Journal of Third Military Medical University 2003;0(08):-
Objective To explore an ideal approach to establish rabbit model of hepatic fibrosis suitable for radiological and serological research. Methods Totally 40 rabbit models of hepatic fibrosis was established by intraperitoneally injecting 5% carbon tetrachloride in oil solution or 100% carbon tetrachloride(both groups,0.1 ml/kg,once a week),and drinking 5% ethanol water.Another 8 rabbits who drank normal water and received a peritoneal injection at 0.1 ml/kg served as control. The rabbits(5 or 2 per time for model group and control) were killed at 6,8,10 and 12 weeks respectively after first injection.Their livers were resected for gross and morphological observation with HE and Masson staining. Results Death of the models usually happened within 4 weeks after first injection and became stable after 6 weeks.Mortality of 5% carbon tetrachloride group was 60%,but 25% in 100% carbon tetrachloride group.Macroscopy and microscopy indicated that liver fibrosis was observed in rat models. Conclusion Long-term intraperitoneal injection of carbon tetrachloride results in rabbit hepatic fibrosis.Injection of 100% carbon tetrachloride intraperitoneal,at 0.1 ml/kg,once a week, combined with and 5% ethanol as drinking water is a suitable approach to establish rabbit model of hepatic fibrosis with low mortality and high success rate.

Chinese Journal of Experimental Ophthalmology 2013;(4):334-338
Background Researches showed that the content of nitric oxide (NO) and nitric oxide synthetase (NOS) increases in blood, aqueous humor and tear of cataract patient. But the function of NO and NOS in cataract formation is still elusive. Objective The aim of this study was to explore the prevention and treatment effect of NOS inhibitor,1-nitro-arginine methyl ester (L-NAME),on galactose cataract. Methods Sixty clean three-week-old Wistar rats were equally and randomly divided into 3 groups.0.9% Normal saline solution (30 ml/kg) was subcutaneously injected every day for 30 days in the rats of the control group, and 50% of D-galactose solution (30 ml/kg) was used in the rats of the model and L-NAME group at the same way.L-NAME eye drops was simultaneously administered in the L-NAME group 3 times per day for 30 days. The eyes of the rats were examined under the slit lamp in 10,20 and 30 days, and the degree of lens opacification was scored.Lenses of the rats were obtained at the end of this experiment for the detect of NO,NOS contents.Flow cytometry was used to assay the caspase-3 level of rat lens.Repeated measurement two factor analysis of variance was used to analyze the difference of lens opacification scores in different groups and different time points, and one-way ANOVA was used to analyze the differences of NO,NOS and caspase-3 contents in lens among the groups. Results Lens opacification appeared in 10 days after injection of 50% D-galactose solution in the rats of the model group and L-NAME group.Lens opacification score was higher among the different groups and different time points (Ftime =435.251,P =0.000 ;Fgroup =395.120,P=0.000).NO content in the lens was (0.45±0.15) μmol/g,(2.67 ± 0.47) μmol/g and (1.68±0.34) μmol/g in the control group, model group and L-NAME group, showing a significant difference (F=58.872,P=0.000).The NOS contents in the lens was (0.0160±0.0020) U/ml,(0.0370±0.0040) U/ml and (0.0270±0.0010) U/ml in the control group, model group and L-NAME group, showing a significant difference (F =66.174,P=0.000).Caspase-3 contents in the lens was (339.4 ± 37.9),(697.7 ± 46.5) and (650.7 ± 53.1),Showing a significant difference among them (F =100.005,P =0.000).Conclusions The increase of NO,NOS and caspase3 levels are associated with lens opacification.Topical administration of L-NAME eye drops can down-regulate NOS content in lens, reduce the NO formation and inhibit the apoptosis of lens epithelial cells.

Objective To investigate the effects of HepG2 and HepG2.2.15 cells steatosis on the mRNA and protein expressions of suppressors of cytokine signaling-3(SOCS-3) and sterol regulatory element binding proteins (SREBP-1c).Methods The cell model of chronic hepatitis B (CHB) combined with nonalcoholic fatty liver disease (NAFLD) was successfully constructed using an oleic acid-induced HepG2 and HepG2.2.15 cells steatosis.Cells were divided into HepG2 cell control group (HepG2 cell control group), HepG2.2.15 cell control group (HepG2.2.15 cell control group), HepG2 cell steatosis group (HepG2 cell steatosis group) and HepG2.2.15 cell steatosis group (HepG2.2.15 cell steatosis group). The expression levels of SOCS-3 and SREBP-1c mRNA were detected by real-time quantitative polymerase chain reaction (PCR).Changes in protein expressions of SOCS-3 and SREBP-1c were measured by western blot. Results SOCS-3 mRNA expression level in HepG2.2.15 cell control group was significantly lower than that in HepG2 cell control group (P<0.01).The level in HepG2 cell steatosis group was also significantly lower than that in HepG2 cell control group (P<0.01).However, the level of SOCS-3 mRNA in HepG2.2.15 cell steatosis group was lower than HepG2.2.15 cell control group with no statistical significance (P=0.173).There was interaction between cells and steatosis (F=25.547, P<0.01).The expression of SREBP-1c mRNA in HepG2.2.15 cell control group was significantly lower than that in HepG2 cell control group (P<0.01), and was significantly higher in HepG2.2.15 cell steatosis group than that in HepG2.2.15 cell control group (P<0.01).There was no significant difference between HepG2 cell steatosis group and HepG2 cell control group (P=1.000).There was interaction between cells and steatosis (F=5.04, P<0.05).Western blot analysis showed that protein levels of SOCS-3 and SREBP-1c in steatosis cells at 48 h and 72 h were significantly higher than those in non-alcoholic steatosis cells. Conclusions Protein expressions of SOCS-3 and SREBP-1c are up-regulated in both steatosis groups.Factorial analysis shows that there is interaction between cells and steatosis.HBV gene could inhibit SOCS-3 mRNA expression and promote the expression of SREBP-1c mRNA in steatosis cells.
