LG with 15% fatal bovine serum (FBS) in the incubator of 37℃ with 5% CO2 for 10-14 days. The passage cells were induced in DMEM-HG with 15% FBS (containing TGF-β1 10 μg/L, 10-7 mol/L dexamethasone, 50 mg/L VitC).MAIN OUTCOME MEASURES :The morphology, growth, as well as proliferation and specific expression of chondrogenic matrix of in vitro cultured MSCs due to specific induction.RESULTS: Totally 20 SD rats were observed and analyzed with no loss SCs grew in visible symmetric colonies, displaying a long-spindle shape,and the morphological characteristics of marrow-derived MSCs had no obvious changes during passage-culture, but its proliferation time was found from a shuttle fibroblastic appearance to polygonal shape, displaying posiHC staining of type Ⅱ collagen of cartilage specific matrix.bronectin adherent screening technique is a convenient, effective and practical method to separate and collect MSCs from rat bone marrows in chondrogenic phenotype when induced by a specific medium and can secrete cartilage specific matrix, and they can be the optimal seed cells for cartilage tissue engineering.
8.Rat tail tendon cell as a model seed cell for tendon tissue engineering study

BACKGROUND:The low output of seed cells and long cycle of traditional ceils culture methods in tendon animal models(rabbits and chicken) restrict the researches of tendon tissue engineering study.OBJECTIVE:To establish an ideal culture protocol of tail tendon in SD rats,to get more seed cells within less time for subsequent engineered tendon construction research.DESIGN,TIME AND SETTING:Controlled observation was performed in the National Key Laboratory of Biotherapy,Department of Stem Cells and Tissue Engineering,Sichuan University between February and November in 2006.MATERIALS:Rat tail tendon cells were harvested from 2 SD rats,aged 7-10 days;human prepuce fibroblasts were offered by National Key Laboratory of Biotherapy,Department of Stem Cells and Tissue Engineering,Sichuan University.METHODS:Tail tendon of SD rats was draw off and cut into pieces,which were then cultured in 10% fetal bovine serum+DFculture medium for getting primary tendoncyte by using suspension tissue culture method. The third generation cells were processed into immuocytochemistry stain with collagen type Ⅰ and Ⅲ,while human prepuce fibroblasts served as controls.Absorbance of stain result was measured by image-pro plus 5.02 for statistical analysis.MAIN OUTCOME MEASURES:Immuocytochemistry stain and absorbance measurement of SD rat tail tendon cells.RESULTS:The second generation of SD rat tail tendon cells were positive for type Ⅰ collagen stain,and negative for type Ⅲ collagen stain;human fibroblast were positive for both Ⅰ and Ⅲ collagen. In the rat tail tendon cells and human flbroblasts,the absorbance value of type Ⅰ collagen expression was dramatically higher than of type Ⅲ collagen(P<0.05). There was no significant differences addressing the absorbance of type Ⅲ collagen expression between type Ⅰ and Ⅲ collagen of SD rat tail tendon cells and blank control group (P>0.05).CONCLUSION:Cells cultured from SD rat tail tendon have biological characteristic of tendon cells. Tissue piece suspensionculture can obtain a quantity of primary or subcuitured cells of rat tail tendon within a short time.
9.Monitoring on extravascular lung water in patients with septic shock and its prognostic value

Objective To analyze the alterative characteristics of the extravascular lung water (EVLW ) in the patients with septic shock and clarify its value on the prognosis of these patients.Methods By the methods of retrospective analysis,according to the ultimate survival,21 patients with septic shock were divided into survivor group (10 cases) and non-survivor group (11 cases).The clinical features of the patients were observed and hemodynamic monitoring was made with PiCCO monitor.The EVLW was measured and the relationship between the EVLW and the prognosis of patients was analyzed.Results On the first,second and third day,EVLW was (12.7 ±1.8),(11.3 ±1.3),(10.1 ±1.3) ml/kg in survivor group,and (14.4 ± 1.0),(14.6 ± 1.4),(14.6 ±1.3) ml/kg in non-survivor group respectively,and there were statistical differences between two groups (P <0.05).However,on the second day after the intensive therapy,EVLW in survivor group dropped significantly(P<0.05),but the non-survivor group only declined slightly,and compared with the result of the first day,there was no obvious difference (P >0.05).Conclusions The EVLW in the patients with septic shock increases significantly.The dynamic changes of the EVLW may be one of the factors for predicting the prognosis of patients with septic shock.
10.Clinical analysis of 62 cases of convulsion associated with acute purulent meningitis of children

Objective The study was conducted to investigate the acute phase of convulsion related problem on the clinical manifestations,imaging and electroencephalograph (EEG) examination of purulent meningitis.Methods Cluster sampling method was employed to select children in our hospital,a total of 301 cases with purulent meningitis was analyzed retrospectively.Among them,62 cases had convulsion.The incidence of convulsion in the acute phase of the purulent meningitis,risk factors,and prognosis were analyzed.Results The convulsion incidence rate of acute purulent meningitis was 20.60%.The partial seizure was eight cases (12.90%).The secondarily generalized seizure following partial seizure was 15 cases (24.19%).The generalized seizure was 32 cases (51.61%).The convulsive status was 7 cases (11.29%).The EEG abnormality was significantly different between the convulsion group and the no convulsion group (P < 0.05).The incidence of brain organic damage was significantly different between two groups (P <0.05).The multivariate unconditional logistic regression analysis showed,cause of disease,first symptom,disturbance of consciousness,obvious signs,and cerebrospinal fluid culture with convulsion were the relevant factors (P < 0.01).Conclusions The most common seizure of purulent meningitis was the generalized seizure.Brain organic damage easily resulted in convulsion of purulent meningitis.The days of hospitalization,cause of disease,first symptom,disturbance of consciousness,obvious signs,and cerebrospinal fluid culture with convulsion were the positively relevant factors.Those positively relevant factors in combination of the clinical manifestations,imaging,and EEG examination in children would play an important role in diagnosis,treatment,and prognosis evaluation of convulsion derived from purulent meningitis.Moreover,convulsion affects the disease recovery in children with purulent meningitis.
